에폭시 몰딩 컴파운드를 위한 에폭시 나노복합재료의 소수성 실리카의 영향 (Influence of Hydrophobic Silica on Physical Properties of Epoxy Nanocomposites for Epoxy Molding Compounds)

  • 본 연구에서는 에폭시 나노복합재료의 수분 흡수, 열적 안정성 및 기계적 특성에 대한 소수성 실리카의 효과에 대해 고찰하였고, 에폭시 수지의 필러로는 디메틸디크로로실란에 의해 소수성으로 처리된 실리카를 사용하였다. 실험 결과, 실리카는 실란 커플링제의 첨가후 용융혼합법에 의하여 에폭시 수지내에서 균일하게 분산되었으며, 나노복합체의 수분 흡수율은 소수성으로 처리된 실리카의 함량 증가와 함께 감소하는 것을 확인하였다. 열분해 온도, 유리전이 온도, 그리고 열팽창 계수를 통한 나노복합재료의 열안정성은 실리카의 첨가와 함께 향상되는 것을 확인하였다. 또한, 인장강도 및 탄성율을 통한 나노복합재료의 기계적 특성은 실리카 함량 증가와 함께 증가하였고, 이는 에폭시 수지내에 고르게 분산된 실리카와 에폭시 수지 간의 강한 물리적 상호작용에 기인하는 것으로 판단된다.

Multilevel Precision-Based Rational Design of Chemical Inhibitors Targeting the Hydrophobic Cleft of Toxoplasma gondii Apical Membrane Antigen 1 (AMA1)

  • Toxoplasma gondii is an intracellular Apicomplexan parasite and a causative agent of toxoplasmosis in human. It causes encephalitis, uveitis, chorioretinitis, and congenital infection. T. gondii invades the host cell by forming a moving junction (MJ) complex. This complex formation is initiated by intermolecular interactions between the two secretory parasitic proteins-namely, apical membrane antigen 1 (AMA1) and rhoptry neck protein 2 (RON2) and is critically essential for the host invasion process. By this study, we propose two potential leads, NSC95522 and NSC179676 that can efficiently target the AMA1 hydrophobic cleft, which is a hotspot for targeting MJ complex formation. The proposed leads are the result of an exhaustive conformational search-based virtual screen with multilevel precision scoring of the docking affinities. These two compounds surpassed all the precision levels of docking and also the stringent post docking and cumulative molecular dynamics evaluations. Moreover, the backbone flexibility of hotspot residues in the hydrophobic cleft, which has been previously reported to be essential for accommodative binding of RON2 to AMA1, was also highly perturbed by these compounds. Furthermore, binding free energy calculations of these two compounds also revealed a significant affinity to AMA1. Machine learning approaches also predicted these two compounds to possess more relevant activities. Hence, these two leads, NSC95522 and NSC179676, may prove to be potential inhibitors targeting AMA1-RON2 complex formation towards combating toxoplasmosis.

Preparation and Characterization of Hydrophobic Coatings from Carnauba Wax/Lignin Blends

  • To realize the infinite possibilities of materials derived from wood, it is necessary to overcome the weak moisture stability of wood. Thus, the development of an eco-friendly hydrophobic coating agent is essential, and of these, woody biomass-based materials are strongly attractive as coatings. In this study, eco-friendly hydrophobic wood coatings were prepared using carnauba wax purified from palm leaves and sprouts, and kraft lignin. The physicochemical properties of the carnauba wax/lignin blends according to the ratio of carnauba wax and lignin were observed by morphology and functional group change. In addition, the coating performance of carnauba wax/lignin blend coatings was confirmed by measuring the contact angle change. It was found that the addition of lignin could accelerate the atomization of wax particles, and that micro-roughness can be realized when applied to the actual wood surface, to ensure that the coating effect over time lasts longer. In addition, it was confirmed that the addition of lignin increases the hydrogen-bond-based interaction with the wood of the coating, thereby providing better coating stability and increasing the durability of the coating solvent under friction. The carnauba wax/lignin paint developed in this way is eco-friendly because all components are made of wood-based raw materials and have an excellent affinity with wood surfaces. Therefore, it is expected to be applicable to the coating process of wood-plastic composites and timber composites.

  • Studies on the effect of the wall-ion, wall-water, water-ion and ion-ion interaction on properties of water and ions in nano-channels have been performed through the use of different kinds of ions or different models of potential energy between wall-ion or wall-water. On this paper, we address the effect of water-wall interaction potential on the properties of confined aqueous solution by using the molecular dynamics (MD) simulations. As the interaction potential energies between water and wall we employed the models of the Weeks-Chandler-Andersen (WCA) and Lennard-Jones (LJ). On the MD simulations, 680 water molecules and 20 ions are included between uniformly charged plates that are separated by 2.6 nm. The water molecules are modeled by using the rigid SPC/E model (simple point charge/Extended) and the ions by the charged Lennard-Jones particle model. We compared the results obtained by using WCA potential with those by LJ potential. We also compared the results (e.g. ion density and electro-static potential distributions) in each of the above cases with those provided by solving the Poisson-Boltzmann equation.

Small Molecules Targeting for ESX-Sur2 Proteins' Interaction

  • It's been known that overexpression of the oncoprotein Her2 (eu/ErbB2), transmembrane receptor protein, occurs in human breast cancer. Her2-positive breast cancer patients who have Her2 overexpression show less therapeutic efficacy with enhanced metathesis and increased resistance to chemotherapy. So far, a humanized monoclonal antibody against Her2 protein called Herceptin is the only drug approved by Food and Drug Administration for treatment of Her2-overexpressing breast tumors. However, antibody therapy of Herceptin may not be ideal method for therapeutic intervention of Her2 protein expression. The therapeutic intervention of Her2 protein expression may be more efficiently achieved by inhibiting the expression of Her2 gene rather than by down-regulating the Her2 protein already overexpressed. Here, we found that the interaction of two proteins of ESX (an epithelial-restricted transcription factor) and DRIP130/CRSP130/Sur2 (a Ras-linked subunit of human mediator complexes) mediates the expression of Her2 gene. The association of ESX with Sur2 is mediated by a small hydrophobic face of 8-amino acid helix in ESX, suggesting that the ESX-Sur2 interaction can be a new novel target for Her2-positive cancer. The process to develop potent ESX-Sur2 interaction inhibitors targeting for Her2-positive cancer therapeutics will be discussed.

  • The protein binding of salicylate analogs has been investigated by equilibrium dialysis. A series of binding experiments were performed in order to elucidate the effects of physicochemical properties of salicylate analogs on the binding with bovine serum albumin. Attempts to correlate affinity constants with capacity factor, steric factor and Hammett ${\sigma}$ values suggested hydrophobic forces to be involved in the binding of salicylate analogs. Steric factor contributes to binding process partly, whereas electronic interaction appears to be insignificant.

  • Cytochrome c (cyt c) binds to acidic membranes at low ionic strength. Replacement of Lys-72 or Lys-87 by Glu reduced the binding affinity of cyt c toward large unilamellar vesicles (LUV) in liquid crystalline phase. The differences were smaller for LUV in gel phase. A fraction of bound cyt c was non-electrostatically associated.(omitted)

Proteolysis of Glucagon Bound to Dimyristoylphosphatidylcholine Vesicle

  • Glucagon was found to interact with DMPC vesicles electrostatically and hydrophobically. It appears that glucagon bound irreversibly to the vesicles through hydrophobic interaction was partially protected from the proteolysis by trypsin. Out of three possible sites, only the peptide bond preceded by Arg-18 was cleaved by a prolonged trypsin treatment. ${\alpha}$-chymotrysin did not affect the vesicle-bound glucagon. Based on these observations, possible structure of irreversibly bound glucagon on the vesicle surface is discussed.
