The objectives of this research are to investigate the mechanism of coagulation affecting UF, find out the effect of metal salt coagulant on membrane fouling. Either rapid mixing + UF or slow mixing + UF process caused much less flux decline. For PACl coagulant, the rate of flux decline was reduced for both hydrophilic and hydrophobic membrane than alum due to higher formation of flocs. In addition, the rate of flux decline for the hydrophobic membrane was significantly greater than for the hydrophilic membrane, regardless of pretreatment conditions. In general, Coagulation pretreatment significantly reduced the fouling of the hydrophilic membrane, but did little decrease the flux reduction of the hydrophobic membrane. When an Al(III) salt is added to water, monomers, polymers, or solid precipitates may form. Different Al(III) coagulants (alum and PACl) show to have different Al species distribution over a rapid mixing condition. During the rapid mixing period, for alum, formation of dissolved Al(III) (monomer and polymer) increases, but for PACl, precipitates of $Al(OH)_{3(s)}$ increases rapidly. This experimental results pointed out that precipitates of $Al(OH)_{3(s)}$ rather than dissolved Al(III) formation is major factor affecting flux decline for the membrane.
Heat treatment condition for the stabilization of foamed glass block through the foaming process of the hydrolized waste glass was investigated and scale-up test for the manufacturing of foamed glass was also attempted for the actual foaming process. Proper heat treatment condition was quenching from the foaming temperature to $550{\sim}600^{\circ}C$ for stabilization, and then annealing from stabilization temperature to $200^{\circ}C$ and holding up at $200^{\circ}C$ for removal thermal stress, and then annealing to the room temperature with cooling speed of $0.3^{\circ}C/min$. Through this heat treatment conditions, foamed glass block with size of $250mm{\times}250mm{\times}90mm$ was produced successfully. The properties of this foamed glass block showed density of $0.28{\pm}0.06g/cm^3$, thermal conductivity of $0.048{\pm}0.005kcal/hm^{\circ}C$, moisture absorption of $0.5{\pm}0.09vol%$, linear expansion coefficient of $(8.6{\pm}0.2){\times}10^{-6}m/m^{\circ}C$($400^{\circ}C$), flexural strength of $15.0{\pm}0.6kg/cm^2$, and compression strength of $39.5{\pm}0.6kg/cm^2$.
Stews were prepared by 2 processes and 4 treatments, and stored for 3 different storage periods. The two processes were beef cooked in a stew and stored in a polyethylene container at $5^{\circ}C(P1)$ and in a barrier bag at $0^{\circ}C(P2)$. The four treatments were beef cooked alone (T1), with onions (T2), with carrots (T3) and with onions and carrots (T4). Stews in P1 were stored for 0, 2 and 4 days and stews in P2 were stored for 0, 2 and 4weeks. Cooking decreased the cephalin content by 39%. the lecithin content by 21% and most of the prolipid fatty acid concentrations as well as the fatty aldehyde levels in the phospholipids of beef from stew. Process or storage did not significantly affect the level of either phospholipids. however cooking beef with carrots seemed to exhibit some protection against hydrolysis of cephalin. P1 stews had a higher TBA-value (p<0.05) than P2 stews, and the TBA-value of P1 stews increased linearly during 4 days storage. The TBA-value was not affected (p<0.05) by treatment for any of the stews and did not change significantly during 4 weeks storage in P2 stews.
Silk protein has been explored to be used for biomedical applications for several decades. However, it has not been used in this field cause to their irreversible crystallization after dissolving in water. The existing methods of silk protein hydrolysis using silkworm cocoon were used with harmful solvents and through a very complicated process. Therefore, we have developed novel methods for the production of water-soluble hydrolysate using silkworm gland. We manufactured two types of silkworm gland-derived hydrolysate (water-soluble SGH, SSGH; total SGH, TSGH) and compared the characteristics with commercial cocoon-derived sericin hydrolysate (CSH). The molecular weight of SGH ranged from 7 to 50 kDa (SSGH) and 5 to 15 kDa (TSGH) within glycine, alanine, and aspartic acid as a main amino acid composition. In contrast, CSH ranged from 15 to 50 kDa within serine and aspartic acid. The results of FTIR implied that SGH was more soluble form than CSH, as shown by the decrease in the ${\beta}$-sheet structure at $1630cm^{-1}$ on amide I peak. In comparison with 10% fetal bovine serum, 0.1% (1 mg/ml) SSGH had equivalent effect on the proliferation of human dermal fibroblasts and mesenchymal stem cells. All results of the SSGH made by novel manufacturing process indicate the SSGH is more preferable as a culture medium supplement than cocoon-derived sericin.
In general after the etching process, waste etching solution contains metals. (ex. Nickel (Ni), Chromium (Cr), Zinc (Zn), etc.) In this work, we proposed a recycling process for waste etching solution and refining from waste liquid contained nickel to make nickel metal nano powder. At first, the neutralization agent was experimentally selected through the hydrolysis of impurities such as iron by adjusting the pH. We selected sodium hydroxide solution as a neutralizing agent, and removed impurities such as iron by pH = 4. And then, metal ions (ex. Manganese (Mn) and Zinc (Zn), etc.) remain as impurities were refined by D2EHPA (Di-(2-ethylhexyl) phosphoric acid). The nickel powders were synthesized by liquid phase reduction method with hydrazine ($N_2H_4$) and sodium hydroxide (NaOH). The resulting nickel chloride solution and nickel metal powder has high purity ( > 99%). The purity of nickel chloride solution and nickel nano powders were measured by EDTA (ethylenediaminetetraacetic) titration method with ICP-OES (inductively coupled plasma optical emission spectrometer). FE-SEM (field emission scanning electron microscopy) was used to investigate the morphology, particle size and crystal structure of the nickel metal nano powder. The structural properties of the nickel nano powder were characterized by XRD (X-ray diffraction) and TEM (transmission electron microscopy).
In this study, enzymatic saccharification of substrates from Alnus hirsuta Ruper (8-14 years). Quercus acutissima Carruthers, Betula platyphylla var. japonica Nera, Populus euramericana Guiner and Platanus orientalis L. were investigated using crude cellulase preparations of Trichoderma viride Pers. ex. Fr. SANK 16374, and conduced on the optimum treated conditions of the cellulase sacchrification and reactivation of residue of digested substrates. The Trichoderma viride cellulase was produced by the submerged culture process and produced in the culture fluid was salted out quantitatively by the use of ammonium sulfate. The method of dilignification from wood (5 species) was treated by the peracetic acid(PA) method. The reducing sugar was determined by the dinitrosalicylic acid (DNS) method. 1. The results of tests carried out for 96 hr. (Figure 1), show conclusively the initial substrates from 5 species ($S_3$) which has been rendered highly reactive form and the mean rate of reducing sugar was 28.3 %. 2. The results of tests carried out for 96 hr., the reactivation of residue of digested substrates (improvement in the quality of the substrate through preheating in air at $190^{\circ}C$. for 45 min. followed by milling was (60 mesh size) at the same substrate level, increased concentrations of cellulase at the same substrate level, and increased concentrations of cellulase increases the rate of hydrolysis considerably. 3. Figure 1. shows conclusively that the residue of digested substrates ($S_1$ dried at $60^{\circ}C$) which has been rendered extremly resistant to cellulase action can be reactivated into a highly reactive form ($S_2$), almost comparable to that of the initial substrates ($S_3$). And the reducing sugar formation did not show statistically significent differences at 5% levels by initial substrates and the residue of digested substrates (preheating in air at $190^{\circ}C$. for 45 min. fallowed by milling was (60 mesh size).
In this study, enzymatic hydrolysis of the substrate of the waste wood of Cortinellus edodes was investigated using crude cellulase preparation of Trichoderma viride Pers. ex. Fr. SANK 16374. The crude cellulase was produced by the submerged culture process and produced in the culture fluid was salted out quantitatively by the use of ammonium sulfate. Reducing sugar was determined by the dinitrosalisylic acid (DNS) method. 1. The chemical composition of the waste wood was crude protein 2.26%, c. fat 2.57%, c. fibre 44.60%, c. ash 5.58% and lignin 13.62%. In amino acid composition, no cystine and methionine was showed, but trace amount of Vitamin A, $B_1$, and $B_2$, niacine and chloride were detected. (Table 1) 2. As heat treatment of the substrate was found to produce the highest reducing sugar yield being reacted for 48hr. with T.v cellulase, the substrate was heated to $190{\pm}5^{\circ}C$. for 45 min. either before or immediately after milling. 3. The substrate heated and ball milled at $190{\pm}5^{\circ}C$. for 45 min. the reducing sugar yield reached to 11.5%. 4. The substrate without any treatment was found to produce the highest reducing sugar yield being reacted 72hr. with T. v cellulase, the reducing sugar yield reached to 10.1%. 5. The rate of reducing sugar per each treated substrate was decreased by the order of the substrated, heated and then ball milled at $190{\pm}5^{\circ}C$. for 45 min. (11.5%)> without any treatment (10.1)> ball milled and heated at $190{\pm}5^{\circ}C$. for 45 min. (6.9%). 6. Saccharification of waste wood has been shown to be possible by heat treated and milling the substrate in contact with cellulase. And it is likely to be recommended that the waste wood may be valuable for raw materials of saccharification.
In this study, the mechanical stability of red mud was improved for its commercial use as a catalyst to effectively decompose HFC-134a, one of the seven major greenhouse gases. Red mud is an industrial waste discharged from aluminum production, but it can be used for the decomposition of HFC-134a. Red mud can be manufactured into a catalyst via the crushing-preparative-compression molding-firing process, and it is possible to improve the catalyst performance and secure mechanical stability through calcination. In order to determine the optimal heat treatment conditions, pellet-shaped compressed red mud samples were calcined at 300, 600, 800 ℃ using a muffle furnace for 5 hours. The mechanical stability was confirmed by the weight loss rate before and after ultra-sonication after the catalyst was immersed in distilled water. The catalyst calcined at 800 ℃ (RM 800) was found to have the best mechanical stability as well as the most catalytic activity. The catalyst performance and durability tests that were performed for 100 hours using the RM 800 catalyst showed thatmore than 99% of 1 mol% HFC-134a was degraded at 650 ℃, and no degradation in catalytic activity was observed. XRD analysis showed tri-calcium aluminate and gehlenite crystalline phases, which enhance mechanical strength and catalytic activity due to the interaction of Ca, Si, and Al after heat treatment at 800 ℃. SEM/EDS analysis of the durability tested catalysts showed no losses in active substances or shape changes due to HFC-134a abasement. Through this research, it is expected that red mud can be commercialized as a catalyst for waste refrigerant treatment due to its high economic feasibility, high decomposition efficiency and mechanical stability.
The process for ethanol production requires lignocellulosic biomass to be hydrolyzed to generate monomeric sugars for the fermentation. During hydrolysis step, a monomeric sugars and a broad range of inhibitory compounds (furan derivatives, weak acids, phenolics) are formed and released. In this study, we investigated the effects of inhibitory compounds on the fermentative performance of Saccharomyces cerevisiae K35 and Pichia stipitis KCCM 12009 in ethanol production, two yeast strains were fermented in the synthetic medium including six inhibitory compounds such as 5-hydroxymethylfurfura (5-HMF), furfural, acetic acid, syringaldehyde, vanillic acid and syringic acid. Ethanol of over 40 g/L was produced by two yeast strains in the absence of inhibitory compounds, respectively. Most inhibitory compounds except acetic acid had a little effect on the ethanol production, but acetic acid showed high inhibition effect on the cell growth and ethanol production.
To examine functionality of depolymerized alginate obtained by hydrolysis of alginate through a heating process at $121^{\circ}C$ on gastrointestinal physiology, the changes of body weight, organ weight and length, pancreatic and small intestinal composition, and light microscopy (LM) observation of small intestinal microvilli's appearances were checked in the rats. Rats were fed diets containing $1\%, 5\%, and 10\%$ of each depolymerized alginate (HAG-10, HAG-50, HAG-100) and alginate for 35 days, The feeding of 5 and $10\%$ HAG-50 and $10\%$ alginate diets for 35 days significantly depressed the body weight gain, but increased the length and weight of the small intestine and cecum in rats (p<0.01). Pancreatic protease activity was decreased significantly (p<0.01) in all groups except lo/o of HAG-10 diets, but the protein content increased in all groups, However, pancreatic amylase and lipase activities as well as DNA and RNA content were not significantly different. The small intestinal protein and the DNA content were the highest in diets fed $5\%$ HAG-50; RNA content increased significantly (p<0.01) in all groups except in the fiber-free diets. Light microscopy (LM) observation showed growth of small intestinal microvilli with numerous ridges; the multiplication of the convolution goblet cells in rats fed with diets containing $5\%$ of HAG-50 were more than others group.
