PIV/OH PLIF 동시 측정을 이용한 동축공기 수소확산화염의 실험적 연구 (Simultaneous PIV/OH PLIF Measurements in Hydrogen Nonpremixed Flames with Coaxial Air)

  • Simultaneous measurements of velocity and OH distribution were made using particle image velocimetry(PIV) and planar laser-induced fluorescence(PLIF) of OH radical in turbulent hydrogen nonpremixed flames with coaxial air. The OH radical was used as an approximate indicator of chemical reaction zone. The OH layer was correlated well with the stoichiometric velocity, $U_s$, instantaneously and on average. In addition, high strain-rate regions almost coincide with the OH distribution. The residence time in flame surface, calculated from the root-mean-square value of the radial velocity, is proportional to $(x/d_F)^{0.7}$. It is found that the mean value of principal strain rate on the OH layer can be scaled with $(x/d_F)^{-0.7}$ and therefore, the product of the residence time and the mean strain rate remains constant over all axial positions.

  • The sonochemical process has been applied as a treatment method to investigate its effect on the decomposition of humic substances (HS). The reaction kinetics and mechanisms in the process of sonochemical treatment for humic substances in wastewater have also been discussed. It was observed that the metal ions such Fe(II) and Mn(II) showed catalytic effects, while Al(III), Ca(II), and Mg(II) had inhibitory effects on the decomposition of humic substances in sonochemical reaction with hydrogen peroxide. Experimental results also showed factors such as hydrogen peroxide dose affected the formation of disinfection by-products. Two trihalomethanes, chloroform and dichlorobromomethane were formed as major disinfection by-products during chlorination. The depolymerization and the radical reaction of HS radicals appear to occur simultaneously. The final step of the reaction is the conversion of organic acids to carbon dioxide.

  • Insampaedok-san is a traditional medicine used for the treatment of colds. We investigated several compounds in Insampeadok-san, and tested their neuroprotective and anti-oxidative activities after fermentation with Lactobacillus. The amounts of four marker compounds (ferulic acid, hesperidin, 6-gingerol and glycyrrhizin) and unidentified compounds in Insampaedok-san (IS) and fermented Insampaedok-san (FIS) were measured and compared by an established HPLC-DAD method. Neuroprotective activity of IS and FIS extracts was evaluated and compared after glutamate-induced neurotoxicity in HT22 cells. Anti-oxidative activity of IS and FIS was also compared in DPPH free radical, hydroxyl radical and hydrogen peroxide scavenging activity assays. Contents of two compounds, ferulic acid and glycyrrhizin were decreased while 6-gingerol was increased by fermentation. FIS showed more potent neuroprotective activity than IS. DPPH, hydroxyl radical and hydrogen peroxide scavenging was slightly increased by FIS when compared to IS. In conclusion, fermentation with Lactobacillus can vary the amounts of the marker compounds in IS and improve neuroprotective and anti-oxidative activities of IS.

  • The ethanol extract of peony root (Paeonia Lactiflora Pall, Paeoniaceae) and its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. And protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were performed. The ethanol extract of peony root (PRE), gallic acid and methyl gallate were shown to possess the significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed the inhibitory effect of lipid peroxidation as expressed by malondialdehyde (MDA) formation. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, assessed by single cell gel electrophoresis. Furthermore, oral administration of 50% PRE (50% ethanol extract), gallic acid and methyl gallate potently inhibited micronucleated reticulocyte (MNRET) formation of mouse peripheral blood induced by KBrO3 treatment in vivo. Therefore, PRE containing gallic acid and methyl gallate may be a useful natural antioxidant by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage.

Theoretical Studies on the Hydrogen Atom Transfer Reaction (Ⅱ)$^*$

  • The hydrogen atom transfer reaction between substituted methane, $CH_3X,$ and its radical, $CH_2X(X=H,F,CH_3,CN,OH\;and\;NH_2$ was studied by MINDO/3 method. The transition state(TS) structure and energy barriers were determined and variation of the transition state and of the reactivity due to the change of X were analyzed based on the potential energy surface characteristics. It was found that the greater the radical stabilization energy. the looser the TS becomes; the TS occurs at about 15% stretch of the C-H bond, which becomes longer as the radical stabilization energy of $CH_2X$ increasers. The intrinsic barrier, ${\Delta}E*_{x.x},$ of the reaction with X was found to increase in the order $H The degree of bond stretch of the C-H bond stretch of the C-H bond at the TS also had the same order indicating that the homolytic bond cleavage of the C-H bond is rate-determining. Orbital interactions at the TS between LUMO of the fragment $C{\ldots}H{\ldots}C$ and the symmetry adapted pair of nonbonding, $n{\pm}(=n_1{\pm}n_2),$ or pi orbitals of the two X atoms were shown to be the dominant contribution in determining tightness or looseness of the TS. The Marcus equation was shown to apply to the MINDO/3 barriers and energy changes of the reaction.

흰목이와 흑목이 버섯의 다당추출 및 유리라디칼 소거활성 비교 (Polysaccharide Extraction and Comparison of Free Radical Scavenging Activities from Tremella fuciformis and Auricularia auricula Fruit Body)

  • The polysaccharides from fruit body of Auricularia auricula and Tremella fuciformis were extracted using hot water, and partially purified through ethanol precipitation and dialysis. Free radical scavenging activities of the crude and purified polysaccharides were examined and compared each other. Free radical scavenging activities of the partially purified polysaccharides were higher than those of crude polysaccharides. DPPH free radical, ABTS radical and SOD-like activities of partially purified polysaccharide at 1 mg/mL of concentration from A. auricula were 61.7, 9.6 and 38.9%, respectively, while those of T. fuciformis were 9.6, 5.7 and 15.3%, respectively. Results of site and non-site specific hydroxyl radical scavenging activities indicated that the partially purified polysaccharide fractions from A. auricula and T. fuciformis exhibited the hydroxyl radical scavenging effect by hydrogen donating ability and iron ion chelating ability. Also, reducing powers of A. auricula and T. fuciformis were 77.1 and 14.7% of BHT (0.1%) as standard, respectively. It was suggested that antioxidant activities of A. auricula were about 1.4~6.4 times higher than those of T. fuciformis due to different levels of polyphenol content.

  • 대기중 광화학반응에서 생산되는 이차 생성물중의 하나인 $H_2O$$_2$(hydrogen peroxide)는 peroxy radicals간의 결합에 의해 생성된다. HO$_2$ + HO$_2$ $\longrightarrow$ $H_2O$$_2$+ $O_2$ 이렇게 생성된 $H_2O$$_2$는 대기중에서 주요 산화제로 작용하며, pH 4.5 이하 수용액 내에서 S(IV)를 S(VI)로 산화시켜 H$_2$SO$_4$(sulfuric acid)를 생성 한다. 또한 $H_2O$$_2$는 대기중에서 odd-hydrogen radicals(OH, HO$_2$, and RO$_2$)의 저장고 역할과 함께 odd-hydrogen radical의 생성과 소멸에 작용하여 대기의 산화력을 반영한다(Lee et al. 2000). Elucidating H/D-Exchange Mechanism of Active Hydrogen in Aniline and Benzene-1,2-dithiol

  • In this study, the hydrogen/deuterium (HDX) exchange mechanism of active hydrogen, nitrogen, and sulfur-containing polycyclic aromatic hydrocarbon (PAH) dissolved in toluene and deuterated methanol by atmospheric pressure photoionization (APPI) is investigated. The comparison of the data obtained using APPI suggests that aniline and benzene-1,2-dithiol contain two exchanging hydrogens. The APPI HDX that best explains the experimental findings was investigated with the use of quantum mechanical calculations. The HDX mechanism is composed of a two-step reaction: in the first step, analyte radical ion gets deuterated, and in the second step, the hydrogen transfer occurs from deuterated analyte to de-deuterated methanol to complete the exchange reaction. The suggested mechanism provides fundamentals for the HDX technique that is important for structural identification with mass spectrometry.   • The degradation efficiency of chlorophenolic compounds in $TiO_2/H_2O_2$ combined system was compared with that of in $TiO_2$ sole system. As a result, the addition of hydrogen peroxide in photocatalytic oxidation reaction greatly enhanced the degradation efficiency of chlorophenolic compounds due to the availability of the hydroxyl radical formed on the $TiO_2$ surface. The hydrogen peroxide under UV illumination produces hydroxyl radicals that appear to be another source of hydroxyl radical formation. These results indicated the $TiO_2/H_2O_2$ combined system shows higher degradation efficiency than the $TiO_2$ sole system. Compared to another oxidation reaction, hydrogen peroxide assisted photocatalytic oxidation is more promising in practical aspect.

Effect of Vitamin E Against the Cytotoxicity of Reactive Oxygen Species on Vascular Endothelial Cells

  • Reactive oxygen species (ROS) is one of the main pathological factors in endothelial disorder. For example, an atherosclerosis is induced by the dysfunction of vascular endothelial cells. The dysfunction of vascular endothelial cells cascades to secrete intercellular adhesion molecule (ICAM)-l substance by ROS. Therefore, The ROS is regraded as an important factor of the injury of vascular endothelial cells and inducement of atherosclerosis. Oxygen radical scavengers playa key role to prevention of many diseases mediated by oxidative stress of ROS. In this study, the toxic effect of ROS on vascular endothelial cells and the effect of antioxidant, vitamin E on bovine pulmonary vascular endothelial cell line (BPVEC) treated with hydrogen peroxide were examined by the colorimetric assay. ROS decreased remarkably cell viability according to the dose- and time-dependent manners. In protective effect of vitamin E on BPVEC treated with hydrogen peroxide, vitamin E increased remarkably cell viability compared with control after BPVEC were treated with $15{\mu}M$ hydrogen peroxide for 6 hours. From these results, it is suggested that ROS has cytotoxicity on cultured BPVEC and oxygen radical scavenger such as vitamin E is very effective in prevention of oxidative stress-induced cytotoxicity.
