Neuroprotective Effect of Hwangryunhaedok-tang Against Glucose-deprivation Induced Cytotoxicity in PC-12 Cells (Glucose-deprivation으로 유발된 PC-12 cell의 세포 고사에 대한 황연해독탕(黃連解毒湯)의 보호 효과)

  • The purpose of the study was to confirm what effect HRHDT treatment had on cell extinction by damage of endoplasmic reticulum induced to PC-12 cell damage by glucose deprivation. The study confirmed what effect it had on forming the condition of glucose deprivation within a culture fluid of PC-12 cell and on a nerve cell's survival rates and tested whether HRHDT could prevent extinction of PC-12 cell by glucose deprivation. Also, the study confirmed what effect HRHDT treatment had on the emitted quantity of LDH by glucose deprivation. To examine PC-12 cell's behavioral change under the condition of glucose deprivation and a protective effect of HRHDT on the change, the study observed PC-12 cell's behavioral change with a microscope. Also, the study confirmed density of calcium ion within cells followed by a culture time in the condition of glucose deprivation with FACS and confirmed what effect HRHDT treatment had on the above density of calcium ion within cells. Finally, the study carried out the western blot and confirmed what effect HRHDT treatment had on revelation of GRP 78 and CHOP protein and a segmental type of aspase 12. In this study, HRHDT rescued PC-12 cells from glucose deprivation-induced cell death. HRHDT also prevents the LDH release, Ca++ accumulation, and morphological change, which was associated with the ER stress. Furthermore, HRHDT reduced the expression of ER chaperone (Grp78 and CHOP) proteins by glucose deprivation in PC-12 cells. These results suggest that HRHDT might provide a useful therapeutic strategy in treatment of the neurodegenerative diseases caused by glucose deprivation injuries.

Antioxidative and Antimicrobial Activities of Herbal Complex Extract from Hwangryunhaedok-tang and Essential Oil of Chamaecyparis obtusa (황련해독탕(黃連解毒湯)·편백(扁柏) 정유 복합추출물의 항산화 및 항균활성에 관한 연구)

  • Objectives : The present study was designed to investigate effects of mixed extracts (9:1, v/v) of Hwangryunhaedoktang, Chamaecyparis obtusa essential oil. We evaluated the antimicrobial and antioxidant activity by manufacturing mixed extracts as the materials for functional medicinal herb cosmetics. Methods : We performed antimicrobial were tested microbes (Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Propionibacterium acnes) by disc diffusion method and measure clear zone. Antioxidant activities were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. Results : Antimicrobial activities of mixed extracts against S. epidermideis, P. acnes, S. aureus, E. coli, and Candida. A were $10.9{\pm}3.0mm$, $16.9{\pm}2.0mm$, $9.2{\pm}2.0mm$, $10.3{\pm}1.0mm$, $11.8{\pm}3.0mm$ respectively. The Chamaecyparis obtusa essential oil had the highest antimicrobial activities against S. epidermideis, P. acnes, S. aureus, E. coli, and Candia. A and clear zone of microbes ware $16.7{\pm}3.0mm$, $28.8{\pm}0.2mm$, $15.9{\pm}2.0mm$, $11.5{\pm}1.0mm$, $16.3{\pm}3.0mm$. Hwangryunhaedoktang extract showed antimicrobial activity but only P. acnes and S. aureus The antioxidant activities of the mixed extracts were tested through the evaluation of DPPH radical scavenging activity. The 100 % mixed extracts were found to have 90 % DPPH radical scavenging activity. The mixed extracts was presented similar antioxidant activities compared with that of ascorbic acid. As a result, A mixture extract is expected to have antimicrobial effects and free radical scavenging activity was found. Conclusions : Accordingly, It can be concluded that mixed extracts has the potential to cosmetic material.

Comparative Effectiveness of Hwangryunhaedok-tang Pharmacopuncture, Essential Bee Venom Pharmacopuncture and Jungsongouhyul Pharmacopuncture for Cervical Pain caused by Traffic Accidents: A Retrospective Observational Study (교통사고 후 발생한 경항통에 대한 황련해독탕약침, 봉독약침, 중성어혈약침 치료의 효과 비교 연구: 후향적 관찰 연구)

  • Objectives The purpose of this study is to compare the effectiveness of Hwangryunhaedok-tang Pharmacopuncture, Bee Venom Pharmacopuncture and Jungsongouhyul Pharmacopuncture on cervical pain caused by traffic accident retrospectively. Methods The retrospective observational chart review was conducted to 52 cases of traffic accident patients who had admitted to Daegu haany University Pohang Korean Hospital from December 1st, 2015 to May 31th, 2017. The patients were classified into three groups by which pharmacopuncture was used as Hwangryunhaedoktang pharmacopuncture treatment group, bee venom pharmacopuncture treatment group and Jungsongouhyul pharmacopuncture treatment group. Additionally, outcomes should include Neck disability index(NDI) and patient's global assessment(PGA) for evaluate the treatment effects. Results Each pharmacopuncture treatment group showed significant decrease of NDI and PGA. But there was no significant difference in NDI and PGA between three groups. Conclusions According to the results, we found out that 3 kinds of pharmacopunctures are considered to be effective and useful for cervical pain patients caused by traffic injury. Further well designed prospective studies are needed.

Acute Toxicity Study of the Hwangryunhaedok-tang Extract in ICR Mice (황련해독탕(黃連解毒湯) 열수 추출물의 급성독성 연구)

  • Objective : The objective of this study was to evaluate the single dose oral toxicity of Hwangryundaedok-tang extract in ICR mice. Methods : 0(control group), 1250, 2500 and 5000 mg/kg of Hwangryundaedok-tang extracts were orally administered to 20 male and 20 female ICR mice. After single oral administration of Hwangryundaedok-tang extract to ICR mice, we observed number of the death, clinical signs, changes of body weights for 14 days. After 14 day of Hwangryundaedok-tang extract administration, all mice were sacrificed and major organs were observed. Results : Compared with the control group, we could not find any toxic signs in the mortalities, clinical signs, body weight changes, necropsy findings and hematological values in all treated groups(1250, 2500 and 5000 mg/kg). Conclusions : $LD_{50}$ value of Hwangryundaedok-tang extracts may be over 5000 mg/kg and it may have no side toxic effect to ICR mice.

A Case of Systemic Acute Erythema Multiforme (전신을 침범한 급성 다형홍반 치험 1례)

  • Objectives : The aim of this study is to report the improvement of symptoms in patients with systemic erythema multiforme through Korean medicine treatment. Methods : We treated 51 years old male patient who diagnosed with erythema multiforme for 15 days with acupuncture, herbal medicine, pharmacopucture, cupping therapy and Hwangryunhaedok-tang wet dressing therapy. We used two herbal medicine; Hwabanhaedok-tang for first 10 days and Danggwieumja for another 5 days. Improvement of symptoms was evaluated by Visual Analogue Scale(VAS) and photography of lesions. Results : Most of the erythema disappeared, leaving only pigmentation. Subjective symptoms of patients such as itching, hot flash, dryness were decreased from VAS 7 to VAS 1-2. Conclusions : We found that Korean medicine treatment including herbal medicine, acupuncture was effective in improving systemic acute erythema multiforme.

Study on Transfer Rate of Hazardous Materials in Herbal Medicine Prescriptions (한방처방 전탕 단계별 위해물질의 이행률에 대한 연구)

  • Objective: To compare the contents and transfer rate of hazardous substances in crude, washing solution, crude after washing, decoction and remnant after boiling. Methods: The heavy metal contents of each step were measured by inductively coupled plasma mass spectrometer (ICP-MS) and mercury analyzer (SP-3DS). In order to analyze pesticides in each sample we used simultaneous multi-residue analysis of pesticides by GC/ECD, which was followed by GC/MSD analysis to confirm the identity of the detected pesticide in each sample. In addition, the contents of sulfur dioxide ($SO_2$) were performed by Monier-Williams distillation method. Results: 1. Contents (mg/kg) of heavy metals were not detected in decoctions of any tested herbal medicine prescriptions. 2. Transfer rates (%) of heavy metals from crude to remnant were as follows: Yijin-tang (As: 46.9, Cd: 50.0 and Pb: 100.0), Oryung-san (As: 80.0, Cd: 100.0 and Pb: 73.8), Hwangryunhaedok-tang (As: 88.9, Cd: 71.4 and Pb: 92.7), Bangpungtongseong-san (As: 100.0, Cd: 17.3 and Pb: 56.1), Oyaksungi-san (As: 47.4, Cd: 175.0 and Pb: 142.4). 3. Contents (mg/kg) of residual pesticides were not detected in any samples. 4. Transfer rate (%) of sulfur dioxide ($SO_2$) from crude to remnant in all samples were as follows: Yijin-tang (25.0), Oryung-san (166.7), Hwangryunhaedok-tang (50.0), Bangpungtongseong-san (181.8), Oyaksungi-san (50.0). Conclusion: Our results showed that the boiled herbal medicine prescriptions which we take are safe from the hazardous substances.

The mean values of heavy metal contents (mg/kg) for the samples were as follows: Socheongryong-tang (before decoction - Pb; 1.115, Cd; 0.179, As; 0.069 and Hg; 0.028, after decoction - Pb; 0.110, Cd; 0.011, As; 0.005 and Hg; 0.002), Insampaedok-san (before decoction - Pb; 1.207, Cd; 0.148, As; 0.171 and Hg; 0.026, after decoction - Pb; 0.075, Cd; 0.006, As; not detected and Hg; O.OOD, Oryung-san (before decoction - Pb; 1.955, Cd; 0.430, As; 0.063 and Hg; 0.027, after decoction - Pb; 0.083, Cd; 0.013, As; 0.006 and Hg; 0.002), Hwangryunhaedok-tang (before decoction - Pb; 1.825, Cd; 0.210, As; 0.050 and Hg; 0.009, after decoction - Pb; 0.107, Cd; 0.010, As; 0.005 and Hg; O.OOD, Bangpungtongseong-san (before decoction - Pb; 1.740, Cd; 0.162, As; 0.585 and Hg; 0.018, after decoction - Pb; 0.041, Cd; 0.006, As; 0.022 and Hg; not detected) and Oyaksungi-san (before decoction - Pb; 1.199, Cd; 0.183, As; 0.321 and Hg; 0.031, after decoction - Pb; 0.096, Cd; 0.008, As; 0.021 and Hg; 0.0004). 2. Contents (mg/kg) of sulfur dioxide (S0$_2$) before a decoction in Socheongryong-tang, Insampaedok-san, Oryung-san, Hwangryunhaedok-tang, Bangpungtongseong-san and Oyaksungi-san exhibited 3.2, 5.7, 4.5, 49.8, 7.8 and 22.4, respectively. However, contents of sulfur dioxide after a decoction in all samples were not detected. 3. Contents (mg/kg) of residual pesticides before/after a decoction in all samples were not detected. Conclusions : These results will be used to establish a criterion of heavy metals, residual pesticides and sulfur dioxide.

Effects of Hwangryunhaedok-tang plus Yukmijihwang-tang add Chogudeung on Blood Pressure and Renal Function in Spontaneously Hypertensive Rats (황연해독탕 합 육미지황탕 가 조구등이 자발적 고혈압 백서의 혈압 및 현장 기능에 미치는 영향)

  • The present study examined the effects of water extract of Hwangruynhaedoktang plus Yukmijihwangtang add Chogudeung (HYC) on the blood pressure and renal function in spontaneously hypertensive rats (SHR). The HYC water extract blocked increase of systolic blood pressure (SBP) in SHR. The urinary volume (UV), sodium (UNaV), potassium (UkV), and chloride (UclV) excretions were significantly increased in SHR administered with high dose of water extract of HYC, but urinary osmolality was not changed. The creatinine clearance (Ccr), which is index of glomerular filtration rate (GFR), was increased by administration of HYC, while solute-free water reabsorption (TcH2O) was not changed. Taken together, these results suggest that the depressor effects of HYC, at least in part, caused by diuretic effect due to increase of glomerular filtration rate.

Antimicrobial Activity of Hwangryunheadok-tang and Extracts of Scutellariae Radix, Phellodendri Cortex, Coptis Rhizome, and Gardenia Jasminoides against Salmonella Typhimurium (黃連解毒湯, 黃連, 黃芩, 黃柏, 梔子의 Salmonella Typhimurium에 대한 항균효과)

  • Objectives: This experiment investigated the antimicrobial activity of Hwangryunheadok-tang and extracts of Scutellariae radix , Phellodendri cortex , Coptis rhizome , and Gardenia jasminoides against Salmonella typhimurium . Methods: After spreading S. typhimurium on a bacterial culture medium plate, antimicrobial activity was tested by dripping diluted Hwangryunheadok-tang or extracts of Scutellariae radix , Phellodendri cortex , Coptis rhizome , or Gardenia jasminoides (80 μl, diluted 100, 50, 10, and 1%) onto the plate, followed by culture for 16 to 72 hours. The minimal inhibitory concentration (MIC) was tested by dripping the minimum dilution that showed antimicrobial activity (80, 60, 40, and 20 μl) and measuring the density. The antimicrobial activity of Hwangryunheadok-tang and four extracts showed continuous antibacterial activity against S. typhimurium throughout the experiment. Result: 1. S. typhimurium . (Standard Microorganism, ATCC) 1) The Hwangryunheadok-tang and extracts of Scutellariae radix , Phellodendri cortex , and Coptis rhizome showed antibacterial activity in the undiluted solutions and in 50% dilutions. Gardenia jasminoides extract showed potency only in the undiluted solution. The antimicrobial potency of the undiluted solution was increased when the volume of inoculation was increased, but no difference was noted when the culture time was extended. All the extracts showed antimicrobial potency against S. typhimurium . 2. S. typhimurium isolated from diarrhea patients 1) When compared to the standard microorganism experiment on S. typhimurium , the MICs of the five extracts were increased. However, whereas the antimicrobial potency of doxycycline was lost entirely against bacteria isolated from patients with diarrhea, the antimicrobial potency of all the extracts was diminished but did not disappear. 2) The antimicrobial activity of Hwangryunheadok-tang and four extracts was continuous even when the culture time was extended to 16, 24, and 72 hours. Conclusions: The Hwangryunheadok-tang and four kinds of extracts have antimicrobial activity against S. typhimurium . Comparison of a standard microorganism with S. typhimurium isolated from diarrhea patients showed that the antimicrobial activity of all the extracts was better than that of antibiotics. Further studies should focus on the value and benefits of Hwangryunheadok-tang , and the four kinds of extracts as clinical treatments.

Case Study on Removing Lentigo Using a Cauterization (낙법(烙法)을 이용한 흑색점 제거 증례)

  • Objectives : This report is intended to suggest that lentigo can be removed using the cauterization. Methods : Handle of the needle, which is distal part to the tip and shaft, was heated with an alcohol lamp to cauterize the lentigo one by one to form a crust, and wet dressing was applied to the treatment sites by using Hwangryunhaedok-tang pharmacopuncture solution. Results : Ten days after the cauterization, the lentigo was cleared from the normal skin without adverse reactions. Conclusions : The cauterization is a useful technique for removing lentigo, but the conventional method is somewhat inconvenient, so a radiofrequency technique or a laser-based cauterization may be more useful Instead of the traditional cauterization.
