Studies on manufacture system of Hwanggum(黃芩) aqua-acupuncture by Boiling water Method (전탕에 의한 약침 황금제제의 제조에 관한 연구)

  • In order to study the reproducibility and effeciency of the manu-pacture system Hwanggum(黃芩) aqua-acupuncuture was prepared. The filtra-tion and dilution have been mainly employed in the study of the manupacture step. These methods were showed possibility of standardization and variety for aqua-acupuncture.

  • In order to study of safety, Hwanggum(黃芩) aqua-acupuncture was prepared by boiling extract and filtration. This study was performed to determine the toxic effects of graded dose levels and effects of Scutellaria Root after repeated administration(14days). From these results, Hwanggum(黃芩) aqua-acupuncture was quite sag in male and female rats.

  • These studies were conducted to investigate the effects of the various aqua-aqupunctures of Hwanggum(Scutellariae Radix) on the proliferation of 3T3-Ll cells. They were tested by means of Sulforhodamin B(SRB) assay. The results were summerized as follows: All tested aqua-aqupunctures inhibited the proliferation of preadipose 3T3-Ll cells. In case of the dilution of Hwanggum aqua-aqupuncture(HG), the results were quite opposite. In 1000 times dilution of HG(${\times}1000$), a low concentration increased the proliferation of preadipose 3T3-Ll cells, but the high($100{\mu}l\;and\;200{\mu}l$) concentration inhibited it. These results suggest that Hwanggum aqua-aqupunctures may be used on the obesity induced by the overgrowth of preadipose 3T3-Ll cells.

Effect of cookies made with soybean/seoritae and Hwanggum using response surface methodology on the blood glucose response in healthy adults (반응표면분석을 이용해 개발한 황금과 대두콩/서리태 분말 혼합 쿠키가 정상인의 혈당반응에 미치는 영향)

  • The goal of this study was to develop an optimal formula for cookies containing soybean/seoritae, Hwanggum, and isomalto-oligosaccharide using response surface methodology to achieve a blood glucose lowering effect. The model showed a good fit with the experimental data [$R^2=0.92$ (soybean) and 0.93 (seoritae)]. However, the p-value of lack of fit was less than 0.05 and ridge analysis was used to determine an optimal formula. The estimated optimal conditions were as follows: soybean cookie: 68.7% soybean, 2.5% Hwanggum, and 75.2% isomalto-oligosaccharide; seoritae cookie: 56.5% soybean, 3.8% Hwanggum, and 56.2% isomalto-oligosaccharide. The area under the curve and glycemic index were significantly lower in the soybean cookie group than in the control. The glycemic load (GL) index of the soybean (19.9) cookie was in the range of a low-glycemic food (<20 GL). These results can be applied to develop a cookie with a blood glucose lowering effect.

The Visible Injury and Physiological Responses of Two Varieties of Glycine max to Ozone (오존에 의한 두 품종 콩의 가시피해 및 생리적 반응)

  • A glass chamber experiment was conducted to evaluate the impact of ozone (O$_3$) on the physiology of two soybean, Glycine max L. cultivars, 'Hwanggum' and 'Jangyub'. Thirty-day old plants with 1-2 nodes were exposed to $O_3$ of <10 and 150 n1 1$^{-1}$ in the chambers for 8 h d$^{-1}$ for 3 days. Net photosynthesis, stomatal conductance, chlorophyll a, chlorophyll b and total chlorophyll contents, and foliar injury (% injured leaves) were measured. Although foliar damage was more severe on Jangyub than on Hwanggum, net photosynthesis was decreased by 60% on Hwanggum and 13% on Jangyub due to the $O_3$ treatment. Stomatal conductance on Jangyub was twice higher than that on Hwanggum and it was not changed by the $O_3$ treatment. Whereas, stomatal conductance on Hwanggum was 60-80% decreased by $O_3$, Chlorophyll contents did not change due to the $O_3$ treatment or variety. We can conclude that the reduction of net photosynthesis by $O_3$ was mainly due to the decreases of stomatal conductance and Rubisco (ribulose-1,5-bisphosphate carboxylase/oxygenase) activity on dark reaction. And foliar injury and chlorophyll content did not contribute to the net photosynthetic decrease. The gas-exchange variables measured 24 h after the termination of $O_3$ fumigation showed that there was no significant recovery within a day. Since the physiological responses on Jangyub were not much affected by the $O_3$ treatment, this variety could be $O_3$ resistant.

Effects of Soybean Varieties on the Physicochemical and Sensory Characteristics of Tofu (대두 품종별 두부의 이화학적, 관능적 특성)

  • The purpose of this study was to evaluate the effects of soybean varieties on the physicochemical and sensory characteristics tofu. Five varieties of soybeans(Daewon, Jinpoung, Hwanggum, Jinmi, and Manni) were selected for these experiments. Proximate composition and total phenolic acids in the soybeans, and the moisture, turbidity, lightness, yield rate, texture characteristics, and sensory characteristics of the tofu were measured. The Hwanggum soybeans had the highest levels of crude lipid and crude ash. There was a significant difference in the moisture and yield rate of the tofu(p<0.05). The crude protein and crude lipid contents of the soybeans affected the hardness and springiness of the tofu. In sensory evaluations, significant differences(p<0.05) were shown in taste, texture, and overall acceptability depending on the tofu by soybean variety. Therefore, one should consider soybean varieties when manufacturing tofu.

Retrospective Study of Crystalluria and Urinary Tract Symptoms in Dogs (개에서 발생한 결정뇨와 요로증후군에 대한 후향적 연구)

  • This retrospective study was performed in 40 cases of canine crystalluria and urinary tract symptoms. Breed, age, sex, diet history and common clinical signs were investigated for patient information. CBC, serum chemistry, urinalysis, urine culture, urine sediment and urinary stone analysis were undertaken for clinicopathological diagnosis. Crystalluria and urinary tract symptoms occurred from 6 months till 14 years in this clinical investigation and the average age was 4.68 years. It had taken an average 7.4 weeks to correct crystalluria and urinary tract symptoms treated by supportive therapy and dietary management. In conclusion, regular clinical pathologic examination, treatment and diet management should be needed to prevent the formation of urolithiasis and urinary tract diseases in dogs with crystalluria and urinary tract symptoms.

Studies on effect and Safety of various Aqua-acupunctures from Scutellariae Radix (다양(多樣)한 황금약침제제(黃芩藥鍼製劑)의 안전성(安定性) 및 효능(效能)에 관한 연구(硏究))

  • In order to detect the safety and effect of various aqua-acupunctures from Scutellariae Radix, the modifications of boiling, filtration and dilution were employed for the manufacture of aqua-acupunctures. We injected 0.2cc of aqua-acupunctures into Joksamri (足三里) of rat, repeatedly. We compared subacute toxicity of them with saline group, distilled water(D.W.) group, acupuncture group and control group. The results were summarized as follows: 1. The groups were all healthy and alive, and there was no special abnormality in physical condition and autopsy. And there were not any toxic symptoms in repeating application of aqua-acupunctures to the rat, including changes of body weight, organ weight, haematological examination and serum biochemical test. 2. There was slight change of body weight in acupuncture group : We could see significance after 3 days(p<0.05) and after 7 days(p<0.001) in body weight loss. After 9 days, all tested groups were suppressed in body weight increment. 3. Result of organ weight : In Palkang aqua-acupuncture(D-2 group), saline group and acupuncture group there were some statistical significance. Especially, acupuncture group revealed significant result in liver and spleen than aqua-acupunctures. From this result, we could suggest that the efficacy of acupuncture was preceded herbal medicine. 4. In serum biochemical test, we examined glucose(GLU), triglyceride(TG) and cholesterol(CHOL). In comparison with control group, the diluted 10 times of Hwanggum aqua-aqupuncture$({\times}10\;group)$ was recognized significant decrease of glucose, but the diluted 100 times of Hwanggum aqua-acupuncture$({\times}100\;group)$, D-2 group, saline group were confirmed significant increment. There was not any meaningful change of CHOL in all of tested group, excepting the acupuncture group was exhibited statistically significant decrease(p<0.05). In TG level all tested group except complex injection of standard compound(CPA group) and HG, there were significant value iii statistically. The diluted solution was more significant decrease than Hwanggum aqua-acupuncture(HG). The mutual relationship of components of aqua-acupunture tended to decrease level of TG, regardless of its concentration. In acupunture guoup, we gained some interesting result In meaningful decrease in 7G. 5. Haematological examination showed significant increment of granulocytes(GR) in all tested groups except Hwanggum aqua-acupuncture. And the diluted solutions of HG expressed very high increment of them(p<0.001). The GR and Mean Corpuscular Volume(MCV) of acupuncture group showed statistical significance.

Comparison of Growth Characteristics and Yield of Soybean Varieties for Soiling Crop Improvement (풋베기용 대두 개량을 위한 품종의 생육특성 및 수량 비교)

  • This Study was carried out to compare growth characteristics and yield of 10 cultivated soybean varieties, Jinpum I, Jinpum II, Hwanggum, Gumjung I, Gumjung II, Seukryangboot, Jangyup, Gumjungol, Bokwang, Manri for improvement of soiling crop. Plant height of Gumjung I(118cm), Gumjung II(114cm) and Jinpum I(114cm) were taller than other varieties based on dough stage. In total fresh weight and DM yield of Jinpum I, Jinpum II, Gumjung II were the highest of all varieties as 23,841kg, 23,499kg and 22,815kg per hectare, and 5,531kg, 5,173kg and 5,236kg per hectare, respectively. The highest leaf/stem ratio of ten soybean varieties were Manri(2.1), Jinpum I(1.9) and Hwanggum(1.9). Except early matured varieties, Seukryangboot(25.9%) and Gumjungol(25.4%), dry matter percent at dough stage were in the range of 22.0~23.4%. According to plant height, fresh weight, dry matter yield, and leaf/stem ratio, Jinpum I and Gumjung II were proper varieties to improve soiling crop.

Identification of Genetic Relationships Among Morus alba Genotypes Based on RAPD and ISSR Fingerprinting

  • Mulberries have importance in the sericulture industry as food for Bombyx mori, silkworm reared for its silk. Korean Morus alba have many cultivars and, for the protection of these cultivars and for utilization in plant-breeding programs, genetic information and the diversity among cultivars are essential. This study with 14 mulberry genotypes was undertaken using RAPD and ISSR fingerprinting to discover the genetic divergences between cultivars. Polymorphism rate among the cultivars produced by RAPD primer was found to be 64.48% and 66.29% relative to ISSR primer. The genetic relationships among the cultivars were identified using a dendrogram constructed with the UPGMA clustering method. Nei's method was used to calculate the genetic dissimilarity coefficients between each pair of genotypes, and the highest dissimilarity coefficient of 0.246 was exhibited between Suwon and Hwanggum cultivars. To determine the efficiency of each primer, a polymorphic index was calculated, and the robustness of the dendrogram was checked using cophenetic correlation coefficient. The results of this study can be utilized for the improvement of mulberry varieties in plant-breeding programs.
