  • The effect of rancid perilla oil on the immune response in mice was studied. ICR male mice were divided into 5 groups and were fed on the experimental diets for 4 weeks. Mice were sensitized and challenged with sheep red blood cell. Immune responses were evaluated by antibody production, Arthus reaction, delayed type hypersensitivity (DTH), Rosette forming cell and macrophage activity. Biochemical items were measured by serum protein and serum albumin. The weight of spleen, thymus and liver were measured. The rancid perilla oil diets decreased humoral and cellular immune responses, the number of peripheral circulating white blood cells and total protein and serum albumin. These results showed that the high rancid perilla oil diet decreased more humoral and cellular immune response, the number of peripheral circulating white blood cells, and total protein and serum albumin than the low rancid perilla oil diet did.

  • Experiments were performed on mice to investigate the effect of Panax ginseng petroleum ether fraction on the immunotoxicity of lead acetate. Lead acetate was administered in the drinking water and ginseng p-ether fraction was injected intraperitoneally, Mice were sensitized and challenged with sheep red blood cell. Humoral immune responses were evaluated by antibody production and Arthus reaction. Pathotoxicological influences were measured by serum protein, alkaline phosphatase and total cholesterol. The weight of liver, spleen and thymus were measured. Lead acetate exposure significantly decreased hemagglutination titer, hemolysin titer, Arthus reaction, spleen and thymus weight. Ginseng p-ether fraction administration significantly restored or potentiated reduced humoral immune response, spleen and thymus weight. Reduced serum A/G ratio, total cholesterol and alkaline phosphatase activity were restored or increased by ginseng p-ether fraction administration.

  • It has been more than a year since severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) first emerged. Many studies have provided insights into the various aspects of the immune response in coronavirus disease 2019 (COVID-19). Especially for antibody treatment and vaccine development, humoral immunity to SARS-CoV-2 has been studied extensively, though there is still much that is unknown and controversial. Here, we introduce key discoveries on the humoral immune responses in COVID-19, including the immune dynamics of antibody responses and correlations with disease severity, neutralizing antibodies and their cross-reactivity, how long the antibody and memory B-cell responses last, aberrant autoreactive antibodies generated in COVID-19 patients, and the efficacy of currently available therapeutic antibodies and vaccines against circulating SARS-CoV-2 variants, and highlight gaps in the current knowledge.

Ginsan Enhances Humoral Antibody Response to Orally Delivered Antigen

  • Background: There have been several reports describing the capability of ginseng extracts as an adjuvant. In this study, we tested if ginsan, a polysaccharide extracted from Panax ginseng, was effective in enhancing antibody response to orally delivered Salmonella antigen. Methods: Ginsan was treated before oral salmonella antigen administration. Salmonella specific antibody was determined by ELISA. mRNA expression was determined by RT-PCR. Cell migration was determined by confocal microscopy and flow cytometry. COX expression was detected by western blot. Results: Ginsan treatment before oral Salmonella antigen delivery significantly increased both secretory and serum antibody production. Ginsan increased the expression of COX in the Peyer's patches. Various genes were screened and we found that CCL3 mRNA expression was increased in the Peyer's patch. Ginsan increased dendritic cells in the Peyer's patch and newly migrated dendritic cells were mostly found in the subepithelial dome region. When COX inhibitors were treated, the expression of CCL3 was reduced. COX inhibitor also antagonized both the migration of dendritic cells and the humoral immune response against oral Salmonella antigen. Conclusion: Ginsan effectively enhances the humoral immune response to orally delivered antigen, mediated by CCL3 via COX. Ginsan may serve as a potent vaccine suppliment for oral immunization.

전씨백출산(錢氏白朮散)이 생쥐의 체액성(體液性) 면역반응(免疫反應)과 세포성(細胞性) 면역반응(免疫反應)에 미치는 효과(效果) (Effect of Junsibaekchulsan on the humoral and cell-mediated immune responses in mouse)

  • Even though appropriate immune response is necessary for the survival of the individual, excessive or insufficient immune response might cause autoimmune or allergic disease respectively. So the immune response must be controlled to the degree that is beneficial for the well being of the individual. This study was undertaken to know the effects of Junsibaekchulsan(JB) on the immune system od the mouse. For the evalulation of the cell-mediated immunity(CMI), delayed-type hypersensitivity against dinitrofluorobenzene(DNFB) were measured, and humoral immunity, hemagglutinin and hemolysin titers against SRBCs(sheep red blood cells) were measured, and rosette formation of spleen cells with SRBCs were measured. For the evaluation of innate immunity, phagocytic activity of macrophages, natural killer cell activity, and reactive nitrogen and oxygen intermediates were measured. The results are as follows: 1. The administration of JB depressed the antibody formation (hemagglutinin and hemolysin) against SRBCs. 2. The administration of JB did not affect the delayed-type hypersensitivity against DNFB. 3. The administration of JB did not affect the cytotoxic activity of natural killer cells. 4. The administration of JB increased the phagocytic activity of macrophages. 5. The administration of JB increased the rosette formating cells of the spleen cells. 6. The exposure of JB induced the secretion of reactive nitrogen intermediates but administration of JB deperssed the production of reactive oxygen intermediates. Administration of JB selectively depressed the humoral immune response without affecting CMI and innate immunity. These results of JB on the immune system might be useful for the treatment of such.

Modulation of Immune Response Induced by Co-Administration of DNA Vaccine Encoding HBV Surface Antigen and HCV Envelope Antigen in BALB/c Mice

  • Plasmid DNA vaccines encoding the hepatitis B virus (HBV) surface and hepatitis C virus (HCV) envelope antigens, respectively, were constructed, and attempt were made to find the possibility of a divalent vaccine against HBV and HCV. The expression of each plasmid in Cos-1 cells was confirmed using immunocytochemistry. To measure the induced immune response by these plasmids in vivo, female BALB/c mice were immunized intramuscularly with $100\;{\mu}g$ of either both or just one of the plasmids. Anti-HBV and HCV-specific antibodies and related cytokines were evaluated to investigate the generation of both humoral and cellular immune responses. As a result, specific anti-HBV and anti-HCV serum antibodies from mice immunized with these plasmids were observed using immunoblot. The levels of IL-2 and RANTES showing a $Th_{1}$ immune response were significantly increased, but there was no change in the level of IL-4 ($Th_{1}$ immune response) in any of the immunized groups. Compared with each plasmid DNA vaccine, the combined vaccine elicited similar immune responses in both humoral and cell-mediated immunities. These results suggest that the combined DNA vaccine can induce not only comparable immunity experimentally without antigenic interference, but also humoral and $Th_{1}$ dominant cellular immune responses. Therefore, they could serve as candidates for a simultaneous bivalent vaccine against HBV and HCV infections.

Comparison of immunoadjuvant activities of four bursal peptides combined with H9N2 avian influenza virus vaccine

  • The bursa of Fabricius (BF) is a central humoral immune organ unique to birds. Four bursal peptides (BP-I, BP-II, BP-III, and BP-IV) have been isolated and identified from the BF. In this study, the immunoadjuvant activities of BPs I to IV were examined in mice immunized with H9N2 avian influenza virus (AIV) vaccine. The results suggested that BP-I effectively enhanced cell-mediated immune responses, increased the secretion of Th1 (interferon gamma)- and Th2 (interleukin-4)-type cytokines, and induced an improved cytotoxic T-lymphocyte (CTL) response to the H9N2 virus. BP-II mainly elevated specific antibody production, especially neutralizing antibodies, and increased Th1- and Th2-type cytokine secretion. BP-III had no significant effect on antibody production or cell-mediated immune responses compared to those in the control group. A strong immune response at both the humoral and cellular levels was induced by BP-IV. Furthermore, a virus challenge experiment followed by H&E staining revealed that BP-I and BP-II promoted removal of the virus and conferred protection in mouse lungs. BP-IV significantly reduced viral titers and histopathological changes and contributed to protection against H9N2 AIV challenge in mouse lungs. This study further elucidated the immunoadjuvant activities of BPs I to IV, providing a novel insight into immunoadjuvants for use in vaccine design.

Effects of Swainsonine on the Humoral Immune Response of Lipopolysaccharide

  • Effects of swainsonine (SW;8${\alpha}$, ${\beta}$-indolizidine-${\alpha}$, $2{\alpha}$8-triol from Locoweed) on the humoral immune responses of lipopolysaccharide (LPS) wer studied in ICR mice. Mice were divided into 4 groups (10 mice/group), and LPS was given to each mouse 1 hr after i.p. injection with 3.7 mg/kg of swainsonine, by i.p. injection twice a week for 14 days at a dose of 2 mg/kg. Humoral immune responses were evaluated by hemagglutination (HA) titer and splenic plaque forming cells (PFC). The results of this study were summarized as follows: Mice administrated each of LPS and SW showed significant enhancement of the weight ratios of spleen to body, HA titer, 2-mercaptoethanol-resistant HA(MER-HA) titer and PFC compared with those in controls. However, the LPS plus SW treatment decreased HA titer, MER-HA titer and PFC corresponding to humoral immunity, as compared with those in the mice treated with LPS alone. These findings indicated that LPS significantly enhanced humoral immune responses, but their enhancement effects were lowered somewhat by SW.

고춧가루 첨가 식이가 흰주의 체액성 면역기능에 미치는 영향 (Effect of Dietary Hot Red Pepper Powder on Humoral Immune Response in Rat)

  • 고추가루 첨가 식이가 체액성 면역기능에 미치는 영향을 조사하였다. 실험동무로 흰쥐를 사용하여 대조군과 2%, 5%, 10% 고추가루 첨가 식이군을 설정한 후 27일간 실험식이로 사육하였다. 실험종료 10일 전에 각군의 흰쥐를 면양적혈구로 세차례 면역시킨 후 항체생성 세포수 및 혈청 중 항체 농도 등 체액성 면역기능을 중심으로 조사하였다. 그 결과, 대조군에 비해 고추가루 첨가 식이군의 항체 생성 세포수, 적혈구 응집반응 및 혈청중 항체 농도가 높은 것으로 나타났으며, 또한 이때 고추가루 첨가식이군의 각 장기조직 중 AsA 함량은 대조군에 비해 높은 것으로 나타났다. 이들 결과로 보아, 고추성분은 체액성 면역기능의 활성화에 기여하는 물질을 함유하고 있을 뿐만이 아니라, AsA의 공급효과가 뛰어난 것이 확인되었다.

Effects of Florfenicol and Chromium (III) on Humoral Immune Response in Chicks

  • One hundred and sixty day-old Hainan chicks were randomly allotted into eight pens to investigate the effect of different dietary concentrations of chromium (Cr) in the form of chromium chloride, and different dosages of florfenicol on humoral immune responses by determining antibody titers to Newcastle disease (ND) vaccines using the hemagglutination inhibition test. The results indicated that ND antibody titers were significantly higher in chicks receiving Cr at low (5 mg/kg feed) and middle (10 mg/kg feed) dose compared with the control (p<0.01). However, ND antibody titers were significantly decreased in chicks receiving Cr at a high dosage of 500 mg/kg feed (p<0.01), though the ND antibody titers of the early days (d 21 and d 28 of age) were higher than that of the control group. It is suggested that excessive Cr intake has detrimental effects on ND antibody production in chicks. No significantly lower response was measured in chicks that received florfenicol at a low dosage of 50 mg/kg feed (p>0.05), but the ND antibody titers were significantly decreased in chicks receiving 200 and 400 mg/kg feed of the drug (p<0.01). The ND antibody titers of group receiving 200 mg/kg feed of florfenicol plus 10 mg/kg Cr were slightly higher than that of the group receiving single florfenicol of 200 mg/kg although, no significant differences were observed between these two treatments. It is suggested that the humoral immune response impaired by florfenicol (200 mg/kg feed) could not be significantly reversed by Cr (10 mg/kg feed).
