Large-Scale Refolding and Enzyme Reaction of Human Preproinsulin for Production of Human Insulin

  • Human insulin is composed of 21 amino acids of an A-chain and 30 amino acids of a B-chain. This is the protein hormone that has the role of blood sugar control. When the recombinant human proinsulin is expressed in Escherichia coli, a serious problem is the formation of an inclusion body. Therefore, the inclusion body must be denatured and refolded under chaotropic agents and suitable reductants. In this study, H27R-proinsulin was refolded from the denatured form with β-mercaptoethanol and urea. The refolding reaction was completed after 15 h at $15^{\circ}C$, whereas the reaction at $25^{\circ}C$ was faster than that at $15^{\circ}C$. The refolding yield at $15^{\circ}C$ was 17% higher than that at $25^{\circ}C$. The refolding reaction could be carried out at a high protein concentration (2 g/l) using direct refolding without sulfonation. The most economical and optimal refolding condition for human preproinsulin was 1.5 g/l protein, 10 mM glycine buffer containing 0.6 M urea, pH 10.6, and 0.3 mM β-mercaptoethanol at $15^{\circ}C$ for 16 h. The maximum refolding yield was 74.8% at $15^{\circ}C$ with 1.5 g/l protein. Moreover, the refolded preproinsulin could be converted into normal mature insulin with two enzymes. The average amount of human insulin was 138.2 g from 200 L of fermentation broth after enzyme reaction with H27R-proinsulin. The direct refolding process for H27R-proinsulin was successfully set up without sulfonation. The step yields for refolding and enzyme reaction were comparatively high. Therefore, our refolding process for production of recombinant insulin may be beneficial to the large-scale production of other biologically active proteins.

  • Psychophysical data, in general, belong to one of the four scale categories : Nominal, Ordinal, Interval, and Ratio Scale. This paper introduces properties of the four scale categories and describes some psychophysical scales that attempt to measure subjective feeling or opinion of human. In addition, guidelines of analyzing and interpreting measured data are suggested. Some examples of analyzing and interpreting paychophysical data inappropriately are presented especially with category scales which have been used most widely in measuring subjective information.

A Study on Parenting Beliefs, Parental Satisfaction, and Child Rearing Stress (부모의 양육신념 및 역할만족도와 양육스트레스에 대한 연구: 3-7세 자녀를 둔 부모를 중심으로)

  • The relationships of parenting beliefs, parental satisfaction, and child rearing stress were examined in a sample 400 parents who have 3-7 aged children. Parents completed three kinds of questionnaires such as parenting beliefs scale, parental satisfaction scale, and child rearing stress scale. Data analysis was by mean, percentile, t-test, oneway-ANOVA, pearson correlation, and multiple regression. The major findings of this study were as follow; First there was a significant difference between parenting beliefs and child rearing stress. Second, there was a significant difference between parental satisfaction and child rearing stress. Third, conflict of parental role in parental satisfaction and problem solving in parent beliefs was the most predicting variables in child rearing stress.

  • Purpose: This empirical study is focused on the relationship between innovation capability (R&D and Human Resource innovation) and performance in knowledge-based service firms. Methods: We build research model to test how each of innovation capability on technology and human resource is influenced on their financial and non-financial performance in the knowledge-based service industries. Based on the previous research, we hypothesized the factors are regarded innovation capabilities of the firms as the scale of R&D and human resources. Because this study is especially targeted to the performance of knowledge-based service firms. With the survey on 424 main knowledge-based service firms, the multi-regression analysis was performed. Results: The result showed that the scale of R&D and human resources capabilities are main factors for knowledge-based service firms' performance, which reflects the current industrial structure. Conclusion: This study empirically demonstrated that human resources are most important to the growth of knowledge-based service firms.

  • As for structural system, which constitutes his construction, there are column inside dimension and span which constitutes plane, and as for constructions for section, there are C.H and slab, and as for constructive factors for envelope, there are window and closing panel of outer wall, and as for opening, there are punching window and wave window. With these constructive parts and opening of envelope, his construction composes volume and mass. The relation of structure and modulor which are shown in his later construction can be divided in two cases that modulor is directly and indirectly adapted for rate and measure calculation of the constructive body. As for indirectly adapted case, we can find it form most of his later construction, it is living place in which human beings life is, and it is adapted mainly for small space. In his construction, he tried to tell human scale and sense of musical rhythm through modulor. In other words, he played sense of space and musical scale by adapting regular and repetitive modulor of opening, and in small space for daily life, he made the size of space into the space which human can perceive. And, if we interpret mudulor in modern meaning, it is an establishment of radius of human act. And, we can make organic and harmonic design of space if we use modulor as origin of human centered measurement calculation, and if we adapt space after dividing by use.

Human Activity Recognition with LSTM Using the Egocentric Coordinate System Key Points

  • As technology advances, there is increasing need for research in different fields where this technology is applied. On of the most researched topic in computer vision is Human activity recognition (HAR), which has widely been implemented in various fields which include healthcare, video surveillance and education. We therefore present in this paper a human activity recognition system based on scale and rotation while employing the Kinect depth sensors to obtain the human skeleton joints. In contrast to previous approaches that use joint angles, in this paper we propose that each limb has an angle with the X, Y, Z axes which we employ as feature vectors. The use of the joint angles makes our system scale invariant. We further calculate the body relative direction in the egocentric coordinates in order to provide the rotation invariance. For the system parameters, we employ 8 limbs with their corresponding angles each having the X, Y, Z axes from the coordinate system as feature vectors. The extracted features are finally trained and tested with the Long short term memory (LSTM) Network which gives us an average accuracy of 98.3%.

  • A full-scale driving simulator is being developed as an effective test-bed for reproducing realistic driving situation in a safe and tightly controlled environment and conducting various human sensibility ergonomics studies. The simulator will be equipped with a 6 degree-of-freedom, high frequency motion base and a 4 channel, wide field-of-view visual system. The simulator will be fully interactive, highly realistic, and yet economical, taking advantages of the fast growing PC technology.

Development of Safety Competences, Behavioral Indicators and Measuring Methods for Preventing Human-Error in Nuclear Power Plants: A Preliminary Study (원전 인적오류 예방을 위한 안전 역량, 행동 지표 및 측정 방법 개발: 예비 연구)

  • The purpose of this study was to develop safety competences, a set of behavioral indicators of each competence and measuring methods of behavioral indicators for preventing human error of nuclear power plants(NPPs). The safety competences and behavioral indicators were derived from the five steps consisted of derivation of preliminary competence items through literature review, content analysis, interview(FGI, BEI), examination of content validity and decision making of final indicators. The results showed that 13 core safety competences and 35 behavior indicators were derived finally. In addition, the methods of measuring safety competences or behavioral indicators such as Behaviorally Anchored Rating Scale (BARS), Behavior Observation Scale (BOS) were developed and suggested.

  • The purpose of this study was to test the validity of Olson Circumplex Model and Beavers Systems Model related to adolescent adjustment. The 830 subjects were selected from the second grade of middle and high schools and adolescents of Juvenile Judge in the city of Taegu. The survey instruments were FACESIII, SFIII, State-Trait Anxiety Inventory, Depression Scale, and Delinquency Scale. Factor Analysis, Cronbach's ${\alpha}$. MANOVA, Scheff'e test were conducted for the data analysis. The major findings of this study were as follows: 1) Family system classification method on Olson Circumplex Model was partially useful in evaluating anxiety, depression, and delinquency of adolescent. 2) Family system classification method on Beavers Systems Model was partially useful in evaluating anxiety and depression of adolescent.
