외인성 insulin-like growth factor-I이 육계의 성장에 미치는 영향 (The effects of exogenous insulin-like growth factor-I on broiler chicken growth)

  • Insulin-like growth factor-I(IGF-I) plays an important role in the regulation of mammalian and poultry growth. IGF-I has many actions in different tissues, which include metabolic, mitogenic, and differentiative actions. IGF-I induces insulin-like effects - such as increased cell glucose uptake and glycogen sysnthesis, however several physiological actions of IGF-I may not have been identified yet. In order to investigate the effect on growth in broiler chicken treated with exogenous insulin-like growth factor-I, 30 chickens were injected $50{\mu}g$ reconbinant human IGF- I (rhIGF- I ) per kg body weight as experimental group and 30 ckickens saline subcutanously as control, 3 times according to ages from 2 to 35 days. We established radioimmunoassay method by which we can measure chicken IGF- I (cIGF- I ) as in rhIGF- I assay. The results obtained were as follows; 1) The dilution curve showed in parallelism between rhIGF- I and cIGF- I in the Sep-pak $C_{18}$ cartridge plasma extracts. 2) The body weight of broiler chicken were significantly increased at 31 days($1,176.50{\pm}99.79g$) and 35 days($1,252.84{\pm}125.21g$) of age in treatment groups, compared with control group($1,011.88{\pm}40.22g,\;1,111.32{\pm}153.67g$). The liver and kidney weights on 35 days$(35.24{\pm}5.18g,\;11.05{\pm}1.47g)$ were significantly higher in rhIGF- I treated group than control group($30.95{\pm}4.04g,\;10.01{\pm}1.60g$) 3) The plasma concentration of IGF- l and total protein in rhIGF- I treated group were $58.17{\pm}1.69ng/ml$, $3.75{\pm}0.62g/dl$ respectively compared with control group $45.70{\pm}1.64ng/ml$, $2.32{\pm}0.53g/dl$. The results suggest that exogenous rhIGF- I increased total body weight, liver and kidney weights in broiler chicken, and it may increase IGF- I and total protein concentration in serum.

  • Transforming growth factor $\beta$1(TGF-$\beta$1)은 여러 가지 생물학적 활성을 가지는 관계로 의학적 치료제로서 사용될 가능성이 크다. 본 연구에서는 혈소판추출, 젤여과, 양이온교환 크로마토그래피 및 역상 HPLC등 네 단계의 정제과정으로 이루어져 있는 정제공정을 이용한 TGF-$\beta$1을 값싸고 효울적으로 정제하였다. 이 과정을 거쳐 최종적으로 얻어진 TGF-$\beta$1은 비환원조건하에서 SDS-PAGE를 행한 결과 구매된 TGF-$\beta$1 표준품과 일치한 위치에서 한 개의 band가 관찰되어 순수하다는 것을 확인하였으며 또한 이것이 Westem blot를 통하여 TGF-$\beta$1 항체와 결합하는 것으로부터 TGF-$\beta$1임을 확인하였다 또한, mink lung epithelial cell line 을 이용한 성장저해 실험을 통해 정제된 TGF-$\beta$1이 구매되TGF-$\beta$1 표준품보다 조금 높은 활성을 가지는 것을 확인하였다 최종적으로 농축혈소판 10단위로부터 약 3.7$\mu$g의 정제된 TGF-$\beta$1이 얻어져 그 최종수율은 약 21%였다.

  • 인간 췌장 유래 pro-carboxypeptidase B(CPB) 유전자를 클로닝한 후 GAL10 promoter 하류에 Saccharomyces cerevisiae mating factor alpha-1 secretion signal $(MF{\alpha}-1)$과 연결하여 $MF{\alpha}1$-pro-CPB를 구축하였다. 구축된 plasmid $pY{\alpha}$-hproCPB(7.72 kb)를 S. cerevisiae 2805에 형질 전환시켰다. 재조합 인간 proCPB(hproCPB)는 galactose 유도로 S. cerevisiae에서 성공적으로 발현되었고, 배양상등액으로 분비되었다. SDS-PAGE와 western blot 분석에 의해, 정제된 hproCPB 단백질이 약 45.9kDa임을 확인하였다 S. cerevisiae $2805/pY{\alpha}$-hproCPB의 발효조 회분배양 시 trypsin 처리에 의해 pro영역을 제거한 후 hCPB의 세포외 활성은 60시간째에 10.16 unit/mL이었다. 또한 재조합 hCPB의 Km 값은 약 0.43 mM 이었다.

Establishment of a Pancreatic Cancer Stem Cell Model Using the SW1990 Human Pancreatic Cancer Cell Line in Nude Mice

  • Aim: To establish a pancreatic cancer stem cell model using human pancreatic cancer cells in nude mice to provide a platform for pancreatic cancer stem cell research. Materials and Methods: To establish pancreatic cancer xenografts using human pancreatic cancer cell line SW1990, nude mice were randomly divided into control and gemcitabine groups. When the tumor grew to a volume of $125mm^3$, they treated with gemcitabine at a dose of 50mg/kg by intraperitoneal injection of 0.2ml in the gemcitabine group, while the mice in control group were treated with the same volume of normal saline. Gemcitabine was given 2 times a week for 3 times. When the model was established, the proliferation of pancreatic cancer stem cells was observed by clone formation assay, and the protein and/or mRNA expression of pancreatic stem cell surface markers including CD24, CD44, CD133, ALDH, transcription factors containing Oct-4, Sox-2, Nanog and Gli, the key nuclear transcription factor in Sonic Hedgehog signaling pathway was detected by Western blot and/or RT-PCR to verify the reliability of this model. Results: This model is feasible and safe. During the establishment, no mice died and the weight of nude mice maintained above 16.5g. The clone forming ability in gemcitabine group was stronger than that of the control group (p<0.01). In gemcitabine group, the protein expression of pancreatic cancer stem cell surface markers including CD44, and ALDH was up-regulated, the protein and mRNA expression of nuclear transcription factor including Oct-4, Sox-2 and Nanog was also significantly increased (P<0.01). In addition, the protein expression of key nuclear transcription factor in Sonic Hedgehog signaling pathway, Gli-1, was significantly enhanced (p<0.01). Conclusions: The pancreatic cancer stem cell model was successfully established using human pancreatic cancer cell line SW1990 in nude mice. Gemcitabine could enrich pancreatic cancer stem cells, simultaneously accompanied by the activation of Sonic Hedgehog signaling pathway.

항해사의 피로가 해양사고에 미치는 영향 평가에 관한 연구 (A Study on the Effects of Marine Accidents by Navigation Officers' Fatigue)

  • 최근 국내에서는 연간 약 600여건의 각종 해양사고가 발생하고 있다. 해양사고의 발생에 관한 국 내외 많은 연구와 분석에 의하면 해양사고의 약 70~80%가 인적요인에 의해 발생하고 있다. 여러 인적요인 중에서도 항해사의 피로는 매우 중요한 요인이다. 항해사의 피로가 해양사고에 중요한 역할을 한다고 인식되고 있음에도 불구하고 항해사의 피로도를 정량적으로 파악할 수 있는 연구는 거의 미미하다. 따라서 본 연구에서는 해양사고 종류별로 인적요인에 의한 발생률을 분석하고, 문헌과 5점 척도를 이용한 설문조사를 통하여 항해사의 피로에 영향을 미치는 중요한 5가지 요인을 추출하였다. 그리고 ISM법을 이용하여 피로요인에 의한 해양사고 발생 위험성의 평가요소를 계층구조화하였다. 마지막으로 AHP법을 이용하여 추출된 각 피로요인의 중요도를 구하고 항해사의 피로에 의해 발생할 가능성이 높은 해양사고의 순위를 결정하였다. 그 결과 수면시간 0.386, 스트레스 0.302, 건강상태 0.139, 휴식시간 0.099, 음주 약물 0.074순으로 높은 것으로 나타났다. 또한, 인명사상사고 0.328, 충돌사고 0.308, 좌초사고 0.195, 침몰사고 0.094, 화재사고 0.075로 나타났다. 따라서 중요도가 높은 요소들을 중심으로 제어방안을 마련해야 할 필요가 있다.

Comparative evaluation of obesity-related parameters in junior sumo wrestlers and children with obesity

  • [Purpose] Exercise is a key factor in preventing obesity and metabolic syndrome. Sumo wrestlers increase their body size from childhood for athletic advantage; however, the risk of metabolic syndrome in junior sumo wrestlers is undetermined. Preventive measures against pediatric obesity should be initiated during childhood to prevent obesity in adulthood, considering its high global incidence. We comparatively evaluated the risk factors for metabolic syndrome in junior sumo wrestlers and children with obesity. [Methods] We enrolled 70 male children (age 9-17 years [sumo group, n = 14] and 9-14 years [other sports and non-exercise groups, n = 28 each]) and evaluated their anthropometric parameters (height, weight, body mass index z-score, obesity rate, waist circumference, waist to height ratio) and hematological parameters (total, low-density, high-density, and non-high-density lipoprotein-cholesterol; triglycerides; plasma glucose, and glycated hemoglobin levels). [Results] The BMI z-score, obesity rate, waist circumference (p < 0.05, along with the non-exercise group), and systolic blood pressure were significantly higher and the high-density cholesterol level was lower in the sumo group than in the other sports group (p < 0.05). The waist to height ratio was significantly higher in the non-exercise group than in the other sports group (p < 0.05). No significant difference was found in other blood lipid, plasma glucose (significantly lower level than the reference range in the sumo group, p < 0.05), and glycated hemoglobin (within the reference range in all groups) levels among the three groups. [Conclusion] Junior sumo wrestlers had a larger body size and higher blood pressure than children with obesity who exercised regularly. This provides direction for future research into targeted preventive interventions against metabolic syndrome for junior sumo wrestlers with large body size.

Rat의 방사선 조사성 구내염에 대한 Recombinant Human Epidermal Growth Factor (rhEGF)의 효과 (Effects of Recombinant Human Epidermal Growth Factor (rhEGF) on Experimental Radiation-Induced Oral Mucositis in Rats)

  • 목적 : 구내염은 두경부종양이 있는 암환자에게 방사선 및 항암제 치료 시술 시 매우 흔하게 발생하는 합병증이다. 본 연구는 Rat 의 방사선성 구내염 모델에서 recombinant human epidermal growth factor (rhEGF)의 효과를 평가하고자 하였다. 대상 및 방법: 25 Gy의 방사선량으로 두부에 단회 조사한 Rat를 무작위로 7마리씩 무처치군, 부형제 처치군, rhEGF 15 또는 $30{\mu}g/day$ 구강 내 처치군으로 나누었으며, 방사선을 조사하지 않은 7 마리의 Rat를 정상시험군으로 나누었다 rhEGF 시료 및 부형제는 1일 3회 Rat 의 구강점막에 매일 도포하였다. Rat의 생존율, 체중변화 및 사료섭취량을 18일 동안 관찰하였으며, 방사선 조사 후 7 일 및 18 일째에 Rat의 구강점막을 조직학적으로 평가하였다. 결과 : 실험종료 시점에서 rhEGF 15 또는 $30{\mu}g/day$ 구강 내 처치군이 모두 33%의 생존율을 보인 것에 비하여, 무처치군 및 부형제 처치군은 모두 0%의 생존율을 보였다. 체중변화에서도 rhEGF 처치군은 방사선 조사 후 2일부터 7일까지 부형제 처치군에 비하여 Rat 의 평균체중이 통계적으로 더욱 무거웠다 사료섭취율은 모든 시험군에서 방사선 조사 후 4 일까지 감소하는 양상을 보이다가, rhEGF 15 또는 $30{\mu}g/day$ 구강 내 처치군에서 14일째에 뚜렷한 사료섭취율의 증가가 관찰되었다. 방사선 조사 후 7 일째의 조직학적 분석 결과, rhEGF 15 또는 $30{\mu}g/day$ 구강 내 처치군의 Rat 에서는 점막 표피층의 각질세포의 종창 및 변성만이 관찰되었던 것에 비하여, 무처치군 및 부형제 처치군에서는 심한 위막성 또는 궤양성 구내염이 관찰되었다. 결론; rhEGF (15 또는 $30{\mu}g/day$ 구강 내 처치군) 처치에서 방사선 조사로 유발시킨 Rat의 구내염 모델에서 유의성 있는 치유 효과를 확인하였으며, 본 시험결과로 rhEGF가 방사선에 의해 유발된 구내염을 치료할 수 있는 임상 제제로써의 가능성을 볼 수 있었다.

Effects of γ-aminobutyric acid-enriched fermented sea tangle (Laminaria japonica) on brain derived neurotrophic factor-related muscle growth and lipolysis in middle aged women

  • This study evaluated the effects of γ-aminobutyric acid (GABA)-enriched fermented sea tangle (GFST), as a functional food, on brain derived neurotrophic factor (BDNF)-related muscle growth and lipolysis, in a sarcopenic obesity high-risk group. Twenty-one middle-aged women (53-63 y) participated in this randomized, double-blind, placebo controlled study. Participants ingested either 1,000 mg of GFST (n = 10) or a sucrose placebo (CON) (n = 11) everyday, for 8 weeks. Subjects were asked to abstain from any regular exercise. Fasting venous blood samples, body composition and muscular strength were measured before and after supplementation period. Collectively, we demonstrated that GFST significantly decreased total fat mass and triglyceride in body composition, as well as significantly increasing serum BDNF (p < 0.001), angiotensin converting enzyme (p < 0.001), human growth hormone and insulin-like growth factor-1 levels (p < 0.05 and p < 0.05, respectively) accompanied by increased total lean mass (p < 0.01). Furthermore, the reported improvements in total work, knee extension and flexion at 60° s−1 (p < 0.05), and peak torque normalized to body weight of knee flexion at 60° s−1 (p < 0.05), support an ergogenic effect of GABA associated with increased growth factor levels. The use of GFST, as a functional food ingredient, to elicit anti-obesity effects and stimulate the release of muscle-related growth factors with increasing serum BDNF levels may provide a protective intervention for age-related degeneration such as sarcopenic obesity.

  • This study investigates the effects of sociocultural influences and body image on appearance enhancement behaviors (facial management, clothing selection, and weight/figure management). For data collection, a questionnaire was administrated to 562 female college students in Gwangju City, Chonnam area and Chonbuk area, Korea, from May 23 to June 10, 2011. To analyze the data, a SPSS 18.0 statistics package was used, and descriptive statistical analysis, frequency analysis, factor analysis, reliability analysis, and regression analysis were conducted. The results were as follows. First, sociocultural influences were divided into three factors: parental influence, media influence, and peer influence. Overall sociocultural influences had positive effects on appearance enhancement behavior. Second, body image was divided into two factors: weight-concern and appearance-concern. Sociocultural influences had significant effects on overall body image. Third, body image appeared to have positive effects on overall appearance enhancement behavior.

  • In need of studies on the kinds and structure of thread, the biggest variable factor in knitwear patterns, this study attempts: to examine the physical properties by thread type to basically establish systematic data in order to utilize various mixture and structure of yarn and to contribute to the development of optical patterns by building a systemic and scientific methods to produce knit wear patterns though a statistical analysis of the relation between the variations and physical properties. The results is as follows: with time, a feature of knit, which causes instability making it difficult to maintain the original shape, related to material properties, the weight and expansibility recovery rate have the greatest influence on the variation of wale lengths, though the amount varies by material. The variation of course contraction is closely related to density, the dense fabrics showing the highest values, due to the bust of the human body, the wale length variation of the front is greater than that of the back, by a regression analysis of material properties and the variations is obtained showing the weight, density and expansibility recovery rate have the greatest influence on the wale extension and course contraction of knit.
