Metabolomic studies using human urine have shown that human metabolism is altered by a variety of environmental, cultural, and physiological factors. Comprehensive information about normal human metabolite profiles is necessary for accurate clinical diagnosis of disease and for disease prevention and treatment. In this study, metabolite correlation analyses, using $^1H$ nuclear magnetic resonance (NMR) spectroscopy coupled with multivariate statistics, were performed on human urine to compare metabolic differences based on gender and/or obesity in healthy human subjects. First, we applied partial least squares discriminant analysis to the NMR spectral data set to verify the data's ability to discriminate by gender and obesity. Then, the differences in metabolite-metabolite correlation between male and female, and between normal and high body mass index (obese) subjects were investigated through pairwise correlations. Creatine and several metabolites, including isoleucine, trans-aconitate, and trimethylamine N-oxide (TMAO), exhibited different quantitative relationships depending on gender. Dimethylamine had a different correlation with glycine and TMAO, based on gender. The correlation of TMAO with amino acids was considerably lower in obese, compared to normal, subjects. We expect that the results will shed light on the metabolic pathways of healthy humans and will assist in the accurate diagnosis of human disease.
We describe an effective method of microchip electrophoresis (ME) based on single strand conformation poly-morphism (SSCP) analysis to rapidly detect the point mutation, Leu72Met, in a human obesity gene. The 207-bp dsDNA in the Leu72Met region, an estimate of the child obesity DNA mutant, was amplified by polymerase chain reaction (PCR) and submitted to a conventional glass microchip analysis with a sieving matrix of 1.75% poly(vinylpyrrolidone) (Mr 1 300 000), 1.0% poly(ethyleneoxide) (Mr 600 000) and 5.0% w/w glycerol. When combined with base stacking (BS) with hydroxide ions, the SSCP-ME provided rapid analysis as well as sensitive detection. The detection sensitivity was effectively enhanced in the OH- concentration range of 0.01-0.025 M NaOH. The sensitivity and speed of this ME-based SSCP methodology for the rapid detection of Leu72Met point mutations makes this an attractive method for diagnosing childhood obesity in a clinical diagnostic laboratory.

Objectives The purpose of this study was to examine the characteristics of diagnostic pattern questionnaire associated with Body Mass Index in premenopausal women. Methods We studied 41 patients visiting Gangdong Kyung Hee University Hospital from 1st April 2011 to 25th May 2011. The subjects were categorized in two groups, overweight & obesity group (n=12) and low weight & normal group (n=29). We studied the difference of diagnostic pattern questionnaire scores between two groups by Independent samples T-test and correlation between diagnostic pattern questionnaire scores of overweight & obesity group by Pearson's correlation coefficient test using SPSS for windows (version 13.0). Results Blood stasis, retention of undigested food, cold pattern scores of overweight & obesity group were non-significantly higher than low weight & normal group. Heat pattern score of overweight & obesity group was significantly higher than low weight & normal group. Cold-heat, phlegm-cold, blood stasis-cold, phlegm-blood stasis, phlegm-retention of undigested food significantly showed positive correlation coefficient in overweight & obesity group. Conclusions The results suggest that obese women tend to show heat pattern. It seems to be that multiple factors such as phlegm, blood stasis, retention of undigested food are causative of obesity.
Background: As data on the relation between obesity and lymph node ratio are missing in the literature, we here aimed to assess the impact of obesity on this parameter and other clinicopathological features of breast cancer cases and patient survival. Materials and Methods: Medical data of 646 patients, all referred to two centers in Tehran, Iran, were reviewed. Factors that showed significant association on univariate analysis were entered in a regression model. Kaplan-Meier and Cox-regression were employed for survival analysis. Results: Obesity was correlated with the expression of estrogen and progesterone receptor (p=0.004 and p=0.039, respectively), metastasis to axillary lymph nodes (p=0.017), higher lymph node rate (p<0.001) and larger tumor size (p<0.001). The effect of obesity was stronger in premenopausal women. There was no association between obesity and expression of human epidermal growth factor receptor. Three factors showed independent association with BMI on multivariate analysis; tumor size, estrogen receptor and lymph node ratio. Obesity was predictive of shorter disease-free survival with a hazard ratio of 3.324 (95%CI: 1.225-9.017) after controlling for the above-mentioned variables. Conclusions: The findings of this study support the idea that obese women experience more advanced disease with higher axillary lymph node ratio, and therefore higher stage at the time of diagnosis. Furthermore, obesity was associated with poorer survival independent of lymph node rate.
Objectives: This study examined trends in the health status and dietary behavior changes by region using the raw data from the 2008 ~ 2019 Community Health Survey. Methods: This study analyzed the data of 2,738,572 people among the raw data of the Community Health Survey from 2008 to 2019. The regional differences in health status and dietary behavior were examined by classifying the regions into capital and non-capital regions, and the non-capital regions were classified into metropolitan cities and provinces. A chi-square test was conducted on the body mass index (BMI), diagnosis of diabetes and hypertension, frequency of eating breakfast, salty taste in usual diet, recognition of nutrition labeling, reading of nutrition labeling, and utilization of nutrition labeling. Results: In determining obesity using the BMI, the normal weight by year decreased, and the obesity rate by year was 34.6% in 2019, which increased by 13% compared to 2008. In addition, the diabetes diagnosis rate and hypertension diagnosis rate continued to increase with the year. Both diabetes and hypertension diagnosis rates were higher in the non-capital regions than in the capital region. Eating breakfast five to seven times per week was most common and showed a significant decreasing trend by year (P < 0.001). The percentage of respondents who said they eat slightly bland foods increased from 19.5% in 2008 to 19.9% in 2010 and then to 22.1% in 2013. The percentage then decreased to 19.9% in 2019, but showed an overall increasing trend (P < 0.001). According to the region, the capital region had a higher percentage than the non-capital region. The nutrition labeling's recognition rate and utilization rate increased yearly, whereas the reading rate decreased. Conclusions: The study results presented the primary data necessary to develop nutrition education programs and establish strategies for local nutrition management projects to improve disease prevention and dietary problems.
In the past gut microbiome has been the main focus of microbiome research. Studies about the microbiome inside oral cavities and other organs are underway. Studies about the relationship between noninfectious diseases and periodontal diseases, and the negative effects of harmful oral microbes on systemic health have been published in the recent past. A lot of attention is being paid towards fostering a healthy oral microbial ecosystem. This study aimed to understand the roles and effects of the microbiome inside the human body can potentially help cure various diseases including inflammatory bowel diseases with no known cure such as Crohn's disease, atopic dermatitis, obesity, cancer, diabetes, brain diseases and oral diseases. The present study examined technological trends in the correlation between the human microbiome and diseases in the human body, interactions between the human body's immunity, the metabolic system, and the microbiome, and research trends in other countries. While it has been proven that human microbiome is closely correlated with human diseases, most studies are still in the early stage of trying to compare the composition of microbiomes between health and patient groups. Since the oral environment is a dynamic environment that changes due to not only food intake but also other external factors such as lifestyle, hygiene, and drug intake, it is necessary to continue in-depth research on the microbiome composition characteristics to understand the complex functions of oral microorganisms. Analyzing the oral microbiome using computational technology may aid in disease diagnosis and prevention.
The metabolic syndrome, by definition, is not a disease but is a clustering of individual metabolic risk factors including abdominal obesity, hyperglycemia, hypertriglyceridemia, hypertension, and low high-density lipoprotein cholesterol levels. These risk factors could dramatically increase the prevalence of type 2 diabetes and cardiovascular disease. The reported prevalence of the metabolic syndrome varies, greatly depending on the definition used, gender, age, socioeconomic status, and the ethnic background of study cohorts. Clinical and epidemiological studies have clearly demonstrated that the metabolic syndrome starts with central obesity. Because the prevalence of obesity has doubly increased worldwide over the past 30 years, the prevalence of the metabolic syndrome has markedly boosted in parallel. Therefore, obesity has been recognized as the leading cause for the metabolic syndrome since it is strongly associated with all metabolic risk factors. High prevalence of the metabolic syndrome is not unique to the USA and Europe and it is also increasing in most Asian countries. Insulin resistance has elucidated most, if not all, of the pathophysiology of the metabolic syndrome because it contributes to hyperglycemia. Furthermore, a major contributor to the development of insulin resistance is an overabundance of circulating fatty acids. Plasma fatty acids are derived mainly from the triglycerides stored in adipose tissues, which are released through the action of the cyclic AMP-dependent enzyme, hormone sensitive lipase. This review summarizes the latest concepts in the definition, pathogenesis, pathophysiology, and diagnosis of the metabolic syndrome, as well as its preventive measures and therapeutic strategies in children and adolescents.
1. Backgrounds and Aims Human being is divided into 4 constitution types(Taeyangin, Soyangin, Taeumin, and Soumin). According to Sasang Constitutional Medicine[SCM] Each constitution has its own incidence, treatment and prevention of disease. The purpose of this study is to find the relation of dietary habits, life styles and Psychosocial stress with obesity in each Sasang Constitution. 2. Methods 975 subjects who have received health examinations and diagnosis of Sasang Constitution at Kyung-hee Medical Center were divided into each constitutional group. Each constitutional group was then divided into obese group and normal group. We evaluated risk factors of obesity such as dietary habits, life styles, and Psychosocial stress. These data were statistically analysed to investigate the relations between risk factors and obesity. And then significant factors were analysed by multinomial logistic regression analysis to get each odds ratio. 3. Results In a multinomial logistic regression analysis adjusted for age and sex, promotion of appetite and overeating or excessive diet elevated risks of obesity in Taeumin, promotion of appetite and high speed of diet elevated risks of obesity in Soyangin, and Psychosocial stress elevated risks of obesity in Soeumin. 4. Conclusions The results suggest that promotion of appetite and overeating or excessive diet may be major risk factors for obesity in Taeumin, promotion of appetite and high speed of diet in Soyangin and Psychosocial stress and dyschezia in Soeumin.

Objectives ; Sasang Constitional Medicine is a part of Korean traditional Medicine, and classify human constitution into four types. Scalp and hair is a part of skin. Skin is one of constitutional charasteristic element. So we researched a correlation between Sasang constitutions and Korean adult scalp and hair condition. This is a basic study to find constitution can be a important factor to care scalp and hair. Methods ; We examined Sasang constitutions and scalp and hair condition of 80 human who aged from 20-50. Scalp disease, dandruff was observed by video micro scope and sebum tape diagnosis. Thickness of hair was measured by Laser Scan Micrometer(LSM-3100). Itching sense and life habit was also investigated. Constitution diagnosis and scalp, hair diagnosis was investigated seperately as a blind test and the data was statistically analyzed with chi-square test, correlation analysis, one-way analysis of varience. Results ; 1. In Sasang Constitution, Taeumin group had significant tendency of oilily scalp. 70% of Taeumin group had oilily scalp. 2. The group of dry or oilily scalp had more itching sense and more sensativity of scalp compaired to neuter scalp group. 3. There was no correlation between height, weight, thickness of hair, itching sense, scalp disease and Sasang constitution. 4. There was correlation between loss of hair and drinking alcohol, obesity, curly hair, soft scalp. Conclusions ; Korean adult scalp and hair condition had correlation with Sasang constitution.
This study attempted to analyze the general and physical characteristics, the status of physical function, the type and number of current diagnosis and self-reported symptoms, healthy life mode and food preference according to self-rated health (SRH) of older adults (135 men and 270 women). It also attempted to assess the factors affecting the SRH standards in agricultural and fishery areas located in southwestern Korea. The subjects considered themselves as being 'good'(57.6%), 'normal'(29.6%) or 'poor'(12.85) SRH, meaning positive self-rating of health and it was found that the correlation between factors such as the gender, current marriage status, monthly wage, the reception of the government's livelihood subsidy and subjective economical status and the SRH were significant. Regardless of the categories of obesity, a large number of the participants rated themselves as 'healthy', but not statistically significant. The subjects who reported poor ADL and IADL capacities, indicators of the status of physical function, were significantly more classified to the 'poor' health category and vice versa (P<0.001). The reported chronic diseases in this study that have lasted more than three months, were lumbago, sciatica, arthritis, high blood pressure and peptic ulcer. The current ratios of smoking, drinking and exercise were 85.3, 39.0 and 18.6%, respectively. The less drinking and exercising there was, the higher the 'good' SRH categories obtained (P<0.05). The respondents who had less chance of eating sour and hot foods estimated their health status as being better.
