본 연구에서는 다양한 사람의 암세포주(A-549, MCF-7, MDA-MB-231, U87-MG, AGS, MKN-74 및 SNU-601 세포)와 정상세포주(MRC-5 섬유아세포 및 사랑니 유래 중간엽성 줄기세포에 토복령 추출물(Smilax china L. extract, SCLE)을 처리하여 세포 사멸 효과를 조사하였다. SCLE 처리 후, MTT 분석에서 여러 암세포주는 정상세포주보다 유의적으로 휠씬 낮은 반억제농도값을 나타내었고, 세포는 세포부착력의 소실로 인한 세포사멸(anoikis)이 관찰되었다. 또한, SCLE를 처리한 A-549, AGS 및 MCF-7 암세포주에서 세포의 생존성과 말단소립 복원효소의 활성도를 조사하였을 때, SCLE 처리 후 4일째에 세포의 생존성과 말단소립 복원효소의 활성도가 현저히 줄어드는 것을 관찰하였다. 또한, SCLE를 처리한 A-549, AGS 및 MCF-7 암세포주에서 세포 주기의 G1기에서 세포 성장이 정지되었고,세포 사멸이 유의적으로 증가하는 것을 알 수 있었다. 그러나, SCLE 처리는 rho 단백질의 활성과 관련 없는 세포부착력의 소실과 세포 사멸이 유도되는 것을 관찰하였다. 이 연구의 결과를 바탕으로 토복령 추출물은 정상 세포보다는 암세포에 특이적으로 세포부착력의 소실과 세포 사멸을 유도하여, 이 추출물에 포함된 물질을 이용한 항암 연구에 응용될 수 있을 것으로 판단된다.
본 실험은 transforming growth factor-${\beta}1$(TGF-${\beta}1$)이 첨가된 chondrogenic induction medium(CIM)을 이용하여 인간지방조직에서 유래된 중간엽 줄기세포(human adipose tissue-derived mesenchymal stem cells, ATMSCs)의 연골형성능과 gelatin-chondroitin-glucosamine scaffold(GCG-scaffold)에 접종시킨 ATMSCs의 연골형성능을 알아보고자 수행하였다. ATMSCs와 생쥐 chondrocyte를 기본배양액과 TGF-${\beta}1$이 첨가되지 않은 CIM1 및 TGF-${\beta}1$이 첨가된 CIM2에서 배양하여 연골형성능을 비교하였다. ATMSCs의 연골형성은 gelatin scaffold(G-scaffold)와 GCG-scaffold를 사용하여 glycosaminoglycan(GAG) 합성과 조직화학적 염색으로 연골기질형성 여부를 확인하였다. 펠렛 배양에서 ATMSCs와 chondrocyte의 GAG합성은 대조군에서 14일의 배양기간 동안 약간 증가하였으나, CIM1과 CIM2배양군은 배양 14일에 대조군에 비해크게 증가하였으며, CIM2 배양군에서 가장 높았다. 그러나 연골기질은 배양 14일에 CIM2 배양군에서만 Safranin O와trichrome에 의해 염색되었다. Well plate에서 배양된 ATMSCs의 증식은 모든 배양군에서 배양 10일까지 계속적인 세포증식이 일어났으며, 대조군에 비해 CIM 배양군이 높았다. ATMSCs 접종 후 플라스크나 scaffold의 세포부착률은 배양시간이 길어짐에 따라 모든 배양군에서 증가하였고, 플라스크에 비해 scaffold에서 더욱 높게 나타났다. Scaffold에 ATMSCs 접종후 GAG 합성은 28일의 배양기간 동안 대조군에서는 별 변화가 없었으나, CIM1, CIM2 배양군은 대조군보다 GAG합성이 증가되었다. CIM2 배양군에서 GAG 합성이 매우 높게 나타났다. 또한 G-scaffold보다 GCG-scaffold에서 GAG 합성이 약간 높게 나타났다. 조직화학적으로 관찰한 ATMSCs의 연골기질형성도 CIM2 배양군의 GCG-scaffold에서 가장높게 나타났다. 본 실험의 결과들을 종합해 볼 때, 펠렛 배양에서 ATMSCs는 생쥐 chondrocyte보다 낮은 연골형성능을 보였고, CIM2 배양군에서만 연골기질이 형성된 것으로 보아 TGF-${\beta}1$이 연골분화에 중요한 요소로 작용한 것이라 사료된다. G-scaffold는 효과적인 연골분화 환경을 제공해 줌으로써 ATMSCs의 세포부착률과 GAG 합성을 증가시키는 것으로보여지며, G-scaffold보다 GCG-scaffold에서 많은 연골기질이 형성된 것은 GCG-scaffold에 첨가된 chondroitin과 glucosamine이연골기질 형성을 촉진시키는 물질로 작용한 것으로 사료된다.
인간중간엽줄기세포는(Human mesenchymal stem cells, hMSC) 골수, 지방, 피부, 근육, 혈액에 존재하며, 뼈, 연골, 지방, 근육, 신경세포로 분화가능성이 보고되어 손상조직의 재생을 위한 재료로서뿐만 아니라 유전자치료의 매개체로 이용될 수 있는 가능성이 제안되고 있다. 인간중간엽줄기세포의 적절한 배양조건에는 소 태아혈청(fetal bovine serum, FBS)이 요구되어지므로 세포치료에는 소 태아혈청이 다수 포함되어 있을 것이며 세포배양 배지 유래 소 태아혈청의 단백질에 의한 면역거부반응이 우려된다. 이미 앞선 연구에서 자가혈청 하에서 인체지방줄기세포 분리와 계속적인 세포배양을 실시하였을 때 인체지방줄기세포의 증식능력과 다 분화 능이 유지되며 면역결핍 생쥐에 골수의 말초혈액에서 유래된 CD34세포 이식 시 안착 능을 촉진함을 보였다. 본 연구에서 인체지방줄기세포가 인체혈청 하에서 배양되었을 때 소 태아혈청 하에서 배양할 때 발현하는 표면항원을 유지함을 확인했으며 microarray를 사용하여 유전자 발현을 비교했다. 유 세포 분석을 통하여 인체혈청 하에서 계속적으로 배양된 인체유래지방줄기세포에서 HLA-DR, CD117, CD29 와 CD44 의 발현이 소 태아혈청 하에서 배양했을 때와 비슷함을 밝혔다. 그러나 인체혈청 하에서 배양된 인체지방줄기세포의 유전자 발현형태와 소 태아혈청 하에서 배양된 세포의 유전자 발현형태 간에는 상당한 차이를 보였다. 그러므로 본 연구는 인체혈청 하에서 배양된 인체지방기질줄기세포가 임상적용을 위한 선행 데이터로써 직접적인 추정을 하기 위해서는 인체지방기질줄기세포 이식연구에 in vivo 동물실험연구가 수행되어져야 함을 제시하고 있다.
Human mesenchymal stem cells (hMSC) are multipotent stromal cells that have great potential to differentiate into a variety of cell types such as osteocytes, chondrocytes, and myocytes. Although there have been many studies on their clinical availability, little is known about how intracellular signals can be modulated by topographic features of the extracellular matrix (ECM). In this study, we investigated whether and how microwavy-patterned extracellular matrix (ECM) could affect the signaling activity of focal adhesion kinase (FAK), a key cellular adhesion protein. The fluorescence resonance energy transfer (FRET)-based FAK biosensor-transfected cells are incubated on microwavy-patterned surfaces and then platelet derived growth factor (PDGF) are treated to trigger FAK signals, followed by monitoring through live-cell FRET imaging in real time. As a result, we report that PDGF-induced FAK was highly activated in cells cultured on microwavy-patterned surface with L or M type, while inhibited by H type-patterned surface. In further studies, PDGF-induced FAK signals are regulated by functional support of actin filaments, microtubules, myosin-related proteins, suggesting that PDGF-induced FAK signals in hMSC upon microwavy surfaces are dependent on cytoskeleton (CSK)-actomyosin networks. Thus, our findings not only provide new insight on molecular mechanisms on how FAK signals can be regulated by distinct topographical cues of the ECM, but also may offer advantages in potential applications for regenerative medicine and tissue engineering.
목 적: 사람의 제대유래 줄기세포 (HUC)와 양막유래 줄기세포 (HAM)를 간유사세포로 분화 시키고자 하였다. 연구방법: 산모의 동의아래 만삭 정상 산모로부터 양막 및 제대를 얻고 줄기세포를 분리, 배양하였다. 분리된 세포의 줄기세포로서의 특성을 규명하기 위하여 RT-PCR, 세포면역화학 분석 그리고 중배엽성세포로의 분화능 실험을 하였다. 또한 이들 줄기세포를 분화 배양액에서 간유사세포로 분화 유도 후, 간세포 특이 유전자 발현, 알부민 ELISA, 알부민에 대한 면역블로팅 및 세포면역화학염색 그리고 PAS 염색을 시행하였다. 결 과: 사람의 양막 및 제대로부터 세포를 분리하여 줄기세포로서의 특성을 조사하고, 이들 세포의 지방세포, 연골세포, 골아세포로의 분화능력을 확인하였다. 동일한 분화 배양액에서 HUC과 HAM을 간유사세포로 분화시킨 결과 HUC이 모든 조건에서 HAM보다 알부민과 요소를 더 많이 합성 분비하였다. 간유사세포로의 분화능력이 더 좋은 것으로 나타난 HUC을 가지고 최적의 분화를 유도한 결과 $1.2{\pm}0.8\;{\mu}g/mL$의 알부민을 합성 분비하였고, $8.9{\pm}1.2\;mg/dL$의 요소를 합성 분비하였다. 결 론: HUC과 HAM 모두 알부민을 분비하는 간유사세포로 분화할 수 있었으며, HUC이 더 잘 유도되는 것으로 관찰되었다. 이러한 결과로써 HUC과 HAM을 간질환 환자의 간세포 이식 치료에 필요한 세포치료제로써 이용할 수 있을 것으로 생각된다.
양막은 다양한 생체재료로 이용되어 왔으며 생체적합성과 환부의 치료효능이 우수한 것으로 알려져 있다. 본 연구에서는 양막을 갈아 양막스폰지를 제조하고 중간엽줄기세포를 배양한 뒤 누드마우스이식을 통하여 콜라젠스폰지와 비교하여 골형성유도능에 대한 연구를 실시하였다. 연구결과 세포의 부착과 증식면에서는 두 시료가 유사하였으나, 체내에 이식한 결과 양막스포지군에서 좀 더 많은 콜라젠 분비와 칼슘이 침착되었음을 확인하였다. 그리고 면역화학염색 결과 골형성 시 필요한 오스테오칼신과 오스테오넥틴이 좀 더 발현된 것을 관찰할 수 있었다. 따라서 양막은 골형성유도를 향상시킬 수 있는 가능성이 있음을 알 수 있었다.
이 연구의 목적은 유치 줄기세포에 대해 다양한 Calcium silicate-based material (CSM)의 세포독성을 비교하고 평가하는 것이다. Retro $MTA^{(R)}$ (RM), $EZ-Seal^{TM}$ (EZ), ENDOCEM $Zr^{(R)}$ (EN)의 powder를 세포 배지를 이용하여 용출시키고 filtering 하였다. 유치 줄기세포가 다양한 농도의 용출액 하 배양되었다. MTS assay를 통해 CSM 용출액이 세포 증식에 미치는 영향을 분석하였고, Flow cytometry analysis를 통해 세포 표현형의 변화 여부를 관찰하였다. 10% 농도의 용출액에서 배양된 유치 줄기세포의 흡광도 값은 RM > EN > EZ 순이었다(p = 0.0439). 그러나 유치 줄기세포는 재료에 관계없이 간엽줄기세포의 표현형을 유지했다. 세 종류의 CSM이 줄기세포 marker를 변형시키진 않았지만, EZ가 RM과 EN보다 세포적합성이 낮음을 알 수 있었다.
골다공증의 진행은 뼈질량 감소와 골절위험 증가를 야기한다. 골다공증은 노인 인구에서 흔하며, 최근 들어 급속한 고령화 사회로 인해 그 환자수도 동반하여 크게 증가하고 있다. 현재 처방되는 골다공증 치료제의 대부분은 파골세포 억제 효과에 기반하여 골흡수를 방지한다. 그러나 이러한 골다공증 치료제는 새로운 뼈형성을 증가시키지는 못하며 수반되는 여러 부작용도 보고되고 있다. 따라서 골다공증의 새로운 제어와 치료법 개발을 위해 성체줄기세포의 골세포 분화유도와 조골세포 활성을 도모하는 재생의학적 접근이 활발히 연구되고 있다. 스타틴(statin) 계열 약물은 혈중 콜레스테롤 강하제로 심혈관 질환에 흔히 처방되는 치료제이다. 흥미롭게도 최근 일련의 연구에서 이러한 스타틴이 조골세포 활성에 긍정적인 영향을 주어 뼈형성을 촉진한다는 보고가 발표되고 있다. 따라서, 본 연구에서는 이러한 스타틴 약물이 인체 골막유래 성체줄기세포의 골세포 분화과정이나 조골세포 활성에 영향이 있는 지를 분석하였다. 현재 임상적으로 처방되는 총 7 종류의 스타틴 약물에 대해, 골막유래 성체줄기세포의 골세포 분화과정에서 조골세포 활성과 관련된 초기와 후기 표지자인 alkaline phosphatase의 활성과 칼슘 침착을 각각 분석하였다. 본 연구에서 일부 스타틴(pitavastatin과 pravastatin)은 약하지만 뼈형성을 증가시키는 효과가 있음을 알 수 있었다. 이러한 연구결과는 스타틴이 골막유래 줄기세포로부터 골세포로의 분화나 조골세포 활성을 조절할 수 있는 물질이 될 수 있으며, 이러한 약물이 골세포분화나 재생의학의 새로운 조절 물질로서 골다공증 치료에 응용될 수 있음을 제시한다.

hBMSCs are multipotent cells that are useful for tissue regeneration to treat degenerative diseases and others for their differentiation ability into chondrocytes, osteoblasts, adipocytes, hepatocytes and neuronal cells. In this study, biodegradable elastic hydrogels consisting of hydrophilic poly(ethylene glycol) (PEG) and hydrophobic poly(${\varepsilon}$-caprolactone) (PCL) scaffolds were evaluated for tissue engineering because of its biocompatibility and the ability to control the release of bioactive peptides. The primary cultured cells from human bone marrow are confirmed as hBMSC by immunohistochemical analysis. Mesenchymal stem cell markers (collagen type I, fibronectin, CD54, $integrin1{\beta}$, and Hu protein) were shown to be positive, while hematopoietic stem cell markers (CD14 and CD45) were shown to be negative. Three different hydrogel scaffolds with different block compositions (PEG:PCL=6:14 and 14:6 by weight) were fabricated using the salt leaching method. The hBMSCs were expanded, seeded on the scaffolds, and cultured up to 8 days under static conditions in Iscove's Modified Dulbecco's Media (IMDM). The growth of MSCs cultured on the hydrogel with PEG/PCL= 6/14 was faster than that of the others. In addition, the morphology of MSCs seemed to be normal and no cytotoxicity was found. The coating of the vascular endothelial growth factor (VEGF) containing scaffold with Matrigel slowed down the release of VEGF in vitro and promoted the angiogenesis when transplanted into BALB/c nude mice. These results suggest that hBMSCs can be supported by a biode gradable hydrogel scaffold for effective cell growth, and enhance the angiogenesis by Matrigel coating.

Regenerative orthopedics needs significant devices to transplant human stem cells into damaged tissue and encourage automatic growth into replacements suitable for the human skeleton. Soft biomaterials have similarities in mechanical, structural and architectural properties to natural extracellular matrix (ECM), but often lack essential ECM molecules and signals. Here we engineer mineralized polysaccharide beads to transform MSCs into osteogenic cells and osteoid tissue for transplantation. Bone morphogenic proteins (BMP-2) and indispensable ECM proteins both directed differentiation inside alginate beads. Laminin and collagen IV basement membrane matrix proteins fixed and organized MSCs onto the alginate matrix, and BMP-2 drove differentiation, osteoid tissue self-assembly, and small-scale mineralization. Augmentation of alginate is necessary, and we showed that a few rationally selected small proteins from the basement membrane (BM) compartment of the ECM were sufficient to up-regulate cell expression of Runx-2 and osteocalcin for osteoid formation, resulting in Alizarin red-positive mineral nodules. More significantly, nested BMP-2 and BM beads added to a non-union skull defect, self-generated osteoid expressing osteopontin (OPN) and osteocalcin (OCN) in a chain along the defect, at only four weeks, establishing a framework for complete regeneration expected in 6 and 12 weeks. Alginate beads are beneficial surgical devices for transplanting therapeutic cells in programmed (by the ECM components and alginate-chitosan properties) reaction environments ideal for promoting bone tissue.

② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
