  • No report has been addressed to the CYP isoforms catalyzing chlorpropamide, a structural analogue of tolbutamide. To evaluate enzyme(s) mediating formation of 2-hydroxycWorpropamide, a major metabolite and identified by LC/Mass and NMR, incubation studies using human liver microsomes and cDNA expressed CYP were performed on the presence or absence of selective inhibitors of each CYP isoform.   • Simple spring-damper-mass models have been widely used to investigate whole-body vortical biodynamic response characteristics of the seated vehicle driver. Most previous models have not considered the effect of wobbling masses; i.e. heart, lungs, liver, intestine, etc. In this study, 4 -DOF seated driver model including one non-rigid mass representing wobbling visceral mass, 5-DOF model including intestine, and 10-DOF model including five lumbar vertebral masses were proposed. The model parameters were identified by a combinatorial optimization technique. simulated annealing method. The objective function was chosen as the sum of error between model response of seat-to-head transmissibility and driving point mechanical impedance and those of experimental data for subjects seated erect without backrest support. The model response showed a good agreement with the experimental response characteristics. Using a 10-DOF model, calculated resonance frequency of lumbar spine at 4Hz was matched well with experimental results of Panjabi et al.

  • This research aims to study the changes In gastrointestinal function attributed to aging In human. The thresholds for recognition and detection of flavors became elevated and salivary gland acinar cells decreased in the old age. But most esophageal function remained relatively Intact. Although gastric emptying time has been slowed with aging, the total intestinal transit time did not differ. Atropic gastritis due to H. pylori in old man decreased secretion of acid and Intrinsic factor and absorbability of calcium and iron. Pancreatic secretion is droned in older persons. Prevalence of gallstones rised with age. Liver size and portal blood flow decreased significantly with age. Mucosal surface area has been reported to be slightly diminished in the aging man. Glucose transporters decreased and Insulin tolerance Increased. Absorption of aromatic amino acid is diminished with age. Dietary protein In that aging human increased fecal nitrogen excretion. Vitamin A tolerance increased. Vitamin D receptor concentration decreased and resistance to 1,25-(OH)2D3 action increased. Permeability of aging small Intestine Increased. Zinc balance dirt not differ Copper absorption appeared not to be significantly affected by age. Neurotensin secretion decreased thus slowed colonic peristaltic movements and Intestinal mucosal growth.

사상의학에서 각 체질별 기운의 방향 특성 (Directional Properties of 4 Type Constitution in Sasang Constitutional Medicine)

  • 1. Objectives The purpose of this study is to find the ground why constitution must be classified into 4 types. 2. Methods I analysed the basic presuppositions to class 4 Type Constitution in Sasang Constitutional Medicine(SCM). And I connected the presuppositions with other theories from the viewpoint of directional properties of human body. 3. Results and Conclusions The most important presupposition to class 4 Type Constitution and apply to clinical therapy in SCM is that there is native strength and weakness of lung's Exhale Dispersing Qi, liver's Inhale Gathering Qi, spleen's Ascending Yin Qi and kidney's Descending Yang Qi. If there is superior tendency of Qi flow direction in human body, 6 directions(upward, downward, forward, backward, outward and inward) will be remarkable. But, in embryonic period, the outer skin is originated from the back(ectoderm) and the alimentary canal is from the front(endoderm). Therefore 4 directions(upward, downward, outward and inward) will be more remarkable, and those directions are included in the former presupposition.

내분비계 장애물질인 Bisphenol A의 free radical 생성을 통한 독성발현 (Cytotoxicity of Environmental Estrogenic Compound, Bisphenol A, via Generation of Free Radicals)

  • Bisphenol A shares similarities in structure, metabolism and action with DES, a known human teratogen and carcinogen. Bisphenol A, a monomer of polycarbonate and epoxy resins, has been detected in canned food and human saliva. The purpose of the this study was to evaluate the cytotoxicity, cell proliferation of bisphenol A In the presence of a rat liver S9 mix, contaning cytochrome P450 enzymes, and Cu (II). In the present study, Bisphenol A in combination with Cu (II) exhibited a enhancement in cytotoxicity which were inhibited by free radical scavengers. The content of malondialdehyde, an end product of lipid peroxidation, was also found to increase with concentration of bisphenol A. Also, we examined the change of CuZn-SOD, Mn-SOD, catalase and GPx activities in the MCF-7 cells exposed to bisphenol A. The activities of CuZn-SOD, CPx, catalase were found to decrease with bisphenol A concentration. Meanwhile, the activity of Mn-SOD was unchanged. This indicated that elevated oxidative stress caused by imbalance between the production and removal of free radicals occurred in cells.

시메티딘과 리팜피신의 상호작용 (Drug interaction of Cimetidine and Rifampicin in normal Human subjects)

  • The intraction between cimetidine and rifampicin was studied pharmacokinetically in normal human subjects. The serum level and the area under the serum concentration curve(AUC) of rifampicin administrated orally were elevated significantly by cimetidine. Volume of distribution, total clearance and elimination rate constant of rifampicin were reduced significanyly by cimetidine. Biological half-life of rifampicin was prolonged significantly by cimetidine. The mechanism of this results is probably related to the inhibition of rifampicin metabolism(deacetyl form) or reduction of blood flow in the liver. It is desirable that dosage regimen of rifampicin shoud be adjusted when combined with cimetidine in clinical pharmacy practice.

오액(五液)에 관한 연구(硏究) (Study on the Five Fluids)

  • "Jinaik(津液)" is essential nutritious body fluid for human to be alive, and it can be divided various kinds according to it's location, character, and function. Five Fluids -sweat, snivel(mucus), slobber(saliva), tear, spittle(saliva)- are kinds of "Jinaik(津液)", and all of them belong to kidney. They are also related with five organs, such as sweat belongs to heart, snivel(mucus) belongs to lung, slobber(saliva) belongs to spleen, tear belongs to liver, spittle(saliva) belongs to kidney. We can find out five organs' physiological and pathological conditions by means of observations on the Five Fluids. Five organs' function comes out upper seven openings -two eyes two ears, nose, mouth and tongue. The Five Fluids provide all the nutrition upper seven openings need, so they can help for human to hear, see, smell, and taste properly.

Biological Effects of Black Colored Soybean

  • It has been believed that supplementation of human diets with soybean products markedly reduces human cancer mortality rates. Specifically the black colored soybean has been considered as having a medicinal effect since ancient times in Korea. Six genotypes of black colored soybean and a normal variety were investigated for their biological effect. While the isoflavone content of black colored soybean was higher than the normal soybean, saponin content of the seeds was not different among varieties. In the case of antioxidant activity, black colored soybean showed over 3 times increase in the active effect than the control variety. When feeding soybean extract, a general change of alcohol resolving effect, such as alcohol dehydrogenase activity, was observed in the blood and liver of rats.

황기의 지질과산화 억제작용 (Antilipidperoxidative activity of Astragalus membranaceus)

  • The root of Astragalus membranaceus (Leguminosae) has been used in traditional chinese prescriptions for strengthning the superficial resistance, promoting pus discharge and tissue regeneration, diuretis and alleviating edema. Lipid peroxidation has been suggested as a major cause of atherosclerosis, cancer, liver disease, and the aging process. In order to investigate the anti-lipid peroxidation activity, Astragali Radix was extracted with $80\%$ MeOH and fractionated with hexane, $CH_{2}Cl_2$, EtOAc, BuOH and Water. The antilipid peroxidation activities of them were determined by human erythrocyte ghost and $CCl_4$-induced lipid peroxidation. $CH_{2}Cl_2$ and EtOAc fractions, especially, isoflavonoids, 7,2'-dihydroxy-3',4'-dimethoxyisoflavan-7-O-${\beta}$-D-glucoside and calyco sin-7-O-${\beta}$-D-glucoside from EtOAc fraction showed anti-lipid peroxidation activities.

  • Cytochrome P-450 3A4 (CYP3A4) is major enzyme in human liver, the role of this is detoxification and metabolizing more than 50% clinical drugs in use. Expression of CYP3A4 is transciptionally regulated by the Pregnenolone X receptor (PXR), of which human form is Steroid and Xenobiotics receptor (SXR). SXR is activated by wide range of endogenous and exogenous compounds, and then induces CYP3A4 gene expression. In the previous study, it has been known that proximal promoter (-864 to +64) does not response to chemical inducers such as pregnenolone 16a-carbonitrile (PCN), Rifampicin, Estrogen in terms of transcription of CYP 3A4 in cultured cells.
