Lactobacillus species in the female genital tract are thought to act as a barrier to infection. Several studies have demonstrated that lactobacilli can adhere to vaginal epithelial cells. However, little is known about how the adherence of lactobacilli to vaginal epithelial cells affects the acidity, cell viability, or proliferation of the lactobacilli themselves or those of vaginal epithelial cells. Lactobacillus acidophilus was co-cultured with immortalized human vaginal epithelial cells (MS74 cell line), and the growth of L. acidophilus and the acidity of the culture medium were measured. MS74 cell density and viability were also assessed by counting cell numbers and observing the cell attachment state. L. acidophilus showed exponential growth for the first 6 hr until 9 hr, and the pH was maintained close to 4.0-5.0 at 24 hr after culture, consistent with previous studies. The growth curve of L. acidophilus or the pH values were relatively unaffected by co-culture with MS74 cells, confirming that L. acidophilus maintains a low pH in the presence of MS74 cells. This co-culture model could therefore potentially be used to mimic vaginal conditions for future in vitro studies. On the other hand, MS74 cells co-cultured with L. acidophilus more firmly attached to the culture plate, and a higher number of cells were present compared to cells cultured in the absence of L. acidophilus. These results indicate that L. acidophilus increases MS74 cell proliferation and viability, suggesting that lactobacilli may contribute to the healthy environment for vaginal epithelial cells.
리포솜은 수십~수백 나노미터 크기의 소포체(small vesicle)로서 약물전달에 유용한 구조체이나 체내 혈류 순환계(blood circulatory system)에서 구조적 불안정성에 의해 체내 순환시간이 짧고 세망내피계(reticuloendothelial system)에 의해 소실되어 이를 개선하기 위한 다양한 연구가 시도되고 있다. 본 연구에서는 이당류(disaccharide)로서 락토오스와 슈크로스가 1,2-디스테아로일-sn-글리세로-3-포스포에탄올아민(1,2-Distearoylsn-glycero-3-phosphoethanolamine, DSPE)과 공유결합된 새로운 당-DSPE 유도체를 합성하고 이를 리포솜의 구성성분으로 하는 리포솜을 제조함으로써 리포솜의 표면이 이당으로 수식된 리포솜을 제조하였다. 제조된 리포솜의 입자크기는 대략 100 nm였으며, 표면전하 값은 당이 수식되지 않은 대조군 리포솜이 -10 mV를 나타내었으나 당-DSPE 유도체를 함유한 리포솜의 경우 리포솜 표면에 존재하는 당의 수산화기에 의하여 표면전하의 값은 -25 mV를 나타내었다. 리포솜 내부에 모델약물인 독소루비신(doxorubicin)의 로딩효율은 약 90%를 나타내었다. 당-DSPE 유도체 함유 리포솜의 in vitro 안정성은 혈청 내에서 단백흡착량의 변화 및 리포솜의 입자크기를 관찰하여 평가하였다. 당-인지질 유도체를 함유한 리포솜은 당-인지질 유도체를 함유하지 않은 리포솜 또는 폴리에틸렌글리콜(PEG) 수식 리포솜에 비해 단백흡착의 양과 혈청 내 입자크기의 변화가 적은 것으로 관찰되었다. 이당이 결합된 DSPE와 이를 함유한 리포솜은 혈액내 안정성이 향상되어 체순환계 내에서 장시간 순환이 가능한 약물전달체로서 유용할 것이라 사료된다.
Objective : To investigate the effects of posterior implant rigidity on spinal kinematics at adjacent levels by utilizing a cadaveric spine model with simulated physiological loading. Methods : Five human lumbar spinal specimens (L3 to S1) were obtained and checked for abnormalities. The fresh specimens were stripped of muscle tissue, with care taken to preserve the spinal ligaments and facet joints. Pedicle screws were implanted in the L4 and L5 vertebrae of each specimen. Specimens were tested under 0 N and 400 N axial loading. Five different posterior rods of various elastic moduli (intact, rubber, low-density polyethylene, aluminum, and titanium) were tested. Segmental range of motion (ROM), center of rotation (COR) and intervertebral disc pressure were investigated. Results : As the rigidity of the posterior rods increased, both the segmental ROM and disc pressure at L4-5 decreased, while those values increased at adjacent levels. Implant stiffness saturation was evident, as the ROM and disc pressure were only marginally increased beyond an implant stiffness of aluminum. Since the disc pressures of adjacent levels were increased by the axial loading, it was shown that the rigidity of the implants influenced the load sharing between the implant and the spinal column. The segmental CORs at the adjacent disc levels translated anteriorly and inferiorly as rigidity of the device increased. Conclusion : These biomechanical findings indicate that the rigidity of the dynamic stabilization implant and physiological loading play significant roles on spinal kinematics at adjacent disc levels, and will aid in further device development.
유산균과 금은화, 발효 금은화는 장누수 증후군과 연관된 장 상피세포 점막 투과성 감소에 대하여 유의한 효과를 나타내었다. 따라서 유산균을 단독으로 사용하는 방법과 유산균을 이용해 발효시킨 금은화를 증가된 장 투과성 및 장내미생물 불균형으로 인한 장누수 증후군과 관련된 일련의 증상들을 치료하고 면역관련 질환 및 만성 염증성 질환에도 응용할 수 있을 것으로 예상되며 이에 대한 앞으로의 추가 연구가 필요할 것으로 사료된다.
rhGH-GST 융합단백질을 사용하여 재조합 대장균 세포 파쇄액으로부터 직접적으로 내포체의 solid-phase 재접힘을 수행할 수 있는 새로운 공정을 개발하였다. 그것은 고체 업자를 제거하는 동시에 초기에 목적딴백질을 흡착 포집할 수 있 는 expanded bed adsorption 크로마토그래피의 장점을 이용한 것이다. 세포 파쇄액 내 용해훤 내포체로부터의 풀린 융합단백질은 expanded bed adsorption 원리에 의해 STREAMLINE DEAE resin에 흡착되고 세포 찌꺼기 등 고체 입자물들은 위 방향 흐름에 의해 효과적으로 제거된다. Urea를 접차적으로 제거함으로써 융합단백질은 고체 matrix 표면에서 재접힘 된 후 염 놓도 구배에 의해 용출된다. 이 새로운 EBA-mediat$\xi$d 재접힘 방법은 응집현상을 획기적으로 줄이고 공정수율윤 향상시킬 뿐 아나라 공정단계 수를 줄일 수 있다. 이 공정은 우리가 알고 있는 한 세계에서 최초로 개발된 공정이며, 현재 single-chain polypeptide, affinity-tagged protein 등과 갈은 다른 행태의 단백질에 EBA를 사용한 재접힘 공정올 적용시키가 위한 연구가 진행되고 있다.
Many dental composites are Bis-GMA based resin which diluted with the more fluid monomer triethylenglycol dimethacrylate(TEGDMA). TEGDMA is often present in exess so that some quantity remains unreacted following photo-initiated polymerization. TEGDMA is a component of some resin composites which contributes to their cytotoxicity. The presence of dentin between resin composite and pulp space reduce the cytotoxicity in vitro. The root system from extrcted human third molar was removed and then a circular occlusal cavity 4mm in diameter was prepared, leaving a remaining dentinal thickness to the roof of the pulpal chamber within the range 1.0-1.5mm. Dentine was treated with 37% phosphoric acid prior to Z 100 placement without using bonding resin(group 1). In group 2, SMP(Scotchbond Multi Purpose) primer, bonding resin prior to Z 100 placement were applied sequently. In group 3, moulds with internal dimensions 4mm diameter by 2mm depth were used to contain the composite alone with an equvalent mass on tooth model, and then they were immersed directly into water. The purpose of this study is to evaluate the release rate and quantity of TEGDMA with or without the application of bonding resin. Both release rate and total cumulative amount of TEGDMA for the three groups were determined using reversed-phase HPLC at times up to 10 days. The results were as follows: 1. All experimental groups showed the highest rate of release was in the first sample period(0-4.32 min) and the rate of release declined exponentially thereafter. 2. The maximum release rate and total cumulative account of TEGDMA in the tooth model of group 1 and group 2 with the use of SMP bonding resin were reduced however ther were no significant differences between these groups(P>0.05). 3. In the first sample period(0-4.32 min), the rate of release of TEGDMA from composite resin in group 3 immersed directly into water was significantly higher than that in group 1 and group 2 of tooth model(P<0.05). Conclusively, TEGDMA diffusion from Z 100 resin was not effectively prevented by the presence of dentin in spite of using the SMP bonding resin.
정상인 말초혈액임파구 및 C57BL/6마우스 비장임파구에 $^{60}Co{\gamma}-rays$를 in vitro상태에서 조사한 후 500개 또는 1000개의 cytokinesis-blocked(CB) lymphocytes의 미세핵(micronuclei)의 발생빈도를 측정하였다. 방사선조사량에 따라 미세핵의 발생빈도는 증가하였으며 linear-quadratic model로 측정한 결과 선량반응곡선의 식은 인체의 경우 $Y=(0.31{\pm}0.049)D+(0.0022{\pm}0.0002)D^2+13.19$($r^2=1.000$)이었으며, 마우스의 경우 $Y=(1.31{\pm}0.264)D+(0.0015{\pm}0.0006)+8.7$($r^2=0.988$)이었다(Y는 1000개의 CB cell 당 미세핵발생빈도, D는 cGy로 표시되는 조사선량). 인체 말초혈액임파구에 대한 마우스 비장임파구의 상대적 생물학적 효과(relative biological effectiveness)는 미세핵의 발생율이 세포당 0.05~0.8의 범위에서 $1.84{\pm}0.48$이었다. 미세핵분석법은 인체 및 동물의 방사선 피폭시 간편하고 빠른 생물학적 선량측정법으로 사용될 수 있을 것이다.
최근 기존 공기 전도형 보청기의 문제점과 불편함을 해결하기 위해 이식형 보청기인 인공중이 (Implantable middle ear hearing devices, IMEHDs)가 미국, 일본, 한국 등에서 활발히 연구 개발되고 있다. 본 논문은 인공중이용 진동체로서 PZT 적층 압전체 기반의 플로팅 매스 트랜스듀서의 설계 및 구현에 관한 것이다. 인공중이용 압전 플로팅 매스 트랜스듀서 (piezoelectric floating mass transducer, PFMT)의 기계적 진동 특성을 구하기 위한 근사적 모델링과 이의 해석을 위한 전기적 변환을 통한 진동 특성 분석을 수행하여 트랜스듀서 설계에 반영하여 구현하였다. 구현된 PFMT를 본 연구팀이 개발한 완전 이식형 인공중이 시스템 시작품에 적용하였으며, PFMT의 성능을 확인하기 위해 PFMT를 사체 이소골에 장착한 후 이소골 진동변위를 측정하는 실험을 행하였다. 이를 통해 본 논문에서 설계 및 구현된 PFMT의 인공중이용 진동 트랜스듀서로서 유효성을 확인하였다.
연구배경 : 대표적 세포내 감염질환인 결핵에 대한 숙주의 방어기전 및 면역반응은 아직도 정확히 이해되지 못하고 있으며 이러한 병리기전을 연구하기 위하여서는 적절한 감염모델이 필요하다. 전혈 (whole blood)은 체액성 면역과 세포성 면역을 모두 포함한 생체의 상태를 반영하므로 다양한 대상에서 면역상태의 차이에 따른 개체간 결핵균 살균력의 차이를 비교 할 수 있는 적절한 모델로 추정된다. 따라서 본 연구의 목적은 인체의 결핵균 전혈 배양모델을 개발하여 궁극적으로 시험관내에서 숙주면역의 정도를 측정하는 대리 표지자를 개발하고자하는 것이다. 방 법 : PPD 양성 정상인을 대상으로 제대혈, 결핵환자, 당뇨 및 폐암환자와 비교하였다. 전혈을 희석하여 결핵균 Mycobacterium avium과 M. tuberculosis $H_{37}Ra$에 낮은 감염률로 감염시키고 $37^{\circ}C$, 5% $CO_2$ 배양기 속의 회전교반기에서 회전시키면서 배양(rotating culture) 하였다. 배양 1 일, 3일 및 4일 뒤 증류수로 긴장저하용해 시킨 후 Middlebrook 7H10/OADC 평판배지에서 결핵균 집락이 형성될 때까지 3-4주간 $37^{\circ}C$, 5% $CO_2$ 배양기에 배양하여 집락수를 계산하였다. 일부실험에서 TNF-${\alpha}$의 분비능을 90%이상 감소시키 기 위하여 methylpredmsolone과 pentoxifylline을 첨가하여 면역조정을 하였으며, CD4+ T-림프구와 CD8+ T-림프구를 magnetic bead에 코팅된 단클론 항체를 사용하여 제거하였다. 결핵균의 수는 용해질 $m{\ell}$ 당 CFU로 계산하였다. 살균력은 ${\Delta}$ log killing ratio로 표시하였다. ${\Delta}$ logKR=$log_{10}$(Final CFU/Initial CFU). 결 과 : 1. 제대혈의 결핵균 살균력이 PPD 양성 대조군에 비하여 다소 감소된 경향을 보였으며, 결핵환자의 결핵균 살균력은 PPD 양성 대조군과 특별한 차이를 보이지 않았다. 또한 당뇨군과 폐암군의 결핵균 살균력도 정상 대조군에 비하여 특별한 차이를 보이지 않았다. 2. Methylprednisolone과 pentoxyfylline을 사용한 면역조정 시에 제대혈, PPD 양성 정상 대조군과 결핵군 모두에서 전혈에서의 결핵균의 살균력이 감소되는 경향을 보였다. 3. CD4+ 및 CD8+ T-림프구 삭제시 log KR가 증가되어 유의하게 결핵균 살균력이 감소되었으며 CD4+ 및 CD8+ T-림프구 동시 삭제시 현저한 상승효과를 보였고 이러한 결과는 Mycobacterium avium보다는 독력이 없는 균인 Mycobacterium tuberculosis $H_{37}Ra$에서 보다 뚜렷하였다. 4. 폐결핵 치료후의 결핵균 살균력은 치료 전과 비교하여 유의하게 ${\Delta}$ logKR가 감소하였으며, 배양 3-4일에도 현저한 결핵균 증식의 억제를 보였다. 결 론 : 인체의 결핵균에 대한 감수성인 개체간의 면역상태를 전혈에서 결핵균 살균력을 통하여 비교할 수는 없었다. 그러나 인체 전혈 모델은 간단하고 임상경과 관찰이 쉬우며 결핵환자에서 치료 전과 후에 현저한 결핵균 살균력의 차이를 보이므로, 최근 결핵연구의 가장 중요한 과제의 하나인 백신 개발에서 그 성과를 판단하는 vaccine trial에 이용할 수 있을 가능성을 시사한다.

Direct injection of CRISPR/Cas9 into zygotes enables the production of genetically modified nonhuman primates (NHPs) essential for modeling specific human diseases, such as Usher syndrome, and for developing novel therapeutic strategies. Usher syndrome is a rare genetic disease that causes loss of hearing, retinal degeneration, and problems with balance, and is attributed to a mutation in MYO7A, a gene that encodes an uncommon myosin motor protein expressed in the inner ear and retinal photoreceptors. To produce an Usher syndrome type 1B (USH1B) rhesus macaque model, we disrupted the MYO7A gene in developing zygotes. Identification of appropriately edited MYO7A embryos for knockout embryo transfer requires sequence analysis of material recovered from a trophectoderm (TE) cell biopsy. However, the TE biopsy procedure is labor intensive and could adversely impact embryo development. Recent studies have reported using cell-free DNA (cfDNA) from embryo culture media to detect aneuploid embryos in human in vitro fertilization (IVF) clinics. The cfDNA is released from the embryo during cell division or cell death, suggesting that cfDNA may be a viable resource for sequence analysis. Moreover, cfDNA collection is not invasive to the embryo and does not require special tools or expertise. We hypothesized that selection of appropriate edited embryos could be performed by analyzing cfDNA for MYO7A editing in embryo culture medium, and that this method would be advantageous for the subsequent generation of genetically modified NHPs. The purpose of this experiment is to determine whether cfDNA can be used to identify the target gene mutation of CRISPR/Cas9 injected embryos. In this study, we were able to obtain and utilize cfDNA to confirm the mutagenesis of MYO7A, but the method will require further optimization to obtain better accuracy before it can replace the TE biopsy approach.

제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
