Chlorella vulgaris Has Preventive Effect on Cadmium Induced Liver Damage in Rats

  • We investigated if Chlorella vulgaris (CV) has protective effects on cadmium (Cd) induced liver damage in male Sprague-Dawley (SD) rats. Forty rats, aged 5 weeks old and weighed 90-110g, were divided into a control (with Cd free water), 50 ppm of $CdCl_2$ in drinking water treated groups (Chlorella 0% diet group (Cd/CV0%), Chlorella 5% diet group (Cd/CV5%) or Chlorella 10% diet group (Cd/CV10%). All the rats had freely access to water and diet for 8 weeks. The results show that body weight gain and relative liver weight had significantly lower in Cd/CV0%-treated group than in Cd/CV-treated groups. Hepatic Cd contents showed significantly less by feeding CV (P<0.05). Cd/CV0%-treated rats had significantly (P<0.05) higher hepatic T-MTs, and Cd-MTs concentrations, compared to Cd/CV5% or Cd/CV10% treated rats. The MT I/II mRNA was expressed in the liver of all experimental rats. Its expression was more increased in Cd/CV5%- or Cd/CV10%-treated rats, compared to control and Cd-treated rats. Thus, this study suggested that CV would have a protective effect on Cd-treated liver injury by the reduction of Cd concentrations and stimulation of Cd-MT binds in the liver. However, more studies are needed to identify the proper mechanism of CV and liver toxicity.

남녀 대학생의 영양지식과 다이어트 행동에 관한 연구 (A Study of the Nutritional Knowledge and Diet Practice Behavior of College Students)

  • The principal objective of this study was to determine the effects of diet knowledge and perception on diet practice behavior among university students. Data were collected between March and April of 2005 from 523 male and female university students in Seoul and Chungbuk, via a self-administered questionnaire. The subjects included 217 male students (41.5%) and 306 female students (58.5%), and 52.0% of the subjects were 21-23 years old. With regard to residence type, 42.6% of the subjects lived in the dormitory, home (29.1%) self-boarding (19.5%) and meal (8.8) were in the order. The average weights and heights of the subjects were as follows: 70.9$\pm$9.0 kg and 174.3$\pm$4.4 cm in the male students and 53.9$\pm$5.9 kg and 161.3$\pm$4.5 cm in the female students. The average BMI values of the male and female students were 23.4$\pm$2.5 and 20.8$\pm$2.1, respectively. The average nutritional knowledge scores were as follows: 1.96 out of 3.0, dietary habit (2.13), nutrition (2.0), exercise (1.98), preferred food (1.90), and chronic disease (1.79). We noted no significant differences between the male and female subjects in terms of scored points. Nutritional knowledge by gender and residence type were shown to be significantly different--the male subjects had higher scores for exercise than did the female subjects. The self-boarding subjects had significantly lower scores with regard to nutritional knowledge and chronic disease. The experience and duration of previous diet practice were also significantly different with regard to gender and residence type. The male subjects tended to conduct diet practice for longer periods than females. The self-boarding students reported more attempts at diet behavior, but for shorter durations. The results show that the nutritional knowledge and diet practice behavior of the subjects were influenced by gender and residence type.

노인의료 복지시설 영양사의 업무활동 및 요구도 분석 (Analysis of the Job Activities and Demand of Dietitian in the Elderly Health-care Facilities)

  • This study was objectively performed to identify dietitians' job in the elderly health-care facilities, to assess facilities and dietitian's demographic characteristics, and to determine performance and importance of dietitian's job including the demand of therapeutic diet development. Survey was conducted by mail and samples were the dietitians working in 376 facilities which capacity is over 50 members from nationwide 583 the elderly health-care facilities. Returned questionnaire was 102 and used for statistic analysis. The distributions of the elderly health-care facilities showed 39 the elderly nursing facilities(38.2%), 32 skilled nursing facilities (32.4%), 13 geriatrics hospital facilities(12.7%) and 9 the elderly cost nursing facilities(8.8%). 60.0 percent of the samples showed its menu price as 1,000 to 1,500 won. A cycle-menu program was in-use at the 79.0% facilities, but only 7.1% facilities have been introduced a selected menu system. 92.9% facilities employed only one dietitian. In the demographic characteristics of dietitian only 14.7% dietitian had a clinical dietitian license and 51.5% of respondents answered at least 1 to 3 months internship program is needed. Job activities of the dietitian in the elderly health-care foodservice were identified as 45 activities with 9 dimensions. Job performance score evaluated dietitian oneself was 4.71 of 7 points. The average importance score that the dietitian evaluated their own job was 5.66 points of 7. The job activities shown higher importance but lower performance were therapeutic diet development for in-patients, menu development suitable for taste of the elderly, and leadership. Job performance score by characteristics of dietitian and their elderly health-care facilities was significantly associated with experience of dietitian in elderly health-care (F=4.480, p<0.05), education of dietitian(F=2.659, p<0.01), number of dietitian(F=2.245, p<0.05), and number of employee in foodservice(F=2.607, p<0.05). Most common diseases of the aged was proved as hypertension(81.7%), diabetes mellitus(71.4%), and dementia(65.0%). The therapeutic diets frequently provided were diabetes mellitus diet, dysphagia diet, low sodium diet, high fiber diet, and high protein diet, in order. For those reasons, dietitian in the elderly health care emphasized that the information about therapeutic diet development such as diabetes mellitus diet, dysphagia diet, low sodium diet and hypertension diet must be continuously developed and provided. The result from this study can be applicable to enlarge and enrich job activities of dietitian in elderly health-care foodservice.

Green perilla leaf extract ameliorates long-term oxidative stress induced by a high-fat diet in aging mice

  • BACKGROUND/OBJECTIVES: Oxidative stress is caused by an imbalance between harmful free radicals and antioxidants. Long-term oxidative stress can lead to an "exhausted" status of antioxidant defense system triggering development of metabolic syndrome and chronic inflammation. Green perilla (Perilla frutescens) is commonly used in Asian cuisines and traditional medicine in southeast Asia. Green perilla possesses numerous beneficial effects including anti-inflammatory and antioxidant functions. To investigate the potentials of green perilla leaf extract (PE) on oxidative stress, we induced oxidative stress by high-fat diet (HFD) in aging mice. MATERIALS/METHODS: C57BL/6J male mice were fed HFD continuously for 53 weeks. Then, mice were divided into three groups for 12 weeks: a normal diet fed reference group (NDcon), high-fat diet fed group (HDcon), and high-fat diet PE treated group (HDPE, 400 mg/kg of body weight). Biochemical analyses of serum and liver tissues were performed to assess metabolic and inflammatory damage and oxidative status. Hepatic gene expression of oxidative stress and inflammation related enzymes were evaluated by quantitative real-time polymerase chain reaction (qRT-PCR). RESULTS: PE improved hepatopathology. PE also improved the lipid profiles and antioxidant enzymes, including hepatic glutathione peroxidase (GPx) and superoxide dismutase (SOD) and catalase (CAT) in serum and liver. Hepatic gene expressions of antioxidant and anti-inflammatory related enzymes, such as SOD-1, CAT, interleukin 4 (IL-4) and nuclear factor erythroid 2-related factor (Nrf2) were significantly enhanced by PE. PE also reduced the levels of hydrogen peroxide (H2O2) and malondialdehyde (MDA) in the serum and liver; moreover, PE suppressed hepatic gene expression involved in pro-inflammatory response; Cyclooxygenase-2 (COX-2), nitric oxide synthase (NOS), interleukin 1 beta (IL-1β), and interleukin 6 (IL-6). CONCLUSIONS: This research opens opportunities for further investigations of PE as a functional food and possible anti-aging agent due to its attenuative effects against oxidative stress, resulting from HFD and aging in the future.

Regulation of cyclooxygenase-2 and mapkinases by isoflavones in ovariectomized and estrogen-supplemented mature female rats fed a high fat-high cholesterol diet

  • The effects of soy-isoflavones, which are phytoestrogens derived from plants with a flavonoid structure, on cyclooxygenase -2 (COX-2) expression, PGE2 production, and mapkinases expression, were investigated in experimentally-induced atherogenic rats by feeding a high fat-high cholesterol diet. Female Sprague-Dawley rats were bilaterally ovariectomized; sham-operated animals were used as controls. Three weeks later, the animals were randomized to the following treatments for an eight-week experimental period: 17$\beta$-estradiol (200$\mu$ g/kg diet), low concentration of isoflavones (0.8g/kg diet), and high concentration of isoflavones (4.0g/kg diet). In the group supplemented with a high dose of isoflavones, COX-2 expression was down-regulated. This down-regulation was accompanied by a reduced expression of pERK1/2. In the second experiment using 48-week old female Sprague-Dawly rats, the effects of isoflavones and estrogen were compared in the basal estrogen-supplementation at the level of 600$\mu$ g/kg diet. Isoflavones induced the marked down-regulation of COX-2 protein and the decrease in $PGE_2$ production in estrogen supplemented states and this was followed by the down-regulation of p38 among mapkinases. The two different mapkinases are involved in the down-regulation of COX-2 depending on estrogen-deficient and estrogen supplemented states. This kind of COX-2 down-regulation by isoflavones was not observed in the different tissue, mammary glands. Further investigations on the relationship between COX-2 and biological activities such as vasodilation by isoflavonesin the absence or the presence of estrogen ave required in vivo system of female rats.

Effect of vegetable oils with different fatty acid composition on high-fat diet-induced obesity and colon inflammation

  • BACKGROUND/OBJECTIVES: Different fatty acids exert different health benefits. This study investigated the potential protective effects of perilla, olive, and safflower oils on high-fat diet-induced obesity and colon inflammation. MATERIALS/METHODS: Five-week old, C57BL/6J mice were assigned to 5 groups: low-fat diet (LFD), high-fat diet (HFD) and high-fat diet supplemented with-perilla oil (HPO), olive oil (HOO), and safflower oil (HSO). After 16 weeks of the experimental period, the mice were sacrificed, and blood and tissues were collected. The serum was analyzed for obesity- and inflammation-related biomarkers. Gene expression of the biomarkers in the liver, adipose tissue, and colon tissue was analyzed. Micro-computed tomography (CT) analysis was performed one week before sacrifice. RESULTS: Treatment with all the three oils significantly improved obesity-induced increases in body weight, liver weight, and epididymal fat weight as well as serum triglyceride and leptin levels. Treatment with perilla oil (PO) and safflower oil (SO) increased adiponectin levels. The micro-CT analysis revealed that PO and SO reduced abdominal fat volume considerably. The mRNA expression of lipogenic genes was reduced in all the three oilsupplemented groups and PO upregulated lipid oxidation in the liver. Supplementation of oils improved macroscopic score, increased colon length, and decreased serum endotoxin and proinflammatory cytokine levels in the colon. The abundance of Bifidobacteria was increased and that of Enterobacteriaceae was reduced in the PO-supplemented group. All three oils reduced proinflammatory cytokine levels, as indicated by the mRNA expression. In addition, PO increased the expression of tight junction proteins. CONCLUSIONS: Taken together, our data indicate that the three oils exert similar anti-obesity effects. Interestingly, compared with olive oil and SO, PO provides better protection against high-fat diet-induced colon inflammation, suggesting that PO consumption helps manage inflammation-related diseases and provides omega-3 fatty acids needed by the body.

Effect of cinacalcet-mediated parathyroid hormone reduction on vitamin D metabolism in high-fat diet-induced obese mice

  • Purpose: Obesity is associated with alterations in vitamin D metabolism and elevation of parathyroid hormone (PTH). Increased PTH level in obesity is likely one of the factors contributing to the dysregulation of vitamin D metabolism. We investigated the effects of lowering the PTH level in high-fat diet-induced obese mice on vitamin D metabolism. Methods: Five-week-old male C57BL/6N mice were fed either with control (10% energy as fat) or high-fat (60% energy as fat) diets ad libitum for 12 weeks, and vehicle or cinacalcet HCl (30 ㎍/g body weight) was gavaged daily during the final week of the experiment. The following groups were studied: CON (control diet + vehicle), HFD (high-fat diet + vehicle), and HFD-CIN (high-fat diet + cinacalcet HCl). PTH, 1,25-dihydroxyvitamin D (1,25[OH]2D), 25-hydroxyvitamin D (25[OH]D), calcium, and phosphate levels in circulation, and the expression of genes related to vitamin D metabolism in the liver and kidneys were determined. Results: Renal 1α-hydroxylase expression in the HFD group was higher than that in the CON group despite the lack of a difference in the PTH levels between the 2 groups. The plasma PTH level in the HFD-CIN group was 60% lower than that in the HFD group (p < 0.05). In parallel, the HFD-CIN group had lower adipose tissue amount (9% lower), renal 1α-hydroxylase expression (48% lower), and plasma 1,25(OH)2D concentration (38% lower) than the HFD group. Conclusion: Lowering the PTH levels in high-fat diet-induced obese mice recovered the expression of renal 1α-hydroxylase and might be associated with lower amounts of white adipose tissue.

동물성(動物性)과 식물성(植物性) 단백질(蛋白質) 섭취(攝取)가 혈청(血淸) 지질(脂質) 및 뇨중(尿中) Methylhistidine에 미치는 영향(影響) (Effect of Plant and Animal Proteins on Serum Lipids and Urinary Methylhistidine in Human)

  • 본(本) 연구자(硏究者)는 21세(歲)부터 25세(歲)까지의 건강(健康)한 여대생(女大生) 8명(名)을 대상(對象)으로 식이중(食餌中)의 단백질(蛋白質) 급원(給源)((동물성)(動物性)과 식물성(植物性) 단백질(蛋白質))을 달리했을 때 혈청중(血淸中)의 cholesterol 농도(濃度)와 triglyceride 농도(濃度) 및 24시간(時間) 뇨중(尿中)의 creatinine 농도(濃度)와 3-methylhistidine의 농도(濃度)에 미치는 영향(影響)을 살펴보았다. 조사대상자(調査對象者)들은 control diet와 meat diet와 textured soy protein (TSP) diet를 각(各) 3일씩 섭취(攝取)하였으며, 각(各) 식이(食餌)의 3일째 되는 날에는 24시간(時間) 뇨(尿)를 채취(採取)하였고, 4일째 되는 날 아침 식사(食事) 전에 채혈(採血)하여 분석(分析)한 결과(結果)는 다음과 같다. 1) 혈청(血淸) cholesterol 농도(濃度)는 meat diet 시에 control diet보다 현저(顯著)하게 증가(增加)하였으며 (P<0.05), TSP diet시에는 control diet 보다 현저(顯著)하게 감소(減少)하였고 (P<0.01) meat diet와 비교(比較)시에도 현저(顯著)하게 감소(減少)되었다(P<0.01). 2) 혈청(血淸) triglyceride 농도(濃度)는 meat diet 시에 control diet 보다 감소(減少) 되었으나, 유의적(有意的)인 차이(差異)는 없었고 meat diet와 비교(比較)시에는 현저(顯著)하게 감소(減少)되었다(P<0.01). 3) 뇨중(尿中)의 creatinine 농도(濃度)는 meat diet 시에 control diet보다 약간 증가(增加)했으나 유의적(有意的)인 차이(差異)는 없었다. TSP diet 시에는 control diet보다 약간 감소(減少)하였으나 유의적(有意的)인 차이(差異)는 없었고 meat diet와 비교(比較)시에는 현저(顯著)하게 감소(減少)되었다(P<0.05). 4) 뇨중(尿中)의 3-methylhistidine 농도(濃度)는 meat diet 시에 control diet보다 약간 증가(增加)했으나, 유의적(有意的)인 차이(差異)는 없었다. 고지방식이로 비만을 유도한 생쥐에서 혈장 케모카인 발현 변화 (Alteration in plasma chemokine profile in a high-fat diet-induced obesity mouse model)

  • 본 연구에서 고지방식이로 비만을 유도한 생쥐는 식이 4주 후 지방조직의 형태학적 변화가 관찰되었고 생식기 지방조직의 무게가 증가하였으며, 혈장 중성지방 및 혈당치도 현저하게 증가하였다. 고지방식이 6주 후 몸무게의 유의한 증가가 관찰되기 시작하였으며, 뒤다리넙적근과 어깨사이 지방조직의 무게는 감소하고, 생식기 지방조직 및 간의 무게는 증가하였다. 지방조직의 형태학적 변화가 시작되는 고지방식이 유도 4주 후 혈장 내 40종의 시토카인 및 케모카인의 변화를 동시에 관찰하여 대조군과 비교해 본 결과, CXCL12 (SDF-1)와 CXCL13 (BLC)의 발현이 가장 현저하게 증가하였으며, G-CSF의 발현도 다소 증가하였다. 혈장 내 염증성 시토카인의 발현은 전반적으로 낮았다. 이상의 결과를 종합하면 고지방식이 유도 비만 초기에 만성 염증 상태로 진입하기 전 혈장 내 CXCL12와 CXCL13의 발현이 현저히 증가하는 것을 밝혔으며, CXCL12와 CXCL13의 증가로 B 세포, T 세포 및 단핵구가 혈관을 빠져나가 지방조직 및 지방조직 주변 림프조직으로 이동하여 지방조직 재형성과 국소 지방조직 면역에 관여할 것으로 보인다.

Effects of Low Calorie Diet and Platycodon Grandiflorum Extract on Fatty Acid Binding Protein Expression in Rats with Diet-induced Obesity

  • Obesity can be defined as a metabolic disease due to an increased fat accumulation in the body caused by an imbalance of calorie intake and output The prevalence of obesity has increased substantially over the past 2-3 decades in developed and developing countries. The health impact of weight gain is so marked that obesity has now been classified as a major global public health problem In order to investigate the effect of diet conversion and oral administration of Platycodon grandiflorum extracts on the treatment of obesity, male Spraque-Dawley rats were divided into four groups: a group converted to normal diet (Control group), a group maintained high fat (30%) diet (H), and two groups with Platycodon grandiflorum extract added to the previously mentioned two groups. All animals were fed high fat diet for 7 weeks to induce the obesity. Then they were divided as mentioned above. Animals were fed experimental diet and Platycodon grandiflorum extract (150 mg/ml/rat/day) for 7 weeks. Body weight, adipose tissue weight (subcutaneous, epididymal, peritoneal fat pads) and serum lipids (total cholesterol and triglyceride) showed some differences among groups. The Platycodon grandiflorum feeding markedly decreased both body weight and adipose tissue weight in control group compared to H, high fat diet maintaining, group. Platycodon grandiflorum extracts significantly decreased the concentrations of serum lipids compared to H group. Fat cell numbers and sizes were significantly reduced in the oriental medicinal herb extract administrated group. Increased fatty acid binding protein (FABP) expression in high fat diet group was decreased by the dietary conversion to normal diet and the oral administration of Platycodon glandiflorum extracts. In contrast, there was no significant effect on FABP expression in the high fat maintenance group. In this study, the conversion from high fat diet to low fat or normal diet had a beneficial effect on body weight loss and serum lipid profiles. Dietary Platycodon glandiflorum extracts had an additive beneficial effect on the prevention and treatment of obesity.
