작업환경을 위한 TLV의 근거 - m-PHENYLENEDIAMINE(1)

  • m-Phenylenediamine의 직업적 노출기준 TLV-TWA $0.1mg/m^3$은 간 손상을 최소화하기 위하여 권고하였다. 또한 이 수준은 피부 자극이나 피부염을 예방하기 위한 기준이다. 흰쥐와 생쥐에게 m-phenylenediamine을 처치한 후의 발암성이 음성적이거나 명확하지 않은 결과로 A4(Not Classifiable as a Human Carcinogen, 사람에게 발암성으로 분류되지 않음)로 권고하였다 m-Phenylenediamine을 생산하는 근로자들에서 피부 알레르기반응이 있었다는 제한적인 자료도 있었지만 피부(Skin)와 감작제(SEN)의 경고주석 그리고 TLV-STEL을 권고하기에는 유용한 자료가 충분하지는 않다.

  • 살균제인 pentachloronitrobenzene(PCNB)의 직업적 노출을 위한 TLV-TWA를 0.5 mg/$m^3$으로 권고하였다. 이 수준은 간에 영향을 미치는 것을 최소화하기 위한 수준이다 피부자극과 피부 흡수에 관한 것은 증명되지 않았다. PCNB에 대한 발암가능성을 흰쥐와 생쥐를 대상으로 생물학적 검정을 한 결과, 생쥐에서 양성이었다. 이러한 결과는 생물학적 검정에 사용한 PCNB가 공업용이었고 불순물로 포함된 hexachlorobenzene에 의해 암이 유발된 것으로 확인되었다. 따라서 FCNB 물질은 발암성 유발 인자(A4;Not Classifiable as a Human Carcinogen)로 권고하였다. PCNB의 Skin, SEN notation, TLV-STEL에 관한 정보는 충분한 데이터가 확보되지 않아 제공되지 못하고 있다.

  • The heavy metal cadmium is a xenobiotic toxicant of environmental and occupational concern and it has been classified as a human carcinogen. Inhalation of cadmium has been implicated in the development of emphysema and pulmonary fibrosis, but, the detailed mechanism by which cadmium induces adverse biological effects is not yet known. Therefore, we undertook the investigation of genes that are induced after cadmium exposure to illustrate the mechanism of cadmium toxicity. (omitted)

  • Dioxins refer to a family of chemicals comprising 75 polychlorinated dibenzo-p-dioxin (PCDD) and 135 polychlorinated dibenzo-p-furan (PCDF) congeners, which may cause skin disorder, human immune system disruption, birth defects, severe hormonal imbalance, and cancer. The effects of exposure of dioxin-like compounds such as PCBs are mediated by binding to the aryl hydrocarbon receptor (AHR), which is a ligand-activated transcription factor. To grasp physicochemical factors affecting human toxicity of dioxins, six geometrical and topological indices, eleven thermodynamic variables, and quantum mechanical descriptors including ESP (electrostatic potential) were analyzed using QSAR and semi-empirical AM1 method. Planar dioxins with high lipophilicity and large surface tension show the probability that negative electrostatic potential in the lateral oxygen may make hydrogen bonding with DNA bases to be a carcinogen.

  • There are numerous and evidential findings that TCDD (2, 3, 7, 8-tetrachlorodibenzo-Pdioxin, or dioxin) is a potential carcinogen and general toxin in rodents. flowever, human risk assessment for dioxin exposure has been a topic of debate, owing in part to the large animal interspecies differences in its toxicity. We review dioxin-related reports indicating its toxicity, toxic effects in animal, and human epidemiologic findings. The intent of this paper does not provide a causal inference about chronic human diseases related to dioxin exposure. This summary would give a valuable clue for a researcher to conduct or design a further dioxin-related study.

  • Chemical toxicants are metabolically converted to numerous metabolites in the body. Toxicokinetic characteristics of metabolites could be therefore used as biomarker of exposure for human risk assessment. Biologically based dose response (BBDR) model was proposed for future direction of risk assessment. However, this area has not been developed well enough for human application. Benzo(a)pyrene (BP), for example, is a well-known environmental carcinogen and may produce more than 100 metabolites and BPDE-DNA adduct, a covalently bound form of DNA with benzo(a)pyrene diolepoxides (BPDES), has been applied to qualitatively or quantitaively estimate human exposure to BP. In addition, di(2-ethylhexyl) phthalate (DEHP), a widely used plasticize. in the polymer industry, is one of endocrine-disrupting chemicals (EDCs) and has been monitored in humans using urinary or serum concentrations of DEHP or its monomer MEHP for exposure and risk assessment. However, it is difficult to estimate the actual level of toxicants using these biomarkers in humans using. This presentation will discuss a methodology of exposure and risk assessment by application of metabolic profiling kinetics.
