Exposure to metalworking fluids (MWFs) has significantly been associated with cancer developed in multi-organs, respiratory diseases and skin diseases. Several carcinogens to humans or animals are contained in MWFs. They have been reported to be mineral oils, polynuclear aromatic hydrocarbons (PAHs), formaldehyde and N-nitrosodiethanolamine (NDELA). The great hazards of MWF have forced the advanced country including United States to regulate carcinogens contained in MWF. In 2001, American Conference of Governmental Industrial Hygienists (ACGIHs) regarded MWF mist as suspected carcinogen to human (A2) and added it to “Notice of Intended Change (NIC)” list of 2001. In spite of the fact that much MWF has widely been used in many industries using machines, Korea has no legal actions for management of MWF. What is worse, even toxicity such as Carcinogenicity has not been reported. KS (Korean Standards) lists 7 advices of MWF but it does net state the hazards to health. It is very hard to control or minimize worker's exposure to MWF containing many carcinogens. Prier to the introduction of MWF to workplace, it is the most effective measure to regulate carcinogens below a certain level. Regulation on the content of PAH seems to be necessary because less amount of PAH in mineral oils improves the quality of MWF. Also, addition of nitrosating groups to MWF should be prohibited to minimize worker's exposure to NDELA. Employers and manufacturers should indicate the Carcinogenicity of all carcinogens in MWFs in Material Safety Data Sheets (MSDS) in order fer workers to recognize Carcinogenicity. Legal actions have to be taken to protect workers from health hazards due to exposure to MWF by further investigation on MWF.
Ethyl carbamate (EC) classified as a probable human carcinogen (Group 2A) is naturally formed in alcoholic beverages and fermented foods during fermentation process and/or during storage. The objective of this study was to analyze EC in 34 food items including 14 alcoholic beverages and 20 fermented foods sold in Korea. Each food was collected from 18 supermarkets in 9 metropolitan cities in Korea, and then made into composite. According to food composition and alcohol content, samples were divided into four matrices such as apple juice, milk, Soju (liquor containing about 20% alcohol), and rice porridge. The maximum EC value of $151.06{\mu}g/kg$ was found in Maesilju (liquor made from Maesil and Soju). Whisky and Bokbunjaju (Korean black raspberry wine) contained $9.90{\mu}g/kg$ and $6.30{\mu}g/kg$, respectively. EC was not detected in other alcoholic beverages. Of 20 fermented foods, Japanese-style soy sauce had highest level of $15.59{\mu}g/kg$ and traditional one contained $4.18{\mu}g/kg$. Soybean paste had $1.18{\mu}g/kg$, however, EC was not found in other fermented foods.

Furan ($C_4H_4O$) is a volatile compound formed mostly during the thermal processing of foods. The toxicity of furan has been well documented previously, and it was classified as "possible human carcinogen (Group 2B)" by the International Agency for Research on Cancer. Various pathways have been reported for the formation of furan, that is, thermal degradation and/or thermal rearrangement of carbohydrates in the presence of amino acids, thermal degradation of certain amino acids, including aspartic acid, threonine, ${\alpha}$-alanine, serine, and cysteine, oxidation of ascorbic acid at higher temperatures, and oxidation of polyunsaturated fatty acids and carotenoids. Owing to the complexity of the formation mechanism, a vast number of studies have been published on monitoring furan in commercial food products and on the potential strategies for reducing furan. Thus, we present a comprehensive review on the current status of commercial food monitoring databases and the possible furan reduction methods. Additionally, we review analytical methods for furan detection and the toxicity of furan.
Formononetin is an isoflavonoid phytoestrogen found in certain foodstuffs such as soy and red clover. In this study, we examined the action of formononetin with the carcinogen activation pathway mediated through the aryl hydrocarbon receptor (AhR) in MCF-7 breast carcinoma cells. Treating the cells with formononetin alone caused the accumulation of CYP1A1 mRNA as well as elevation in CYP1A1-specific 7-ethoxyresorufin O-deethylase (EROD) activity in a dose dependent manner. However, a concomitant treatment with 7,12-dimethylbenz[a]anthracene (DMBA) and formononetin markedly reduced both the DMBA-inducible EROD activity and CYP1A1 mRNA level. Under the same conditions, formononetin inhibited the DMBA-induced AhR transactivation, as shown by reporter gene analysis using a xenobiotic responsive element (XRE). Additionally, formononetin inhibited both DMBA-inducible nuclear localization of the aryl hydrocarbon receptor (AhR) and metabolic activation of DMBA, as measured by the formation of the DMBA-DNA adducts. Furthermore, formononetin competed with the prototypical AhR ligand, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), for binding to the AhR in an isolated rat cytosol. These results suggest that formononetin might be considered as a natural ligand to bind on AhR and consequently produces a potent protective effect against DMBA-induced genotoxicity. Therefore, that's the potential to act as a chemopreventive agent that is related to its effect on AhR pathway as antagonist/agonist.

Bromate is a disinfection by-product generated mainly from the oxidation of bromide during the ozonation and disinfection process in order to remove pathogenic microorganism of drinking water, and classified as a possible human carcinogen by International Agency for Research of Cancer (IARC) and World Health Organization (WHO). For the purpose of determining the trace level concentration of bromate, several sensitive techniques are applied mostly based on suppressed conductivity detection and UV/Visible detection after postcolumn reaction (PCR). In this study, the suppressed conductivity detection method and the PCR-UV/Visible detection method through the triiodide reaction were compared to analyze the trace bromate in water samples and estimated for the availability of these analytical methods. In addtion, the state-of-the-art techniques was applied for the determination of trace level bromate in various water matrices, i.e., soft drinking water, hard drinking water, mineral water, swimming pool water, and raw water. In comparison of two analytical methods, it was found that the conductivity detection had the suitable advantage to simultaneously analyze bromate and inorganic anions, however, the bromate might not be precisely quantified due to the matrix effect especially by chloride ion. On the other hand, the trace bromate was analyzed effectively by the method of PCR-UV/Visible detection through triiodide reaction to satisfactorily minimize the matrix interference of chloride ion in various water samples, showing the good linearity and reproducibility. Furthermore, the method detection limit (MDL) and recovery were 0.161 ㎍/L and 101.0-108.1 %, respectively, with a better availability compared to conductivity detection.
Some of the petroleum products contain benzene which is well known as a confirmed human carcinogen. For example, gasoline products contain benzene ranging up to several percents by weight. High exposures to the benzene and other organic solvents would be likely to occur during intermittent tasks and or processes rather than continuous jobs such as sampling, repair, inspection, and loading/unloading jobs. The work time for these jobs is various. However, most of work time is very short and the representative time interval is 15 minutes. Thus, it is preferable to do exposure assessment for 15 minute time weighted average which is known as a short time exposure level(STEL) by ACGIH rather than for 8-hours TWA. It is particularly significant to the exposure monitoring for benzene since it has been known that the exposure rate plays an important role to provoke the leukemia. Due to the large variations, a number of processes/tasks, the traditional sampling technique for organic solvents with the use of the charcoal and sampling pumps is not appropriate. Limited number of samples can be obtained due to the shortage of sampling pumps. Passive samplers can eliminate these limitations. However, low sampling rates resulted in collection of small amount of the target analysts in the passive samplers. This is originated the nature of passive samplers. Field applications were made with use of passive samplers to compare with the charcoal tube methods for 15 minutes. Gasoline loading processes to the tank lorry trucks at the loading stations in the petroleum products storage area. Good agreements between the results of passive samplers and those of the charcoal tubes were achieved. However, it was found that special cautions were necessary during the analysis at very low concentration levels.

Background: Phenethyl isothiocyanate (PEITC), the most comprehensively studied aromatic isothiocyanate, has been shown to act as an anti-cancer agent mainly through modulation of biotransformation enzymes responsible for metabolizing carcinogens in the human body. Humans are often exposed to carcinogenic factors, some of which through the diet, such as polycyclic aromatic hydrocarbon benzo[a]pyrene via the consumption of over-cooked meats. Inhibition of the enzymes responsible for the bioactivation of this carcinogen, for example CYP1A1, the major enzyme required for polycyclic aromatic hydrocarbons (PAHs) bioactivation, is recognized as a chemoprevention strategy. Objective: To evaluate the inhibitory effects of PEITC against benzo[a]pyrene-induced rise in rat liver CYP1A1 mRNA and apoprotein levels. Materials and Methods: Precision cut rat liver slices were treated with benzo[a]pyrene at 1 and $5{\mu}M$ in the presence of PEITC ($1-25{\mu}M$) for 24 hours, followed by determination of CYP1A1 mRNA and apoprotein levels using quantitative polymerase chain reaction and immunoblotting. Results: Findings revealed that PEITC inhibited benzo[a]pyrene-induced rise in rat liver CYP1A1 mRNA in a dose-dependent manner as well as the apoprotein levels of CYP1A. Conclusions: It was demonstrated that PEITC can directly inhibit the bioactivation of benzo[a]pyrene, indicating chemopreventive potential.
Objectives: Diesel engine exhaust (DE) accounts for a significant percentage of air pollutants that are associated with various health outcomes including mortality, asthma, chronic bronchitis, respiratory tract infection, etc. In June, 2012, the International Agency for Research on Cancer (IARC) released the assessment results that classified DE as "carcinogenic to humans" (Group 1). This review is therefore focused on the lung cancer risks of DE. Methods: Literatures were searched using PubMed with key words of "diesel exhaust", "lung cancer", and other related terms for the period between 1990 and 2012. A total of 295 articles were searched and sixteen epidemiologic studies were identified as potentially relevant. Results: Sixteen epidemiologic studies about the lung cancer risks of workers exposed to DE in various occupations were summarized in two tables, 1) retrospective cohort studies and 2) case-control studies. Increased lung cancer risk, although not always smoking adjusted, was observed in 6 out of 8 retrospective cohort studies and 4 of 8 case-control studies. Conclusions: Diesel fuel is widely used in Korea. Exposure to DE is confirmed to be a human carcinogen by IARC. Noncancer health risks of DE also need careful attention as DE is a major source of fine-particle pollution. Along with the efforts for reducing the DE emission through improvements of diesel engines and fuel, and the use of alternative fuels, comprehensive health risk assessment of DE should be conducted to minimize the adverse health effects.

Rapidly progressive interstitial renal fibrosis has recently been reported in young women who have been on a slimming regimen including chinese herbs. Aristolochic acid, suspected as the causal factor of this renal disease, is a well known carcinogen. It has been known that Madouling (Aristolochiae fructus) contains aristolochic acid. The objective of this study was to investigate the effects of Madouling, Madouling-tang, which are the extract mixture from 10 different chinese herbs including Madouling, and aristolochic acid on reproductive and developmental toxicity. Female rats were administered orally with the extracts of Madouling, madouling-tang, and aristolochic acid from 14 days before mating to day 17 of gestation. Madouling (8mg/kg) decreased fertility in the 8mg/kg group, but Madouling-tang and aristolochic acids did not. Significant decrease of mean fetal body weights were observed in the 16mg/kg group of aristolochic acids. External, visceral and skeletal malformation of fetuses were not observed with treatment. Histopathological examination showed the discrete damage of kidney in the 8mg/kg group of Madouling and 16mg/kg groups of aristolochic acid. In whole embryo culture, Madouling and Madouling-tang caused the retardation of growth and development of embryo in the dose of 1 $\mu$g/ml and 0.02 $\mu\textrm{g}$/kg, respectively while aristolochic acids showed the similar effect in the dose of 300 $\mu\textrm{g}$/kg. These results indicate that Madouling, up to 0.05mg/kg (prescription dose to human) has no adverse effects on the fertility, reproduction and development of Sprague-Dawley rats.

This study covers the investigation of the actual condition in the workplace to produce plastics products using synthetic resins and the investigation on the trends amount of the domestic production of thermoplastic resins. To analyze the monomers included in thermoplastic resins frequently used in the workplace, we analyzed thermal characteristics for test compounds using thermogravimetric analysis and did the qualitative analysis using Pyrolyzer GC-MSD & TDS GC-MSD. And then we classified the health hazard of monomers based on GHS classification criteria using information toxicity & carcinogenicity. The number of the workplace to produce plastics products among all domestic manufacturers of 73,884 was 4,391 (5.94%). The number of workers to produce plastics products among all workers of 2,522,750 in all domestic manufacturers was 104,971 (4.16%). The amount of production per year for thermoplastic resins is in the order of PP, HDPE, LDPE, PVC, ABS, PS and such compounds was producing over 1 Million ton per year each. The classification result based on GHS classification criteria for 22 main compounds included thermoplastic resins says 2 compounds of acrylonitrile, naphthalene are in Acute oral category 3 and benzene is in Acute dermal category 1. The classification results of health hazard of carcinogenicity based on IARC & ACGIH carcinogen classification says 2 compounds of benzene, vinyl chloride are in category 1A (known to be human carcinogens).
