Hematopoietic Efficacy of Recombinant Human Erythropoietin (DWP413) (Recombinant Human Erythropoietin (DWP413)의 적혈구 생성 효과)

  • Efficacy and in vivo bioassay of recombinant human erythropoietin (rh-EPO, DWP413) was investigated. Efficacy studies on erythropoiesis were conducted in normal, cisplatin-induced anemic rats and acute hemorrhage - induced anemic rats. Animals were treated intravenously with DWP413 for 5 days, the changes in the number of red blood cells (RBC), hematocrit value (Hct), hemoglobin concentration (Hb) and reticulocyte were examined. In normal rats, at the doses of 50, 250, and 1250 IU/kg/day, in cisplatin-induced anemic rats, at the doses of 50, 100 and 200 IU/kg/day, RBC, Hb, Hct and reticulocyte were increased dose-depen-dently. And in acute hemorrhage-induced anemic rats, DWP413 (150, 450 and 1350 IU/kg/day) significantly increased RBC, Hb, Hct and reticulocytes. In histopathological findings of kidney, cisplatin alone treated rats expressed severe glomerulus and tubular damage. But in the DWP413 treated rats after cisplatin treatment, these were not remarkable compared to cisplatin alone treated rats. In vivo bioassay, DWP413 had 102.43% of bioactivity compared to erythropoietin BRP(Biological Reference Product, European Directorate for the Quality of Medicines). These results suggest that DWP413 might be useful for the therapy of anemia induce by renal failure and acute blood loss.

  • The detection of pulmonary blood vessels is very difficult owing to their complex tree structures and different widths. In this paper, We propose a new detection algorithm. The motivation of this algorithm is that Han is the best detector. So, this algorithm is developed to imitate the human searching process. To realize it, the algorithm consist of two components. One is Pyramid Images whose one pixel is median value of four pixels of the previous low level. Searching gradually from high level to low level, We concentrate on global and main information of structure at the first. Then based on it, We search the detailed data in low level. The other is fuzzy logic which makes it easy to convert searching process expressed as human language into numeric multi_value. This algorithm showes speedy and robust results. But the more study on both human searching process and the detection of small part is needed.

  • Efficacy and general pharmacology of recombinant human erythropoietin (rHu-EPO), were investigated. Efficacy studies were conducted in normal, cisplatin- and acute hemorrhage-induced anemic rats. Normal and anemic animals were treated intravenously with rHu-EPO for 5 days and the changes in the numbers of red blood cells (RBC), hemoglobin (Hb), hematocrit (Hct) and reticulocytes (Ret) were examined. In normal rats, rHu-EPO significantly increased RBC, Hb, Hct and Ret at doses of 50∼ 1250 IU/kg/day. Cisplatin-induced anemic rats showed significant increase of RBC, Hb, Hct and Ret in a dose-dependent manner after administration of rHu-EPO (50∼200 lU/kg/day). And in acute hemorrhage-induced anemic rats, rHu-EPO(4∼100 Iu/kg/day) accelerated recovery of anemia with significant increase in Ret. These changes disappeared gradually after cessation of the treatment. The general pharmacological effects of rHu-EPO were investigated in mice, rats, guinea-pigs, rabbits and cats. rHu-EPO had no influences on central nervous, cardiovascular, gastrointestinal and blood coagulation system. It also had no influence on the contraction of phrenic nerve-diaphragm preparation. rHu-EPO produced significant increase of urine volume at a dose of 7000 IU/kg. These results suggest that rHu-EPO might be useful for the therapy of anemia without serious side effect.

Effect of Leaf Mustard (Brassica juncea) on Lipid Metabolism of Rat with Chroethanol Administration (갓(Brassica juncea) 첨가식이가 만성적인 알콜 투여시 흰쥐의 지방대사에 미치는 영향)

  • This study was undertaken to investigate the effects of mustard leaf diet on the lipid metabolism under chronic alcohol administration. Sprague-Dawley male rats were fed either AIN-76 diet(control), control with ethanol, AIN-76 diet plus 5% mustard leaf(mustard leaf), or mustard leaf with ethanol lot 30 days. On the 21st day, all of the rats were given an oral dose of ethanol and blood-ethanol concentration were monitored for the next 5 hours. Lipid and enzyme determinations in serum and liver were carried out after 30 days. The results obtained were summarized as following: 1) Supplementing 5% of mustard leaf did not recover the body weight loss due to chronic alcohol administration. 2) There were no significant differences in blood ethanol concentrations among the experimental groups. 3) Mustard leaf diet decreased the plasma total cholesterol, triglyceride levels increased due to the chronic alcohol administration, but not HDL-, LDL-cholesterol, and liver lipids. 4) Mustard leaf diet decreased ${\gamma}$ -GTP level increased by chronic alcohol administration. Overall, these data suggest that mustard lear can have a recovery function, which was not via ethanol metabolism on the symptoms of alcohol related diseases.(Korean J Human Ecology 1(1) : 94~102, 1998)

  • Medical equipment of high frequency has been presently applied to various parts of human body and novel treatment effects have been shown by it. Recently there are many cases that high frequency apparatus has been used in dermatology as the interest in beauty has been growing. Especially used in skin, blood volume is increased by keeping surface temperature of skin at around $43^{\circ}C$ and domodex folliculorum are treated by applying high electric field. It is the aspect we consider that increasing blood volume and domodex folliculorum treatment take effect on hair loss. In this study, high frequency electrotherapy apparatus was proposed to be applied to hair loss treatment. For that, human body was experimented with varying output voltage and frequency. The adjustable switching signal for treatment was obtained from 100[kHz] to 1000[kHz] by using DSP. Stable output specific was also obtained by feedback control to protect human body. The best hair growth conditions were the output voltage of 1.2[kV], the frequency of near 300[kHz] and the maximum current of 2[mA]. We apply this high frequency electrotherapy apparatus to two bald men, 46-year-old and 45-year-old. After treatment for a month they had thick and strong hair.

  • Human state in human-machine systems highly affects the overall system performance, and should be detected and monitored. Physiological cues are essential indicators of human state and useful for the purpose of monitoring. The study presented in this paper was focused on developing a bio-inspired sensing system, i.e., Nano-Skin, to non-intrusively measure physiological cues on human-machine contact surfaces to detect human state. The paper is presented in three parts. The first part is to analyze the relationship between human state and physiological cues, and to introduce the conceptual design of Nano-Skin. Generally, heart rate, skin conductance, skin temperature, operating force, blood alcohol concentration, sweat rate, and electromyography are closely related with human state. They can be measured through human-machine contact surfaces using Nano-Skin. The second part is to discuss the technologies for skin temperature measurement. The third part is to introduce the design and manufacture of the Nano-Skin for skin temperature measurement. Experiments were performed to verify the performance of the Nano-Skin in temperature measurement. Overall, the study concludes that Nano-Skin is a promising product for measuring physiological cues on human-machine contact surfaces to detect human state.

Effect of Panax ginseng Extract on Growth Responses of Human Intestinal Bacteria and Bacterial Metabolism

  • The growth responses of a variety of human Intestinal bacteria to extracts of Pun(1.vKy'n.ieny and five other oriental medicinal Araliaceae were evaluated in vitro and in vivo. The extracts enhanced the growth of Bifidobncterilim breve and B. longum in Media with or without carbon sources, suggesting the bifid factor (5) might be involved in the phenomenon. This effect was most pronounced with water extract of p. ginseng, the growth of 27 bifidobacteria strains belonging to B. ndolexcentium, H. longlrm, and 1. breve and B. iniuntis being greatly stimurated, whereas seven B. bifidum strains and other bacteria such as clostridia and 5.fcherirhia coli had little or no ability to utilizes it (or growth. Methanol extracts of p, ginseng were found to selectively inhibit growth of various clostridia including C. perfringens and C. Paraputrificum, but this effect was not observed on other bacteria including bifidobacteria. The effect of ginseng extract intake(600 mg/day for two weeks) on the fecal microflora, pH, volatile fatty acids, ammonia, putrefactive products, and -glucuronidase, -glucosidase and nitroreductase activities, and on the blood components (triglyceride, total cholesterol and ammonia) were investigated using seven healthy human volunteers. The total concentration of fecal microflora including Bri'idobucterilim app. during the period of ginseng extract intake was significantly unaffected from the proceeding and sub sequent control periods. However, the frequency of occurrence of subjects having C. perfringens was significantly decreased. The fecal pH value was also significantly decreased, suggesting that the intake might increase the activity of Bifidobacterium spp. Other biochemical properties in faces did not changed significantly. The levels of ammonia and triglycerid in blood were decreased with ginseng extract intake. These results may be an indication of at least one of the pharmacological actions of P ginseng as an adaptogen.

  • Polychlorinated dibenzo-p-dioxins(PCDDs), dibenzofurans(PCDFs), and biphenyls(PCBs) were analyzed by HRGC/HRMS in human blood samples from the Siwha industrial area in Korea. The PCDD/F and PCB concentrations in workers were higher than those in residents. The average TEQ concentrations of PCDD/Fs in workers and residents were 40.3 pg I-TEQs/g lipid and 16.6 pg I-TEQs/g lipid, respectively, and the total PCB concentrations were 174.1 ng/g lipid and 151.0 ng/g lipid, respectively. When applied to principal component analysis, the PCDD/F congener distributions of workers were differentiated from those of residents. But, all of them were similar to the distribution in several Korean foods. Estimated daily intake(EDI) of each person was calculated, and then only 40% to the total people were within the tolerable daily intake range(1-4pg I-TEQ/kg bw/day) suggested by WHO(1997).

  • The morphological development of the carotid body was studied by electron microscope in human fetuses from 40mm to 260mm crown rump length (10-30 weeks of gestational age). At 40mm fetus, the carotid body was composed of cluster of primitive glomus cells, primitive supporting cells, unmyelinated nerve fibers, and blood capillaries. In connective tissue between internal and external carotid arteries adjacent to the superior cervical sympathetic ganglion, two types of glomus cells through all prenatal period were found. Dark cells contained a dense cytoplasm with conspicuous large dense-cored granules, whereas light cells had a less dense cytoplasm with dense-cored granules. The light cells contained dense-cored granules that were smaller and less abundant than those in the dark cells. The primitive supporting cells appeared star-shaped with attenuated cytoplasmic extensions intervening between the adjacent glomus cells. Synaptic contact between the axon terminals and soma of the glomus cells were first observed at 40mm fetus. In 80-100mm fetus, the carotid body contained tightly packed collection of glomus cells and supporting cells which surrounded the abundant thin-walled blood vessels. Intercellular junctions between the glomus cells and adjacent cells were commonly seen. Nerve endings on the glomus cells have the form of small boutons and the other from of large calyces. During the second half of the fetal period, the glomus cells were completely enveloped by supporting cells and nerve terminals. At 260mm, the morphological features of carotid body were similar to those of human adult. The result of this study demonstrates that there are differences between the carotid body and aorticopulmonary bodies, especially with respect to their synaptic complexes, abundant blood capillaries, and two glomus cell types.

  • We have expressed human erythropoietin (EPO) in transgenic mice using a recombinant EPO cDNA combined with a partial TPO construct. The gene was microinjected using standard techniques and five mice were detected as transgenic by PCR and further used as founders. The life span of the transgenic founders was much shorter than that of their normal littermates. Most of the tissues of the transgenic founders contained human EPO transcripts as judged by RT-PCR. Especially high expression levels were seen in the liver and lung. EPO protein levels in serum were examined by ELISA and ranged from 266-414 mIU/ml. The number of red blood cell, white blood cell and hemoglobin in the hEPO transgenic mice was higher than in normal mice. These results indicate that overexpression of hEPO is deleterious and can provoke lung failure and erythrocytosis.
