Phytoestrogens, especially Yak-kong or soybean-derived isoflavones have been traditionally used as a supplement of estrogen for preventing postmemopausal osteoporosis in oriental folk medicine. In our previous study, the treatment of Yak-kong and soybean increased estrogen receptor-a (ERa) expression and proliferation of MG-63 osteoblastic cells. In contrast, the increase of estrogen receptor-$\beta$ (ER$\beta$) expression in proliferating MG-63 cells with Yak-kong and soybean treatment was less pronounced, which suggested that ER$\beta$ may play a role rather in the regulation of bone cell differentiation To determine the role of ER$\beta$ in Yak-kong or soybean mediated regulation of bone cell differentiation, we established MG-63 cell lines stably expressing either ER$\beta$ or antisense ER$\beta$ RNAs. Increased expression of ER$\beta$ did not affect ERa expression and proliferation of MG-63 cells. However, increased expression of ER$\beta$ in MG-63 cells (ER$\beta$-MG63 cells) selectively enhanced Yak-kong or soybean induced expression of osteoprotegerin (OPG), a novel soluble glycoprotein which is secreted from osteoblasts and mediates the signal for osteoclast differentiation. Inhibition of ER$\beta$ expression by antisense ER$\beta$ RNAs (As-ER$\beta$-MG63) caused these cells to insensitize Yak-kong or soybean induced expression of OPG but increased MG-63 cell proliferation. Furthermore, the comparable effects between Yak-kong and the combined treatment of genistein and daidzein at $0.5{\times}l0^{-8}$ M, which is a concentration of these two isoflavones similar to Yak-kong at 0.001 mg/mL, on OPG expression in ER$\beta$-MG63 cell demonstrate that the enhanced expression of OPG with Yak-kong treatment is mediated by the synergistic effect of low leveled isoflavones in the extracts. Together, coupled with low level of ER expression in osteoclasts, our data demonstrate that ER$\beta$ in osteoblasts plays an important role in Yak-kong and soybean mediated inhibition of osteoclast differentiation indirectly by enhancing the expression of OPG.
The anticarcingenic effect of methanol extracts from such cruciferous vegetables as cabbage, red cabbage, Korean cabbage, kale, cauliflower, broccoli, radish root, leafy radish, rape leaves and shepherd’s purse on the growth of human K-562 leukemia cells, MG-63 osteosarcoma cells, HT-29 colon cancer cells and AGS gastric cancer cells were studied. All of cruciferous vegetables inhibited more than 70% of the growth of K-52 leukemia cells and more than50% fo rhe growth fo AGS gastric cancer cells. Particularly, kale, broccoli and shepherd’s purse showed inhibition rates of 93.5%, 93,5% and 96.3% on the growth of AGS gastric cancer cells, respectively. In case of HT-29 colon cancer cells, the methanol extracts of cabbage, kale and shepherd’purse exhibited 82.4%, 72.15, 79.4% and 95.6% of inhibitory effects, respectively. The cabbage, kale, cauliflower and shepherd’s purse extracts also highly suppressed the proliferation of MG-63 cells. Generally the 10 cruciferous vegetable we studied strongly decreased the growth of various human cancer cells in vitro, however, kale and shepherd’s showed the most effective vegetable among them.

We investigated the inhibitory effects of phytol and methyl 11,14,17 eicosatrienoic acid (methyl ETA, n 3, 20 : 3) separated from perilla leaves on the growth of human cancer cells. Phytol inhibited significantly the growth of HT 29 human colon cancer cells, MG 63 osteosarcoma cells and AZ 521 gastric cancer cells. Although the activity of methyl ETA was lower than that of phytol, it was also observed to have the inhibitory effect on three human cancer cell lines. Furthermore, the DNA synthesis of the MG 63 osteosarcoma cells was markedly decreased by the addition of the phytol and methyl ETA. These results suggest that phytol and methyl ETA identified from the perilla leaves may play a role on the growth inhibition of the human cancer cells.
A variety of titanium (Ti) and its alloys are used in the clinical procedures of bone regeneration for periodontal and dental implant therapies. This study was performed to determine the effect of different surface dental implant materials on biologic responses of a MG-63 human osteoblast-like cell line. MG-63 cells were cultured on Ti coated with hydroxyapatite (HA), calcium metaphosphate (CMP), anodized (A), which compared with non-coated Ti (control). The appearances of surface of dental implant materials and the morphology of these cells were assessed by scanning electron microscopy (SEM). The gene expression profiles of MG-63 cells cultured on Ti were examined by human cDNA microarray (1,152 elements). The expression of several genes was up- and down-regulated by different surfaces of dental implant materials. Interesting, the genes correlated with cellular adhesion and extra cellular matrix (ECM) formation were enhanced, in accordance surface morphology of the dental implant materials used.
Phytoestrogens, especially soy-derived isoflavones, are receiving great scrutiny as a food supplement for preventing hormone dependent diseases such as cardiovascular diseases, cancer, and osteoporosis. These beneficial effects of phytoestrogens are caused by functioning as partial agonists or antagonists of estrogens. In contrast to the common usage of soy bean, Yak-kong(Rhynchosia Molubilis ; ) has been used as supplements of estrogen fir preventing postmenopausal osteoporosis in Oriental medicine. To investigate estrogenic effects of Yak-kong and soy bean on the proliferation of MG-63 osteoblastic cells, each bean was extracted with 70% methanol and dried by freeze-drying. Yak-kong treatment of MG-63 cells resulted in an increase of cell proliferation to a maximum of 76% compared to 68% of soy bean treatment. Treatment of MG-63 cells with Yak-kong extract also resulted in an increase of transactivation of an ERE(estrogen response element)-luciferase reporter plasmid and IGF-I expression selectively. Despite increased effects of both bean treatments on the expression of estrogen receptor $\alpha$(ER$\alpha$) and $\beta$(ER$\beta$), soy bean treatment decreased transactivation of an ERE-luciferase reporter plasmid and did not further enhance IGF-I expression. Together, our data demonstrates that the greater estrogenic response of Yak-kong extract for MG-63 cell proliferation is mediated by ER derived transactivation of ERE and selective induction of IGF-I expression.
Carthamus tinctorius L. and Eucommia umoides Oliver are often used in traditional herbal medicines for reducing damage to the liver, kidney, bone and muscle. In the present study, we investigated cell viability and alkaline phosphatase activity in the human osteoblast-like MG-63 cell line with methanol extracts of Carthami Semen (CS) and Eucommiae Cortex (EC) alone or in a mixture (CS+EC). Osteoblast cell viability was evaluated using the MTS assay and alkaline phosphatase activity assays. The cell viability and alkaline phosphatase activity significantly increased in MG-63 osteoblast cells treated with the CS+EC mixture. These findings suggest the CS+EC mixture may have beneficial effects on bone health through the proliferation of osteoblast cells.
Purpose: The aim of this study was to investigate the relationship between cell adhesion and anoikis evasion among human osteosarcoma cells (MG-63), and to further study the molecular mechanisms. Materials and Methods: Human osteosarcoma cells (MG-63) were assessed for apoptosis, and caspase-3, E-cadherin and ${\beta}$-catenin expression in EDTA and control non-EDTA groups. Results: MG-63 cells were predominantly aggregated when in suspension, and the suspended cells were more dispersed in the EDTA group. Following culture in suspension for 24 h, 48 h, or 72 h, the rates of apoptosis were $34.88%{\pm}3.64%$, $59.3%{\pm}7.22%$ and $78.5%{\pm}5.21%$ in the experimental group and $7.34%{\pm}2.13%$, $14.7%{\pm}3.69%$, and $21.4%{\pm}3.60%$ in the control group, respectively. Caspase-3 expression progressively increased and E-cadherin and ${\beta}$-catenin were decreased in the experimental group, whereas there was no change in the control group. Conclusions: MG-63 cells could avoid anoikis through cell adhesion, and E-cadherin might play a role in this process.
This study investigated the antioxidant and anti-inflammatory effects of Galla Rhois (Rhus verniciflua) on LPS (lipopolysaccharide)-stimulated human osteoblastic cells (MG-63). The aim of this study was to evaluate the LPS induced nitric oxide (NO) production and antioxidant radical in human osteoblast-like MG-63 cells. Therefore, the present work indicate that Galla Rhois extracts may be an ideal candidate for further research into their use for dental caries prevention component as well as, natural plant-based products. This suggested that 80% methanol and hexane extracts of Galla Rhois were inhibited NO generation and 1,1-diphenyl-2-pirylhydrazyl (DPPH) radical. Therefore, 80% methanol and hexane extracts of Galla Rhois may be utilized as a good source of protection against inflammation and oxidative stress. This study is intended to pursue wellness convergence in quality of life in the excavations in natural ingredients to create a variety of oral products with fewer side effects, cheap oral products for the dental treatment.
This study investigated the effect of snail extract on the growth parameters of old female rats (27 weeks). Rats were administered orally with snail extract at a dose of 100 mg/kg, 200 mg/kg, chondroitin sulfate 10 mg/kg and 0.9% saline (control) for 8 weeks. Bone mineral density (BMD) and serum concentrations of insulin-like growth factor 1 (IGF-1) and insulinlike growth factor-binding protein 3 (IGFBP-3) were significantly higher in rats exposed to snail extract for 8 weeks. MG-63 cells (human osteoblast-like cells) were treated with snail extract for 48 h. Their differentiation and proliferation was investigated with Western blot and morphological changes observed via immunofluorescence staining of ${\beta}-catenin$. Treatment with snail extract significantly increased the levels of growth factors including ${\beta}-catenin$ and IGF-1. The snail extract affected osteoblast formation. Morphological changes in MG-63 cells were observed via immunofluorescence staining. Treatment with snail extract increased the expression of ${\beta}-catenin$ in MG-63 cells. Results suggest that the treatment of MG-63 cells with snail extract increased the longitudinal growth and growth factor levels. Snail extract may be pharmacologically effective in osteogenic differentiation in vitro and represents a potential therapeutic agent for bone formation.
Effect of solvent extracts and juice supernatants from kimchis on the growth of various human cancer cells was studied, comparing with the actions on the normal cells. Inhibitory effect of kimchi extracts on[3H] thymidine incorporation n cancer cells was also investigated. The methanol extract, hexane extract and methanol soluble fraction (MSF) of 3-week fermented kimchi did not have growth inhibitory effect on Ac2F rat normal liver cells at the concentrations of 0.5~2%. However, marked decrease in the growth of AGS human gastric cancer cells was shown by the treatment of those extacts. The juice from the kimchi samples also suppressed the growth of K-562 human leukemia cells and MG-63 human osteosarcoma cells. Especially, the juice of 3-week fermented kimchi exhibited the strong growth inhibitory effect in MG-63 human osteosarcoma cells. At the photomicrographs, growth inhibition and morphological change of the cells treated with kimchi juice were observed. And the solvent extracts of 3-week fermented kimchi suppressed the growth of cancer morethan the extracts or juices from fresh and 6-week fermented kimchi. When AGS human gastric cancer cels were treated with the extracts of 3-week fermented kimchi, [3H] thymidine incorporation in the cells also decreased. These results showed that kimchi extracts and juices had growth inhibitory effects on human osteosarcoma, leukemia and gastric cancer cells, but had no toxicity to the normal cells. We suggest that kimchi might have anticancer effect in part due to inhibition of the growth and DNA synthesis of cancer cells.
