Analysis of Fatty Acid Compositions and Biodiesel Properties of Seeds of Woody Oil Plants in Korea (국내 목본 유지식물 종자의 지방산 조성 및 바이오디젤 특성 분석)

  • In order to evaluate their potential as sources of biodiesel, oil content and fatty acid composition of seeds and fatty acid methyl ester (FAME) properties from seven woody oil plants in Korea were analysed. The oil content of seed of all woody plant species ranged from 15.1 (Ligustrum lucidum) to 70.3% (Camellia japonica) by dry weight. Fatty acid composition consisted mainly of oleic acid, linoleic acid, linolenic acid, palmitic acid and stearic acid, with oleic acid being the most abundant. The content of unsaturated fatty acids of all species was higher than saturated fatty acids. Oxidation stability of seed oils of all woody plants ranged from 2.25 to 8.62 hours/$110^{\circ}C$. Fatty acid methyl ester of Styrax japonica has been found to have the highest iodine value, indicating that unsaturated fatty acid content is higher than other seed oils. Cold filter plug point(CFPP) was varied over a wide range from $0^{\circ}C$ to $-13^{\circ}C$. The cold fluidity of FAME of Chionanthus retusa were excellent.

Oxidative Stress in C100 Cells Induced by Combined Treatmentof Benzo(a)pyrene and/or 2,3,7,8-Tetrachlorodibenzo-p-dioxin(TCDD)

  • When an organism is exposed to various toxicants chronically, reactive oxygen species(ROS) are accumulated and eventually result in several biological effects from gene expression to cell death. In the present study we investigated the oxidative damage of 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin(TCDD) and/or benzo(a)pyrene (B(a)P) in C100 cells. C100 cells treated with TCDD(30 nM) and B(a)P($3{\mu}M$) underwent diverse oxidative stress as determined through thiobarbituric acid-reactive substances(TBARS) formation, DNA fragmentation, DNA single strand break(SSB) assay, immunohistochemical staining of 8-hydroxy-2'-deoxyguanosine(8-OHdG), and mRNA expressions of antioxidant enzymatic genes such as Cu/Zn-SOD gene, GPx(glutathione peroxidase 5) gene, and catalase gene. Lipid peroxidation in C100 cells was determined through measuing the formation of TBARS. For theat, the cells were pretreated with TCDD(30 nM) and/or B(a)P($3{\mu}M$) for 0.5, 1, 2 and 4 days. TBARS formation was increased in TCDD(30 nM) and B(a)P($3{\mu}M$) and mixture($30nM\;TCDD+3{\mu}M\;B(a)P$) and positive control treatment groups comparing to the controls. Mixture treatment induced more DNA fragmentation than the single treatment group at day 6. Also, SSB in all treatment groups was clearly observed when compared with the negative control group. As with the expression of antioxidant enzyme, GPx 5mRNA, B(a)P alone and mixture($30nM\;TCDD+3{\mu}M\;B(a)P$) treatment were higher comparing to those of the negative control and TCDD treatment groups. Our results suggest that exposure of C100 cells to mixture of TCDD and B(a)P leads to significant oxidative damage comparing to the exposures to the individual chemicals. Mechanisms of action are discussed. Additional studies are needed to elucidate the detailed mechanism of mixture-induced toxicity.

Helicobacter pylori inhibited cell proliferation in human periodontal ligament fibroblasts through the Cdc25C/CDK1/cyclinB1 signaling cascade

  • Purpose: Several studies have shown that the oral cavity is a secondary location for Helicobacter pylori colonization and that H. pylori is associated with the severity of periodontitis. This study investigated whether H. pylori had an effect on the periodontium. We established an invasion model of a standard strain of H. pylori in human periodontal ligament fibroblasts (hPDLFs), and evaluated the effects of H. pylori on cell proliferation and cell cycle progression. Methods: Different concentrations of H. pylori were used to infect hPDLFs, with 6 hours of co-culture. The multiplicity of infection in the low- and high-concentration groups was 10:1 and 100:1, respectively. The Cell Counting Kit-8 method and Ki-67 immunofluorescence were used to detect cell proliferation. Flow cytometry, quantitative real-time polymerase chain reaction, and western blots were used to detect cell cycle progression. In the high-concentration group, the invasion of H. pylori was observed by transmission electron microscopy. Results: It was found that H. pylori invaded the fibroblasts, with cytoplasmic localization. Analyses of cell proliferation and flow cytometry showed that H. pylori inhibited the proliferation of periodontal fibroblasts by causing G2 phase arrest. The inhibition of proliferation and G2 phase arrest were more obvious in the high-concentration group. In the low-concentration group, the G2 phase regulatory factors cyclin dependent kinase 1 (CDK1) and cell division cycle 25C (Cdc25C) were upregulated, while cyclin B1 was inhibited. However, in the high-concentration group, cyclin B1 was upregulated and CDK1 was inhibited. Furthermore, the deactivated states of tyrosine phosphorylation of CDK1 (CDK1-Y15) and serine phosphorylation of Cdc25C (Cdc25C-S216) were upregulated after H. pylori infection. Conclusions: In our model, H. pylori inhibited the proliferation of hPDLFs and exerted an invasive effect, causing G2 phase arrest via the Cdc25C/CDK1/cyclin B1 signaling cascade. Its inhibitory effect on proliferation was stronger in the high-concentration group.

  • Polycyclic Aromatic Hydrocarbons(PAHs) contamination arises from several sources including processing of food(smoking, direct drying, cooking) and environmental contamination of air, water, or soil, the later being considered as the most important. In this study, to establish the analytical method for some PAHs[benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, indeno(1,2,3-c,d)pyrene] in fish, alkali digestion time, extraction solvents, elution volume of florisil cartridge for clean-up have been optimized. The methodology involved saponification and extraction with n-hexane, clean-up on Sep-Pak florisil cartridges and determination by HPLC/FLD(High Performance Liquid Chromatography/Fluorescence Detector). Overall method recoveries for 8 PAHs spiked into these products ranged from 90 to 106%.

Air Cleaning Unit using Combination of $TiO_2$ Photocatalyst and Pulsed Discharge Plasma (산화티타늄 광촉매와 펄스 방전 플라즈마 조합에 의한 공기정화장치)

  • The purpose of this work is to develop a high-efficiency air cleaning system for air pollutants such as particulate and gaseous state in indoor environments. In order to enhance a removal efficiency of gaseous state pollutants, we suggested that pulsed discharge plasma be combined with $TiO_2$ photocatalyst (photocatalytic plasma air cleaning unit). We investigated experimentally the basic characteristics of photocatalytic plasma air cleaning unit and measured air pollutants removal efficiency. The wavelength of light radiated from pulsed discharge plasma under the atmospheric condition was 310~380nm. Its energy is enough to excite the $TiO_2$ photocatalyst and it makes a photochemical reaction in the surface of $TiO_2$ photocatalyst. The removal quantity of trimethylamine$((CH_3)_3N)\; was\; 130mg/m^34 which is twice quantity of pulsed discharge plasma without $TiO_2$ phtocatalyst unit. From the result of gas analysis using FT-IR, nitric oxide was not detected and trimethylamine was decomposed to $H_2O\; and \;CO_2$. And trimethylamine removal efficiency was 95%. These experimental results indicate that photocatalytic plasma air cleaning unit is a potential method in removing the pollutants.

  • SiC-based ceramic matrix composites show many advantages over their monolithic ceramic counterparts, which makes them potential candidates for applications in various fields. Depending strongly on the chemical composition and microstructure of the fiber reinforcement, matrix as well as the fiber/matrix interphase in the material, the properties of ceramic matrix composites(CMCs) are highly tailorable. In this paper, the latest progresses in the interphase design, matrix modification and fiber reinforcement decoration of CMCs are reviewed, their effects on the properties of the CMCs are introduced.

  • Chroma key technique which composes images by separating an object from its background in specific color has restrictions on color and space. Especially, unlike general chroma key technique, image composition for stereo 3D display requires natural image composition method in 3D space. The thesis attempted to compose images in 3D space using depth keying method which uses high resolution depth information. High resolution depth map was obtained through camera calibration between the DSLR and Kinect sensor. 3D mesh model was created by the high resolution depth information and mapped with RGB color value. Object was converted into point cloud type in 3D space after separating it from its background according to depth information. The image in which 3D virtual background and object are composed obtained and played stereo 3D images using a virtual camera.

Production responses of Holstein dairy cows when fed supplemental fat containing saturated free fatty acids: a meta-analysis

  • Objective: A meta-analysis was conducted to evaluate the effects of supplemental fat containing saturated free fatty acids (FA) on milk performance of Holstein dairy cows. Methods: A database was developed from 21 studies published between 1991 and 2016 that included 502 dairy cows and a total of 29 to 30 comparisons between dietary treatment and control without fat supplementation. Only saturated free FA (>80% of total FA) was considered as the supplemental fat. Concentration of the supplemental fat was not higher than 3.5% of diet dry matter (DM). Dairy cows were offered total mixed ration, and fed individually. Statistical analysis was conducted using random- or mixed-effects models with Metafor package in R. Results: Sub-group analysis showed that there were no differences in studies between randomized block design and Latin square/crossover design for dry matter intake (DMI) and milk production responses to the supplemental fat (all response variables, $p{\geq}0.344$). The supplemental fat across all studies improved milk yield, milk fat concentration and yield, and milk protein yield by 1.684 kg/d (p<0.001), 0.095 percent unit (p = 0.003), 0.072 kg/d (p<0.001), and 0.036 kg/d (p<0.001), respectively, but tended to decrease milk protein concentration (mean difference = -0.022 percent unit; p = 0.063) while DMI (mean difference = 0.061 kg/d; p = 0.768) remained unchanged. The assessment of heterogeneity suggested that no substantial heterogeneity occurred among all studies for DMI and milk production responses to the supplemental fat (all response variables, $I^2{\leq}24.1%$; $p{\geq}0.166$). Conclusion: The effects of saturated free FA were quantitatively evaluated. Higher milk production and yields of milk fat and protein, with DMI remaining unchanged, indicated that saturated free FA, supplemented at ${\leq}3.5%$ dietary DM from commercially available fat sources, likely improved the efficiency of milk production. Nevertheless, more studies are needed to assess the variation of production responses to different saturated free FA, either C16:0 or C18:0 alone, or in combination with potentially optimal ratio, when supplemented in dairy cow diets.

An Evaluation of the Phenomenon of Heat-Island Effect by Multi-Criteria Methods (다항목 평가기법의 적응을 통한 열섬현상의 평가)

  • The purpose of this research was to present multi-criteria which were related to the heat island and find methods which decreased heat island affection on the ecological landscape planning. The results of this study were as follows. According to the analysis of surface temperatures, the first grade was the outside-city like a mountain and its temperature was less than $13.0^{\circ}C$. The fifth grade was the downtown, industrial area and its temperature was more than $26.9^{\circ}C$. Therefore, the result was seen the serious heat-island effect. The results of field survey, the closer to the first grade, the higher the value of green coverage. The closer to the fifth grade, the higher the value of impermeability surface, paving materials and colors. According to the correlation analysis, the temperature had high correlation with impermeability surface, paving materials and colors. According to the simple regression analysis, permeability surface, green coverage, topography, impermeability surface, paving materials f: colors, human impact related with surface temperatures. To plan for the decrease of Heat-Island Effect needed the extension of green space, decrease of impermeability surface. This research suggested data for urban green plan and decrease of heat island effect, but there was a limit to get the objective method for grade classification because of lacking in the basic data, the research of multi-criteria will be accomplished continuously.

Egg Development and Larvae and Juveniles Morphology of Carp, Cyprinus carpio in Korean

  • This study was conducted to observe egg and larvae morphological development of carp to obtain basic data for resource conservation and taxonomic research. Brood carp used in the research (total length 67.3-75.5 cm, average $71.0{\pm}3.45cm$) were bred in a circular rearing aquarium ($600{\times}300{\times}100cm$) using a running water system from January to July, 2015. Breeding water temperature was maintained at $23.0-25.0^{\circ}C$(average $24.0^{\circ}C$). Fertilized carp eggs were translucent and globular, and their size was 1.75-1.89 mm (average $1.82{\pm}0.06mm$). Blastoderms formed 10 min after fertilization and reached the two-cell stage 30 min after fertilization. Then, the embryo turned dark and exhibited melanophores, and blood started flowing from the heart across the egg yolk at 42 hrs and 50 min after fertilization. Hatching began 70 hrs and 26 min after fertilization larvae emerged through the egg membrane, starting from the head. The length of prelarvae immediately after hatching was 5.23-5.38 mm (average $5.31{\pm}0.11mm$) the mouth and anus were closed, and the pectoral fin was formed. Postlarvae at 18 days after hatching had a total length of 11.9-13.9 mm (average $12.9{\pm}1.40mm$), separate anal fin and back membranes, and fin ray. Juveniles fish at 35 days after hatching had a total length of 29.9-30.2 mm (average $30.1{\pm}0.13mm$), with the body covered with scales, and the same number of fin rays, color, and shape as their broodstork.
