Two d10 Metal Coordination Polymers Based on 1H-1,2,4-Triazole: Synthesis, Structure and Fluorescence

  • The reactions of 1H-1,2,4-triazole (Htr) with $MX_2$ ($ZnCl_2$ for 1; $CdBr_2$ for 2) resulted in two coordination polymers, [Zn(tr)Cl]$_n$ (1) and $[Cd(Htr)_2Br_2]_n$ (2). The structural analyses indicate that 1 and 2 feature a 2D layer and 1D triple chain, respectively. In 1, neighouring Zn atoms are connected by ${\mu}_3-1$ ${\kappa}N$: 2 ${\kappa}N$: $4{\kappa}N-tr^-$ anionic ligand into 6- and 16-membered rings, further grow into a 2D sheet. Cd atoms in 2 are bonded by two ${\mu}_2-Br^-$ bridges and neutral ${\mu}_2$-1 ${\kappa}N$: 2 ${\kappa}N$-Htr to form a 1D triple chain. The fluorescent characterizations of 1, 2 and the free Htr ligand feature simlilar emission peakes at 444, 446 and 423 nm respectively, which can be assigned to intra-ligand ${\pi}-{\pi}^*$ transition of (H)tr. The energy gaps of 5.90 eV for 1, 5.16 eV for 2, and 5.93 eV for Htr suggest that the compounds behave as insulators.

IL-23 Inhibits Trophoblast Proliferation, Migration, and EMT via Activating p38 MAPK Signaling Pathway to Promote Recurrent Spontaneous Abortion

  • As a vital problem in reproductive health, recurrent spontaneous abortion (RSA) affects about 1% of women. We performed this study with an aim to explore the molecular mechanism of interleukin-23 (IL-23) and find optimal or effective methods to improve RSA. First, ELISA was applied to evaluate the expressions of IL-23 and its receptor in HTR-8/SVneo cells after IL-23 treatment. CCK-8, TUNEL, wound healing and transwell assays were employed to assess the proliferation, apoptosis, migration and invasion of HTR-8/SVneo cells, respectively. Additionally, the expressions of apoptosis-, migration-, epithelial-mesenchymal transition- (EMT-) and p38 MAPK signaling pathway-related proteins were measured by western blotting. To further investigate the relationship between IL-23 and p38 MAPK signaling pathway, HTR-8/SVneo cells were treated for 1 h with p38 MAPK inhibitor SB239063, followed by a series of cellular experiments on proliferation, apoptosis, migration and invasion, as aforementioned. The results showed that IL-23 and its receptors were greatly elevated in IL-23-treated HTR-8/SVneo cells. Additionally, IL-23 demonstrated suppressive effects on the proliferation, apoptosis, migration, invasion and EMT of IL-23-treated HTR-8/SVneo cells. More importantly, the molecular mechanism of IL-23 was revealed in this study; that is to say, IL-23 inhibited the proliferation, apoptosis, migration, invasion and EMT of IL-23-treated HTR-8/SVneo cells via activating p38 MAPK signaling pathway. In conclusion, IL-23 inhibits trophoblast proliferation, migration, and EMT via activating p38 MAPK signaling pathway, suggesting that IL-23 might be a novel target for the improvement of RSA.

Association of the TREML2 and HTR1E Genetic Polymorphisms with Osteoporosis

  • Osteoporosis is one of the diseases caused by accumulation of effects from complex interactions between genetic and environmental factors. Aging is the major cause for osteoporosis, which normally increases skeletal fragility and bone fracture especially among the elder. "Omics" refers to a specialized research field dealing with high-throughput biological data, such as genomics, transcriptomics, proteomics or metabolomics. Integration of data from multi-omics has been approved to be a powerful strategy to colligate biological phenomenon with multiple aspects. Actually, integrative analyses of "omics" datasets were used to present pathogenesis of specific diseases or casual biomarkers including susceptible genes. In this study, we evaluated the proposed relationship of novel susceptible genes (TREML2, HTR1E, and GLO1) with osteoporosis, which genes were obtained using multi-omics integration analyses. To this end, SNPs of the susceptible genes in the Korean female cohort were analyzed. As a result, one SNP of HTR1E and five SNPs of TREML2 were identified to associate with osteoporosis. The highest significant SNP was $rs6938076^*$ of TREML2 (OR=0.63, CI: 0.45~0.89, recessive P=0.009). Consequently, the susceptible genes identified through the multi-omics analyses were confirmed to have association with osteoporosis. Therefore, multi-omics analysis might be a powerful tool to find new genes associated with a disease. We further identified that TREML2 has more associated with osteoporosis in females than did HTR1E.

A STUDY ON TISSUE RESPONSES OF VARIOUS ALLOPLASTIC MATERIALS FOR RIDGE AUGMENTATION (수종 치조제 이물성형재의 조직반응에 관한 연구)

  • Loss of teeth causes the inevitable reduction of residual ridge. Among the various methods solving this problem, hydroxyapatite proved to be useful for correction of ridge defect and irregularity. The purpose of this study is to evaluate the tissue responses of two types Of hydroxyapatites and resin polymer. Calcitite 2040 (Calcitek Inc.), Interpore 200 (Interpore Int.), and HTR polymer (HTR Sciences) were implanted into the jaw of an adult dog. The procedure was designed to obtain the results of 1 week, 2 week, 4 week, and 12 week-intervals. And after 12 weeks from the first operation day, the dog was sacrificed and evaluated histologically by light microscope. The results were as follows : 1. The mucosa was healed after two weeks. 2. After 1 week, there were acute inflammatory cells, but diminished after 2 weeks and were not seen after 12 weeks. 3. The hydroxyapatites implanted in soft tissues were surrounded by fibrous connective tissue. And some foreign body giant cells were found. 4. Calcitite and Interpore particles implanted subperiosteally were surrounded by newly formed bone after 12 months. And direct contact between bone and particles was noted. 5. The HTR particles implanted in soft tissues were encapsulated by fibrous connective tissues. The sample where the particles contacted directly to bone could not obtained by some probable insufficiencies of surgical technique or care of the animal. And the residue of HTR particles was digested by macrophage.

Deriving Optimal Conditions of Hydrothermal Reaction for Stabilizing Heavy Metals in Contaminated Dredged Soil (오염준설토의 중금속 안정화를 위한 Hydrothermal Reaction의 최적 조건 도출)

  • Hydrothermal Reaction (HTR) was applied for the stabilization of contaminated soil with heavy metals, and then the test determined the optimal conditions for HTR. After HTR, the concentration of heavy metals in the contaminated soil increased. However, it was observed that the leachability potential significantly decreased as determined by TCLP and SPLP tests. This decrease was attributed to a decline in fractions 1-2 and an increase in fractions 3-4 as revealed by sequential extraction procedure. Due to the mineralogical characteristics of the dredged soil, distinct changes were not evident in the five-stage fraction. Therefore, it is deemed necessary to understand the chemical and mineralogical characteristics of the target soil for HTR application in order to selectively address contaminants. Comparison among operating conditions determined the optimal condition to be at 240℃ for one hour.

THE BENCHMARK CALCULATIONS OF THE GAMMA+ CODE WITH THE HTR-10 SAFETY DEMONSTRATION EXPERIMENTS

  • KAERI (Korea Atomic Energy Research Institute) has developed the GAMMA+ code for a thermo-fluid and safety analysis of a VHTR (Very High Temperature Gas-Cooled Reactor). A key safety issue of the VHTR design is to demonstrate its inherent safety features for an automatic reactor power trip and power stabilization during an anticipated transient without scram (ATWS) accident such as a loss of forced cooling by a trip of the helium circulator (LOFC) or a reactivity insertion by a control rod withdrawal (CRW). This paper intends to show the ATWS assessment capability of the GAMMA+ code which can simulate the reactor power response by solving the point-kinetic equations with six-group delayed neutrons, by considering the reactivity changes due to the effects of a core temperature variation, xenon transients, and reactivity insertions. The present benchmark calculations are performed by using the safety demonstration experiments of the 10 MW high temperature gas cooled-test module (HTR-10) in China. The calculation results of the power response transients and the solid core temperature behavior are compared with the experimental data of a LOFC ATWS test and two CRW ATWS tests by using a 1mk-control rod and a 5mk-control rod, respectively. The GAMMA+ code predicts the power response transients very well for the LOFC and CRW ATWS tests in HTR-10.

Lodging Pattern of Rice Plant in Broadcast-Seeded and Hand -Transplanted Cultivation (벼 담수표면직파재배와 손이앙재배의 도복발생 발생 양상)

  • Broadcast-seeded rice in submerged paddy frequently lodge in the field. In general, the causes of lodging in rice cultivation differ with different cultural methods. This study was conducted to investigate the causes of lodging in broadcast-seeded rice (BSR) and hand-transplanted rice (HTR) under four nitrogen (N) levels. Lodging in BSR was mainly a root lodging due to shallow root distribution, while that in HTR showed a bending type owing to deep rooting system. At the upper soil layer (0-5cm from the surface of ground) the root distribution of BSR (65.2%) was much larger than that of HTR (51.6%), whereas at the 5-10cm soil layer the root distribution of BSR (18.5%) was much smaller than that of HTR (28.0%). The depth of buried culm base was much shallower in BSR (1.2cm) than in HTR (4.0cm). The plant height, fresh weight, lodging index, culm diameter and thickness in HTR were much greater than those in BSR, and the breaking strength of lower internode was similar in the two cultivation methods indicating that HTR would have more lodging causes than BSR. In spite of . the more advantages to lodging resistance in BSR it severely lodged in the field. The main lodging-inducing factors of BSR were the shallow root distribution and shallow depth of buried culm base. Besides these, the higher ratio of gravity center of culm was an important factor. This result suggested that for the fundamental prevention of lodging in BSR, an ideotype of rice plant with ' a deep-rooted behavior ' should be developed.

Association Study between Serotonin 1A Receptor Gene rs6295 Polymorphism and Tardive Dyskinesia in Patients with Schizophrenia (조현병 환자에서의 지연성 운동이상 발생과 세로토닌 1A 수용체유전자 rs6295 다형성과의 연관성)

  • Objectives: Tardive dyskinesia (TD) is a movement disorder that is characterized by hyperkinetic movements. Previous studies have suggested that the serotonergic systems are correlated with TD vulnerability. In this study, the association between a single-nucleotide polymorphism (SNP) of the serotonin 1A receptor gene (HTR1A) rs6295 and TD was investigated. Methods: We investigated whether HTR1A rs6295 SNP is associated with antipsychotic-induced TD in 280 Korean patients with schizophrenia. Patients with schizophrenia having TD (n=105) and those without TD (n=175) were matched for their antipsychotic exposures and other relevant variables. The HTR1A rs6295 SNP was analyzed using polymerase chain reaction (PCR)-based methods. Results: There was no significant difference in the distribution of genotypic (χ2=2.70, p=0.26) and allelic (χ2=1.87, p=0.17) frequencies between the patient groups with TD and without TD. There was no significant difference in total abnormal involuntary movement scale score (F=0.39, p=0.68) among the genotype group either. Conclusion: Although there were no differences in genotypic and allelic frequency between patient groups with and without TD, further studies on association of TD with other SNPs of HTRA1 are needed to understand the pathophysiological mechanism of TD.

LSP Congestion Control methods in ATM based MPLS on BcN

  • ATM based MPLS(Multiprotocol Label Switching) is discussed for its provisioning QOS commitment capabilities, Traffic engineering and smooth migration for BcN in Korea. At this time, due to the comprehensive nature of ATM protocol, ATM has been adapted as the backbone system for carrying Internet traffic[1,2,3,4]. This paper presents preventive congestion control mechanisms for detecting HTR(Hard-To-Reach) LSP(Label Switched Path) in ATM based MPLS systems. In particular, we have introduced a HTR LSP detection method using network signaling information in an ATM layer. MPLS related studies can cover LSP failures in a physical layer fault, it can not impact network congestion status. Here we will present the research results for introducing HTR LSP detection methods and control mechanisms and this mechanism can be implementing as SOC for high speed processing a packet header. We concluded that it showed faster congestion avoidance abilities with a more reduced system load and maximized the efficiency of network resources by restricting ineffective machine attempts.

Adenosine A3 Receptor Mediates ERK1/2- and JNK-Dependent TNF-α Production in Toxoplasma gondii-Infected HTR8/SVneo Human Extravillous Trophoblast Cells

  • Toxoplasma gondii is an intracellular parasite that causes severe disease when the infection occurs during pregnancy. Adenosine is a purine nucleoside involved in numerous physiological processes; however, the role of adenosine receptors in T. gondii-induced trophoblast cell function has not been investigated until now. The goal of the present study was to evaluate the intracellular signaling pathways regulated by adenosine receptors using a HTR-8/SVneo trophoblast cell model of T. gondii infection. HTR8/SVneo human extravillous trophoblast cells were infected with or without T. gondii and then evaluated for cell morphology, intracellular proliferation of the parasite, adenosine receptor expression, TNF-α production and mitogen-activated protein (MAP) kinase signaling pathways triggered by adenosine A3 receptor (A3AR). HTR8/SVneo cells infected with T. gondii exhibited an altered cytoskeletal changes, an increased infection rate and reduced viability in an infection time-dependent manner. T. gondii significantly promoted increased TNF-α production, A3AR protein levels and p38, ERK1/2 and JNK phosphorylation compared to those observed in uninfected control cells. Moreover, the inhibition of A3AR by A3AR siRNA transfection apparently suppressed the T. gondii infection-mediated upregulation of TNF-α, A3AR production and MAPK activation. In addition, T. gondii-promoted TNF-α secretion was dramatically attenuated by pretreatment with PD098059 or SP600125. These results indicate that A3AR-mediated activation of ERK1/2 and JNK positively regulates TNF-α secretion in T. gondii-infected HTR8/SVneo cells.
