The Inhibitory Effects of Houttuynia cordata $T_{HUNB}$ against Cadmium induced Cytotoxicity (II) (어성초의 카드뮴에 대한 독성억제효과 (II))

  • This study was condo더ed to investigate the antitoxic component in aqueous extract of Houttuynia cordata $T_{HUNB}$. The results were as follows: Generally, detoxification effects by Houttuynia cordata $T_{HUNB}$ extract increased in proportion to the extract concentrations in rats. When 40 mBtg dosage of Houttuynia cordata $T_{HUNB}$ extract was administrated, Houttynia cordata $T_{HUNB}$ extract showed the highest antitoxic effects in metallothionein induction. After the extract treatment, body weights increased in proportion to the extract concentrations. However, after 3 weeks, the body weight decreased insignificantly. From the above results, Houttuynia cordate $T_{HUNB}$ extract increased metallothionein concentration and decreased the toxicity of cadmium in rats. In vitro the antitoxic activity of aqueous extract of Houttuynia cordata $T_{HUNB}$ on NIH 373 fibroblasts was evaluated by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetra-zoliumbromide) and SRB (sulforhodamine B protein) assays. The light microscopic study was carried out to observe morphological changes of the treated cells. These results were obtained as follows; The concentration of 10$^{-2}$ mg/ml of Houttuynia cordata $T_{HUNB}$ extract was shown significant antitoxic activity The number of NIH 373 fibroblasts were increased and tend to regenerate. These results suggest that Houttuynia cordata THUNB extract retains a potential antitoxic activity.oxic activity.

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata T/sub HUNB/ extracts on A549 (lung cancer), MDA-MB231 (breast cancer), SNU-C4 (colon cancer) and B16 (mouse melanoma) cell lines. We have determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The 150 ㎍/㎖ concentration of methanol extract (63.81 %) of Houttuynia cordata T/sub HUNB/ was shown significantly antitoxic activity on A549 cell lines. The order of cytotoxicity fractions of methanol from Houttuynia cordata T/sub HUNB/ extracts against cancer cell lines in vitro is as follows : hexane fraction layer > chloroform fraction layer > ethyl acetate fraction layer > buthanol fraction layer > water fraction layer. These results suggest that the hexane fraction of methanol extract from Houttuynia cordata T/sub HUNB/ extract may be a valuable choice for the development of antitumor agents.

The $150{\mu}g/ml$ concentration of chloroform extract of Houttuynia cordata $T_{HUNB}$ was shown significantly antitoxic activity on MDA-MB231$(65.96{\pm}5.68%)$ and SNU-C4$(55.94{\pm}7.39%)$ cell lines.

Comparison of Quercetin and Souble Tannin in Houttuynia cordata $T_{HUNB}.$ according to Growth Stages and Plant Parts (어성초 생육 시기 및 부위별 Quercetin과 Tannin 함량 비교)

  • Concentration of quercetin and soluble tannin in Houttuynia cordata were determined at different growth stages as well as plant parts and application amounts of compost. Both contents of quercetin and tannin at growth stages were decreased before flowering time but slightly increased after flowering. The both contents of components in Houttuynia cordata collected on April 20 were shown to be markedly increased compared to those collected in May or June. The contents of quercetin and tannin at different organs were the highest in the flower. In the experiment of application amounts of compost, as application amounts is increased, fresh weight of aerial part was increased but the contents of quercetin and tannin were decreased. The contents of quercetin and tannin were the highest with 0.67% and 2.3%, respectively, in the compost non-treatment.

Properties of Cheongkukjang Prepared with Admixed Medicinal Herb Powder (생약초(표고버섯, 더덕, 어성초)를 첨가한 청국장의 특성)

  • Cheongkukjang was prepared by addition of medicinal herb powder (from Lentinus edodes, Codonopsis lanceolata BENTH et HOOK, or Houttuynia cordata THUNB) to improve Cheongkukjang quality and functional properties. Redness and yellowness (measured using Hunter’s color values) increased with increasing amounts of medicinal herb powder, whereas lightness (the third Hunter's color value) and pH decreased. Crude protein and lipid levels of Cheongkukjang decreased with increasing amounts of added medicinal herb powder, whereas powder addition did not affect either the moisture or crude ash content of Cheongkukjang. The major organic acids of Cheongkukjang were lactic acid, acetic acid, succinic acid, and citric acid, and acid levels increased as the proportion of medicinal herb powder in the Cheongkukjang preparation increased. The major amino acids of Cheongkukjang were glutamic acid, aspartic acid, leucine, arginine, and lysine and the principal fatty acids were linoleic acid, oleic acid, palmitic acid, and linolenic acid. Addition of medicinal herb powder to Cheongkukjang increased both crude saponin and quercetin contents. Sensory scores of Cheongkukjang containing 2% (w/v) medicinal herb powder were optimal in terms of both quality characteristics and sensory evaluation.
