A Human Fungal Pathogen Cryptococcus neoformans Expresses Three Distinct Iron Permease Homologs

  • Iron plays a key role in host-pathogen interactions. Microbial pathogens require iron for survival and virulence, whereas mammalian hosts sequester and withhold iron as a means of nutritional immunity. We previously identified two paralogous genes, CFT1 and CFT2, which encode homologs of a fungal iron permease, Cft1 and Cft2, respectively, in the human fungal pathogen Cryptococcus neoformans. Cft1 was shown to play a role in the high-affinity reductive iron uptake system, and was required for transferrin utilization and full virulence in mammalian hosts. However, no role of Cft2 has been suggested yet. Here, we identified the third gene, CFT3, that produces an additional fungal iron permease homolog in C. neoformans, and we also generated the cft3 mutant for functional characterization. We aimed to reveal distinct functions of Cft1, Cft2 and Cft3 by analyzing phenotypes of the mutants lacking CFT1, CFT2 and CFT3, respectively. The endogenous promoter of CFT1, CFT2 and CFT3 was replaced with the inducible GAL7 promoter in the wild-type strain or in the cft1 mutant for gain-of-function analysis. Using these strains, we were able to find that CFT2 is required for growth in low-iron conditions in the absence of CFT1 and that overexpression of CFT2 compensates for deficiency of the cft1 mutant in iron uptake and various cellular stress conditions. However, unlike CFT2, no clear phenotypic characteristic of the cft3 mutant and the strain overexpressing CFT3 was observed. Overall, our data suggested a redundant role of Cft2 in the high-affinity iron uptake and stress responses in C. neoformans.

Decoding the intestinal microbiota repertoire of sow and weaned pigs using culturomic and metagenomic approaches

  • To elucidate the role and mechanism of microbes, we combined culture-dependent and culture-independent approaches to investigate differences in gut bacterial composition between sows and weaned pigs. Under anaerobic conditions, several nonselective and selective media were used for isolation from fecal samples. All isolated bacteria were identified and classified through 16S rRNA sequencing, and the microbiota composition of the fecal samples was analyzed by metagenomics using next generation sequencing (NGS) technology. A total of 278 and 149 colonies were acquired from the sow and weaned pig fecal samples, respectively. Culturomics analysis revealed that diverse bacterial genus and species belonged to Firmicutes, Actinobacteria, Proteobacteria, and Bacteroidetes were isolated from sow and weaned pigs. When comparing culture-dependent and culture-independent analyses, 191 bacterial species and 2 archaeal bacterial species were detected through culture-independent analysis, and a total of 23 bacteria were isolated through a culture-dependent approach, of which 65% were not detected by metagenomics. In conclusion, culturomics and metagenomics should be properly combined to fully understand the intestinal microbiota, and livestock-derived microbial resources should be informed by culturomic approaches to understand and utilize the mechanism of host-microbe interactions.

Evaluation of Durum Wheat Genotypes for Resistance against Root Rot Disease Caused by Moroccan Fusarium culmorum Isolates

  • Fusarium culmorum is one of the most important causal agents of root rot of wheat. In this study, 10 F. culmorum isolates were collected from farms located in five agro-ecological regions of Morocco. These were used to challenge 20 durum wheat genotypes via artificial inoculation of plant roots under controlled conditions. The isolate virulence was determined by three traits (roots browning index, stem browning index, and severity of root rot). An alpha-lattice design with three replicates was used, and the resulting ANOVA revealed a significant (P < 0.01) effect of isolate (I), genotype (G), and G × I interaction. A total of four response types were observed (R, MR, MS, and S) revealing that different genes in both the pathogen and the host were activated in 53% of interactions. Most genotypes were susceptible to eight or more isolates, while the Moroccan cultivar Marouan was reported resistant to three isolates and moderately resistant to three others. Similarly, the Australian breeding line SSD1479-117 was reported resistant to two isolates and moderately resistant to four others. The ICARDA elites Icaverve, Berghisyr, Berghisyr2, Amina, and Icaverve2 were identified as moderately resistant. Principal component analysis based on the genotypes responses defined two major clusters and two sub-clusters for the 10 F. culmorum isolates. Isolate Fc9 collected in Khemis Zemamra was the most virulent while isolate Fc3 collected in Haj-Kaddour was the least virulent. This work provides initial results for the discovery of differential reactions between the durum lines and isolates and the identification of novel sources of resistance.

보철물 장착 환자의 구강 칸디다균 분포 (The Distribution of Oral Candida Species in Patient with Prosthetic Appliance)

  • 본 연구는 보철물 장착 환자의 구강 내 C. albicans 분포에 대한 연구로 구강 Candida 감염증의 주 원인균인 C. albicans의 분포를 측정한 후 감염 위험정도의 분석을 통해 보철물 장착 환자들에게 구강건강관리의 중요성에 대한 자료로 제공하고자 연구대상자의 성별, 흡연 및 음주유무, 구강 내 보철물 종류 및 장착 기간, 1일 칫솔질 횟수에 따른 C. albicans 집락 정도를 조사하였다. 연구대상자들 중 11.6%에서 감염 1단계 이상의 분포를 보여 구강 Candida 감염 가능성을 확인하였다. 또한 타액 채취 부위별 C. albicans 분포를 분석한 결과 상악보다 하악의 설면에서, 입천장보다 혀 등이 상대적으로 높은 단계의 분포를 보였다. 성별, 흡연 및 음주유무에 따른 C. albicans 분포는 여자보다 남자가, 흡연을 하는 경우보다 흡연을 하지 않는 경우가, 음주를 하지 않는 경우보다 음주를 하는 경우에 구강 Candida 감염 단계 평균 수치가 높게 나타났다. C. albicans의 단계별 분포는 구강 Candida 감염 3단계의 분포가 여자는 관찰되지 않았지만 남자는 1.4%의 분포가 관찰되었으며, 구강 Candida 감염 2단계 이상의 분포가 흡연 및 음주를 하는 경우에는 관찰되지 않았지만 하지 않는 경우에는 각각 3.3%, 3.6%의 분포가 관찰되었다. 구강 내 보철물 중 가철성 교정장치보다 의치 혹은 임플란트를 장착한 경우 C. albicans의 분포에 더 영향을 미치고 있으며, 보철물을 1년 이상 장착한 경우에 기간이 길어질수록 구강 Candida 감염 1단계 이상의 분포가 점점 높아졌다. 단, 7~9년 장착한 경우에는 100.0%의 0단계의 분포를 보였다. 1일 칫솔질 횟수에 따른 구강 Candida 감염 단계 평균 수치를 통해 구강위생관리와 C. albicans의 분포의 상관관계를 확인하고자 하였으나 칫솔질 횟수는 직접적으로 작용하지 않는 것으로 보였다. 이상의 결과로 볼 때 보철물 장착 환자의 성별, 구강 내 부위, 보철물 종류, 보철물 장착 기간이 구강 내 C. albicans의 분포에 영향을 미치고 있는 것으로 나타났다. 향후 관련된 여러 요인들을 모두 추가, 보완하거나 다각적으로 검토하여 심도 있는 연구를 통해 구강건강을 유지 및 증진시킬 수 있는 자료를 확보하는 것이 필요하다고 생각된다.
