브로콜리 품종 및 부위에 따른 항균활성과 항산화 효과 (Antimicrobial and Antioxidant Activities in Different Parts and Cultivars of Broccoli)

  • 본 연구에서는 브로콜리 품종 및 부위에 따른 항균활성 및 항산화 효과를 조사하였다. 브로콜리 11개 품종에 대해 그람 양성균 Staphylococcus aureus, Listeria monocytogenes 2종과 그람 음성균 Escherichia coli O157:H7, Salmonella enteritidis 2종에 대한 항균활성을 조사한 결과, S. aureus에 대해 AMG 품종의 화뢰 추출물이 12.83mm, 잎 추출물 13.00 mm로 가장 높은 항균활성을 보였으나 부위별로는 큰 차이를 보이지 않았다. L. monocytogenes의 경우, 화뢰 추출물은 NJ 품종이 13.58mm, 잎 추출물은 YDR 품종이 13.92mm로 가장 큰 항균활성을 보였으며, 저해환의 평균값이 화뢰는 12.85mm, 잎은 13.17mm로 다른 4종의 균 중에서 가장 높은 값을 보였다. E. coli O157:H7에 대해서는 Grd 품종의 화뢰와 잎 추출물 모두 가장 높은 항균활성을 보였으나, 부위별로는 차이가 나타나지 않았으며, S. enteritidis는 화뢰추출물은 NJ 품종이 12.67mm, 잎 추출물은 NY, 0c 품종이 12.75mm의 저해환을 보였으며, 잎 추출물이 화뢰 추출물에 비해 높은 항균활성을 보여주었다. 품종별 항산화 효과를 살펴보면, 화뢰 추출물은 NJ 품종이 39.90%, 잎 추출물은 0c 품종이 43.64%로 가장 높았으며, 부위별 평균값은 화뢰추출물 23.35%, 잎 추출물 35.91%를 나타내어 잎 추출물의 항산화 효과가 약 1.5배 높게 나타났다. NJ 품종을 제외하고 모든 품종에서 잎 추출물의 전자공여능이 화뢰 추출물보다 높은 값을 나타내었다.

일당귀 잎의 포장방법이 저장 중 품질 및 생리활성에 미치는 영향 (Changes in the Quality and Physiological Activity of Angelica acutiloba Leaves in Various Packaging Materials during Storage)

  • 본 연구는 유통현장에서 다양한 포장 형태로 이용되는 일당 귀 잎의 포장방법별 선도유지 효과를 구명하고자 하였다. 현장에서 이용되는 일당귀 잎의 1 kg 단위로 포장 형태를 골판지상자에 바로 일당귀 잎을 포장한 무처리를 대조구로 하여 $20{\mu}m$ HDPE 천공 필름, $20{\mu}m$ HDPE 천공 필름+파라핀왁스 코팅 종이와 같이 3가지로 분류하였다. $4^{\circ}C$ 저온에서 20일간 저장하고 20일 후 상온으로 옮겨 2일간 추가 유통하면서 중량감소율, 색도, 외관품위, 항산화활성과 총 플라보노이드 함량 변화를 조사하였다. 그 결과, 포장 형태에 따른 일당귀 잎의 중량 감소는 봉지 형태의 $20{\mu}m$ HDPE 천공 필름 포장과 $20{\mu}m$ HDPE 천공 필름+파라핀왁스 코팅 종이 포장에 의해 가장 적었으며 저장 중의 엽색, 외관 및 중량의 변화폭도 적어 상품성이 20일간 유지되었다. 대조구로 사용한 일반 골판지상자에 포장한 일당귀 잎은 저장 15일 째 상품성을 상실하였다. 항산화활성과 총 플라보노이드 함량은 $4^{\circ}C$에 20일간 저온저장 및 상온에 2일간 추가 유통하는 동안 $20{\mu}m$ HDPE 천공 필름+파라핀왁스 코팅종이 처리가 다른 처리에 비하여 높게 유지되어 생리활성 물질 함량 보존에 유효하였으며, 국내 유통현장에서 사용하기에 가장 적절한 포장방법으로 나타났다.

Revalidating the Factor Structure of Types of Horticultural Therapy Activities with Confirmatory Factor Analysis

  • Background and objective: Horticultural activity is one of the most basic elements of horticultural therapy, which brings about therapeutic effects for participants through various plant-related activities. The main objective of this study was to verify the results of previous research, which suggested six types of activities from the exploratory factor analysis. Methods: To meet the purpose of this study, a questionnaire was designed to determine the preferences for 6 types of the horticultural therapy activities. The survey was conducted on 703 people from March 7 to June 20, 2019. The data of 674 cases were used into the final analysis, excluding unreliable responses. Descriptive statistics, and reliability analysis were performed using IBM SPSS Statistics 25, and confirmatory factor analysis was performed using IBM SPSS Amos 21. Results: First, horticultural therapy activities were classified into 6 types from the exploratory factor analysis, as conducted in previous research. The confirmatory factor analysis provided that the fit of the final model was satisfactory with χ2 = 1,300.590 (p < .001), RMR = .045, GFI = .876, RMSEA = .062, NFI = .914, TLI = .905, CFI = .914. Conclusion: This result revalidated that the mode with 6 types of horticultural therapy activities from previous research is appropriate criteria for the classification of horticultural activities. The model could be used to design more systematic horticultural therapy programs that meet the needs or circumstances of the subject, or that are suitable for necessary therapeutic intervention methods.

Transgenic Lettuce Expressing Chalcone Isomerase Gene of Chinese Cabbage Increased Levels of Flavonoids and Polyphenols

  • Flavonoid are large group of the polyphenolic compounds which are distinguished by an aromatic or phenolic ring structure and the phenolic compounds are induced by microbial infection, ultraviolet radiation, temperature and chemical stress. They are known for their antioxidant activity, anti-allergic, anti-inflammatory, anti-microbial and anti-cancer activities. In this study, changes in flavonoid content were investigated using heterologous chalcone isomerase (CHI) expression system. Also, phenolic compounds level was measured to examine the relation between flavonoids and phenols contents. Explants of lettuce (Lactuca sativa L.) were transformed with Agrobacterium tumefaciens LBA 4404 strain containing pFLH-CHI (derived from pPZP2Ha3) vector constructed with CHI gene from Brassica rapa. The putative transgenic plants were confirmed by genomic DNA PCR analysis. Also the transcription levels of the gene were analyzed by semi-quantitative RT-PCR with gene specific primers. The total flavonoid contents were increased at $T_0$ and $T_1$ generations over 1.4 and 4.0 fold, respectively. Total phenol contents also increased at $T_1$ generation. These results indicate that CHI gene plays an important role to regulate the accumulation of flavonoids and its component changes.

Effects of feeding leaf positions on the growth and fruit quality in muskmelon plants showing leaf yellowing symptoms

  • This study was conducted to evaluate the influence of feeding leaf positions on the growth, net formation of fruits, and occurrence of leaf yellowing symptoms (LYS) in muskmelon plants. Plants having five or ten more leaves above the fruit-bearing node produced the greater biomass than those of plants having equal or five less leaves above the fruit-bearing node. The number of leaves above the fruit-bearing node also influenced on the occurrence of LYS. The number of plants with LYS decreased as the number of leaves borne on the nodes above the fruit-bearing node increased. The LYS infected ratio of BL-5 treatment were the greatest, while fruit weight of BL+5 treatment were the greatest among all the tested treatments. In addition, the net formation of BL-5 treatment showed the poorest. Results indicated that maintaining the higher number of leaves over the fruit-bearing node might be feasible the practical method for coping physiological damages from yellowing symptoms.

Electromyographic Analysis of Upper and Lower Limb Muscles during Gardening Tasks

  • Movements of the upper and lower limb muscles during five common gardening tasks were analyzed by using electromyography (EMG). Twenty adults aged in their twenties (mean age, $24.8{\pm}2.4$ years) were recruited. On two separate occasions, subjects visited a garden plot to perform digging, raking, troweling, weeding, and hoeing; all tasks were performed three times with 20 s intervals for each trial. To measure muscle activation during the five gardening tasks, surface EMG was used. Bipolar surface EMG electrodes were attached to eight upper limb muscles (bilateral anterior deltoid, biceps brachialis, brachioradialis, and flexor carpi ulnaris) or eight lower limb muscles (bilateral vastus lateralis, vastus medialis, biceps femoris, and gastrocnemius) on both sides of the body, for a total of 16 muscles. During the five tasks, photographs were taken of movement phases using a digital video camera. The right flexor carpi ulnaris and brachioradialis showed higher activation than the other upper and lower limb muscles measured during the tasks. All 16 upper and lower limb muscles were actively used only during digging. According to movement analysis of each activity, digging was classified into four movement phases, whereas raking, troweling, weeding, and hoeing each were divided into three movement phases. In each activity, there were high-impact phases in terms of muscle activation; the flexor carpi ulnaris and brachioradialis were identified as major muscles in each impact phase. This analysis may be used to generate biomechanical profiles of gardening tasks for practitioners when designing efficient gardening interventions for physical health or rehabilitation.

The Role of Visitor's Positive Emotions on Satisfaction and Loyalty with the Perception of Perceived Restorative Environment of Healing Garden

  • Background and objective: The purpose of this study is to investigate the effects of visitors' positive emotions on satisfaction and loyalty with the perception of restorative environment of a healing garden created in an urban agriculture expo. Methods: The psychological indicators to the images of the healing garden were analyzed by the visitors' demographic variables and the three factors of plant cultivation activity level: plant cultivation experience, plant preference, and plant-related event. Results: Between age groups and occupational groups, significant differences were found statistically. The Perceived Restorativeness Scale(PRS) showed significantly differences between age groups in repose, fascination and legibility. The Positive Affect & Negative Affect Schedule(PANAS) showed statistically significant differences between age groups in positive emotions. In addition, we investigated the correlation between the PANAS and the three factors of plant cultivation experience level, the four factors of the PRS, satisfaction and loyalty. The three factors of plant cultivation experience level, the four factors of the PRS, satisfaction and loyalty showed a positive correlation with positive emotions and were inversely correlated with negative emotions significantly. Multiple regression analysis with dummy variables was conducted to examine the effects of plant cultivation activity level, attention restoration, and the PANAS on healing garden visitors' satisfaction and loyalty. As a result, among the four factors of the PRS, fascination and positive affectivity were significant variables that affect healing garden visitors' satisfaction and loyalty. Conclusion: The results indicated that the higher the attention restoration of visitors due to the fascination of the healing garden and the higher their positive affectivity and the more they have plant-related memories, the higher their impact on healing garden visitors' satisfaction and loyalty. Therefore, fascinating natural environments or greenery landscapes like healing gardens where people can contact plants would reduce negative emotions such as anger and sadness but to increase positive emotions such as pleasure, joy and satisfaction.

무화과 펙틴에스테라제의 부분 정제 및 in situ 상태에서의 활성 특성 (Partial Purification of Fig Pectinesterase and Characterization of its in situ Activity)

  • 무화과 PE를 추출하여 ammonium sulfate로 분획 투석한 후 Q-Sepharose column 및 CM-cation exchanger column을 이용한 chromatography와 HPLC에 의해 1개의 음이온성 PE와 2개의 양이온성 PE로 분리되었으며, 이들은 모두 전기영동상에 분자량 27,000정도의 밀접한 두 개의 band로 나타난 부분 정제된 단백질이었다. 이 효소 단백질들은 보관 중에 활성을 급격히 상실되므로 현실적인 이용성을 고려하여 분말화한 시료 현탁액을 이용한 in situ PE의 특성을 조사하였다. 그 결과 분말 시료는 냉동 저장뿐 만 아니라 $5^{\circ}C$에서도 장기간 저장할 수 있었으며, 최적 pH는 8.5, 최적온도는 $50^{\circ}C$이였고, 활성화 에너지는 7,671 cal $mol^{-1}\;K^{-1}$ 이었으며 $55^{\circ}C$까지는 열 안정성을 유지하였다. 또한 $0.2{\sim}0.4$ M NaCl에서 활성이 촉진되었으며 PE의 용출은 0.8 M 이상의 NaCl 에서 효과적이였고 $0{\sim}1.0$ M NaCl까지의 범위에서는 특별히 안정성에 영향을 미치지 않았다.

털부처꽃(Lythrum Salicaria L.) 뿌리로부터 항산화 물질의 분리 및 구조동정 (Isolation and Identification of Antioxidant Compound from the Lythrum Salicaria L. Roots)

  • 털부처꽃(Lythrum salicaria L.) 뿌리는 실온에서 80% MeOH로 추출하고 이 추출물을 EtOAc 분획, n-BuOH 분획, $H_2O$ 분획으로 나누었다. 2,2-dipicryl-1-phenylhydrazyl (DPPH) radical 소거능을 이용한 항산화 활성을 측정하여 활성 추적 분획(Activity guided fractionation)에 따라 항산화 활성이 높은 EtOAc 분획에 대하여 silica gel 및 octadecyl $SiO_2$ column chromatography를 반복하여 항산화 활성을 나타내는 화합물 1과 major 화합물 2 및 3을 분리, 정제하였다. NMR, IR, 및 MS 등의 spectrum을 해석하여, Myricetin-3-O-${\beta}$-D-glucopyranoside (1), oleanolic acid (2) 및 betulinic acid (3)로 구조를 결정하였다. 화합물 1은 DPPH radical 소거능을 이용한 항산화 활성을 측정 한 결과 $25{\mu}g/mL$ 농도에서 $74.47{\pm}1.64%$의 저해 값을 가지는 것으로 나타났다.
