Background and objective: This study was conducted to find out the status of plant utilization in the current textbooks by analyzing the plants by grade and subject in the national textbooks for all elementary school grades in the 2015 revised curriculum in Korea. Methods: The data collected was analyzed using Microsoft Office Excel to obtain the frequency and ratio of collected plant data and SPSS for Windows 26.0 to determine learning content areas by grade and the R program was used to visualize the learning content areas. Results: A total of 232 species of plants were presented 1,047 times in the national textbooks. Based on an analysis of the plants presented by grade, the species that continued to increase in the lower grades tended to decrease in the fifth and sixth grades, the upper grades of elementary school. As for the number and frequency of plant species by subject, Korean Language had the highest number and frequency of plant species. The types of presentation of plants in textbooks were mainly text, followed by illustrations and photos of plants, which were largely used in first grade textbooks. In addition, as for the area of learning contents in which plants are used, in the lower grades, plants were used in the linguistic domain, and in the upper grades, in the botanical and environmental domains of the natural sciences. Herbaceous plants were presented more than woody plants, and according to an analysis of the plants based on the classification of crops, horticultural crops were presented the most, followed by food crops. Out of horticultural crops, flowering plants were found the most diversity with 63 species, but the plants that appeared most frequently were fruit trees that are commonly encountered in real life. Conclusion: As a result of this study, various plant species were included in elementary school textbooks, but most of them were horticultural crops encountered in real life depending on their use. Nevertheless, plant species with high frequency have continued a similar trend of frequency from the previous curriculums. Therefore, in the next curriculum, plant learning materials should be reflected according to social changes and students' preference for plants.
The purpose of this study was to understand the current status of education and programs and to identify the items that urban residents need most for garden management. First of all, there were 45 sites (40.5%) with gardens operated by city and county local governments nationwide, and 41(91.1%) of them had dedicated staff (1.8 persons on average). The average area of gardens was 18,623 m2, garden area per person was 20.27m2, the average number of participants was 683, and the average period of use was 8.69 months. In addition to gardening activities, 14 (31.1%) out of 45 sites were operating small group meetings, with an average of 2.29 meetings and 67 participants. In the satisfaction survey after gardening activities, 88.9% of 18 sites were satisfied. According to the questionnaire about education and programs related to garden users, an average of four sessions were conducted per education. In terms of education, the contents were in the following order: basic education on garden cultivation (33.9%) > prior education on garden operation (28.9%) > pest control (14.0%) > eco-friendly management (11.6%) > pesticides and Positive List System (9.9%) > others (1.7%). Over 95% of the respondents were generally satisfied. Regarding the perception survey on which items are needed to develop garden management guidelines, the most necessary items were in the order of crop management (38.7%) > public garden etiquettes among users (27.9%) > pest control (14.4%) > weed management (13.5%) > activities using harvest (5.4%). The contents that are to be included in the guidelines were in the order of garden planning and crop selection (17.2%) > cultivation techniques and schedule (16.5%) > pest and soil management (15.7%) > introduction of garden crops and gardening models (12.7%) > garden etiquettes (10.7%). In establishing urban garden management plans by region, the results will have high utility value as the basic data for continuous garden operation by setting a direction that meets the regional characteristics as well as the needs of urban residents.
Using the abundant available information about the tomato genome, we developed DNA markers that are linked to disease resistant loci and performed marker-assisted selection (MAS) to construct multi-disease resistant lines and varieties. Resistance markers of Ty-1, T2, and I2, which are linked to disease resistance to Tomato yellow leaf curl virus (TYLCV), Tomato mosaic virus (ToMV), and Fusarium wilt, respectively, were developed in a co-dominant fashion. DNA sequences near the resistance loci of TYLCV, ToMV, and Fusarium wilt were used for primer design. Reported candidate markers for powdery mildew-resistance were screened and the 32.5Cla marker was selected. All four markers (Ty-1, T2, I2, and 32.5Cla) were converted to cleavage amplification polymorphisms (CAPS) markers. Then, the CAPS markers were applied to 96 tomato lines to determine the phenetic relationships among the lines. This information yielded clusters of breeding lines illustrating the distribution of resistant and susceptible characters among lines. These data were utilized further in a MAS program for several generations, and a total of ten varieties and ten inbred lines were constructed. Among four traits, three were introduced to develop varieties and breeding lines through the MAS program; several cultivars possessed up to seven disease resistant traits. These resistant trait-related markers that were developed for the tomato MAS program could be used to select early stage seedlings, saving time and cost, and to construct multi-disease resistant lines and varieties.
During the course of a study to develop a spraying program at 15-day spray intervals, two important findings were identified allowing for further reduction of spray frequency by increasing the spray interval. In evaluating the contribution of fungicides from a 15-day spray interval program, control of white rot, which is of prime importance in Korea, was not affected, in spite of the extended spray interval caused by omitting the fungicides during the season. In another experiment assessing the duration of the protective activities of several key fungicides used in the 15-day spray interval program, infection control was maintained for almost 30 days for some fungicide. Based on these two findings, a basic spraying program with a 25-day spray interval was developed. This program was modified for four successive years to improve the control efficacy against bitter rot and Marssonina blotch, which sometimes causes as much damage as white rot.
본 연구에서는 폴리스틸렌 재질의 96-well 마이크로플레이트와 ELISA 판독기를 이용하여 고추의 카로티노이드 함량을 간편하게 분석할 수 있는 방법을 개발하고자 하였다. 분광광도계를 이용하지 않고 450nm 필터가 장착된 ELISA 판독기로도 카로티노이드 분석이 가능하였으나, 카로티노이드 추출 시 사용하는 아세톤으로 인해 마이크로플레이트의 표면이 불투명하게 변색되었다. 그러나 아세톤 색소추출물을 메탄올로 10배 희석한 용액을 사용할 경우, 플레이트 표면과 용액 내 카로티노이드 성분의 변화 없이 카로티노이드 측정이 가능하였고, ASTA-20.1법으로 측정된 결과와도 높은 상관관계를 보였다. 한편, 카로티노이드 추출 조건으로는 고춧가루 0.1g을 아세톤 10mL에 첨가하고 실온에서 4시간 동안 교반하는 것이 적합하였다. 본 실험의 ELISA 판독기를 이용한 색소분석법(Microplate법으로 명명)은 대량의 시료를 처리할 수 있을 뿐만 아니라 아세톤 폐액의 발생량을 ASTA-20.1법의 1/10 수준으로 줄일 수 있어 실제 고추 육종에서 고색소 계통을 선발할 때 매우 유용할 것으로 판단된다.
국내에 보유하고 있는 여주 유전자원을 대상으로 과실특성과 charantin 함량을 분석하고, 추후 유전자원을 다양한 용도로 활용하고자 수행하였다. 2010년에 선발된 수량이 높은 계통 2점의 charantin 함량을 HPLC 분석한 결과, 착과 후 일수에 따른 함량의 변이를 보였는데, BG1은 착과 후 24일 15일순으로 charantin 함량이 높게 나타나 착과 후반부에 charantin 함량이 높게 나타난 반면 BG7은 착과 후 일수가 13일에 가장 높았고, 16일 19일 순으로 charantin 함량이 높게 나타나 착과 전반부에 charantin 함량이 높아지는 것으로 나타났다. Charantin 성분 함량이 높은 여주품종 선발 및 수확시기 규명을 위하여 한국, 중국, 인도, 일본 등에서 도입한 유전자원 33점과 시판종 1점중 수량이 높은 13점의 과실특성 및 charantin 함량을 HPLC 분석한 결과 charantin 함량이 높은 104615, K169995, 엔에스454을 선발하였다. 따라서 수량이 높은 13점의 여주자원중 charantin 함량이 높았던 K169995나 NS454은 새로운 품종 육성이나 가공용으로 활용할 수 있으며, 수량이 많았던 K161952나 K051434 등은 과실 생산 목적으로 활용할 수 있을 것으로 생각되고, 착과후 시기별로 용도에 따라 수확을 하여 이용하면 좋을 것으로 생각된다.
본 연구는 공동생활시설에서 거주하는 지적장애인을 대상으로 원예치료가 일상생활동작 및 대인관계 향상에 미치는 영향에 대해 알아보고자 하였다. 원예치료대상자는 제주시 용담동 J시설에 거주하는 지적장애인 대조군 8명, 실험군 8명 이었다. 원예치료 프로그램은 2009년 3월부터 2009년 7월 중순까지 주 1회 2시간씩 총 20회 실시하였다. 일상생활동작 평가 결과 대조군의 경우 식사동작을 제외하고 모든 항목의 기능이 실험기간 동안 변화가 없거나 오히려 떨어졌다. 실험군의 경우 대조구와는 반대로 원예치료에 의해서 이동동작을 제외한 일상생활동작평가의 모든 항목이 유의하게 향상되었다. 대인관계 평가 결과 대조군은 실험 전 42.25점에서 실험 후에도 42.25점으로 변화가 없었으나, 실험군의 경우 대조구와는 달리 실험 전 41.75점에서 실험 후 50.37점으로 8.62점이 증가하여 99% 수준에서 고도로 유의한 변화를 보였다. 그룹활동 평가 결과 신체적 지각적 능력, 사회적 교류, 인지능력, 감정상태, 직업적 관심이 통계적으로 고도로 유의하게(99%) 향상되었다. 이상의 결과로부터 원예치료 프로그램이 공동 생활시설내 지적 장애인들의 일상생활동작 및 대인관계 향상에 효과적인 것으로 나타났다.
2020년 전라북도 완주 소재 약용작물 전시포장에서 재배중인 범부채(Iris domestica) 140개체 가운데 3개체의 잎에서 괴사증상이 발견되었다. Reverse transcription polymerase chain reaction 검정결과 증상을 나타내는 3개체가 tomato spotted wilt virus (TSWV)에 감염된 것으로 확인되었다. 증상주로부터 분리한 TSWV 분리주 'Blackberry lily-kr1'의 전체 염기서열을 결정하였으며, 유전자 은행에 있는 다른 분리주들과 L, M, S 분절유전체 부위의 염기서열 상동성을 비교하였다. 'Blackberry lily-kr1' 분리주는 L 분절 유전체는 우리나라에서 보고한 'JJ' 분리주(MF159046) 또는 'HJ' (LC273305) 분리주와 상동성이 높았으며, M과 S 분절 유전체는 'JJ' 분리주(MF159058과 KY021439)와 가장 상동성이 높았다. MEGA X 프로그램의 maximum likelihood 방법을 이용하여 'Blackberry lily-kr1'의 다른 TSWV 분리주들과의 계통학적 연관성에 관한 분석을 한 결과 L, M, S 분절유전체 모두 고추에서 분리한 'JJ' 분리주 및 환삼덩굴(Humulus japonicas)에서 분리한 'HJ' 분리주와 높은 연관성을 보였다. 건전한 I. domestica 식물에 'Blackberry lily-kr1'을 감염시킨 Nicoatiana rustica 식물 즙액을 접종한 결과 50일 후에 잎에 괴사 또는 이중 고리형태의 괴사 증상이 형성되었다. 이는 노지에서 재배하는 범부채에서 관찰된 괴사증상이 TSWV 감염에 의한 것임을 시사하는 결과이다. 이 연구는 우리나라에서 범부채에서 TSWV 발생에 관한 최초의 보고이다.
