Effects of Horse Meat Hydrolysate on Oxidative Stress, Proinflammatory Cytokines, and the Ubiquitin-Proteasomal System of C2C12 Cells

  • Sarcopenia, the age-related muscle atrophy, is a serious concern as it is associated with frailty, reduced physical functions, and increased mortality risk. Protein supplementation is essential for preserving muscle mass, and horse meat can be an excellent source of proteins. Since sarcopenia occurs under conditions of oxidative stress, this study aimed to investigate the potential anti-muscle atrophy effect of horse meat hydrolysate using C2C12 cells. A horse meat hydrolysate less than 3 kDa (A4<3kDa) significantly increased the viability of C2C12 myoblasts against H2O2-induced cytotoxicity. Exposure of C2C12 myoblasts to lipopolysaccharide led to an elevation of cellular reactive oxygen species levels and mRNA expression of proinflammatory cytokines, including tumor necrosis factor-α and interleukin 6, and these effects were attenuated by A4<3kDa treatment. Additionally, A4<3kDa activated protein synthesis-related proteins through the protein kinase B/mechanistic target of rapamycin pathway, while decreasing the expression of activity and degradation-related proteins, such as Forkhead box O3, muscle RING finger protein-1, and Atrogin-1 in dexamethasone-treated C2C12 myotubes. Therefore, the natural material A4<3kDa has the potential of protecting against muscle atrophy, while further in vivo study is needed.

The Differences in Chemical Composition, Physical Quality Traits and Nutritional Values of Horse Meat as Affected by Various Retail Cut Types

  • The effects of retail cut type on chemical, quality and nutritional characteristics of horse meat were studied. Jeju female breed horses (n = 9) at 32-mo-old were slaughtered and the carcasses at 24 h post-mortem were fabricated into 10 retail cuts including: tender-loin, loin, strip-loin, shoulder-chuck-roll, shoulder-clod, top-round, outside-round, brisket, short-plate-brisket, and shank. The results revealed that all of parameters (chemical, meat quality and nutritional composition) examined significantly (p<0.05) differed between the cuts. The chemical composition range (minimum to maximum) of cuts was found as such: moisture 65.06% to 71.69%; protein 19.07% to 21.28%; collagen 1.40% to 2.45%; fat 2.56% to 12.14% and cholesterol 55.76 to 79.50 mg/100 g. Shoulder-chuck-roll had the highest pH and water-holding capacity, while top-round had the highest cooking loss. Shear force ranged between the cuts from $2.80kg/cm^2$ to $4.98kg/cm^2$. The Cu, Fe, and Zn contents ranged between the cuts from 1.52 mg/kg to 2.75 mg/kg, 21.25 mg/kg to 30.85 mg/kg, and 16.51 mg/kg to 40.42 mg/kg, respectively. Additionally, most of the cuts studied showed favorable polyunsaturated fatty acid/saturated fatty acid, n-3/n-6 and essential amino acid/non-essential amino acid ratios.

  • A quick and reliable method for identifying meat origin is developed to ensure species origin of livestock products for consumers. The present study examined the identification of meat sources (duck, chicken, goat, deer, pig, cattle, sheep, and horse) using PCR by exploiting the mitochondrial 12S rRNA and mitochondrial cytochrome b genes. Species-specific primers were designed for some or all mitochondrial 12S rRNA nucleotide sequences to identify meat samples from duck, chicken, goat, and deer. Mitochondrial cytochrome b genes from pig, cattle, sheep, and horse were used to construct species-specific primers, which were used to amplify DNA from different meat samples. Primer sets developed in this study were found to be superior for detecting meat origin when compared to other available methods, for which the discrimination of meat origin was not equally applicable in some cases. Our new development of species-specific primer sets could be multiplexed in a single PCR reaction to significantly reduce the time and labor required for determining meat samples of unknown origin from the 8 species. Therefore, the technique developed in this study can be used efficiently to trace the meat origin in a commercial venture and help consumers to preserve their rights knowing origin of meat products for social, religious or health consciousness.

Genetic diversity and relationship of Halla horse based on polymorphisms in microsatellites

  • Halla horse is crossbreed between Jeju and Thoroughbred horses and is used for riding, racing and meat production. Thus, molecular genetic studies are needed to establish and preserve the industrially valuable Halla horses. This study aimed to analyses the genetic diversity and population structure through 12 microsatellite (MS) markers for Halla and putatively related 3 breeds (Jeju, Mongolian and Thoroughbred horses). On average, the number of alleles, observed heterozygosity (Hobs), expected heterozygosity (Hexp), and polymorphic information content (PIC) among all horses were 10, 0.767, 0.799, and 0.771, respectively. Neighbor-joining tree and STRUCTURE analysis showed that Halla horses were between Thoroughbred and Jeju horses, tend to more influenced by Thoroughbred horses. Therefore, these results could be considered for use as the basic genetic breed relationships resource among the horse breeds (Jeju, Mongolian, and Thoroughbred horses) related to the origins of the Halla horse.

말고기의 육질 등급에 따른 부위별 일반성분, 이화학적 특성 및 지방산 성분 비교 (Comparison of Chemical Composition, Physico-chemical Properties and Fatty Acid Composition of Horse Meat by Different Grade and Cuts)

  • 본 연구는 말고기 등급(1등급과 2등급)에 따른 품질을 비교하여 향후 부위별 및 등급별 구분 판매시 등급간 육질 특성의 기초자료로 활용하기 위해 이화학적, 관능적, 지방산조성 분석을 실시하였다. 등심에 대한 일반성분분석 결과에서는 1등급이 2등급에 비해 높은(p<0.05) 조지방 함량을 보인 반면, 전단력은 1등급이 5.87 ($kg/cm^2$)으로 2등급 10.86 ($kg/cm^2$)에 비해 유의적으로 낮았다(p<0.05). 육질등급별 등심, 목심, 넓적다리에 대한 보수력 측정결과 등급간 및 부위간에 유의적 차이가 없었고(p>0.05), 육색에 대한 육질등급별 부위간 비교에서는 $L^*$값, $a^*$값 및 $b^*$값에서 차이가 없었으나, 2등급의 경우 등심, 목심, 넓적다리의 $L^*$, $a^*$$b^*$값은 유의적 차이가 있는 것으로 나타났다(p<0.05). 지방산 조성에 대한 분석결과 등심에서 palmitoleic acid (C16:1)는 1등급이 11.39%, 2등급이 5.36%으로 1등급이 2등급에 비해 유의적으로 높았다(p<0.05). 반면 stearic acid (C18:0)는 1등급이 2등급에 비해 유의적으로 낮게 나타났으나(p>0.05), oleic acid (C18:1)는 등급간에 유의적 차이(p<0.05)가 없는 것으로 나타났다. 관능검사결과 육질 등급간에는 전체적인 기호도는 유의적인 차이가 없는 것으로 나타났다(p>0.05). 이상의 결과에서 냉동된 말고기의 육질 1등급과 2등급에서는 조지방 함량, 전단력, palmitoleic acid (C16:1)와 stearic acid (C18:0)는 등급별 육질을 판별할 수 있는 요소로 이용이 가능할 것으로 사료된다. 또한 품질을 향상시키고 부위 및 등급에 따른 차별화를 위해서는 40개월 이상의 장기간 사육은 바람직하지 않을 것으로 사료된다.

Cervus nippon yesoensis (Japanese Yeso Deer) 사슴육에서의 색소 및 색소 안정성 (Color and Its Stability in Venison from Cervus nippon yesoensis(Japanese Yeso Deer))

  • Color and its stability in venison, longissumus dorsi (LD) and quadriceps femoris (QF) muscles, from 8 wild Cervus nippon yesoensis (Japanese Yeso Deer) were investigated by means of the CIE L$^{*}a{*}b{*}$ measurement and autoxidation rate recorded using partially purified myoglobin. It was observed a common feature of the change of three mean values($L^{*}$, $a^{*}$ and $b^{*}$) in both LD and QF that mean value increased at 1 or 2 day post-mortem and then decreased during storage. The differences between 1 and 7 days was the largest in $a^{*}$ value than those in $L^{*}$ and $b^{*}$ values. The mean differences among storage days were only significant in $a^{*}$ except for $b^{*}$ of LD. It was same tendency that the mean difference of CIE $L^{*}$, $a^{*}$ and $b^{*}$ values during refrigerator storage was larger in $a^{*}$ than both in $L^{*}$ and $b^{*}$ reported in beef(Sekikawa et al., 1995) and venison(Stevenson et al., 1989) during storage. The smaller $a^{*}$ value was indicated that bright red of meat changed to dull red, brown red causing met-Mb formation. To compare of color stability with respects to the Mb autoxidation rate, we measured this rate of deer and horse muscles, because horse Mb was considered to have the fastest autoxidation rate among domestic animals, and we used crude Mb and pH 6.0, which might be reflected to the intact meat. Mean value of the autoxidation rate measured in this study in deer was 0.037 and that was 0.026 in horse(sigma). Although there was no significant mean difference and were different Mb purity between deer(A409/A 280 nm = 4.0) and horse(5.6), in generally Mb purity was the higher and the faster autoxidation rate, but this rate in deer was faster than in horse. These results might indicate that venison meat discolors at faster rate compared with beef.

Identification of Meat Species Using PCR-RFLP Marker of Cytochrome b Gene

  • Food labeling regulations require that the meat species in various meat products are accurately declared to the consumer. Substitution or adulteration of costly meat with a cheaper one is one of the most common problems in the meat industry. In this study, PCR-restriction fragment length polymorphism(RFLP) method of the mitochondrial cytochrome b(mt cyt b) gene has been applied for identification of the origin of six mammalian meat species(beef, port horse, goat, mutton and deer) and three poultry meat species(chicken, turkey and duck) as raw materials for meat products. PCR was used to amplify a variable region of mt cyt b gene. Meat species differentiation was determined by digestion of the amplified products with a 359 bp fragment using HaeIII and HinfI restriction enzymes, which generated species-specific RFLP patterns. This PCR-RFLP DNA marker of mt cyt b gene could be very useful for the accurate and reliable identification and discrimination of animal meat species in routine analysis.

Colour Changes in Meat of Foals as Affected by Slaughtering Age and Post-thawing Time

  • The aim of the present work was to investigate how colour changes of foal meat can vary after thawing out in relation to the slaughtering age of the horses and to the post-thawing time. Eighteen Italian Heavy Draught Horse (IHDH) foals were used for the trial. They were subdivided in three groups according to their slaughtering age (6, 11 and 18 months). Two different surfaces were investigated for each sample: a fresh cut surface (daily renewed cutting surface: DRCS), and not-renewed cutting surface (NRCS). The redness of both investigated surfaces increased with slaughtering age (p<0.01). Moreover, this parameter decreased during post-thawing time (p<0.01) only on the NRCS, probably due to the myoglobin oxidation processes. Colour is an important visual cue denoting perceived quality by consumers. So, by a chromatic perspective the thawed meat of IHDH foals slaughtered at 6 and 11 months proved to be that which best meets the market requirements.

숙성이 제주마 등심의 육질과 관능적 특성에 미치는 영향 (Meat Quality and Sensory Characteristics in Longissimus Muscle of Jeju Horse as Influenced by Ageing)

  • 연도증진의 가장 일반적인 방법인 숙성을 통해 말고기의 육질과 관능적 특성이 어떻게 변화하는지를 조사하여 말고기의 적정 숙성기간을 제시하고, 국내 말고기의 냉장 유통에 대비한 기초자료를 확보하고자 제주마 24두의 등심을 4℃에서 25일간 숙성시키면서 육질과 관능적 특성을 조사하였다. 말고기 pH는 숙성기간에 따라 차이가 없었으며, 전단력은 저장 12일까지 유의적으로 감소하다가(P<0.05) 12일 이후 차이를 나타내지 않았다. 보수력, 가열감량 및 포장감량은 숙성기간이 경과할수록 증가하는 경향이였으며, 육색특성은 숙성기간이 경과할수록 명도(L*)와 적색도(a*) 값이 증가하는 경향을 보였다. 관능적 특성 중 풍미는 저장 7일에 가장 높은 수치를 나타내었고, 다즙성은 숙성 12일에 패널에 의해 가장 좋은 점수를 받았으며, 연도는 숙성 7일에 유의적인 증가를 보였으며(P<0.05) 이후 숙성기간에는 차이가 없었다. 숙성기간별 말고기에 대한 전체기호도는 숙성 7일에 유의적인 증가를 보인 후 숙성 18일까지 지속하다가 25일에 다소 감소하였다(P<0.05). 이상의 결과로 볼 때 말고기 등심의 최적숙성기간은 도축 후 9~14일로 나타났으며, 도축 후 20일 이후에는 기호성과 육질이 저하되는 결과를 보였다.

Meat Species Identification using Loop-mediated Isothermal Amplification Assay Targeting Species-specific Mitochondrial DNA

  • Meat source fraud and adulteration scandals have led to consumer demands for accurate meat identification methods. Nucleotide amplification assays have been proposed as an alternative method to protein-based assays for meat identification. In this study, we designed Loop-mediated isothermal amplification (LAMP) assays targeting species-specific mitochondrial DNA to identify and discriminate eight meat species; cattle, pig, horse, goat, sheep, chicken, duck, and turkey. The LAMP primer sets were designed and the target genes were discriminated according to their unique annealing temperature generated by annealing curve analysis. Their unique annealing temperatures were found to be $85.56{\pm}0.07^{\circ}C$ for cattle, $84.96{\pm}0.08^{\circ}C$ for pig, and $85.99{\pm}0.05^{\circ}C$ for horse in the BSE-LAMP set (Bos taurus, Sus scrofa domesticus and Equus caballus); $84.91{\pm}0.11^{\circ}C$ for goat and $83.90{\pm}0.11^{\circ}C$ for sheep in the CO-LAMP set (Capra hircus and Ovis aries); and $86.31{\pm}0.23^{\circ}C$ for chicken, $88.66{\pm}0.12^{\circ}C$ for duck, and $84.49{\pm}0.08^{\circ}C$ for turkey in the GAM-LAMP set (Gallus gallus, Anas platyrhynchos and Meleagris gallopavo). No cross-reactivity was observed in each set. The limits of detection (LODs) of the LAMP assays in raw and cooked meat were determined from $10pg/{\mu}L$ to $100fg/{\mu}L$ levels, and LODs in raw and cooked meat admixtures were determined from 0.01% to 0.0001% levels. The assays were performed within 30 min and showed greater sensitivity than that of the PCR assays. These novel LAMP assays provide a simple, rapid, accurate, and sensitive technology for discrimination of eight meat species.
