Effects of Horse Bone Powder Extract on Longitudinal Bone Growth in Adolescent Male Rats (마골(馬骨) 추출물의 성장기 흰쥐 장골 길이 성장에 대한 효과)

  • Objectives : This study was designed to investigate the effects of horse bone powder extract on the growth of longitudinal bone in adolescent male rats. Methods: Longitudinal bone growth was measured by fluorescent microscopy. To examine the effects on the growth plate metabolism, the total height of growth plate, the induction of local insulin-like growth factor-1 (IGF-1), and bone morphogenetic protein-2 (BMP-2) were measured. Results: Horse bone powder extract enhanced longitudinal bone growth and total height of the growth plate. Also, it promoted the induction of local IGF-1 and BMP-2 of the growth plate. Conclusions: This study shows that the horse bone powder extract effects longitudinal bone growth in adolescent rats and might be used for both stunted adolescents and inherent growth failure patients.

Effects of Horse Bone Extracts on the Induced Postmenopausal Osteoporosis in Rats (백서의 폐경기 골다공증 모델에서 말뼈 추출물의 효과)

  • Osteoporosis is a metabolic bone disease associated with an imbalance of bone remodeling. Osteoporosis is characterized by decreased bone mass and increased bone fractures. In this study, we investigated the effects of horse bone extracts (HBEs) in vivo. Horse bone was extracted with 80% alcohol (HBE-A) at $100^{\circ}C$ or water (HBE-W) at $120^{\circ}C$. Animal model of postmenopausal osteoporosis was used, in which osteoporosis was induced by ovariectomy of female S.D. rats (female rats were divided into 5 groups), and HBEs were administered to ovariectomized rats every day for 8 weeks. After 8 weeks, the rats were sacrificed and the following osteoporotic factors were measured: body weight, bone mineral density (BMD), uterine/body weight ratio, serum estradiol (E2), and serum alkaline phosphatase (ALP). The results showed that the administration of HBE-W decreased the changes of body weight in ovariectomized rats. HBE-W increased the uterine/body weight ratio and BMD. In addition, HBEs decreased the ALP. Therefore, HBEs may be used for the prevention or treatment of bone disease.

Nutritional Composition in Bone Extracts from Jeju Crossbred Horses at Different Slaughter Ages

  • The effects of slaughter age (28, 32, or 38 mon) on the proximate composition, collagen content, fatty acid composition, amino acid content, and mineral contents of horse leg bone (femur and tibia) extracts (HLBE) obtained from Jeju crossbred horses (n=14) in Korea. HLBE was extracted for 24 h with boiling water. At those ages, the respective proximate compositions of the HLBE were found as follows: 5.20-6.42% crude protein; fat 0.61-1.65% crude; and 0.10-0.22% crude ash. At 32 and 38 mon, the HLBE showed higher levels of both crude protein and collagen than at 28 mon. The major fatty acids of the horse leg bone extract powder (HLBP) were palmitic acid and oleic acid. Palmitoleic acid and the essential fatty acids were higher in the HLBP at 38 mon compared to that at 28 mon. Nearly all the amino acids were found at higher levels in the HLBP at 38 mon than at 28 mon, except histidine. The P, K, Zn, Se and Fe contents in the HLBP increased significantly with age. These results suggest that some nutrients in the HLBE increase with age, and that extracts from horses older than 32 mon would be more nutritious for human consumers.

In vitro Anti-oxidative and Anti-inflammatory Activities of Horse-bone Extract via Up-regulation of Heme-oxygenase 1 (말뼈추출물의 Hemeoxygenase-1의 발현 조절을 통한 시험관내 항염증 효과)

  • Few studies have been reported that horse-bone extract(HBE) can prevent and treatment of bone diseases. However, HBE' therapeutic activities are still not fully understood. This study determined whether HBE up-regulates hemeoxygenase 1(HO-1) and this mediates its anti-inflammatory effect in murine macrophages.Nitric oxide(NO) assay, MTT assay and DPPH assay were performed. In addition, Western blotting and real time PCR were used to determine protein expression, and gene expression, respectively. HBE significantly inhibited NO production without observable cytotoxicity. In addition, HBE attenuated inducible nitric oxide synthase (iNOS), cyclooxygenase-2(COX-2) and phospho (p)-ERK protein expressions in LPS(0.1㎍/ml) stimulated RAW264.7 cells. On the other hand, HBE alone up-regulated HO-1 and Nrf-2 expressions, which mediated HBE's anti-inflammatory effect in RAW264.7 cells. Finally, HBE up-regulated HO-1 and impaired ERK1/2 signaling pathways, and thus it may provide protection against cellular oxidation and inflammation.
