Production and Molecular Cytogenetic Identification of Wheat-Barley Hybrids and Translocations

  • New winter wheat winter barley hybrids were produced (Mv9 kr1 Igri, Mv9 kr1 Osnova, Asakazekomugi Manas). The wheat-barley hybrids showed entire male sterility and were multiplied in tissue culture. Chromosome configurations were studied with GISH in meiosis in the Mv9 krl x Igri hybrid and in its progenies multiplied in vitro. Chromosome pairing between wheat and barley has been observed in some cells in the hybrids multiplied in vitro. Backcross plants with 43 and 44 chromosomes were selected with the aim of developing new wheat-barley addition lines. Wheat-barley translocations were demonstrated with GISH in backcross progenies originating from in vitro regenerated wheat (Triticum aestivum L. cv. Chinese Spring) x barley (Hordeum vulgare L. cv. Betzes) hybrids. Five different translocations were observed. Sequential N-banding and GISH analyses were performed to further identify the translocations. The N-banding pattern of the Robertsonian translocation suggests that this chromosome consists of the short arm of barley chromosome 4H translocated to the long arm of 2B of wheat. Plants with four different homozygous translocations were selected from the following BC2F3 generation.

Molecular Analysis of Korean Isolate of Barley mild mosaic virus (Iks Isolate)

  • The complete nucleotide sequences of both RNA of an isolated Barley mild mosaic virus (BaMMV) from Iksan, Korea, have been determined. RNA1 was 7273 nucleotides long and encodes for a polyprotein of 2261 amino acids, which contains the eight putative functional proteins. RNA2 was 3520 nucleotides long and encodes for a polyprotein of 894 amino acids, which contains two functional proteins. Results of multiple sequence alignment showed 92.9% similarity with Na1 isolate, followed by Sil, UK(F), Asl1, Remis M and UK(M) isolates, respectively. Comparison of the BaMMV-Iks polyproteins with the corresponding proteins of BaMMV-Na1 isolates showed 95% amino acid sequence identity. The phylogenetic analysis revealed that Iks isolate was closely related to Na1 strain and have a common origin.

Effect of Nanobubbles on the Seed Germination (초미세버블이 종자발아에 미치는 영향)

  • For the last few decades, much research has been tried to improve crop’s characteristics such as crop yield, quality, seeding period and environmental adaptability. In this paper, the effect of nanobubbles on the germination rate of barley seed is experimentally investigated. The air nanobubble was generated in water by gas-liquid mixing method. The results were shown that the mean diameter and concentration of nanobubble fabricated in DI water are 133 ㎚ and 8.59✕108 particles/㎖, respectively. Also, the seed germination rate for the nanobubble water was approximately 40 % higher than that of pure water.

  • Prolamins are the main seed storage proteins in cereals. Gluten proteins seem to be prolamins because their primary structure have the meaningful quantity of proline and glutamine amino acid residues. Gluten proteins are found in crops such as wheat (Triticum aestivum), barley (Hordeum vulgare), and rye (Secale cereale) which are major food crops in the Triticeae tribe. Glutenin and gliadin, hordein, and secalin are typical gluten proteins found in wheat, barley, and rye, respectively. Gluten affect grain quality so that many researches, such as isolation or characterization of their genes, have been carried out. To improve the quality of grains in the Triticeae tribe, it is necessary to understand the relationship within their gluten proteins and their evolutionary changes. The sequences of nucleotides and amino acids of gluten protein including glutenins, gliadins, hordeins, and secalins were retrieved from NCBI (https://www.ncbi.nlm.nih.gov/) and Uniprot (http://www.uniprot.org/). The sequence analysis and the phylogenetic analysis of gluten proteins were performed with various website tools. The results demonstrated that gluten proteins were grouped with their homology and were mostly corresponded with the previous reports. However, some genes were moved, duplicated, or disappeared as evolutionary process. The obtained data will encourage the breeding programs of wheat, barley, rye, and other crops in the Triticeae tribe.

Accumulation of Transcripts Abundance after Barley Inoculation with Cochliobolus sativus

  • Spot blotch caused by the hemibiotrophic pathogen Cochliobolus sativus has been the major yield-reducing factor for barley production during the last decade. Monitoring transcriptional reorganization triggered in response to this fungus is an essential first step for the functional analysis of genes involved in the process. To characterize the defense responses initiated by barley resistant and susceptible cultivars, a survey of transcript abundance at early time points of C. sativus inoculation was conducted. A notable number of transcripts exhibiting significant differential accumulations in the resistant and susceptible cultivars were detected compared to the non-inoculated controls. At the p-value of 0.0001, transcripts were divided into three general categories; defense, regulatory and unknown function, and the resistant cultivar had the greatest number of common transcripts at different time points. Quantities of differentially accumulated gene transcripts in both cultivars were identified at 24 h post infection, the approximate time when the pathogen changes trophic lifestyles. The unique and common accumulated transcripts might be of considerable interest for enhancing effective resistance to C. sativus.

A Gene Encoding $\beta$-amylase from Saprolegnia parasitica and Its Expression in Saccharomyces cerevisiae

  • The ${\beta}$-Amylase cDNA fragment from the oomcete Saprolegnia parasitica was cloned by reverse transcription-polymerase chain reaction (RT-PCR) using degenerate oligonucleotide primers derived from conserved ${\beta}$-amylase sequences. The 5'and 3'regions of the $\beta$-amylase gene were amplified using the rapid amplification of cDNA ends (rACE) system. It consisted of an open reading frame of 1,350 bp for a protein of 450 amino acids. Comparison between the genomic and cDNA sequences revealed that the intron was not present in the coding region. The deduced amino acid sequence of the ${\beta}$-amylase gene had a 97% similarity to the ${\beta}$-amylase of Saprolegnia ferax, followed by 41% similarity to those of Arabidopsis thaliana, Hordeum vulgare, and Zea mays. The ${\beta}$-amylase gene was also expressed in Saccharomyces cerevisiae by placing it under the control of the alcohol dehydrogenase gene (ADC1) promoter.

Screening and Characterization of $\alpha$-Amylase Inhibitors from Cereals and Legumes in Korea (한국산 곡류와 두류 중 $\alpha$-Amylase 저해물질의 검색 및 특성)

  • To investigate characterization of the ${\alpha}$-amylase inhibitors from cereals and legumes produced in Korea, inhibitory activities against ${\alpha}$-amylase with the inhibitor from barley(Hordeum vulgare), wheat(Triticum aestivun), black bean(Glycine max), bean(Cajanus cajon) and pea(Pisum sativum) were measured. Among the samples tested, inhibitors from naked barley and black bean(sabong) which showed the highest inhibitor activities of cereals and legumes, respectively, were characterized according to treatment condition. The results obtained were summarized as follows. During the germination of naked barley and black bean, ${\alpha}$-amylase activities were gradually increased but inhibitory activities against ${\alpha}$-amylases were decreased. Both activities were gradually decreased when naked barley and black bean were stored. More than 50% of activities of the inhibitors from naked barley and black bean were remained at 100$^{\circ}C$ for 15 min and 20 min, respectively, indicating that the inhibitor from black bean was more stable to heat than that of barley.

The Effects of Sulfite on the Greening of Etiolated Barley (Mordeum vuigare L.) Seedling (Sulfite가 보리(Hordeum vulgare L.) 유식물의 녹화에 미치는 영향)

  • To investigate the effects of sulfite on the chloroplast development, etiolated barley seedlings were treated with 100 mM sulfite solution every 3 hour by spraying during 96 hours greening Period. The effects were determined by chlorophyll a, b and carotenoids contents, photosynthetic electron transport activity, chlorophyll fluorescence yield and fluorescence quenching parameters. The contents of chlorophyll a and carotenoids were decreased than that of control by treatment of salfite over 48 hours greening. PS II Is more sensitive to sulfite than PS I Is. And by the addition of DPC to the chloroplasts of the barley seedling treated with sulfite, the photoreduction of DCPIP was not recovered. In greening with suite treated barley leaves, Fo, Fv and Nlh ratio were decreased with little difference from that of control. But qP, qNP and qR were lowed in comparison with those of controls whereas qE was markedly higher than that of control. Especially, It is Interesting that qR was decreased markedly compared to that of control. The results in the change of PS I activity, Nf and qP suggest that the strate of Inhibition by suite Is carbon dioxide reduction cycle.

Phytotoxic Effect of Lettuce (Lactuca sativa L.) Leaf Extract on Seedling Growth of Crops and Weeds

  • Lettuce (Lactuca sativa L.) is known to contain water-soluble substances that are biologically active. Aqueous or methanol extracts and residues from leaves of lettuce plants were assayed to determine their allelopathic effects, and the causative allelochemicals from fractions were quantified by means of HPLC analysis and bioassayed. Extracts from oven-dried leaf samples were more phytotoxic than those from freeze-dried samples. Leaf extracts of 40 g L$^{-1}$ were completely inhibitory on root growth of alfalfa (Medicago sativa L.), while root growths of barley (Hordeum vulgare L.) and soybean (Glycine max L.) were less sensitive. Early seedling growth of both alfalfa and barnyard grass (Echinochloa crus-galli) was significantly reduced by methanol leaf extracts. The major allelopathic substances analyzed by HPLC were coumarin, trans-cinnamic acid, o-coumaric acid, p-coumaric acid and chlorogenic acid. Of them p-coumaric acid was found as the greatest amount (8.9 mg 100 g$^{-1}$ ) in the EtOAc fraction; only coumarin was found in all the fractions. Hexane and EtOAc fractions of L. sativa reduced alfalfa root growth more than did BuOH and water fractions. These results suggest that lettuce had potent herbicidal activity, and that its activity differed depending on type and amount of causative compounds by fraction.

Barley Haploid Production Using Interspecific Crosses between Hordeum vulgare and H. bulbosum (야생종 H. bulbosum을 이용한 보리 반수체 육성)

  • The experiments were conducted to establish the effective barley haploid production system using interspecific crosses. Three spring barley cultivars. Bruce. Klages and Rodeo were used for this experiment. 1, 687 florets of three barley cultivars were crossed with bulbosum pollens. 1, 079 seeds were harvested and obtained 834 embryos so that seed set rate and embryo production rate were 64% and 77%, respectively. IAA effect was superior to NAA for root development and 1 ppm concentration of IAA gave the best result among five concentrations; 0ppm, 1ppm., 5ppm, 10ppm and 30ppm.
