  • Objective: This research investigated the effect of administering chromium (Cr) and meloxicam (MEL) on growth performance, cortisol and blood metabolite, and behaviors in young, regrouped heifers. Methods: Fifty Holstein dairy heifers (body weight [BW] 198±32.7 kg and 6.5±0.82 months of age) were randomly assigned to non-regrouped group or four regrouped groups. Non-regrouped animals were held in the same pen throughout the entire experimental period (NL: non-regrouping and administration of lactose monohydrate [LM; placebo]). For regrouping groups, two or three heifers maintained in four different pens for 2 weeks were regrouped into a new pen and assigned to one of four groups: regrouping and LM administration (RL); regrouping and Cr administration (RC); regrouping and MEL administration (RM), and regrouping and Cr and MEL administration (RCM). LM (1 mg/kg BW), Cr (0.5 mg Cr picolinate/kg dry matter intake), and MEL (1 mg/kg BW) were orally administered immediately before regrouping. Blood was collected before regrouping (0 h) and at 3, 9, and 24 h and 7 and 14 d thereafter. Behaviors were recorded for 7 consecutive days after regrouping. Results: Average daily gain was lower (p<0.05) in RL than NL heifers, but was higher (p<0.05) in RM, RC, and RCM than RL heifers. RL heifers had higher (p<0.05) cortisol than NL heifers on d 1 after regrouping. The cortisol concentrations in RC, RM, and RCM groups were lower (p<0.05) than in RL treatment 1 d after regrouping. Displacement behavior was greater (p<0.05) in RL group than all other groups at 2, 3, and 6 d after regrouping. Conclusion: Regrouping caused temporal stress, reduced growth performance, and increased displacement behavior in heifers. Administering Cr and MEL recovered the retarded growth rate and reduced displacement behavior, thereby alleviating regrouping stress.

  • Three fistulated nonlactating Holstein cows were used to examine the change in ruminal characteristics when fed diets of 100% corn silage, 50% corn silage + 50% wet brewers' grain(WBG) and 100% WBG, and also twenty Holstein heifers averaging 156 $\PM$ 3.2kg BW were randomly assingned to determine the adequate feeding level of concentrate with the following treatments ; Only WBG, concentrate to 0.5, 1.0 and 1.5% of BW under feeding WBG ad libitum. Tne main results were as follows : I. Ruminal pH was minimized 3 hrs after feeding and then gradually increased. The mean ruminal pH was highest in feeding only WBG (6.72) and lowest in only corn silage(6.0) 2. Ruminal $NH_3$-N concentration was maximized 1 hr after feeding and then gradually decreased regardless of diets, n7e average ruminal $NH_3$-N content was highest in diet of 100% WBG(12.9 rngldl) and lowest in 100% corn silage(5.9 mg/dl) 3. Acetic acid percent among VFA was highest when WBG replaced 50% corn silage and acetic to propionic acid ratio was higher in order of feeding only corn silage, 50% corn silage + 50% WBG and only WBG. 4. Ruminal bacteria and protozoa number in diet of 50% corn silage + 50% WBG were 8.14 $\times$ > $10^5$ and 1.45 $\times$ $10^5$, respectively, which were most of all treatments. 5. Daily gain and feed efficiency of Holstein heifer in feeding concentrate to 0.5% of BW and forage hay to about 20% of total DM intake when fed free WBG were better

  • This study was conducted to determine the differences in the growth and milk production performance of Holstein Crossbreds (Korean Native Cattle(♀)${\times}$Holstein(♂)) and Korean Native Cattle produced at the Livestock Experiment Station of the Office of Rural Developement from 1973 to 1978. The number of heifers and cows used in this experiment were 15 head of Korean Native Cattle and 11 head pf Holstein Crossbreds Cattle. Body weight and body measurements were taken at birth, 6, 12, 24 and 36 months of age, however, body measurements were not taken at birth. Milk production was checked from the 11 th day to 180th day after calving. The data was analyzed using the least square procedure in order to estimate the effect of the mating group, year of birth, calving season and parity. The results obtained from this study were as follows: 1. The body weights of the Holstein Crossbreds were heavier than the body weights of purebred Korean Native Cattle. The body weight of the Holstein Crossbreds averaged 28.09kg, 146.64kg, 254.48kg, 392.04kg and 454.46kg at birth, 6, 12, 24 and 36 months of age, respectively. However, the body weights of purebreds Korean Native Cattle averaged 22.45kg, 132.82kg, 220.68kg, 363.54kg and 365.54kg at the same ages. 2. The year of birth affected on body weight at each point during the growing stage, except birth, heifers born in the spring and autumn were heavier than the others, but calving season did not affect on body weight during the growing stage except at birth and 6 months. 3. Parity showed significant differences on body weight in the growing stage. Calves from the 5th parity had a tendency to be heavier than the other calves. 4. The year of birth, calving season and parity at calving had no affect on the change of body measurements, but the wither height, hip height, chest depth, chest girth and hip width were significantly greater in the Holstein Crossbreds at 24 months of age. 5. Mating groups had a significant affect on milk production during the growing stage. Year of birth and calving season did not affect milk production, but parity was significant from 11 days after calving. 6. The least-squares means used to determine the daily milk production were 3.60 and 8.26kg/day for Korean Native Cattle and the Holstein Crossbreds, respectively.

  • The present study was conducted to examine the feasibility of utilising bamboo (Bambusa arundinacea) shoot shell (BSS) in ruminants. Chemical composition, rumen degradability and some antinutritional compounds were determined for fresh and boiled BSSs to evaluate its feed value and safety. Thirty-two Holstein heifers were allocated to four groups and used to investigate the response in growth rate to supplementing ammoniated rice straw with fresh shell (phase 1) or silage of boiled BSS (phase 2). All animals were offered ammoniated straw ad libitum with 1kg of cotton seed meal (phase 1) or 0.5 kg of cotton seed meal and 0.5 kg of concentrate mixture (phase 2) per head per day. The BSS was supplemented at levels of 0, 3, 6 or 9 kg/d (phase 1) and 0, 5, 10 or 15 kg/d (phase 2) (as fed basis). The BSS was very high in moisture content, and its contents of crude protein and neutral detergent fiber were 13~16% DM and 65~76% DM, respectively; boiling resulting in higher moisture and protein. No hydrocyanic acid was detected in both BSSs and content of tannins was negligible. Rumen degradability of BSS was reasonably high, and with boiling the rapidly degradable fraction decreased, and potentially degradable component increased. Silage of the boiled BSS was slightly lower in both rapidly and slowly degraded fractions than the fresh BSS. Animals consumed all supplemented BSSs without any adverse health problems. Intake of ammoniated straw decreased with the increasing levels of BSS, but total intake was higher in almost all supplementary groups than in the non-BSS. Heifers had a higher growth rate in phase 1 with fresh BSS than in phase 2 with ensiled shell, and daily weight gains were 622, 629, 744 or 690 g in phase 1, and 578, 575, 677 or 635 g in phase 2 at four BSS levels, respectively. For both phases growth rate was significantly higher for the animals in groups 3 and 4 than those in groups 1 and 2 (p<0.01), with little difference between groups 1 and 2 (p>0.05) but significant difference between groups 3 and 4 (p<0.05). Supplementation with BSS also resulted in an improved feed conversion rate, with the least concentrate consumption in group 3 for both phases. It is concluded that the BSS has a high potential nutritional value as indicated by its medium protein content, reasonably high rumen degradability, and that inclusion of BSS in ammoniated rice straw diet is not only safe to animals, but also may improve growth rate of ruminants and feed conversion rate. It may be disadvantageous to use high amounts of BSS in ammoniated straw diets.

  • Induction of twinning was attempted by transfer of two whole- or demi-embryos in Holstein cows and heifers. Cows were superovulated with follicle stimulating hormone(FSH) administered twice daily in intra-muscular injection. On day of 6.5 to 7 post-estrus, embryos were collected nonsurgically, Normal morulae and early balstocysts were obtained from superovulated cows. The embryos were hisected with a micro-blade made from a razor. Twenty seven pairs of half embryos were successfully produced and 23 pairs of half embryos were transferred to recipients( Group A). Twenty cows were treated with low unit of FSH (3mg${\times}$2,2mg${\times}$2 ; Group B1, 2mg${\times}$2, 1mg${\times}$2, 1mg${\times}$2 ; Group 32) to induce double or triple ovulations. The cows of Group B1 and B2 were Inseminated artificially(A1) at following estrus. Twenty four heifers were bred by A1 and received an additional embryo into the uterine horn contralateral to the corpus luteum(CL) 6 days later(Group C). One embryo was transferred into each uterine hem of 16 heifers 6 days after estrus(Group D). Fourteen heifers were received two embryos into the uterine horn ipsilateral to the CL at day 6 of estrous cycle(Group I). Pregnancy rates at 60 days in Group A, B1, B2, C, D and E were 34.8%, 70.0%, 60.0%, 66.7%, 62.5% and 57.1%, respectively. Twinning rates were 8.7%, 20.0%, 10.0%, 16.7%, 18.7% and 21.4%, respectively. The present experiments demonstrate that FSH treatment( Group 31) and ipsilateral transfer( Group I ) of two whole-embryos are more useful methods to produce twins than the others.

  • This study was carried out to evaluate the effect of early pregnant cow as donor for Ovum Pick-Up (OPU) derived oocyte aspiration and embryo production in Holstein heifers. Four non-pregnant and 2 pregnant Holstein heifers were used as donor and then carried out total 17 OPU session for 10 weeks (2 times per week). Recovered cumulus-oocyte-complexes (COCs) were classified into 4 grade by oocyte cytoplasm and cumulus cells and matured in vitro in TCM-199 supplemented with 10% FBS, 10 mg/ml FSH and 1 mg/ml estradiol in 5% $CO_2$ and over 99% humidity for 24 h. After 24 h co-incubation with post-thaw sperm, the presumed zygotes were cultured in CR1aa medium with 4 mg/ml BSA for 3 days and then changed CR1aa medium with 10% of FBS for another 3~4 days. The Mean number of aspirated follicles and collected oocytes in the early stage pregnant and non-pregnant heifers were $13.0{\pm}4.3$ and $10.6{\pm}3.9$, $5.4{\pm}3.4$ and $7.7{\pm}3.6$ per session, respectively. Rate of collected oocyte from aspirated follicles were 59.2% and 50.5%, respectively. The average number of good quality oocytes (Grade I and II) in the early stage pregnant and non-pregnant heifers was $3.7{\pm}2.7$ and $4.9{\pm}2.6$ (Mean${\pm}$SD). Cleavage and blastocyst developmental rates in Grade I and II were 22.2% and 25.5%, and then $1.7{\pm}0.9$ and $1.4{\pm}1.1$ blastocyst per session, respectively. In conclusion, OPU technology can be used in early stage pregnant and non-pregnant heifers without any problem and so applied OPU derived embryo production to maximize the ability of genetically valuable females.

  • In order to study the changes of pregnancy rates according to the factors, age, season, repeated utilization, synchrony, of recipients (29 holstein heifers, one Korean native heifer and 3 holstein cows) were transfered one embryo (from morulae stage to advanced blastocyst) to each, and the results are as follows ; 1. The effect of age of recipient on pregnancy rate in cows and 18-24months of aged group are 100% and 78%, respectively. Under 14months of aged group recorded the worst, 67%. 2. In winter (Nov.-Jan.) and spring (Feb.-Apr.), the pregnancy rate were better than others 100%, 83%, respectively. In the autumn (Aug.-Oct.), it was only 50%, the worst. 3. Among 31 recipients which were utilized at the first, 25 were pregnanted (80.7%), but in the second utilization after the failure of the first transfering, 2 heifers, none of pregnancy was obtained. 4. Pregnancy rates related to synchronization of estrus between donor and recipients are;-(before donor)12hrs. (100%), -6hrs. (86%), +( after donor)6hrs. (67%), + 12hrs. (79%), and +over 12hrs. (50%), respectively. 5. Differences in the pregnancy rate of the recipients +over 12hrs. were significantly large, so in order to ger good pregnancy rate in embryo transfer, the differences of the synchronization must be reduced inner ${\pm}12hrs$. In general, recipients estrus before donor showed better pregnancy rate than estrus after donor.

  • Objective: The effect of flavonoids from alfalfa on the microbial flora was determined using molecular techniques of 16S ribosome deoxyribonucleic acid (rDNA) analysis. Methods: Four primiparous Holstein heifers fitted with ruminal cannulas were used in a $4{\times}4$ Latin square design and fed a total mixed ration to which alfalfa flavonoids extract (AFE) was added at the rates of 0 (A, control), 20 (B), 60 (C), or 100 (D) mg per kg of heifer BW. Results: The number of operational taxonomic units in heifers given higher levels of flavonoid extract (C and D) was higher than for the two other treatments. The Shannon, Ace, and Chao indices for treatment C were significantly higher than for the other treatments (p<0.05). The number of phyla and genera increased linearly with increasing dietary supplementation of AFE (p<0.05). The principal co-ordinates analysis plot showed substantial differences in the microbial flora for the four treatments. The microbial flora in treatment A was similar to that in B, C, and D were similar by the weighted analysis. The richness of Tenericutes at the phylum level tended to increase with increasing AFE (p = 0.10). The proportion of Euryarchaeota at the phylum level increased linearly, whereas the proportion of Fusobacteria decreased linearly with increasing AFE supplementation (p = 0.04). The percentage of Mogibacterium, Pyramidobacter, and Asteroleplasma at the genus level decreased linearly with increasing AFE (p<0.05). The abundance of Spirochaeta, Succinivibrio, and Suttonella at the genus level tended to decrease linearly with increasing AFE (0.05

  • Four Holstein heifers were used in the present study to investigate the effect of standing and lying behaviors on heat production and physiological responses under low (L), medium (M) and high (H) levels of TDN intake. Rectal temperature (RT), respiration rate (RR), heart rate (HR), heat production (HP) and animals behavior (standing time, ST) were measured continuously for 5h (11:00-16:00) periods. There was a tendency for increased RT with TDN intake, and no difference in RT was observed between standing and lying. The RR, HR and HP during standing were significantly higher (p<0.05) than during lying. The difference between standing and lying HP were 4.41, 4.68 and $5.37kJ/kg^{0.75}$ h for L, M and H of TDN intake, respectively. These values showed that standing HP was 18.6-20.9% higher than lying HP. A multiple regression analysis of HP ($kJ/kg^{0.75}$ h) on TDN intake ($g/kg^{0.75}$ d) and ST (min/h) was HP = 7.75 + 0.28 TDN intake + 0.12 ST (R = 0.84). This analysis showed that the total HP not only depend on feed intake levels, but also depends on animal behavior. It was suggested that the change in HP due to the change in feed intake and animal behavior would influence the level of heat loss which was indicated by the changes in the RR and HR.
