In-situ Calibration of the Hydroperoxyl Radical Using an Immobilized TiO2 Photocatalyst in the Atmosphere

  • The present study is the first report of utilizing $TiO_2$ photocatalyst to analytically calibrate the hydroperoxyl radical ($HO_2\;^{\cdot}$). An in-situ calibration method of $HO_2\;^{\cdot}$ is proposed for air monitoring by using an 2-methyl-6-(pmethoxyphenyl)- 3,7-dihydroimidazo-[1,2-a]pyrazin-3-one (MCLA)-chemiluminescence (CL) technique. In this method, $HO_2\;^{\cdot}$($pK_a$ = 4.80) is produced by the ultraviolet (UV) photolysis of immobilized $TiO_2$ using a constant flow rate of air equilibrated water, in which $HO_2\;^{\cdot}$ is controlled by using various lengths of knotted tubing reactor (KTR). The principle of the proposed calibration is based on the experimentally determined halflife ($t_{1/2}$) of $HO_2\;^{\cdot}$ and its empirically observed pH-dependent rate constant, $k_{obs}$, at a given pH. The concentration of $HO_2\;^{\cdot}$/$O_2\;^{\cdot}$− is increased as pH increases. This pH dependence is due to the different disproportionative reactivities between $HO_2\;^{\cdot}$/$O_2\;^{\cdot}$− and $HO_2\;^{\cdot}$/$O_2\;^{\cdot}$−. Experimental results indicate the practical feasibility of the approach, producing very promising method.

  • The spectrophotometric methods for the determination of Nd, Pr, Sm, Eu, Ho, Er were developed, using the multicomponent method of absorption spectra cointerference. The absorption spectra of individual Nd, Pr, Sm, Eu, Ho, Er, mixture of Sm, Eu, Ho, Er in hydrochloric acid solutions were investigated, and also a band register which includes the molar absorptions for all trivalent Nd, Pr, Sm, Eu, Ho and Er absorption band maxima were measured. The spectrophotometric determination of Nd, Pr, Sm, Eu, Ho and Er were investigated using the analytically significant absorption bands at 444.2, 575.1, 401.7, 394.2, 416.6, and 379.7 nm, respectively.

Determinant Role of the Severity of Hypoxia in the Induction of Reoxygenation Injury in Cat Lung (고양이 허파에서 산소재유입 손상을 결정하는 저산소증의 정도)

  • Although reoxygenation is the best way to salvage hypoxic tissues, reduced oxygen species (ROS) generated during reoxygenation are blown to cause further tissue injuries and the induction of heat shock proteins (HSPs). The present study was undertaken to determine any causal relationship between the severity of hypoxia and the opposite outcomes, either beneficial or detrimental, of the subsequent reoxygenation by measuring the HSP72. To this aim, one group (6 male cats, $2.5{\sim}3.5\;kg$) was subjected to a 5-min episode of hypoventilation (H, ventilation rate: 5/min) for the induction of slight hypoxia and the other group (6 male cats, $2.4{\sim}3.7\;kg$) was subjected to a 5-min episode of apnea (A) for severe hypoxia. Each 3 animals from both groups received a 10-min episode of ventilation with $(95%\;O_2\;(0)$, whereas the remainder did not. After these procedures, all animals were allowed to be ventilated within physiological range for 1, 4, or 8 hours (1H, 1HO, 4H, 4HO, 8H, 8HO, 1A, 1AO, 4A, 4AO, 8A and 8AO groups). Control animals did not receive any manipulation. The arterial blood $pCO_2$ was significantly higher just after apnea than hypoventilation, while $pCO_2$ and pH were significantly lower just after apnea than hypoventilation. Western blot analysis revealed that the magnitude of HSP72 synthesis is larger in 1H, 4H and 8H groups than in 1HO, 4H and 8HO groups, respectively. In contrast, 1AO, 4AO and 8AO groups more induced HSP72 than 1A, 4A and 8A groups, respectively. These results suggest that the reoxygenation is beneficial after slight hypoxia but detrimental after severe hypoxia.

The Protective Effects of Chilgi-tang on Oxidative Stress by Glucose Deprivation in Neuro 2A Cells (Glucose deprivation으로 유발된 Neuro 2A 세포의 산화적 손상에 대한 칠기탕(七氣湯)의 보호효과)

  • Objective: The water extract of Chilgi-tang (CGT) has been traditionally used in treatment of heart diseases caused by stress in Oriental Medicine. However, little is known about the mechanism by which CGT rescues neuronal cells from injury damage. Therefore, this study was designed to evaluate the protective effects of CGT on Neuro-2A cells by glucose deprivation-induced cell death. Methods: We investigated how cell death induced by glucose deprivation was associated with increased reactive oxygen species (ROS) generation. Result: The CGT treatment prior to glucose deprivation insult significantly reduced the number of cell deaths and the glucose deprivation-induced increase in ROS. Nitric oxide (NO) was also attenuated by CGT treatment. In addition, we demonstrated that the anti-cell death effect of CGT was blocked by heme oxygenase-1 (HO-1) activation. Finally, pretreatment of cells with a hemin, HO-1 inducer, reduced glucose deprivation-induced cell death. In contrast, pretreatment of cells with a ZnPP, HO-1 activity inhibitor, attenuated CGT-induced inhibition of cell death. Conclusions: These findings indicate that ROS plays an important role in glucose deprivation-induced cell death and that CGT may prevent glucose deprivation-induced cell death by inhibiting the ROS generation through HO-1 activation in Neuro-2A cells.

Diagnostic Role of Bile Pigment Components in Biliary Tract Cancer

  • Bile pigment, bilirubin, and biliverdin concentrations may change as a results of biliary tract cancer (BTC) altering the mechanisms of radical oxidation and heme breakdown. We explored whether changes in bile pigment components could help distinguish BTC from benign biliary illness by evaluating alterations in patients with BTC. We collected bile fluid from 15 patients with a common bile duct stone (CBD group) and 63 individuals with BTC (BTC group). We examined the bile fluid's bilirubin, biliverdin reductase (BVR), heme oxygenase (HO-1), and bacterial taxonomic abundance. Serum bilirubin levels had no impact on the amounts of bile HO-1, BVR, or bilirubin. In comparison to the control group, the BTC group had considerably higher amounts of HO-1, BVR, and bilirubin in the bile. The areas under the curve for the receiver operating characteristic curve analyses of the BVR and HO-1 were 0.832 (p<0.001) and 0.891 (p<0.001), respectively. Firmicutes was the most prevalent phylum in both CBD and BTC, according to a taxonomic abundance analysis, however the Firmicutes/Bacteroidetes ratio was substantially greater in the BTC group than in the CBD group. The findings of this study showed that, regardless of the existence of obstructive jaundice, biliary carcinogenesis impacts heme degradation and bile pigmentation, and that the bile pigment components HO-1, BVR, and bilirubin in bile fluid have a diagnostic significance in BTC. In tissue biopsies for the diagnosis of BTC, particularly for distinguishing BTC from benign biliary strictures, bile pigment components can be used as additional biomarkers.

  • In this study, we consider the transportation problem in port container terminals. It aims to determine positions in yards to place the containers at the adequate times. The containers on ship must be unloaded one by one from top to bottom, and placed in the main yard in order to reduce additional cost required for unnecessary unloading when getting out by customer with given timetable. The cost for transportation at container terminals could be reduced by a new approach in scheduling: move the containers from ship and stack them onto main yard that minimizes cost of yard crane operation when unloading for customer.
