The influence of ultrasound and adenosine 5'-monophosphate marination on tenderness and structure of myofibrillar proteins of beef

  • Objective: The aim was to investigate the influence of ultrasound and adenosine 5'-monophosphate (AMP) marination (UAMP) on tenderness and structure of myofibrillar proteins of beef. Methods: Five groups, the untreated meat (Control), deionized water marination (DW), ultrasound followed by DW (UDW), AMP marination (AMP), and ultrasound followed by AMP (UAMP) were studied. Myofibrillar fragmentation, cooking loss, shear force, thermograms, histological observation of meats and myofibrillar proteins properties were investigated in these different treatments. Results: The results showed that UAMP significantly increased myofibrillar fragmentation index from 152 (Control), 231 (AMP), and 307 (UDW) to 355 (p<0.05), respectively. The lowest cooking loss, shear force and peak denaturation temperature were observed in UAMP. In histological observation, UDW and UAMP had more fragmented muscular bundles than the others. Furthermore, a drastic increase in ${\alpha}$-helix and decrease in ${\beta}$-sheet of myofibrillar proteins was observed in UAMP, implying the disaggregation of protein samples. The synchronous fluorescence spectra of myofibrillar proteins in UAMP suggested the combination of ultrasound and AMP could accelerate the unfolding molecular structure and destroying hydrophobic interactions. The results of circular dichroism and synchronous fluorescence spectra for myofibrillar proteins coincided with the microstructures of beef. Conclusion: The results indicate that ultrasound combined with AMP improved meat tenderness not only by disruption in muscle integrity, increasing water retention, but also altering their spatial structure of myofibrillar proteins.

Screening for candidate genes related with histological microstructure, meat quality and carcass characteristic in pig based on RNA-seq data

  • Objective: The aim of the present study was to identify genetic variants based on RNA-seq data, obtained via transcriptome sequencing of muscle tissue of pigs differing in muscle histological structure, and to verify the variants' effect on histological microstructure and production traits in a larger pig population. Methods: RNA-seq data was used to identify the panel of single nucleotide polymorphisms (SNPs) significantly related with percentage and diameter of each fiber type (I, IIA, IIB). Detected polymorphisms were mapped to quantitative trait loci (QTLs) regions. Next, the association study was performed on 944 animals representing five breeds (Landrace, Large White, Pietrain, Duroc, and native Puławska breed) in order to evaluate the relationship of selected SNPs and histological characteristics, meat quality and carcasses traits. Results: Mapping of detected genetic variants to QTL regions showed that chromosome 14 was the most overrepresented with the identification of four QTLs related to percentage of fiber types I and IIA. The association study performed on a 293 longissimus muscle samples confirmed a significant positive effect of transforming acidic coiled-coil-containing protein 2 (TACC2) polymorphisms on fiber diameter, while SNP within forkhead box O1 (FOXO1) locus was associated with decrease of diameter of fiber types IIA and IIB. Moreover, subsequent general linear model analysis showed significant relationship of FOXO1, delta 4-desaturase, sphingolipid 1 (DEGS1), and troponin T2 (TNNT2) genes with loin 'eye' area, FOXO1 with loin weight, as well as FOXO1 and TACC2 with lean meat percentage. Furthermore, the intramuscular fat content was positively associated (p<0.01) with occurrence of polymorphisms within DEGS1, TNNT2 genes and negatively with occurrence of TACC2 polymorphism. Conclusion: This study's results indicate that the SNP calling analysis based on RNA-seq data can be used to search candidate genes and establish the genetic basis of phenotypic traits. The presented results can be used for future studies evaluating the use of selected SNPs as genetic markers related to muscle histological profile and production traits in pig breeding.

The Influence of Hen Aging on Eggshell Ultrastructure and Shell Mineral Components

  • The eggshell, which is a complex and highly ordered structure, is very important factor for food safety and egg marketing. This study investigated the changes in eggshell structure and shell components in relationship to hen age. For this study, we examined the histological change of the endometrium of the 30-, 60-, and 72-wk-old commercial layers, and analyzed the ultrastructure and ionic composition of their eggshells. The results showed that histological deformation, fibrosis, atrophy and elimination of micro-villi in the uterus endometrium were found through microscopic observation that was associated with increasing hen age. Concentration of blood-ion components such as $Ca^{2+}$, $Na^+$, $K^+$, and $Cl^-$ ions did not change with age. Along with the results from the ultrastructure analysis of the eggshell, the palisade layer ratio and the density of mammillary knobs were significantly decreased in older hens. In addition, the type B mammillary knobs were frequently observed with increasing hen age. In the mineral element assay from the eggshell, $Ca^{2+}$, $S^{2-}$, and $Co^{2+}$ significantly decreased with increasing hen age, whereas $Na^+$, $K^+$, and $V^{2+}$ significantly increased. Therefore, the damages of endometrial tissue inhibit the processes of ion transmission and the crystallization of eggshell formation, resulting in a large and non-uniform mammillary knob formation. This means the conditions of endometrial cells affect the formation of the eggshell structure. In conclusion, hen aging causes the weakness of the eggshell and degrades the eggshell quality.

Melatonin Induced Changes in Specific Growth Rate, Gonadal Maturity, Lipid and Protein Production in Nile Tilapia Oreochromis niloticus (Linnaeus 1758)

  • We have investigated the effect of melatonin (MLT) on specific growth rate (SGR% $day^{-1}$), condition factor (k), gonado-somatic-index (GSI), histological structures of gonads, serum as well as gonadal protein and lipid in Nile tilapia Oreochromis niloticus. MLT treatment in the dose of 25 ${\mu}g/L$ for three weeks reduced SGR% $day^{-1}$ ($0.9{\pm}0.04$) as compared to control ($1.23{\pm}0.026$). The GSI value was significantly (p<0.05) reduced to $1.77{\pm}0.253$ from control where it was $2.56{\pm}0.25$. Serum protein level increased from $9.33{\pm}2.90$ mg/ml (control) to $11.67{\pm}1.45$ mg/ml after MLT treatment while there was depressed serum triglycerides ($86.16{\pm}1.078$ mg/dl) and cholesterol ($126.66{\pm}0.88$ mg/dl) as compared to control values where these were $123.0{\pm}1.23$ mg/dl and $132.0{\pm}1.65$ mg/dl respectively. Histological structure of ovary showed small eggs of early perinucleolus stage after MLT treatment while testicular structure of control and MLT treated fish was more or less similar. It is concluded that exogenous melatonin suppressed SGR% $day^{-1}$, GSI, ovarian cellular activity, protein and lipid biosynthesis, in tilapia suggesting that melatonin is useful in manipulating the gonadal maturity in fishes.

Copper Toxicity on Survival, Respiration and Organ Structure of Tegillarca granosa (Bivalvia: Arcidae) (꼬막, Tegillarca granosa의 생존, 호흡 및 기관계 구조에 미치는 구리 (Cu) 의 독성)

  • This study was conducted to find out the changes of survival, respiration and organ structure of Tegillarca granosa exposed to copper (Cu). Experimental period was four weeks. Experimental groups were composed of one control condition and three copper exposure conditions (0.125, 0.250 and 0.500 mg/L). The results of the study confirmed that copper induces reduction of survival rate and respiration rate and histopathology of organ structure of the bivalve. In the copper concentration of 0.500 mg/L, mortality was 66.7% after Cu exposure of 4 weeks. Respiration rate was observed exposure groups lower than control decline by 18%. Histological analysis of organ system illustrated degeneration of epithelial layer and connective tissue layer of the mantle. Also, histological degenerations as epithelial atrophy and disappearance of lateral cilia are recognized in the gill and it was observed expansion of hemolymph sinus, disruption of epithelial layer, acidification of mucous and degeneration of muscle fiber bundles in the foot. In the digestive diverticulum, it was showed atrophy and destruction of basophilic cell and epithelial cell in the digestive tubules.

Development Changes in the External Structure of the Head and the Histological Structure of the Eye in Artificially Reared Japanese Eel, Anguilla japonica, Leptocephalus and Glass Eel (극동산 뱀장어(Anguilla japonica) 인공 자어와 실뱀장어의 두부 변화 및 안구의 조직학적 변화)

  • Knowledge of morphological changes in eel larvae is very important for artificial rearing of eel larvae. In this study, we investigated the morphological structure of the head region and histological changes of the eye retina in artificially reared larvae at various stages and in glass eel just after metamorphosis. Structural changes were observed in the upper jaw (maxilla) and the lower jaw (mandible) after 100 dah (day after hatchery) and after metamorphosis. Teeth had degenerated by the time of completion of metamorphosis. Major histological changes observed in the eye retina were the formation of the outer plexiform layer and the outer nuclear layer from 100 dah larva and a change in the rod cell layer after metamorphosis. The cornea was not observed at 10 dah in the eel larva. More information is needed on the early developmental stages of eel larvae to enable mass production of glass eels. The results obtained in the present research will be useful when developing novel rearing programs for eel larvae.

Histological Study on Cutaneous Albinism of Korean Slender Gudgeon Squalidus gracilis majimae from Korea (긴몰개 Squalidus gracilis majimae의 피부백색증에 대한 조직학적 연구)

  • Histological examination of an individual of the Korean slender gudgeon Squalidus gracilis majimae having cutaneous albinism produced rarely in the wild was made and compared with a normal individual. The external body of the albino was colorless, differing from the normal individual, which has dense brownish black spots over its body surface. To make it clear through histological study, we observed eight skin regions: dorsal, lateral, ventral, upper caudal peduncle, lower caudal peduncle, dorsal fin, anal fin, and the eyes. These regional skins were the same in fundamental structure between albinic and normal gudgeon, but there were significant differences in distribution and development of pigment cells (melanins). In the normal gudgeon, the pigment cells were well developed over the regional skins except on the skin from the ventral region. However, it was confirmed in the albino that the pigment cells were vestigial over the upper regions of the eye and body but absent in the ventral region, lower caudal peduncle, and anal fin.

Histological study of the primo vascular system on the falciform ligament (Falciform ligament(간겸상인대)에서 관찰되는 프리모 조직의 조직학적 특성 연구)

  • Objectives : Primo vascular system is known to new circulatory system and suggested as a anatomical structure of meridian system. Primo-vessels are present throughout the whole body. The purpose of this study is to identify primo tissues taken from falciform ligament and to compare with organ surface primo tissue, blood vessel and lymph vessel. Methods : Male Sprague-Dawley rats (8weeks old, 250~320g) used for this study. The medial line of the abdomen was dissected and searched for primo tissues in falciform ligament and on the internal organs using stereomicroscope. We performed serial cross section and histological investigations. The tissues were stained with hematoxylin-eosin and Masson's trichrome. Results : 1. The primo tissues attached on the falciform ligament had white color and length of 5~8mm. 2. We could observe primo tissues are classified with ligament tissues. 3. Histologically, primo tissue on falciform ligament and organ surface primo tissue could be considered same tissue. Conclusions : In this study, we observed primo tissue discovered on the falciform ligament. And we also histologically compared such discovered primo tissue with organ surface primo tissue. Consequently, we could consider that two tissues have histological similarity and possibility of connection in one network system.

Toxicity of Bis(tri-n-butyltin)oxide (TBTO) on the Organ Structure of the Flounder, Paralichthys olivaceus (넙치(Paralichthys olivaceus)의 기관계 구조에 미치는 bis(tri-n-butyltin)oxide (TBTO) 독성)

  • The histopathologieal effects of his(tri-n-butyltin)oxide (TBTO) on the flounder, Paralichthys otivaceus were examined by means of histological methods. The experimental fishes were exposed to 0.17, 0.36, 0.60, 3.20, 6.30, 12.50 ${\mu}g \;L^{-1}$TBTO concentrations for 42 days. Histopathological change of the fish exposed to TBTO is dependent on the exposure duration and concentration. In the lower concentrations early histological changes included activated mucous cells and chloride cells, capillary hyperemia and epithelial hyperplasia in the gill; hepatocyte activation, degeneration of bile duct and pancyeatic zymogen reduction in the hepatopancreas; and capillary hyperemia, appearance of eosinophilic cell and melano-macrophagocytes in the kidney. At the higher concentrations histological changes of dysfunctionality included epithelial lifting and deformation of the lamellae in the gill; pycnosis and cytoplasmic degeneration of hepatocyte; pycnosis of haemopoietic cell and deformation of renal tubules and glomerulus in the kidney. It is indicated that TBTO induced histopathological changes in the fish as other aquatic pollutants.
