Dose Dependence of the Severity of Radiation-Induced Thymic Lymphoma in Mice

  • The dose dependence of the severity of radiation-induced thymic lymphoma in C57BL/6J mice was studied. Mice were exposed to fractionated irradiation at the total doses of 4.0, 6.0 and 8.0 Gy (four irradiations at 8-day intervals) starting from 33 days after birth. Pathological and histological changes of each mouse were observed after periodical sacrifice at day 75, 100, 125, 150, 175, 200, 250, 300 after the first irradiation. The severity of cancers were classified into 4 stages by clinical signs with respect to the enlargement of the thymus, spleen, liver, the progression of the cancer in the thymus, and the metastasis to the spleen, liver, lung and the lymphatic nodes. Among the 490 mice observed, 146 mice had thymic lymphoma. A clear dose-effect relationship was observed as well as the dose-response relationship. Also, periodical observation showed that thymic lymphoma was first induced in mice sacrificed at day 100 (130days old), and metastasize in the order of spleen, lung, liver and then the lymphatic nodes. The results suggest that radiation may be involved not only as a tumor initiator but also as a tumor promoter, and a tumor progression-enhancing agent.

Recovery Rate and Histological Changes in the Gills of Juvenile Abalone Haliotis discus hannai by Exposure Time of Different Water Temperatures and Salinities (수온 및 염분별 노출시간에 따른 북방전복, Haliotis discus hannai 치패의 회복률 및 아가미의 조직학적 변화)

  • This study looked into recovery rate and histological changes in the gills of juvenile abalone Haliotis discus hannai by exposure time (3, 6, 12, 24 and 48 h) of different water temperatures (15, 20 and $25^{\circ}C$) and salinities (30, 25, 20 and 15 psu) to understand reasons for the death of abalone exposed by low salinity water. In each water temperature, abalone spats that were exposed to low salinity water (less than 20 psu) for over 6 hours showed decrease in survival rate during recovery and those were exposed at the salinity of 15 psu for more than 24 hours all died. Histological observation showed expansion or damage of gills of the species which were exposed at less than 20 psu for over 6 hours. In case of abalones exposed at the salinity of 15 psu for over 24 hours, most gill tissues were destroyed. This result was glaringly obvious at a higher water temperature, lower salinity and longer exposure time. Accordingly, suffocation caused by damage of gills considered one of direct causes of the death.

  • As a series of study on the removal of astringency and production of tannin substances, this experiment was conducted to investigate changes of tannin contents and microscopic observation on leaves of persimmon(Diospyros kaki L.). Changes of tannin contents of leaves and fruits in Daegu Bansi(astringent variety) were of similar tendency in content of total and soluble tannin during growth. It seemed therefore that tannin contents of leaves in astringent variety were closely concerned with tannin of fruits. But Fuyu(sweet variety) was different from each other during its growth. Histological form of leaves in microscopic observation had similar tendency between astringent and sweet varieties. It was observed that tannin substances were contained in epidermis cells of levels, that tannin substances were transfered in phloems of nerves system, and that tannin cell existed too in the vicinity of nerves system.

Intravascular Tumour Targeting of Aclarubicin-loaded Gelatin Microspheres Preparation biocompatibility and biodegradability

  • This study is to evaluate the potential use of aclarubicin-loaded gelatin microspheres as an intravascular biodegradable drug delivery system for the regional cancer therapy. The diameter of the microspheres prepared by water in oil emulsion polymerization could be controlled by adjusting the stirring rate in the range of 10-50 $\mu$m : D(in $\mu$m) = -73.8 log (rpm) + 262.7. The addition of proteolytic enzyme increased the in vitro aclarubicin release but it did not change the amount of the initial burst release which reached about 45%. Microspheres injected intravenously into the mouse tail vein embolized only to the lung when observed by fluorescence microscopy. From histological examination following injection of gelatin microspheres into mouse femoral muscle, mild inflammation was observed from the appearance of neutrophils after 2 days and rapid repair process was confirmed thereafter. Biodegradation process of gelatin microspheres lodged on the pulmonary capillary bed was followed up by microscopic observation; degradation was taking place by about 36 hrs, followed by severe damage on the spheerical shape and microspheres was no longer found 10 days after injection.

New Phytoformula (CAS) Containing the Roots of Cyathula officinalis, Achyranthes japonica and Sophora subprostrata Inhibits Collagen-induced Arthritis in Mice

  • The combined new phytoformula (CAS), a mixture (5 : 3 : 2, w/w/w) of the ethanol extracts of the roots of C. officinalis and A. japonica, and the n-butanol fraction of the S. subprostrata strongly inhibited arthritic severity score as well as IL-6 production in serum of collagen-induced arthritic mice. Histological observation also indicated that the CAS-treated group showed less breakdown of joint cartilage of the collagen-treated mice. In contrast, C. officinalis alone or a combination of A. japonica and S. subprostrata did not show significant inhibitory action on the same animal model. Thus, it is thought that CAS possesses a synergistic inhibitory action on arthritic condition. All these results strongly suggest that CAS may be a potential anti-arthritic agent.

  • A 3-year old male Maltese, which had enlarged and ballooned superficial lymph-nodes, was admitted to the Veterinary Teaching Hospital, College of Veterinary Medicine, Kyung-pook National University. Radiographic findings indicated cranial mediastinal mass, interstitial pneumonitis, conspicuous splenomegaly and liver enlargement. The patient showed anorexia, depression and anemia. It was autopsied following its death 2 days after admittance. There were copious hemoperitoneum, lymphomatous mass in the spleen, torsion of the mesenterium and stenosis of the duodenum. On the microscopic observation, there was the increase of the amount of lymphocytic cytoplasm and numbers of nucleus-devided lymphocytes followed by their diffused infiltration in lymph nodule. Based on the physical, histological and pathological findings, this case was diagnosed as a multicentric form of lymphosarcoma.

  • Effects of nonylphenol (NP) on gonadal development and growth of olive flounder, Paralichthys olivaceus were investigated. NP treatment was carried out to fry fish (during 55 to 64 days after hatching) using oral adminstration at nominal NP concentrations of 50, 100 and 150 tig/g BW Gonad before NP treatment was sexually undifferentiated as observed with mostly gonia cells. At 159 days after hatching, ovarian lamella of ovary were filled with oogonia and perinucleolus oocytes. On the other hand, testicular lobules of the testis were occupied by spermatogonia, spermatocytes and spermatids. Histological observation of ovary and testis development was any different between the control and NP treated groups. However, growth was significantly increased in NP treated groups than control groups (P< 0.05). These results considerate that NP has any effect for sex differentiation and gonadal development, but act for early growth in olive flounder.

  • Mulberry heart disease was associated with vitamin E and selenium deficiency of pigs. Anatomical findings of this disease were hydropericadium, extensive patchy hemorrhage of epicardium, endocardium, and discoloration of dark red color of myocardium. In histological findings were characterized by acute myocardial degeneration, extensive hemorrhage, fibrinoid degeneration of arterioles, PAS positive material deposition of arterioles and capillary thrombosis. In affected herds, feed and serum tocopherol and selenium concentration were less than normal values.

  • The development of delayed-type hypersensitivity(DTH) reaction to Staphylococcus aureus in mice was studied, Mice received 3 injections of $10^8$ viable S. aureus subcutaneously showed a marked footpad swelling when mice were challenged with $10\;{\mu}g$ staphylococcal protein antigen into footpad(The percent increase of footpad thickness at 24 h after challenge wsa 35% approximately). Histological observation of footpad of immunized mice showed a marked thickness of subcutaneous tissue due to edematous reaction and massive infiltration of lymphocytes and neutrophils which are characteristic cells in DTH reaction. Intensity of DTH reaction of mice immunized with viable bacteria was much higher than that of mice immunized with staphylococcal protein or heat-killed bacteria. The DTH reaction to S. aureus could be transferred to normal recipient mice by both spleen cells and lymph node cells.

  • The clinical and histopathological observation of leucocytozoonosis was carried out during July, 1982 at a poultry compound in Gyeongbuk province and the results were summarized as follows: 1. Leucocytozoonosis was observed in 36,900 of 59,900 chickens (61.6%) and the mortality rate was 18.3% (6,760 of 36,900 chickens infected). 2. Clinical findings were anorexia, dyspnea, paleness of comb, greenish diarrhea and reduction of egg production. 3. Gross lesions were yellowish white spots scattered on parenchymal organs and swelling of liver and spleen. In addition, petechiation and ecchymosis of small intestine, egg yolk and subcutaneous tissues were observed. 4. Histological findings were infiltration of mononuclear cells in liver, heart and lung, appearance of megaloschizonts in heart, liver, proventriculus and pancreas, proliferation of foreign body giant cells in heart, and hyperemia and swelling of all parenchymal organs.
