감초 추출물의 Dextran Sulfate Sodium 유도 마우스 궤양성 대장염 억제 효과 (Anti-Inflammatory Effects of Glycyrrhiza glabra Linne Extract in a Dextran Sulfate Sodium-Induced Colitis Mouse Model)

  • The aim of this study was to evaluate the anti-inlfammatory effects of Glycyrrhiza glabra Linne extract on ulcerative colitis induced by 3% dextran sulfate sodium in mice. The experimental animals were divided into six groups: control(normal), DSS-induced colitis(control), 1 mg/kg, 10 mg/kg, and 100 mg/kg of Glycyrrhiza glabra Linne extract, and 150 mg/kg 5-aminosalicylic acid(5-ASA)(positive control). We evaluated the pathological disease activity index(DAI), change in weight, colon mucosa damage and myeloperoxidase(MPO) in colon mucosa. Treatment with 10 mg/kg and 100 mg/kg of Glycyrrhiza glabra Linne extract led to significant loss of body weight, the decrease of MPO activity and clinical symptoms such as DAI and histological change. In particular, 100 mg/kg Glycyrrhiza glabra Linne extract led to markedly greater improvement than 150 mg/kg 5-ASA treatment. These results suggest that Glycyrrhiza glabra mediated anti-inflammatory action on colorectal sites may be a useful therapeutic approach to ulcerative colitis.

양릉천(陽陵泉) 오미자(五味子)약침이 고지방식이로 유발된 고지혈증 흰쥐에 미치는 영향 (A Study on the Effect of Herbal-acupuncture with Schizandrae Fructus Solution on Hyperlipidemia in Rats Induced by High Fat Diet)

  • Objectives : The purpose of this study is to observe the effects of herbal acupuncture with Schizandrae Fructus solution at $GB_{34}$(Yangneungcheon) on hyperlipidemia in rats. Methods : In the present study, herbal acupuncture with Schizandrae Fructus solution was administered into hyperlipidemic mice. The author performed several experimental items to analyze the levels of various components and enzymes in serum, liver, as well as the histological changes of liver and aorta. Results : 1. In the Schizandrae Fructus herbal-acupuncture group, serum total cholesterol and Atherogenic index were significantly decreased, and the ratios of HDL to total cholesterol, and phospholipid to total cholesterol in serum were significantly increased as compared with those of the control group. 2. In the Schizandrae Fructus herbal-acupuncture group, HMG-CoA reductase activity was significantly decreased as compared with those of the control group. 3. Schizandrae Fructus herbal-acupuncture reduced deposit of lipid in the liver tissue. Conclusions : From the above results, it is suggested that Schizandrae Fructus herbal-acupuncture at $GB_{34}$ has a therapeutic effect on hyperlipidemia.

금은화(金銀花)가 Bleomycin에 의한 폐섬유화(肺纖維化)에 미치는 영향(影響) (The Effects of Lonicerae Flos on bleomycin-induced lung fibrosis)

  • Objective : Idiopathic Lung Fibrosis(IPF) is chronic fibrotic interstitial pneumonia. The pathogenesis is unclear. Lonicerae Flos is known to prevent the inflammation and reinforce the immune system. The effects of Lonicerae Flos on bleomycin-induced lung fibrosis is evaluated. Material and Methods: Lonicerae Flos extract was given to the Normal rats, control(bleomycin) rats everyday and treated(bleomycin and lonicerae flos) rats 21.0 mg per body weight 109 for 14 days. 14 days after, we observed the change of leukocyte count and percentage of IFN-gamma and IL-4 in BALF. and that of Semiquantative histological index(SHI). Results : Compared to control rats, Lonicerae Flos decreased leukocyte count(P<0.01) lymphocyte, neutrophil percentage(P<0.05), SHI(P<0.01), IFN-gamma and IL-4(P<0.05) in Treated rats. Otherwise, macrophage percentage was increased(P<0.01) in Treated rats. Conclusion : This study showed that Lonicerae Flos reduced the change of inflammatory cells and cytokines in bleomycin-induced lung fibrosis and reduced the fibrosis of tissue. And, we needed many other distinct researches on lung fibrosis.

황기가 C57BL/6J 생쥐의 Bleomycin유발 폐섬유화에 미치는 영향 (The Effects of Astragali Radix on Bleomycin-induced Lung Fibrosis)

  • Backgrounds & Objectives: Many acute and chronic lung disorders with variable degrees of pulmonary inflammation and fibrosis are collectively referred to as interstitial lung diseases. Idiopathic pulmonary fibrosis (IPF) is one of several idiopathic interstitial pneumonias with the pathogenesis unclear. Astragali Radix is known to inhibit the Th2 immune response. The effects of Astragali Radix on bleomycin-induced lung fibrosis were evaluated. Materials and Methods: Astragali Radix extract was daily given to the normal rats, control (bleomycin) and treated (bleomycin and Astragali Radix extract, 24.0 mg/10g body weight) rats for 14 days. After 14 days, we observed the change of total leukocyte count and percentage, IFN-gamma and IL-4 in BALF (Bronchoalveolar lavage fluid), and of semiquantitative histological index (SHI). Results: Compared to the control group, Astragali Radix decreased total leukocyte count (p<0.05), lymphocyte (p<0.05), neutrophil (no significance) percentage, SHI (p<0.05), IFN-gamma and IL-4 (p<0.05). Otherwise, macrophage percentage was increased (p<0.01). Conclusion: This study showed that Astragali Radix reduced the incidence of inflammatory cells and cytokines and prevented the fibrosis of tissue in bleomycin-induced lung fibrosis rats.

HK-1001 약침이 흰쥐의 고지혈증에 미치는 영향 (The Effect of HK-1001 Pharmacopuncture on Hyperlipidemia Induced Rats)

  • Objective&Methods The purpose of this study is to observe the effects of HK-1001 Pharmacopuncture at GB34(Yangleungchean) on hyperlipidemia in rats. The author performed several experimental items to analyze the levels of various components and enzymes in serum, urine and liver, as well as the histological changes of liver and aorta. Results 1. HK-1001 Pharmacopuncture infusion solution increased the cell viability rate, DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. The levels of total cholesterol, free cholesterol, LDL-cholesterol, phospholipid in serum and AI(atherogenic index) were decreased, and the ratio of HDL to TCL(HDL/TCL) and the level of TG in serum were increased as compared with those of the control group. 3. In the HK-1001 group, serum GOT was significantly lower than those of the HG group and the saline group, and serum ALP was significantly higher than that of the HG group. 4. Hepatic GSH and catalase activities were significantly increased as compared with those of the saline group. Conclusion From the above results, it is suggested that HK-1001 Pharmacopuncture at GB34 has a therapeutic effect on hyperlipidemia.

풍륭(豊隆) 전침(電鍼)의 항산화(抗酸化) 효과(效果)에 대한 실험적(實驗的) 연구(硏究) (A Study on Anti-oxidative Effect of Electroacupuncture at Pungnyung($ST_{40}$) in Rats)

  • Objectives : The purpose of this study was to observe the anti-oxidative effects of electroacupuncture at Pungyung($ST_{40}$) in rats. Methods : The author performed several experiments, including measurements of body weightrelative liver weightslevels of albumin, total bilirubin, LDL-cholesterol, LDH, GOT and GPT in serum levels of SOD, glutathione, catalase, NO and MDA in the liver and histological analysis of the liver. Results : 1. Liver index was decreased in the $ST_{40}-EA$ group compared with the holder group and the control group. 2. LDL-cholesterol was significantly decreased in the $ST_{40}-EA$ group compared with the group. 3. SOD activity was significantly increased in the $ST_{40}-EA$ group and $ST_{40}-NR$ group compared with the control group. Glutathione activity was significantly increased in the $ST_{40}-EA$ group compared with the control group, the holder group, and the Sham-EA group. Catalase activity was significantly increased in the $ST_{40}-EA$ group compared with the control group and the holder group. Conclusions : These results suggest that electroacupuncture at $ST_{40}$ has an antioxidant effect in rats.

hMSH2 and nm23 Expression in Sporadic Colorectal Cancer and its Clinical Significance

  • Objective: To study the expression of the mismatch repair proteins hMSH2 and nm23 in sporadic colorectal cancer, determine any inter-relationship, and further investigate any clinical significance. Methods: Expression of hMSH2 and nm23 proteins was assessed in 87 colorectal cancer tissues by SP immunohistochemistry, with analysis of survival using follow-up data. Results: In the sporadic colorectal cancer tissues, nm23 protein expression appeared independent of the histological type (P>0.05), but correlated with the invasion depth and lymphatic metastasis (P<0.05). In contrast, hMSH2 protein expression was not significantly correlated with these clinicopathologic features (P>0.05), although it positively correlated with that of nm23 protein in the sporadic colorectal cancers (rs=0.635, P<0.05). Combined expression of the two was found to be related with invasion depth, lymphatic metastasis and prognosis of sporadic colorectal cancer (P<0.05). Conclusion: nm23 protein level was related with the degree of malignancy, and could be used as an index to predict the invasion and metastasis potential. The expression of hMSH2 protein is positively correlated that of nm23 protein, and the combined expression of the two has certain guiding significance for the prognosis of sporadic colorectal cancer.

동해안 참가자미, Limanda herzensteini의 생식주기 (Reproductive Cycle of the Brown sole, Limanda herzensteini in Eastern Waters of Korea)

  • 이 연구에서는 자연산 참가자미의 주 어획지역인 강원도 주문진 인근해역에서 1998년 9월부터 1999년 10월까지 연간 생식소중량지수의 변화를 조사하였으며, 조직학적 조사에 의한 생식소 발달과정과 생식주기를 밝혔다. 정소는 정소소엽 형태이며, 각각의 정소엽은 여러 개의 소낭구조를 가진다. 각 소낭내의 생식세포들은 같은 단계의 발달상태를 보인다. 난소는 원추형의 낭상으로 체강벽에서 연결되는 난소간막에 의해 부착되어 있으며, 내부는 결체성 조직인 다수의 난소박판으로 구성되며, 이곳에서 난원세포가 유래한다. 수컷의 GSI는 1월에 가장 높았으며, 암컷의 GSI는 3월에 가장 높은 값을 보였다. 생식주기는 성장기(6∼9월), 성숙기(10∼12월), 완숙 및 산란기(1∼3월)그리고 회복 및 휴지기(4∼5월)로 나눌 수 있었다. 난모세포의 발달양식은 난군동기발달형에 속하였다.

Effects of Photoperiod Manipulation on Gonadal Activity of the Damselfish, Chromis notata

  • This study investigated the effect of different photoperiods (14L: 10D, 12L:12D and 10L:14D) on the gonadal development and GtH mRNA expression in the pituitary of damselfish. The results showed that gonadosomatic index (GSI) was significantly lower in shot photoperiod (10L:14D), in comparison with other photoperiodic group during the spawning season. After 60 days treatment, histological analysis of gonad tissue showed that the gonad of 10L:14D and 12L: 12D treatment groups were resting phase with spermatogonia and perinucleolus stage oocytes but the gonad of 14L:10D treatment group was still ripe phase with spermatozoa and mature stage oocyte. The $FSH{\beta}$ and $LH{\beta}$ mRNA expression in pituitary drastically decreased shot photoperiod treatment from July (spawning period). These results suggest that photoperiod is considered to be the most effective environmental factor in controlling the reproductive cycle of damselfish.

Administration of Aqueous Extract of Schizandra chinensis Fruit Inhibits the Experimental Colitis in Mice

  • Schizandra chinensis fruits (SC) have been used as a traditional Oriental medicine for treatments of many stress-induced diseases. In the present study, we investigated the protective effect of SC aqueous extract(SC-Ex) in the inflammatory diseases of intestine using a mouse model of ulcerative colitis. An experimental colitis was induced by daily treatment with 5% dextran sulfate sodium (DSS). SC-Ex was orally administered from day 2 of DSS treatment in a dose-dependent manner. Administration of SC-Ex reduced significantly clinic signs of DSS-induced colitis, including body weight loss, shorten colon length, increased disease activity index, and histological colon injury. Moreover, SC-Ex suppressed significantly not only the activities of myeloperoxidase (MPO) and chymase, but also the expressions of $TNF-{\acute{a}}$ and COX-2 in DSS-treated colon tissues. Inhibitory effect of SC-Ex was effective at a dose over 20 mg/kg. Our results indicate that SC-Ex may possess therapeutic effect on the development of DSS-induced colitis.
