Effects of TongBiEum(TBE) on Hyperlipidemia Induced by a Sucrose-rich Diet

  • Objectives : This study aimed to elucidate the effects of TBE on hyperlipidemia. Methods : We studied the effects of TBE on hyperlipidemia through gene expressions related with lipid metabolism and serum triglyceride as well as total and HDL-cholesterol levels, and perceived histological changes. Results : The present studies demonstrate that TBE can reduce the rise in plasma cholesterol and TG levels induced by a high-cholesterol diet and also reverse pre-established hypercholesterolemia and hypertriglycemia. In the TBE group total cholesterol levels decreased, TG levels decreased, but HDL-cholesterol levels also decreased. In the analysis of absolute and relative liver weight, TBE inhibited the weight gain induced by a high-cholesterol diet. In the histological observations, lipid droplet and apoptotic change in the TBE treated group were less compared with the control group. In the serum biochemical analysis, a difference of serum AST and ALT changes among groups was not shown, but TG and total cholesterol levels were less and HDL level decreased compared with the control group. In the gene expression related with TG and cholesterol metabolism, DGAT decreased slightly but ACAT decreased more as compared with control and Lipidil groups. Conclusion : From this study, we can infer that TBE possesses a hypolipidemic effect by inhibiting the intestinal absorption and storage of exogenous and endogenous cholesterol.

Profile of Colorectal Polyps: a Retrospective Study from King Fahad Hospital, Madinah, Saudi Arabia

  • Aim: To evaluate the predominant colorectal polyps in the Almadinah region of Saudi Arabia. Materials and Methods: In this iretrospective study, we analyzed pathology reports of colonoscopies performed in King Fahad Hospital, Madinah, Saudi Arabia during the period 2006 to 2013. Data based on patient age, gender, size, site and type of polyps and the degree of dysplasia were analyzed by software SPSS 17 and compared with other published studies from different geographic regions of the world. Results: During these years, 224 patients had colonic polyps, of whom 149 (66.5%) were men and 75 (33.5%) were women. The most common types of polyps were adenomatous (166), followed by hyperplastic polyps (24), juvenile (18), inflammatory (13), lipomatous (2) and one patient with Peutz-Jegher polyps. Tubulovillous adenoma was the commonest adenomatous polyp (102), followed by tubular (41) and villous (23) types. The sigmoid colon was the most commonly involved region (36.6%). Dysplasia was significantly associated with female patients who had large size tubulovillous polyps located in the left colon. Conclusions: The type and distribution of colorectal polyps in Saudi Arabia is very similar to Western countries. Patient gender, and size, histological type and location of polyps are closely related to dysplastic change in colonic polyps.

The Study on the Anti-Allergic Rhinitis Effects of Gamigyejitang (focus on histological changes) (가미계지탕(加味桂枝湯)이 알레르기 비염에 미치는 효과에 대한 연구)

  • This study is for Gamigyejitang's effects on the histological change s of allergic rhinitis. for this purpose, the changes of the mucosa epithelium and the submucosa tissue of nose were observed and the effects on neutrophil count and eosinophil count in blood were estimated. The rats were divided into normal group, control group and experimental group. Control group was administered normal saline and experimental group was administerd Gamigyejitang. The results obtained were as follows: 1. The epithelium layer of experimental group was restored to normal thickness and state nearly. 2. The submucosa tissue of experimental group was restored to normal state nearly. 3. The neutrophil count and eosinophil count were decreased in the experimental group(Gamigyejitang treated group) as compared with the control group but not significantly. According to above results, it is supposed that Gamigyejitang has considerable effect on allergic rhinitis and could be applicated to treat the nasal mucosa edema and several symptoms.

The Effects of Jujadokseo-hwan on the Activation of Brain and Neuroprotactive Effects (주자독서환의 뇌기능 활성 및 신경세포 보호효과)

  • Objectives This study is designed to investigate the effects of Jujadokseo-hwan on the brain ability and inducing oxidative stresses. Methods We measured the changes of regional cerebral blood flow and mean arterial blood pressure. Then we analyzed histological examination, immunohistochemistric response and anti-oxidant activity of Jujadokseo-hwan. Results 1. Treatment of Jujadokseo-hwan significantly increased regional cerebral blood flow but decreased mean arterial blood pressure. 2. Treatment of Jujadokseo-hwan-induced increase of regional cerebral blood flow was significantly inhibited by pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. 3. In histological examination through TTC stain, group I was no change, but group II showed that discolored in the most cortical part. Group III showed that decreased discolor in the cortical part. 4. In immunohistochemistric response of BDNF, group II showed that lower response effect. Group III showed that increase response effect. 5. Treatment of Jujadokseo-hwan increased proliferation rates of Glial cell effectively 6. Treatment of Jujadokseo-hwan accelerated proliferation rates of C6 cells in vitro. In addition, protective effects on cell death induced by paraquat, rotenone and hydrogen peroxide. In addition, activity of SOD were increased by treatment with Jujadokseo-hwan. Conclusions In conclusion, Jujadokseo-hwan can improve of the brain ability, learning ability, memory ability and induce ischemic brain injuries.

Effects of Chrysanthemum boreale M. Water Extract on Serum Liver Enzyme Activities and Kupffer Cells of Carbon Tetrachloride-Induced Rats

  • Effects of water extract obtained from Chrysanthemum boreale M. (CE) on serum enzyme activities and Kupffer cells of carbon tetrachloride ($CCl_4$)-induced rats were investigated. Thirty-two healthy male Sprague-Dawley rats were divided into normal (N), $CCl_4$-induced (T), CE-supplemented (C), and $CCl_4$-induced and CE-supplemented (TC) groups. $CCl_4$ injection significantly increased aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, and alkaline phosphatase activities in serum. Significant increases in total cholesterol and triglyceride concentrations were also observed in $CCl_4$-induced rats. Oral administration of CE at 300 mg/kg body weight significantly decreased serum enzyme levels and suppressed $CCl_4$ hepatotoxicity-induced lipid profile changes. Histological findings showed fatty change, fibrosis and increased number of Kupffer cells in T group. Electron microscopic examination showed increased lysosome content and dilation of rough endoplasmic reticulum within Kupffer cells in T group, whereas CE supplement attenuated liver injury in $CCl_4$-induced liver. These results indicated CE could significantly alleviate CC4-induced hepatotoxicity injury.

Mortality of the Horned Turban Shell, Batillus cornutus Caused by Vibrio spp (소라(Batillus cornutus)의 비브리오균 감염에 의한 폐사)

  • Mass mortality of the horned turban shell, Batillus cornutus was investigated using histological and bacteriological methods. Some pathogenic bacteria were isolated from mortal or inactive individuals. The pathogenic agents causing mortality of the horned turban shells were as Vibrio alginolyticus and V. anguillarum. Laboratory experiment indicated that optimal growth temperature of two bacteria was 25 to 30$^{\circ}C$ and 3% of NaCl. Histological examination of the horned turban shells showed that gill necrosis is one of the major symptom of infected individuals. It was believed that sudden increase of those two bacterial agents due to environmental change cause mortality of horned turban shells.

Effect of Salvia miltiorrhiza Extract in Urethane-induced Lung Tumorigenesis in A/J Mice (단삼(丹蔘) 추출액이 Urethan으로 유발된 생쥐의 폐암에 미치는 영향)

  • Objectives: The aim of this study was to determine the effects of Salvia miltiorrhiza (SM) extract in urethane-induced lung tumorigenesis in A/J mice. Methods : We examined change of body weight, histological, apoptosis, immunohistochemical and gene expression of cyclooxygenase (COX-2) in lung tumors. Mice were divided into 3 groups: normal, saline, and experimental group administered SM extract after injection with urethane. Results : Histological observation showed shrunken alveoli in the control group, but recovered from damage in the SM extract administered group. The COX-2 positive materials were observed in the smooth muscle of terminal bronchiole and alveoli from the control group, but these positive materials decreased in the SM extract treatment group. The results of electron microscopical observation, dilated capillary and degenerated endothelia were observed in the control group. The apoptotic nuclei increased more in the control group compared with the normal and SM extract administered groups. Serial sections of the whole lung showed solid and papillary tumors. The size and number of tumors decreased in the SM groups compared with the control groups. Conclusions : These results suggest the possibility that SM may exert an anti-tumor effect on urethane-induced lung tumorigenesis.

Effects of Simulated Acid Rain on Histology, Water Status and Growth of Pinus densiflora (인공산성빗물이 소나무의 조직, 수분수지 및 생장에 미치는 영향)

  • To clarify the effects of acid precipitates on histological damage, water status, and growth of Pinus densiflora green house experiment applyin simulated acid rain was carried out. Contact angle of water droplet on needles of P. densiflora seedlings treated with simulated acid rain of different pHs simulated acid rain was, the more rapid transpiration was. Leaf water potential after water withdrawal was also reduced rapidly in proportion to acidity of simulated acid rain. Height growth of P. densiflora seedlings treated with simulated acid rain of pH 2 decreased, while growth of seedlings treated with that of pH 3 and 4 increased comparing with that treated with normal rain of pH 5.6. pH of cultivated soil in pH 2 plot was acidified with the amount of simulated acid rain applied but that in pH 3 and 4 plots did not show any directional change. From those results, it could be interpreted that decrease of height growth in pH 2 plot was originated from multiple effects of water deficit from rapid transpiration and soil acidification. On the other hand, increased of height growth in pH 3 and 4 plots would be originated from the supply of N and S included in simulated acid rain.

Effects of N-3 Fatty Acids on Renal Function in Rats of Different Ages (N-3rP 지방산이 나이가 다른 흰주의 신장 기능에 미치는 영향)

  • This study was performed to investigate the effects of n-3 fatty acids on renal function in male Sprague-Dawley rats of different ages 5-, 15- and 19-months old. The rats were fed a 20%(w/w) lipid diet containing 10% fish oil, compared with control animals fed a 20% lipid diet without fish oil for 4 weeks. The results were as follows: kidney weights were significantly higher in fish oil-fed rats compared to control rats. Plasma levels of total lipid, total cholesterol, and triglyceride markedly increased, with aging and LDL-cholesterol showing a significantly lower level in fish oil-fed rats than control rats. The urinary protein and glomerular filtration rate (GFR) increased with aging. GFR was higher in fish oil-fed rats. However, urinary protein was the same in the two groups. Renal medulla thromboxane B$_2$(TXB$_2$)tended to be lower in fish oil-fed 19-month-old rats. Urinary TXB$_2$and PGE$_2$were found to be higher proteinuria. Light microscopic examination showed interstitial inflammation, tubular atrophy, interstitial fibrosis and glomerular mesangium increase. Although glomerular sclerosis increased with aging, fish oil in the diet had no effect on histological changes. In conclusion, plasma lipid, urinary protein excretion and renal histological change showed a significant increase with aging. The reduction of TXB$_2$in the medulla and increase of GFR caused by fish oil indicated n-3 fatty acid could affect renal function in line with the hypolipidemic effect.

Effects of Forsythiae Fructus, Ulmi Cortex and JwaGwiEum in Allergic Rhinitis Induced by Ovalbumin in Mice (난알부민으로 유발된 생쥐의 알레르기 비염에 연교, 유근피, 좌귀음이 미치는 영향)

  • Objectives In the past, studies have been shown that Forsythiae Fructus (FF), Ulmi Cortex (UC) and JwaGwiEum (JGE) are effective in treating allergic reactions. However, no report shows the difference in effects of FF, UC and JGE in allergic rhinitis (AR). Therefore, the purpose of this study was to differentiate the effects of FF, UC and JGE in AR induced by ovalbumin (OVA) in mice. Methods From this experiment, the effects of FF, UC and JGE were several - changes in body weight, hematologic changes such as WBC, RBC, Hb and PLT counts, immunological changes such as levels of histamine, IgE, IL-4 and expressions of iNOS & COX-2 mRNA. Moreover, histological change of nasal mucosa was also investigated. Results FF administration group and JGE administration group significantly inhibited level of IgE compared to the control group. Also, FF administration group, UC administration group and JGE administration group inhibited IL-4 and expressions of iNOS & COX-2 mRNA. In histological assessment, FF administration group, UC administration group and JGE administration group showed reduced amount of subepithelial edema, desquamated mucosa and hyperplasia of basement membrane from OVA.
