인삼 분비도관의 조직화학적 염색에 의한 연근판별 (Identification of Root Age by Histochemical Staining of Secretory Duct Layers in Ginseng)

  • Identification of the age of ginseng root is very important in commercial market as well as in research field. However, any criterion abut it has not been clearly established yet. We studied to find a clear morphological key for identification of ginseng root\\`s age using the histochemical staining method. Fresh sections of 3, 4, 5, and 6 year old roots were stained with 1% nile blue, observed under the light microscopy, and compared each other. The number of secretory duct layers(SDL) is a useful key to confirm the age of ginseng root as follow; three-year-old root has two, four-year-old one has three, fie-year-old one has four, and six-year-old one has five resin duct layers on each cortical region of roots. Secretory ducts are thought to be formed by the vascular cambium every year. Unlike the surrounding parenchyma cells, secretory epithelial cells lack starch grains in the cytoplasm.

연령에 따른 청둥오리 장관 내분비세포에 대한 면역조직화학적 연구 (Immunohistochemical studies in the intestine of the duck, Anas platyrhynchos platyrhyncos Linne, with ages)

  • The relative frequency and distribution of bovine chromogranin(BCG)-, serotonin-, somatostatin(SOM)- and S-100 protein-immunoreactive cells in the intestinal tract of the duck (Anas platyrhynchos platyrhyncos, Linne) with ages were studied using immunohistochemical methods. BCG-immunoreactive cells were detected in duodenum on 23 days of incubation. Thereafter these cells were occurred throughout the intestine. SOM-immunoreactive cells were detected in the small intestine from hatching to 9 weeks. Thereafter they were also observed in colon. Serotonin-immunoreactive cells were detected throughout the intestinal tracts after hatching. These immunoreactive cells were increased with ages except that BCG-immunoreactive cells were decreased with ages from 6 weeks after hatching. BCG- and serotonin-immunoreactive cells were most frequently detected in colon regions and SOM-immunoreactive cells in duodenum regions, but no S-100 protein-immunoreactive cells were detected in this study.

  • 발생중의 무당개구리에서의 전신 및 중신 alkaline phosphatase 활성을 Gomori 변법을 써서 관찰하였다. 전신 분비세관의 분화와 함께 세관 刷子綠에 출현한 alkaline phosphataseghkf성은  蓋완성기에 최고도에 달하였다가 제3후지 형성기에 전신의 퇴화와 함께 효소활성은 감소 소실되었다.  蓋완성기에 출현한 중신세관에서의 alkaline phosphatase 활성은 변태기를 통하여 강한 양성반응을 보였다.

  • Barley is considered as one of the most important cereals for Korean in terms of production and utilization. In this review we concentrate mainly on the chemical structure of husk, aleurone layer, endosperm and embryo (especially endosperm). The relationships among the structure, characteristics and role of chemical components in barley kernel are also discussed. The nature of the fine-structure and minor components is not fully identified, however, a lot of investigation and progress in this area probably contributes the development of new technique for better utilization of barley in the future.

  • 1. The purpose of study To study the effectiveness of Yangkyuksanhawtang(YST) against Allergic Contact Dermatitis(ACD) I observed the change of cutaneous shape, histochemistry, immunohistochemistry, and the distribution of apoptotic cells. 2. The material and method of study I divided 4-month-old rats into three groups of 10, which are a contrastive group of having applied Acetone olive oil only, ACD group to have intentionally activated Allergic Contact Dermatitis by DNCB and YST group to give medication of Yangkyuksanhawtang extract. And I observed each group of mice after 24, 48 and 72 hours. 3. The result and conclusion of study The change of cutaneous shape and hypersensitive reaction of immunity was significantly decreased by YST in ACD rats. I consider that YST suppressed hypersensitive reaction of immunity and inflammation in ACD.

동자개 (Pseudobagrus fulvidraco)와 밀자개 (Leiocassis nitidus)에 대한 피부점액세포의 조직화학 (Bagridae, Siluriformes) (Histochemistry of the Mucous Cells in the Skin of Pseudobagrus fulvidraco and Leiocassis nitidus (Bagridae, Siluriformes))

  • 동자개과 (Bagridae) 어류에 속하는 동자개 (P. fulvidraco)와 밀자개 (L. nitidus)의 3부위 (등면, 측면 그리고 배면)의 피부 점액세포를 광학현미경하에 조직화학적인 방법으로 조사했다. 각 종의 피부에 존재하는 점액세포를 PAS, AB-PAS 그리고 HID의 염색방법을 통해 비교해 본 결과, 두 종간에 점액세포의 성분적 차이를 발견할 수 없었고, 그러나 조사 부위에서 점액세포의 위치, 형태, 크기 그리고 수에 있어서 차이가 존재했다. 동자개의 점액세포들은 편평상피세포층과 상피의 alarm substance cells (ASCs)층에 존재했다. 그러나 밀자개의 점액세포들은 편평상피세포층에만 존재했다. 상피의 ASC 층에 존재하는 점액세포는 편평상피세포층에 존재하는 것보다 상대적으로 더 컸다. 편평상피세포층에서 두 종간 각 부위의 점액세포의 평균수는 이분산 T-test에 의해 인정되었고, 등분산 T-test에 의한 동자개의 두 층의 점액세포간의 평균수는 차이가 인정되지 않았다. 그리고 이분산 T-test에 의한 동자개와 밀자개 상피의 ASC 평균수는 등면과 배면에서만 차이가 인정되었다.

흡연이 흰쥐 복부 피지샘의 glycoconjugates에 미치는 영향에 대한 lectin 조직화학 (Lectin Histochemistry on the Effects of Smoking on Glycoconjugates of Rat Sebaceous Glands)

  • 실내 환경 오염원으로 각종 암 발병과 관련이 있는 흡연이 피부 피지샘의 glycoconjugates에 미치는 영향을 lectin 조직화학으로 연구하였다. Sprague-Dawley계 수컷 흰쥐에 1일 1회 10분간 1일군, 2일군, 3일군 및 5일군으로 나누어 직?간접 흡연 방법으로 폭로시켜 피지샘 형태는 PAS 염색, 피지샘의 glycoconjugates 양상을 9종의 biotinylated lectin (DBA, SBA, PNA, BSL-1, WGA, RCA-1, UEA-1, Con A 및 LCA)으로 연구하였다. 대조군에 비해 흡연군에서는 피지샘꽈리의 크기 감소, 피지샘꽈리 상부의 파괴, 중앙부 피지세포의 공포화 및 미성숙 피지샘꽈리가 관찰되었다. 대조군에서 피지샘 glycoconjugates에는 염색성에 차이가 있으나 기저세포는 BSL-1, PNA 및 WGA에만 반응하고 BSL-1의 반응이 더 강하며, 피지세포는 PNA, WGA, Con A, BSL-1 및 SBA에만 반응하고 PNA 및 Con A의 반응이 더 강했다. 흡연군의 렉틴 반응에서 특이적인 변화가 관찰되었다. 흡연군의 피지샘 기저세포의 PNA 반응은 증가되었고, BSL-1는 감소되었다가 회복되었으며, WGA는 소실되었다. 피지세포의 Con A는 증가하였으나 PNA, BSL-1, WGA 및 SBA는 감소 또는 소실되었다. 대조군에 관찰되지 않던 흡연군의 미성숙 샘꽈리 PNA, on A 및 BSL-1이 반응하였으며 PNA 및 Con A의 반응이 더 현저하였다. 결론적으로 흡연은 피지샘의 형태 및 glycoconjugates 대사에 심한 영향을 미치는 것으로 사료된다.

구척(狗脊)이 흰쥐의 척수압박에 의한 신경세포 손상에 미치는 영향 (Effects of Root of Cibotii Rhizoma on Neuronal Damage of Spinal Cord Contusion Injury in Rats)

  • Objectives : This study was performed to evaluate the effects of root of Cibotii rhizoma(CR) ethanol extract on the tissue and neuronal damage of the spinal cord injury(SCI). Methods : SCI was induced by mechanical contusion following laminectomy of 10th thoracic vertebra in Sprague-Dawley rats. CR was orally given once a day for 7 days after SCI. Tissue damage and nerve fiber degeneration were examined with cresyl violet and luxol fast blue(LFS) histochemistry. HSP72(as neuronal damage marker), MAP2(as nerve fiber degeneration marker), c-Fos(immediate early gene), and Bax(pro-apoptotic molecule) expressions were examined using immuno-histochemistry. Individual immuno-positive cells expressing HSP72, MAP2, c-Fos and Bax were observed on the damaged level and the upper thoracic and lower lumbar spinal segments. Results : 1. CR reduced degeneration of nerve fibers and motor neuron shrinkage in the ventral horn of the lower lumbar spinal segment, but generally it did not seem to ameliorate the tissue injury following SCI. 2. CR reduced demyelination in the ventral and lateral funiculus of the lower lumbar spinal segment. 3. CR reduced HSP72 expression on the neurons in the peri-central canal gray matter adjacent to the damaged region. 4. CR strengthened MAP2 expression on the motor neurons in the ventral horn and on nerve fibers in the lateral funiculus of the lower lumbar spinal segment. 5. CR reduced c-Fos positive cells in the peri-lesion and the dorsal horn of the damaged level and in the ventral horn of the lower lumbar spinal segment. 6. CR reduced Bax positive cells in the peri-lesion and the dorsal horn of the damaged level and in the ventral horn of the lower lumbar spinal segment. Conclusions : These results suggest that CR plays an inhibitory role against secondary neuronal damage and nerve fiber degeneration. following SCI.

한국고유종 각시붕어 Rhodeus uyekii 후각기관의 형태 및 조직, 세포화학적 특성 연구 (Morphology, Histology, and Histochemistry of the Olfactory Organ in Korean Endemic Fish, Rhodeus uyekii (Pices, Cyprinidae))

  • 한국 고유종 각시붕어 Rhodeus uyekii의 후각기관과 서식처의 물리 및 화학적 특성 그리고 생태적 습성과의 연관성을 알아보기 위해 실체현미경, 광학현미경, 주사전자현미경을 이용하여 해부 및 조직, 세포화학적 특성을 조사하였다. 후각기관의 외부형태는 반원의 전비공과 후비공, 비경계판으로 구성되는데, 특히 전비공의 앞쪽에는 아치형의 조직이 돌출되어 있다. 비강 내에는 14~15개의 후판으로 구성되는 로제트 구조를 보유하며 후판은 감각상피와 비감각상피로 구분된다. 감각상피는 후감각뉴런, 지지세포, 기저세포, 림프구, 형질세포로 구성되는 반면에 비감각세포는 층상상피세포, 점액세포, 비감각섬모세포, 미확인세포가 1종류 확인된다. 점액세포의 세포화학적 특성은 산성 및 중성의 황점액소(sulfomucine)로 나타났다.

Immunocytochemical Localization of Glutamatergic Neurons in the Lateral Reticular Nucleus Projecting to Ansiform (Crus I and II) and Paramedian Cerebellar Lobules of the Rat

  • I examined the projection of glutamatergic neurons in the lateral reticular nucleus into ansiform (crus l and ll) and paramedian lobules in the rat cerebellum using immunocytochemical methods with antiserum against glutamate combined with WGA-HRP histochemistry. The projections of glutamatergic neurons from the lateral reticular nucleus to crus l were most extensive in number among the three injection cases and the majority of projections originated at the dorsal to dorsomedial region of the ipsilateral magnocellular nucleus. Glutamate-immunoreactive cells projecting to crus ll were less extensive in number than those projecting to crus l and were mainly localized at the dorsomedial portion of the ipsilateral magnocellular nucleus. Double-labelled neurons projecting to crus l or crux ll were also located at ipsilateral subtrigeminal as well as contralateral magnocellular nuclei. Glutamatergic neurons projecting to paramedian lobules were moderate in number and mainly located at the dorsal area of the ipsilateral magnocellular nucleus. A few double-labelled cells were also found at ipsilateral subtrigeminal or contralateral magnocellular nuclei. The present study suggests that glutamate-immunoreactive neurons at the dorsal to dorsomedial magnocellular division of the lateral reticular nucleus may participate in the excitatory control of target neuronal activities at ipsilateral, posterior hemispheric lobules of the rat cerebellum.
