Effect of Decreased Locomotor Activity on Hindlimb Muscles in a Rat Model of Parkinson's Disease (파킨슨병 모델 쥐에서 보행활동저하가 뒷다리근에 미치는 영향)

  • Purpose: The purpose of this study was to examine effects of decreased locomotor activity on mass, Type I and II fiber cross-sectional areas of ipsilateral and contralateral hindlimb muscles 21 days after establishing the Parkinson's disease rat model. Methods: The rat model was established by direct injection of 6-hydroxydopamine (6-OHDA, 50 ${mu}g$) into the left substantia nigra after stereotaxic surgery. Adult male Sprague-Dawley rats were assigned to one of two groups; the Parkinson's disease group (PD; n=17) and a sham group (S; n=8). Locomotor activity was assessed before and 21 days after the experiment. At 22 days after establishing the rat model, all rats were anesthetized and soleus and plantaris muscles were dissected from both ipsilateral and contralateral sides. The brain was dissected to identify dopaminergic neuronal death of substantia nigra in the PD group. Results: The PD group at 21 days after establishing the Parkinson's disease rat model showed significant decrease in locomotor activity compared with the S group. Weights and Type I and II fiber cross-sectional areas of the contralateral soleus muscle of the PD group were significantly lower than those of the S group. Conclusion: Contralateral soleus muscle atrophy occurs 21 days after establishing the Parkinson's disease rat model.

Digital Squamous Cell Carcinoma in a Miniature Schnauzer (Miniature Schnauzer에서 발생한 발가락 편평세포암종)

  • An 11-year-old, female Miniature Schnauzer dog was presented with recurrent skin ulcer of the second metatarsal region in the right hindlimb following metatarsal resection. Physical examination revealed an ulcerated and bleeding lesion of the second metatarsal region in the right hindlimb. Impression smears of the ulcerative lesion confirmed numerous degenerated neutrophils and mixed bacterial infection. Initially, the dog was treated with antibiotics and povidone-iodine flushing for the control of deep pyoderma. Because the skin lesion had been deteriorated over time despite of topical and systemic treatments, skin biopsy was performed. Histopathologic examination indicated squamous cell carcinoma based on the features of multiple nests of squamous neoplastic cells and mitotic figures. Although amputation of the right hindlimb was performed, the dog was expired five months later because of tumor metastasis to the lung and the popliteal lymph node. This is the first case report describes malignant digital squamous cell carcinoma in Korea.

Combination stem cell therapy using dental pulp stem cells and human umbilical vein endothelial cells for critical hindlimb ischemia

  • Narrowing of arteries supplying blood to the limbs provokes critical hindlimb ischemia (CLI). Although CLI results in irreversible sequelae, such as amputation, few therapeutic options induce the formation of new functional blood vessels. Based on the proangiogenic potentials of stem cells, in this study, it was examined whether a combination of dental pulp stem cells (DPSCs) and human umbilical vein endothelial cells (HUVECs) could result in enhanced therapeutic effects of stem cells for CLI compared with those of DPSCs or HUVECs alone. The DPSCs+ HUVECs combination therapy resulted in significantly higher blood flow and lower ischemia damage than DPSCs or HUVECs alone. The improved therapeutic effects in the DPSCs+ HUVECs group were accompanied by a significantly higher number of microvessels in the ischemic tissue than in the other groups. In vitro proliferation and tube formation assay showed that VEGF in the conditioned media of DPSCs induced proliferation and vessel-like tube formation of HUVECs. Altogether, our results demonstrated that the combination of DPSCs and HUVECs had significantly better therapeutic effects on CLI via VEGF-mediated crosstalk. This combinational strategy could be used to develop novel clinical protocols for CLI proangiogenic regenerative treatments.

Effects of Chaenomelis fructus on the Muscle Atrophy Induced by Hindlimb Suspinsion in Rats (목과(木瓜)가 흰쥐의 후지체공(後肢滯空)에 의한 근강축(筋萎縮)에 미치는 영향(影響))

  • This experimental Study was designed to investigate the effect of Chaenomelis Fructus on the muscle atrophy induced by hindlimb suspension in rats. The result are follows; 1. Chaenomelis Fructus significantl inhibited the increase of the activity of CK in serum. 2. Chaenomelis Fructus significantly inhibited the increase of the quantity of creatine in serum. 3. Chaenomelis Fructus significantl inhibited the increase of the activity of aldolase in serum. 4. Chaenomelis Fructus significantly inhibited the increase of the activity of LDH in serum. 5. Chaenomelis Fructus significantl inhibited the increase of the activity of AST in serum. 6. Chaenomelis Fructus inhibited the increase of the activity of ALT in serum. Based on the these results, it is concluded that the clinical application of Fructus chaenomelis can help cure muscle atrophy.

  • We investigated whether controlled dynamization enhanced regenerative bone healing of the tibia in 12 adult mongrel dogs. An average of 2 cm (∼ 10% of bone length) lengthening was obtained. The left hindlimb was allowed dynamization of 1 mm, 2 mm, and 3 mm in magnitude when 1 cm (∼ 5%) of lengthening was achieved, whereas the right hindlimb was not dynamized and served as a control. Teratogenic and Embryotoxic Effects of Clomiphene Citrate in Developing Mice

  • The objective of this study was to assess the teratogenic and embryotoxic effects of clomiphene citrate in mice. The pregnant mice were administered a single dose of clomiphene citrate at different concentrations i.e 1.0, 2.0, 4.0 and 6.0 ${\mu}g/g$ BW on day 8 of gestation. Fetuses recovered on day 18 of gestation were analyzed on morphological, morphometric and histological basis. Morphological observations showed defects like open eyelids, anophthalmia, fore and hindlimb micromelia, meromelia, amelia, sacral hygroma, hydrocephaly, hemorrhagic spots, kyphosis and clubbed feet. Morphometric analysis indicated a significant (p<0.001) reduction in fetal body weight, crown rump length, head circumference, eye circumference, forelimb and hindlimb lengths and tail size against controls. The histological observations showed brain defects like hydrocephaly, enlarged ventricles and undifferentiated neuroglial cells in cerebellum. Cleft palate, underdeveloped pharynx and atrophy of jaw muscles were the common anatomical defects of pharyngeal region. It is concluded that the concentrations of clomiphene citrate used during the present study proved teratogenic in mice fetuses.

Application of Traditional Acupuncture on Canine Intervertebral Disc Disease (개에서 추간판 질환에 대한 침술치료 증례)

  • A four-year-old female Pekingese dog was presented with intermittent ambulatory hindlimb lameness and severe back pain. The course of the disease was acute and static and 5 days passed by until presentation. In neurological examination, severe back pain and slightly decreased proprioception in right hindlimb were shown. In radiographic examination, radiopacity was increased at T10-11 intervertebral disc space. According to the history taking, neurological examination and radiographic examination, it was tentatively diagnosed as intervertebral disc disease (IVDD). Conservative treatment was performed including cage rest, medication with prednisolone and traditional acupuncture. Acupoints were GV-6, GV-7, BL-18, BL-19. BL-40 and GB-34. Dry needlings rested for 20 minutes. Just after the first acupuncture treatment back pain was significantly decreased. With additional acupuncture treatment, all the clinical signs were not observed. It is considered that conservative therapy including traditional acupuncture, medication and cage rest might be the first choice of therapeutic method for Grade I or II IVDD in dogs.

Polymeria with an Extra Hindlimb at the Pelvic Region, Occured in Korean Native Calf (한우 송아지의 후지에 발생한 과다사지)

  • Congenital anomalies occur at low levels in Korean antive cattle. PCR and Serum neutrlizing test against BVD, Akabane virus, Ibaraki virus, Kasba virus were examined. The results were all negative. Gross anatomical analysis was carried aout. Congenital abnormal newborn calf had 2 atropic left hindlimbs, which was nor moved autonomosly. The length of 2 atropic hindlimbs was 72 em and 48cm respectively. Both of them were curved severely at joints. Affected limbs were developed in connective tissue but lack of muscular tissue. In the abdominal region right kidney was normal. But there were two left kidneys which was half size compared with normal kidney. One of them had polycysts. In skeletal region, affected left limb is defective of head of femur and tibia.
