Effect of $\beta$-glucan Extracted from Youngji Mushroom on the Growth Performance of Weaning Pigs (영지버섯에서 추출한 $\beta$-glucan 이 자돈의 생산능력에 미치는 영향)

  • This experiment was selected a $\beta$-glucan producing mushroom strain and developed industrial media, and used to $\beta$-glucan as an alternative for antibiotics in weaned pigs. Yields of mycelial biomass and extracellular $\beta$-glucan from Youngji (Ganoderma lucidum) mushroom was 8.52g/L and 4.49g/L respectively. Also, we prepared optimum formula for mushroom cultivations. A total of 144 pigs ($8.6{\pm}0.9$ kg average body weight, weaned $20{\pm}3$ days of age) were allotted to 4 different treatment groups and replicated 4 times with 8 pigs per replicate in randomized complete block design. Treatments were T1) NC (negative control, basal diet), T2) PC (positive control, basal diet+0.255% antibiotics), T3) NC+0.2% $\beta$-glucan and T4) PC+0.2% $\beta$-glucan. The T2 and T4 treatments were significantly higher in feed efficiency by antibiotics group (p<0.05), however, there was no significant differences in terms of average daily gain (ADG) and average daily feed intake (ADFI) during phase I (0~14 days). In phase II (15~28 days), Pigs fed with antibiotics and $\beta$-glucan (T4) had greater ADG than other treatments (p<0.05), while no differences were observed in ADFI and feed efficiency. During the whole experiment period, the ADG of T4 treatment was higher than other treatments. Pigs fed with $\beta$-glucan (T3 and T4) had greater diarrhea score and moisture content than other treatments (p<0.05). Pigs fed with $\beta$-glucan (T3 and T4) had greater moisture content than other treatments (p<0.05). However, there was no significant differences in diarrhea score and mortality of weaned pigs. There was marginal reductions in feed cost measured feed cost per weight gain used in antibiotics and $\beta$-glucan added diet during phase I. In the second phase, the treatment supplemented with antibiotics had a significantly lower feed cost per weight gain compared to the other treatments. The results from these experiments suggests that $\beta$-glucan is likely able to improve the growth performance, and reduce feed cost although they do not have similar effects like antibiotics in weaning pigs.

Effects of Dietary β-Glucan on Short Chain Fatty Acids Composition and Intestinal Environment in Rats (식이 베타-글루칸이 흰쥐의 장내 단쇄지방산 조성 및 장내환경 개선에 미치는 영향)

  • The effects of dietary ${\beta}$-glucan, obtained from bacterial fermentation, on the intestinal mass, short chain fatty acids, lactate production and pH in Sprague-Dawley (SD) rats were evaluated. SD rats fed with 0% (control group), 1% or 5% ${\beta}$-glucan supplemented diets (w/w) for 3 weeks. The presence of ${\beta}$-glucan in the diets resulted in a significant increase in colonic contents in a dose dependent manner. The amount of short chain fatty acids increased in rats fed ${\beta}$-glucan diets. Rats fed the 5% ${\beta}$-glucan diets had higher levels of acetate, propionate and butyrate by 1.8, 1.7 and 3.0 fold of the control group in the cecum, and 2.2, 2.9 and 3.1 fold of the control group in the colon, respectively. The ${\beta}$-glucan diets also significantly increased the levels of cecal and colonic lactate by 1.4~3.4 fold, when compared to the control diet, indicating that dietary ${\beta}$-glucan stimulated the growth of lactic acid bacteria within the intestine. These results suggest that dietary ${\beta}$-glucan, by providing short chain fatty acids and reducing the cecal and colonic pH, may be beneficial in improving gut health, and provide evidence for the use of ${\beta}$-glucan as a dietary supplement for human consumption.

Determination of Eleutherosides and β-Glucan Content from Different Parts and Cultivating Areas of A. senticosus and A. koreanum (가시오가피와 탐라오가피의 산지별 및 부위별 Eleutheroside B, E 및 β-Glucan 함량 분석)

  • The present study was performed to quantitatively analyze eleutherosides (B and E) and ${\beta}$-glucan in different plant parts of three cultivars (Chungnam, Gangwon, and Jeju) of Acanthopanax senticosus and Acanthopanax koreanum using HPLC and a commercial enzyme kit. Our results showed high linearity in the calibration curves as the coefficients of correlation ($R^2$) were 0.998 (eleutheroside B) and 0.999 (eleutheroside E), respectively. Eleutheroside B and E were found in stem extracts of A. koreanum cultivated in Jeju (1,122 ${\mu}g/g$, eleutheroside B) and A. senticosus cultivated in Chungnam (2,536 ${\mu}g/g$, eleutheroside E), respectively. However, eleutheroside B was not detected in any part of A. senticosus cultivated in Chungnam. For ${\beta}$-glucan contents, stems of A. senticosus and A. koreanum showed higher than other parts. Furthermore, the ${\beta}$-glucan content in stems of A. koreanum cultivated in Gangwon was significantly higher than in those of other cultivars. These results show that the contents of eleutheroside B, E, and ${\beta}$-glucan were higher in stem extracts of A. senticosus and A. koreanum than other parts. Moreover, our results suggest that the contents of eleutheroside B, E, and ${\beta}$-glucan in A. senticosus and A. koreanum are influenced by cultivation area and the selected part.

Quality Characteristics of Sponge Cake Added with ${\beta}-Glucan$ during Storage (${\beta}-Glucan$ 첨가 스폰지 케이크의 저장 중 품질 특성)

  • The quality characteristics of sponge cake with added ${\beta}-glucan$(2, 4 and 6%) were evaluated. Rapid decreases in moisture content in the sponge cake were observed during storage($20^{\circ}C$, 70% relative humidity) in control and 2% ${\beta}-glucan$ cakes by day 3 of storage, while the moisture content in the 4% and 6% ${\beta}-glucan-added$ sponge cake slowly decreased until day 12 of storage. During storage, hardness was much lower in groups with added ${\beta}-glucan$ than in control. On day 1 L, a and b values of both crust and crumb of sponge cake with added ${\beta}-glucan$ were not significantly different from control, although the a and b values decreased significantly with storage days. Sensory test revealed that the scores for over-all acceptability were much higher in sponge cake with added ${\beta}-glucan$, and the same results were obtained on day 5. Based on these results, ${\beta}-glucan$ addition maintained moisture content and had an overall good effect on sponge cake.

Effects of ${\beta}-glucan-enriched$ Barley Fraction on the Lipid and Cholesterol Contents of Plasma and Feces in Rat (보리 ${\beta}-glucan$ 농축획분 섭취가 흰쥐 혈장과 변의 지질 및 콜레스테롤 함량에 미치는 영향)

  • Cholesterol-lowering effects of ${\beta}-glucan-enriched$ barley fraction were investigated in rats fed 0.5% cholesterol and barley as a fiber source. Male rats of Sprague-Dawley strain were divided into six groups and fed different diets for 5 weeks: normal (cholesterol-free), control (cellulose 5%), fiber-free, and three groups containing ${\beta}-glucan-enriched$ barley fractions. Although plasma cholesterol and triglyceride levels were not significantly different among different diet groups, ${\beta}-glucan-enriched$ barley fraction-fed groups had higher HDL-cholesterol concentrations than the cellulose control group after 5 weeks of experiment. The fecal excretion of cholesterol and triglyceride was significantly increased by barley diets. Fecal concentrations of cholesterol and triglyceride in cellulose control group were 38.2 and 2.6 g/day, respectively, whereas those of barley-fed groups were $52.4{\sim}59.2$ and $6.8{\sim}9.5$ g/day, respectively, during the experimental period.

Effects of Extrusion and Enzyme Treatment on Extraction of β-Glucan from Agaricus blazei Murill (압출성형과 효소처리가 신령버섯 β-Glucan의 추출에 미치는 영향)

  • This study analyzed changes in ${\beta}$-glucan content in Agaricus blazei Murill concentrates according to extrusion and extraction conditions. Screw speed and feed rate were fixed to 250 rpm, and 100 g/min, respectively. Moisture contents (20 and 30%) and barrel temperature (130 and $140^{\circ}C$) were adjusted. ${\beta}$-Glucan content of the extruded sample at a moisture content of 20% and barrel temperature of $130^{\circ}C$ was higher compared to other extrusion conditions. ${\beta}$-Glucan content of the extruded sample at a moisture content of 20% and barrel temperature of $130^{\circ}C$ treated with Rohament CL enzyme was higher compared to Viscozyme L, and Plantase TL enzyme treatments under the same extrusion conditions. In conclusion, extrusion and pretreatment with Rohament CL enzyme enhanced yield of ${\beta}$-glucan extract.

  • The activity of Daucus carota L. root $\beta$-glucan synthetase II was observed to increase in the presence of polyamines such as putrescine, spermidine and spermine in vitro, whereas the activity of Daucus carota L. root $\beta$-glucan synthetase I was not affected by the polyamine. The activity $\beta$-glucan synthetase II from Daucus carota L. root protoplasts cultured on medium containing 10-6 M polyamines such as putrescine, spermidine and spermine was observed to be higher than that of the control. Daucus carota L. root protoplasts were observed to have the activities of arginine and ornithine decarboxylases and it was noted that they could produce polyamines, which might have an effect on $\beta$-glucan synthetase II activity.

Physiological Function in vitro of ${\beta}-Glucan$ Isolated from Barley (보리가루에서 분리한 ${\beta}-Glucan$의 생리적 기능성)

  • In order to prove physiological function of ${\beta}-Glucan$ isolated from barley flour by enzymatic method, in vitro experiments simulating the passive membrane transport of gastrointestinal tract were carried out using dialysis membrane. The yield of ${\beta}-Glucan$ from barley flour was $6.2{\%}$ and its constituents were determined to give $81.6{\%}$ total dietary fiber, $72.9{\%}$ soluble dietary fiber, $8.7{\%}$ insoluble dietary fiber, $8.5{\%}$ moisture, $2.5{\%}$ protein and $7.4{\%}$ ash. The water holding capacity of the ${\beta}-Glucan$ preparation was 6 g water/g dry material. The glucose retardation index after 30 minute dialysis was $13.5{\%}$ in the presence of $3{\%}$ ${\beta}-Glucan$. As the dialysis period became longer, the retarding effect toward glucose absorption decreased and the effect was close to zero after 2 hour dialysis. The bile acid retardation index after 30 minute dialysis was 3, 12 and $18{\%}$ in the presence of 1, 3 and $5{\%}$ ${\beta}-Glucan$, respectively. The effect was higher than the glucose retardation index and decreased as the dialysis time elapsed.

Quality Characteristics of Pound Cake Added to $\beta$-Glucan During Storage ($\beta$-Glucan 첨가 파운드 케이크의 저장 중 품질 특성)

  • The aim of this study was to evaluate the quality characteristics of pound cake made of flour substituted with $\beta$-glucan (3, 6 and $9\%$), which is a functional fuod material produced from Agrobacterium Spp. R259 KCTC 10197BP. A rapid decrease of moisture content during storage at $20^{\circ}C$, $70\%$ relative humidity, was observed in the control while the moisture content in 3 and $6\%$ $\beta$-glucan added pound cake was not decreased until the 12th day of storage. During storage, the hardness of the control cake was increased whereas that of $\beta$-glucan $6\%$ and $9\%$ added pound cake was not changed until the 6th day of storage. The addition of $\beta$-glucan was therefore conflmed to delay the staling of cake. During storage, the Hunter color L ud b values of the crust and crumb colors of the $\beta$-glucan added pound cake were not significantly different from those of the control, although the a value did increase with longer storage. The sensory results showed that the scores of over-all acceptability of the 3 and $6\%$ $\beta$-glucan added pound cake at the 6th day of storage days were higher than that of the control. These study results confirmed that the addition of $\beta$-glucan to pound cake maintained the moisture content and delayed hardness during storage.

The effects of $\beta$-1,3/1,6-linked glucan in the diet on immune responses of olive flounder, Paralichthys olivaceus by oral admimistration ($\beta$-1,3 / 1,6-linked glucan 첨가사료가 넙치의 비특이적 면역반응에 미치는 영향)

  • The effects of dietary $\beta$-1,3/1,6-linked glucan administration on nonspecific immune responses, hematology and disease resistance against Edwardsiella tarda in olive flounder, Paralichthys olivaceus were evaluated. Fish were fed the experimental diets supplemented with 0 %, 0.01 %, 0.05 %, 0.1 %, and 0.5 % of $\beta$-glucan to a commercial diet for 7 weeks. The test was performed in two ways such as fed on each $\beta$-glucan concentrations daily and every other week alternately. The body weight gain from the fish fed on daily the 0.05 % supplemented diet and fed on alternately the 0.1 % supplemented diet of $\beta$-glucan were significantly higher than the control. Both lysozyme activity and intracellular superoxide anion production of kidney phagocytes were higher in the all experimental groups than in the control. But there was no large difference in hematology among each group. The relative percent survival rate (RPS) after an artificial challenge with $4{\times}10^6$ cells of E. tarda per fish was higher than the control, except for that fed on daily the 0.01 % supplemented diet.
