Relationship between Intersequence Pauses, Laying Persistency and Concentration of Prolactin during the Productive Period in White Leghorn Hens

  • Prolactin is considered to influence the taking of pauses in between ovulatory sequences in White Leghorn hens. Therefore modulating concentrations of prolactin using bromocriptine - a dopamine agonist during early life (17 to 36 weeks of age) could overcome the inhibitory effects of high concentration of prolactin on ovarian activity. The effect of modulation of prolactin concentration on egg production, sequence length and inter sequence pauses were studied by analyzing the oviposition records from 19 to 72 weeks were studied and compared with untreated controls. Bromocriptine administered subcutaneously (100 $\mu$g kg$^{-1}$ body weight or orally through feed (640 $\mu$g day$^{-1}$ bird$^{-1}$) resulted in a steady and sustained decrease in prolactin levels (p<0.01) during and after the withdrawal of treatment up to one reproductive cycle (72 weeks of age). The treated birds had comparatively longer sequences (p<0.01) and fewer pauses (p<0.01). Egg production increased (p<0.01) by fourteen per cent through subcutaneous administration and eleven per cent through oral feeding, over the control birds. It is concluded that the physiological pauses that occur during ovulatory sequences can be disrupted effectively using bromocriptine. Prolactin levels are modulated which may interfere with the follicular recruitment and subsequent oviposition thereby improve egg laying potential of the bird.

  • A. niger의 GOD(Glucose Oxidase) 대량생산과 효율적인 분비를 protein의 대량생산에 많이 사용되는 strain인 S. cerevisiae에서 시도하였다. S. cerevisiae의 ADH1과 GAL 10 promotor, 그리고 ${alpha}$-MF signal sequence와 A. oryzae의 ${alpha}$-amylase signal sequence 및 S. cerevisiae의 GAL7과 A. niger의 GOD terminator를 이용하여 4개의 expression vector를 합성한 후 S. cerevisiae 2805에 auxotroph 방법으로 형질변환시켰다. 변이체들을 배양하여 세포내와 세포외의 GOD활성도를 분석한 결과 GAL 10 promotor가 삽입된 pGAL변이체들이 ADH1 promotor가 삽입된 pADH 변이체들 보다 GOD 생산성이 높았다. GAL 10 promotor와 A. oryzae의 ${alpha}$-amylase signal sequence가 삽입된 pGALGO2에서 115시간 배양시 GOD의 생산이 가장 높았다($GOD_{total}$: 10.3 unit/mL, $GOD_{ex}$: 8.7 unit/mL). 이 수치는 같은 promotor인 GAL 10 promotor와 ${alpha}$-MF signal sequence가 삽입된 pGALGO1보다 3배정도 높다. 이 결과는 ADH 1 promotor를 사용하였을 경우에도 일치하였다. 또한 A. oryzae의 ${alpha}$-amylase signal sequence가 S. cerevisiae의 ${alpha}$-MF signal sequence보다 GOD를 더 효과적으로 분비시켰다. 상기 결과로 미루어 보면 signal sequence가 단백질의 분비 외에도 단백질 합성에도 많은 영향을 주는 것으로 추측된다. pGALGO1과 pGALGO2의 GOD분비효율은 각각 89%, 84%이었다. S. cerevisiae에서는 일반적으로 과당화가 일어나기 때문에 S. cerevisiae에서 합성된 재조합 GOD의 분자량은 250 kDa으로 A. niger의 GOD(170 kDa)보다 더 컸다.

Load-Balance-Independent High Efficiency Single-Inductor Multiple-Output (SIMO) DC-DC Converters

  • A single-inductor multiple-output (SIMO) DC-DC converter providing buck and boost outputs with a new switching sequence is presented. In the proposed switching sequence, which does not require any additional blocks, input energy is delivered to outputs continuously by flowing current through the inductor, which leads to high conversion efficiency regardless of the balance between the buck and boost output loads. Furthermore, instead of multiple output loop compensation, only the freewheeling current feedback loop is compensated, which minimizes the number of off-chip components and nullifies the need for the equivalent series resistance (ESR) of the output capacitor for loop compensation. Therefore, power conversion efficiency and output voltage ripples can be improved and minimized, respectively. Implemented in a 0.35-${\mu}m$ CMOS, the proposed SIMO DC-DC converter achieves high conversion efficiency regardless of the load balance between the two outputs with maximum efficiency reaching up to 82% under heavy loads.

DNA Structural Perturbation Induced by the CPI-Derived DNA Interstrand Cross-linker : Molecular Mechanisms for the Sequence Specific Recognition

  • The highly potent cytotoxic DNA-DNA cross-linker consists of two cyclopropa[c]pyrrolo[3,4-3]indol-4(5H)-ones insoles [(+)-CPI-I] joined by a bisamido pyrrole (abbreviated to "Pyrrole"). The Pyrrole is a synthetic analog of Bizelesin, which is currently in phase II clinical trials due to its excellent in vivo antitumor activity. The Pyrrole has 10 times more potent cytotoxicity than Bizelesin and mostly form DNA-DNA interstrand cross-links through the N3 of adenines spaced 7 bp apart. The Pyrrole requires a centrally positioned GC base pair for high cross-linking reactivity (i.e., $5^1$-T$AT_2$A*-$3^1$), while Bizelesin prefers purely AT-rich sequences (i.e., $5^1$-T$AT_4$A*-$3^1$, where /(equation omitted) represents the cross-strand adenine alkylation and A* represents an adenine alkylation) (Park et al., 1996). In this study, the high-field $^1$H-NMR and rMD studies are conducted on the 1 1-mer DNA duplex adduct of the Pyrrole where the 5′(equation omitted)TAGTTA*-3′sequence is cross-linked by the drug. A severe structural perturbation is observed in the intervening sequences of cross-linking site, while a normal B-DNA structure is maintained in the region next to the drug-modified adenines. Based upon these observations, we propose that the interplay between the bisamido pyrrole unit of the drug and central C/C base pair (hydrogen-bonding interactions) is involved in the process of cross-linking reaction, and sequence specificity is the outcome of those interactions. This study suggests a mechanism for the sequence specific cross-linking reaction of the Pyrrole, and provides a further insight to develop new DNA sequence selective and distortive cross-linking agents.

  • 본 논문에서는 자동표정인식을 위하여 얼굴 이미지 배열의 가운데 이미지를 예측하는 새롭고 간단한 자기주도학습 방법을 제안한다. 자동표정인식은 딥러닝 모델을 통해 높은 성능을 달성할 수 있으나 일반적으로 큰 비용과 시간이 투자된 대용량의 데이터 세트가 필요하고, 데이터 세트의 크기와 알고리즘의 성능이 비례한다. 제안하는 방법은 추가적인 데이터 세트 구축 없이 기존의 데이터 세트를 활용하여 자기주도학습을 통해 얼굴의 잠재적인 심층표현방법을 학습하고 학습된 파라미터를 전이시켜 자동표정인식의 성능을 향상한다. 제안한 방법은 CK+와 AFEW 8.0 두가지 데이터 세트에 대하여 높은 성능 향상을 보여주었고, 간단한 방법으로 큰 효과를 얻을 수 있음을 보여주었다.

Applied Computational Tools for Crop Genome Research

  • A major goal of agricultural biotechnology is the discovery of genes or genetic loci which are associated with characteristics beneficial to crop production. This knowledge of genetic loci may then be applied to improve crop breeding. Agriculturally important genes may also benefit crop production through transgenic technologies. Recent years have seen an application of high throughput technologies to agricultural biotechnology leading to the production of large amounts of genomic data. The challenge today is the effective structuring of this data to permit researchers to search, filter and importantly, make robust associations within a wide variety of datasets. At the Plant Biotechnology Centre, Primary Industries Research Victoria in Melbourne, Australia, we have developed a series of tools and computational pipelines to assist in the processing and structuring of genomic data to aid its application to agricultural biotechnology resear-ch. These tools include a sequence database, ASTRA, for the processing and annotation of expressed sequence tag data. Tools have also been developed for the discovery of simple sequence repeat (SSR) and single nucleotide polymorphism (SNP) molecular markers from large sequence datasets. Application of these tools to Brassica research has assisted in the production of genetic and comparative physical maps as well as candidate gene discovery for a range of agronomically important traits.

Nucleotide Sequence of Leghemoglobin cDNA from Canavalia lineata

  • Poly(A)+ RNA was selected from Canavalia lineata root nodule RNA through oligo(dT) cellulose column and used for construction of a cDNA library using λgt10-EcoRI arms. The size of the library was 7.2$\times$105 pfu/mL. A full length leghemoglobin (Lb) cDNA clone, pCILb1(687 bp) isolated with soybean Lb probe, contained one open reading frame (ORF) of 447 bp with 54 bp plus 186 bp at 5' and 3' untranslated region, respectively. A consensus sequence of plant translation start region (AAAATGGG) was found at 5' untranslated region, and two polyadenylation-related sequence (AATAAA, AATAAG) and a conserved motif between them (gACTTGTT) were found upstream of poly(A)+ tail consisted of 13 (A)s at 3' untranslated region. The ORF encoded a polypeptide consisted of 149 amino acids with a molecular weight of 16.2 kD. Deduced amino acid sequences showed high degree of homology values with those of other Lbs ranging from 66% (Casuarina glauca) to 85% (Glycine max).

  • We present a tabu search (TS) algorithm to minimize maximum lateness on a single machine in the presence of sequence dependent setup times and dynamic job arrivals. The TS algorithm starts with a feasible schedule generated by a modified ATCS (Apparent Tardiness Cost with Setups) rule, then through a series of search steps it improves the initial schedule. Results of extensive computational experiments show that the TS algorithm significantly outperforms a well-known RHP heuristic by Ovacik and Uzsoy, both on the solutions quality and the computation time. The performance advantage is particularly pronounced when there is high competition among jobs for machine capacity.

  • The DS-SS (Direct Sequence Spread Spec1nun) transmitter is part of a low data rate (∼150 kbps - burst rate and 64 bps - average data rate) wireless communication system. It is traditionally implemented using Digital Signal processing chip (DSP). However, with rapid increase in variety of services through cell phones, such as, web access, video transfer, online games etc. demand for higher rate is increasing steadily. Since the chip rate and thereby the sampling rate requirements of the system are fairly high, the transmitter should implemented using Field programmable Gate Arrays FPGAs instead of a DSP. This paper shows the steps taken to get a working prototype of the transmitter unit on a FPGA based platform.

  • In this paper a new method for head gesture recognition is proposed. A the first stage, face image data are transformed into low dimensional vectors by principal component analysis (PCA), which utilizes the high correlation between face pose images. The a self organization map(SM) is trained by the transformed face vectors, in such a that the nodes at similar locations respond to similar poses. A sequence of poses which comprises each model gesture goes through PCA and SOM, and the result is stored in the database. At the recognition stage any sequence of frames goes through the PCA and SOM, and the result is compared with the model gesture stored in the database. To improve robustness of classification, probabilistic relaxation labeling(PRL) is used, which utilizes the contextural information imbedded in the adjacent poses.
