Effect of NADH-Dependent Enzymes Related to Oxygen Metabolism on Elimination of Oxygen-Stress of Bifidobacteria (NADH요구 산소대사관련 효소가 bifidobacteria의 산소스트레스 제거에 미치는 영향)

  • Selection of oxygen-tolerant strains and elucidation of their oxygen tolerance mechanism were crucial for effective use of bifidobacteria. Oxygen-tolerant bifidobacteria were able to significantly remove environmental oxygen (oxygen removal activity) as compared to oxygen-sensitive strains. Most oxygen removal activity was inhibited by heat treatment and exposure to extreme pH (2.0) of bifidobacterial cell. NADH oxidase was major enzyme related to oxygen removal activity. Oxygen-tolerant bifidobacteria possessed high NADH peroxidase activity level to detoxify $H_2O_2$ formed from reaction of NADH oxidase. Addition of oxygen to anaerobic culture broth significantly increased activities of HADH oxidase and NADH peroxidase within 1hr and rapid increment of oxygen concentration was prevented. Results showed NADH oxidase and NADH peroxidase of oxygen-tolerant bifidobacteria played important roles in elimination of oxygen and oxygen metabolite $(H_2O_2)$.

Development of low power type sensor for the DO concentration measurement by clark electrode (Clark전극에 의한 DO 농도측정을 위한 절전형 센서개발에 관한 연구)

  • A method is described for the design and fabrication of the sensor interface circuits on the Clark electrodes for the dissolved oxygen(DO). The discussion includes a method for the +5 V single-supply driving for the sensor circuits, which has low power comsumption for the front-end electronics. DO probe under test is composed of the Clark electrode with silver anode, gold cathode and the electrolyte of half saturated KCI solution and the FEP teflon memtrance for the oxygen penetration. Typical polarograms for the DO probes by using this sensor circuit reveals high accuracy over 99% of the I to V conversion. Partial pressure of oxygen obtained from the polarograms are well suited to the results calculated. It is expected that the proposed sensor circuits can be utilized into the customized IC for the battery-driven small-size DO meters.

  • In this study, plasma spraying has been used to produce $TiO_2$ polycrystalline coatings from $TiO_2$ powders. The physical and chemical properties of plasma sprayed $TiO_2$ coatings depend greatly on plasma spraying conditions. The electrical resistivity, oxygen concentration, photocurrent and crystal structure of plasma sprayed $TiO_2$ coating has been studied. The results are as follows: 1. The oxygen loss and electrical conductivity of $TiO_2$ plasma sprayed coatings increased by low pressure and high amount of auxiliary gas, hydrogen in plasma spraying. 2. Oxygen loss increase electrical conductivity, and decrease photocurrent of $TiO_2$ plasma sprayed coatings. 3. Photocurrent of $TiO_2$ plasma sprayed coatings manufactured in atmospheric pressure is higher than that of low pressure.

  • Non-enzymatic glycosylation and fragmentation of collagen molecule were investigated by irradiating Ultraviolet A(UV-A) with or without scavengers of reactive oxygen species (ROS) in the presence of glucose. Non-enzymatic glycosylation was increased by UV-A at high concentration of glucose. It was reduced in the presence of the scavengers of superoxide radical and singlet oxygen, but not reduced in the presence of hydroxy radical scavenger. Fragmentation of collagen was increased by UV-A, but it was decreased in the presence of all ROS scavengers tested. Superoxide radical and singlet oxygen produced by autoxidation of glucose without UV-A may encounter the initial phase of glycosylation. Data presented here suggest that UV-A affects only on the fragmentation process, but all ROS except hydroxy radical act on both processes. It appears that hydroxy radical does not act on the glycosylation process.

Method for Improvement of Reduction Reactivity at High Temperature in a Chemical-Looping Combustor (매체순환식 가스연소기에서 고온 환원반응성 증대 방법)

  • When we use NiO based particle as an oxygen carrier in a chemical looping combustion system, the fuel conversion and the $CO_2$ selectivity decreased with increasing reaction temperature within high temperature range (> $900^{\circ}C$) due to the increment of exhaust CO concentration from reduction reactor. To improve reduction reactivity at high temperature, the applicable metal oxide component was selected by calculation of the equilibrium CO concentration of metal oxide components. After that, feasibility of reduction reactivity improvement at high temperature was checked by using solid mixture of the selected metal oxide particle and NiO based oxygen carrier. The reactivity was measured and investigated using batch type fluidized bed. The solid mixture of $Co_3O_4/CoAl_2O_4$(10%) and OCN706-1100(90%) showed higher fuel conversion, higher $CO_2$ selectivity and lower CO concentration than OCN706-1100(100%) cases. Consequently, we could conclude that improvement of reduction reactivity at high temperature range by adding some $Co_3O_4$ based oxygen carrier was feasible.

  • Prestressed Concrete steel Cylinder Pipe (PCCP) is extensively used as seawater pipes for cooling in nuclear power plants. The internal surface of PCCP is exposed to seawater, while the external surface is in direct contact with underground soil. Therefore, materials and strategies that would reduce the corrosion of its cylindrical steel body and external steel wiring need to be employed. To prevent against the failure of PCCP, operators provided a cathodic protection to the pre-stressing wires. The efficiency of cathodic protection is governed by the anodic performance of the system. A mixed metal oxide (MMO) electrode was developed to meet criteria of low over potential and high corrosion resistance. Increasing coating cycles improved the performance of the anode, but cycling should be minimized due to high materials cost. In this work, the effects of $RuCl_3$ concentration on the electrochemical properties and lifespan of MMO anode were evaluated. With increasing concentration of $RuCl_3$, the oxygen evolution potential lowered and polarization resistance were also reduced but demonstrated an increase in passive current density and oxygen evolution current density. To improve the electrochemical properties of the MMO anode, $RuCl_3$ concentration was increased. As a result, the number of required coating cycles were reduced substantially and the MMO anode achieved an excellent lifespan of over 80 years. Thus, we concluded that the relationship between $RuCl_3$ concentration and coating cycles can be summarized as follows: No. of coating cycle = 0.48*[$RuCl_3$ concentration, $M]^{-0.97}$.

  • The closed cycle diesel system is operated in closed circuit system where there is non air breathing with working fluid consists of combination of oxygen, argon and recycled exhaust gas far obtaining underwater or underground power sources. Experimental apparatus using the MTU8V183SE92 high pressurized engine adapted for closed cycle running, capable of operating at the system pressure of maximum 5 bar is constructed with ACAP as data acquisition system in order to operate equally in the open cycle in surface or the closed cycle in submerged conditions. The general features and the characteristics of combustion of HP(high pressure) diesel engine, specially designed and manufactured only for CCDE, are investigated. The test results of performance of HP diesel engine in closed cycle system are presented with respect to power and boost pressure and compared with those of low pressure diesel engine. Studies of the Effects of Hexachlorophene on the $C^{14}-Glucose$ and $C^{14}-Glucose$ Metabolism of Clonorchis Sinensis (Hexachlorophene의 간흡충(肝吸?)의 $C-^{14}$ Glucose 및 $C-^{14}$ Glycine 대사(代謝)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • Hexachlorophene was reported previously to have a powerful parasiticidal effects on Clonorchis sinensis in vitro and in vivo, but the mechanism of its effect was not known. In the present report it was observed that there was an influence of hexachlorophene on the oxygen consumption, the glycolysis, the glycogenesis and the protein synthesis of C. sinensis. A hundred mg. of C. sinensis collected from the biliary tracts of the infested rabbits was incubated in 2 ml of K.R.P. medium with vavious concentration of hexachlorophene, $glucose-1-C^{14}$ and $glycine-1-C^{14}$ in a 25 ml incubation flask with central well. The oxygen consumption was observed by Warburg manometer, the glycogenolysis by measurement of radioactivities of extracted glycogen and protein from C. sinensis incubated with $C^{14}-glucose$ or$C^{14}-glycine$. 1) The oxygen consumption by C. sinensis was markedly inhibited during all stages of incubation in concentration of $10^{-4}$ and $10^{-5}g/ml$ of hexachlorophene, but in $10^{-6}$, slightly increased initially and gradually decreased after 3 hours of incubation. 2) Hexachlorophene inhibited the glycolysis by C. sincnsis markedly in the concentration of $10^{-4}$, $10^{-5}$, $10^{-6}$ and $10^{-7}g/ml$. 3) The protein synthesis by C. sinensis from glycine was inhibited in the concentration of $10^{-5}$, $10^{-6}$ and $10^{-7}g/ml$ of hexachlorophene. 4. The glycogen synthesis by C. sinensis in each concentration of $10^{-4}$, $10^{-5}$ and $10^{-6}g/ml$ of hexachlorophene was inhibited markedly. The speed of inhibition was more rapid in high concentration than in low, and in low concentration even the glycogen itself which had synthesized in their stage in their body was consumed in later stage. 5) The effects of oxygen consumption, glycolysis and glycogen synthesis were not influenced in the concentration of $10^{-5}g/ml$ of chloroquine phosphate, whereas hexachlorophene and dithiazanine inhibited markedly in same concentration, and the former was more potent than the latter.

  • This study performs the pilot-plant experiments to evaluate the effect of the oxygen enrichment on the co-incineration of municipal solid waste and organic sludge from a wastewater treatment facility. The design capacity of the stoker-type incinerator pilot-plant is 150 kg/h. Combustion chamber temperatures were measured as well as the stack gas concentrations, i.e., NOx, CO, and the residual oxygen. The maximum ratio of organic sludge waste to the total waste input is 30%. Also the oxygen-enriched air with 23% of oxygen in supplied air is used for stable combustion. As the co-incineration ratio of the sludge increased up to 30% of the total waste input, the primary and the secondary combustion chamber temperature was decreased $to900^{\circ}C$ (primary combustion chamber), $750^{\circ}C$(secondary combustion chamber), respectively, approximately $200^{\circ}C$ below the incineration temperature of the domestic waste only (primary: $1,100^{\circ}C$, secondary: $950^{\circ}C$). However, if the supplied air was enriched to 22% oxygen content in air, the incinerator temperature was high enough to burn the waste mixture with 30% sludge, which has the heating value of 1,600 kcal/kg.

  • Because of the low temperature operation, proton exchange membrane (PEM) fuel cell has a water phase transition. Therefore, water management is an important operation issue in a PEM fuel cell because the liquid water in the fuel cell causes electrode flooding that can lower the cell performance under high current density conditions. In this study, in order to understand the reactant distributions in the cathode channels of the PEM fuel cell, an experimental technique that can measure the species concentrations of reactant gases by using gas chromatograph (GC) is applied for an operating PEM fuel cell. The oxygen distribution along the cathode flow channels of PEM fuel cell is mainly investigated with various operating conditions. Also, the relations between cathode flooding and oxygen concentrations and oxygen consumption pattern along the cathode channel configurations of the unit cell adopted for this study are discussed using GC measurement and visualization experiment of cathode flooding. It is found that the amount of oxygen consumption is very sensitive to various operating conditions of the fuel cell and was much affected by the flooding occurrence in cathode channels.
