A Study on Variation of Colony Forming Units of Heterotrophic Bacteria by Input Ratios of Bulking Materials in Aerobic Composting of Food Wastes (음식물류폐기물의 호기성 퇴비화에 있어서 팽화재 투입비에 따른 타가영양세균의 균락형성단위의 변화에 관한 연구)

  • This study was performed to evaluate the effects of input ratios of bulking material in aerobic composting of food wastes on variation of colony forming units of heterotrophic bacteria. Wood chips were used as a bulking material. Volume ratios of food wastes to wood chips in reactor of Control, WC-1 and WC-2 were 10/0, 10/5 and 10/10, respectively. Reactors were operated for 24 days with 1hour stirring by 1rpm and 2 hours of the forced aeration per day. WC-2 reached high temperature range faster than WC-1, and the maximum temperature of WC-2 was higher than that of WC-1. This means that the reaction velocity of composting of WC-2 was faster than that of WC-1. Judging from the profile of pH changes, composting of WC-1 proceeded slowly and continuously. Composting of WC-2 proceeded rapidly in the former half reaction period, and kept steady state of high pH in the latter half reaction period. Namely, composting of WC-2 was nearly completed in the former half reaction period. In the case of WC-1 and WC-2. the maximum temperature was followed by the rapid pH increase in 2-3 days, and this was followed by the maximum Colony Forming Units(CFU) in 3 days. But, these three items of WC-2 always appeared faster and higher than those of WC-1.

Circadian Rhythms of Melatonin, Thyroid-Stimulating Hormone and Body Temperature: Relationships among those Rhythms and Effect of Sleep-Wake Cycle

  • Plasma melatonin, thyroid-stimulating hormone (TSH) and body temperature were measured simultaneously and continuously before and after the sleep-wake cycle was shifted in 4 healthy males and changes in the circadian rhythm itself and in the phase relationship among these circadian rhythms were determined. Normal sleep-wake cycle (sleep hours: 2300-0700) was delayed by 10 h (sleep hours: 0900-1700) during the experiment. Even after this shift the typical melatonin rhythm was maintained: low during daytime and high during night. The melatonin rhythm was gradually delayed day by day. The TSH rhythm was also maintained fundamentally during 3 consecutive days of altered sleep-wake cycle. The phase was also delayed gradually but remarkably. The daily rhythm of body temperature was changed by the alteration of sleep-wake cycle. The body temperature began to decrease at the similar clock time as in the control but the decline during night awake period was less steep and the lowered body temperature persisted during sleep. The hormonal profiles during the days of shifted sleep/wake cycle suggest that plasma melatonin and TSH rhythms are basically regulated by an endogenous biological clock. The parallel phase shift of melatonin and TSH upon the change in sleep-wake cycle suggests that a common unitary pacemaker probably regulates these two rhythms. The reversal phase relationship between body temperature and melatonin suggests that melatonin may have a hypothermic effect on body temperature. The altered body temperature rhythm suggests that the awake status during night may inhibit the circadian decrease in body temperature and that sleep sustains the lowered body temperature. It is probable but uncertain that there ave causal relationships among sleep, melatonin, TSH, and body temperature.

A Study on the Spalling Properties of High-Performance Concrete with the Kinds of Aggregate and Polypropylene Fiber Contents (골재종류 및 폴리프로필렌 섬유 혼입률 변화에 따른 고성능 콘크리트의 폭열 특성에 관한 연구)

  • A spalling is defined as the damages of concrete exposed to high temperature during the fire by causing cracks and localized bursting of small pieces of concrete. It is reported that spalling is caused by the vapor pressure and polypropylene(PP) fiber has an important role in protecting from spalling. This paper is a study on the properties and spalling resistance of high-performance concrete with the kinds of aggregate and the contents of PP fiber. According to the experimental results, concrete contained no PP fiber take place in the form of the surface spalling and the failure of specimenns after fire test regardless of the kinds of aggregate. Concrete contained more than 0.05% of PP fiber with the aggregate of basalt does not take place the spalling, while the concrete using granite and limestone does the surface spalling. It is found that residual compressive strength after exposed at high temperature has 50~60% of its original strength. Although specimens after exposed at high temperature is cured at water for 28days, they do not recover their original strength.

Comparative Analysis of the Correlation Between Local Meteorological Elements and the Concentration of Airborne Fungi in the Beopjusa and Seonamsa Temples

  • This study involved a comparative analysis of the correlation between meteorological elements and the concentration of airborne fungi(CFU) in relation to biological damage to two temples on piedmonts, which is a cultural heritage building. The work compared Beopjusa temple in Boeun(Chungcheongbuk-do Province) and Seonamsa temple in Suncheon(Jeollanam-do Province). Twelve meteorological elements and the CFU were measured and the Pearson correlation analysis was used to determine the degree of the relationship between them. The results showed that Beopjusa temple had high wind speed, high total horizontal radiation, high evaporation, and large number of days with precipitation. Seonamsa temple had high air temperature, high relative humidity, high dew point temperature, high sea level pressure, high precipitation, and high CFU. The CFU at Beopjusa temple did not have a linear correlation with meteorological elements, but at Seonamsa temple it was highly positive correlated with the number of days with precipitation, relative humidity, and precipitation, and was highly negative correlated with total horizontal radiation. In addition, Beopju and Seonamsa temple had a common linear relationship between factors not affected by the topographical conditions, and had a individual linear correlation between factors affected by the topographical conditions.

Effects of Shelf Temperature on the Quality of Red Pepper (Capsicum annuum L.) after Low Temperature Storage (홍고추의 저온저장 후 모의 유통온도가 품질에 미치는 영향)

  • The study was conducted to investigate quality changes of red pepper by different shelf temperatures after storage at $7^{\circ}C$ for 30 days. Red pepper fruits produced high volume of $C_2H_4$ and $CO_2$ when alternating temperatures exceeded $10^{\circ}C$. Total sugar contents were not different among all the treatments. In contrast, sucrose content decreased about half at higher shelf temperature conditions than at $7^{\circ}C$. Capsaicinoid content was not influenced by alternating temperature, but ascorbic acid content tended to decrease by high shelf temperature after storage at $7^{\circ}C$. Decay did not occur under $13^{\circ}C$, while it occurred in 22.2% of hits at $15^{\circ}C$ and 68.8% at room temperature, respectively. From this results, it is suggested that the optimum shelf temperature of red pepper after low temperature storage should be under $13^{\circ}C$ and that fruits of red pepper stored at $7^{\circ}C$ for 30 days could be sold within 10 days on the shelf.

Effect of Fermentation Temperature on the Physicochemical Properties of Mustard Leaf(Brassica juncea) Kimchi during Various Storage Days (발효숙성온도를 달리한 갓 김치의 저장중 이화학적 특성 변화)

  • The effect of fermentation temperature on the changes of pH, acidity, salt content, color and vitamin C of mustard leaf kimchi during various storage days was investigated. The conditions of fermentation temperature were set at $4^{\circ}C$ (sample A) and $20^{\circ}C$ (sample B), and $4^{\circ}C$ after keeping at $20^{\circ}C$ for 12 hours(sample C) and $20^{\circ}C$ for 36 hours(sample D). As the fermentation proceeded, pH of sample stored at low temperature(sample A) was drop ped gradually from initial pH of 5.24 but there was great pH drop in the sample stored at high temperature(sample B, D). The salt content of the sample B at high temperature increased remarkably, and then the values showed D > A > C. The Humter values of L and a increased at the optimum ripening period, the higher the initial fermentation temperature(B) and the later the initial fermentation time at $20^{\circ}C$ those values, then decreased. The Hunter value of b constantly increased until day of 108. As fermentation time passed, the content of total vitamin C decreased to the range of 9.0mg% to 14.0mg% up to 24 days of fermentation, and at the optimum ripening period, it increased to the range of 14.0mg% to 22.0mg%, and at the fermentation period(until day 108), it decreased gradually.

  • This paper presents fundamental experiment for the properties of high performance concrete in its fresh and hardened state made with ground granulated blast-furnace (GGBF) slag. The result is that the effect of decreasing xoncrete temperature is to the mixing ratio of GGBF slag, but it presents disadvantage in the slump loss phase. In addition to, we know that the splitting tensile strength, compressive strength and elastic modulus of concrete mixed with high fineness GGBF slag are increased at age 28days.

Effect of temperature on fractional precipitation for paclitaxel purification

  • Fractional precipitation is a simple, efficient method for pre-purifying paclitaxel from plant cell cultures. The storage temperature of factional precipitation was optimized in terms of the yield and purity of paclitaxel with a fixed methanol concentration in water (61.5%, v/v), paclitaxel content in the crude extract (0.5%, w/v), and storage time (3 days). The greatest yield $({\sim}84%)$ was obtained with storage at a constant temperature $(0^{\circ}C)$ for 3 days. Conversely, the highest purity $({\sim}79%)$ was obtained with stepwise reduction in temperature over 3 days. For a constant storage temperature, the maximum purity $({\sim}67%)$ was obtained at $0^{\circ}C$. There was little difference in the yield of paclitaxel between -20 and $12^{\circ}C$. This pre-purification process serves to minimize solvent usage, and the size and complexity of the high-performance liquid chromatography (HPLC) operation required for paclitaxel purification.

Difference in Physiological Responses to Environmental Stress in Protox Inhibitor Herbicide-Resistant Transgenic Rice and Non-transgenic Rice (Protox 저해형 제초제 저항성 형질전환벼와 비형질전환벼의 환경스트레스에 대한 생리적 반응 차이)

  • The objective of this research was to confirm the difference in physiological responses to environmental stresses such as chilling, high temperature, NaCl, and chemical stress (paraquat) in Protox inhibitor resistant-transgenic rice (MX, PX, and AP37) and its non-transgenic counterpart (WT). Transgenic and non-transgenic rice plants were exposed to a chilling temperature of $5^{\circ}C$ for 1 day or a high temperature of $45^{\circ}C$ for 4 days and allowed to recover at $25^{\circ}C$ for 6 days after the chilling treatment or 8 days after the high temperature treatment. Leaf injury, shoot fresh weight, porphyrin biosynthesis substances, and chlorophyll content were investigated in transgenic and non-transgenic rice at 6 days after 0.5% and 1% NaCl treatments or at 5 days after 0~300 ${\mu}M$ paraquat treatments. No significant difference in leaf injury and shoot fresh weight were observed between transgenic and non-transgenic rice during chilling and recovery. Plant height and shoot fresh weight were also similar between transgenic and non-transgenic rice during the high temperature and recovery period (0~5 days). However, plant height and shoot fresh weight in transgenic rice line MX and PX were lower than in non-transgenic rice at 6 days for recovery. Leaf injury, chlorophyll, and Mg-Proto IX ME contents had no significant difference between transgenic rice and non-transgenic rice after NaCl treatment, but Proto IX content for AP37 and shoot fresh weight for PX and AP37 in 0.5% NaCl treatment were significantly reduced compared with non-transgenic rice. There was no difference in leaf injury and shoot fresh weight when comparing transgenic rice and non-transgenic rice after paraquat treatment. Although transgenic rice and non-transgenic rice showed a little difference at a particular measurement period in certain environmental stresses, there was generally no difference in physiological responses between transgenic rice and non-transgenic rice.

Evaluation of pig behavior changes related to temperature, relative humidity, volatile organic compounds, and illuminance

  • The objective of this study was evaluation of pig behavior changes related to temperature, relative humidity, volatile organic compounds (VOCs), and illuminance. A total of 24 growing pigs ([Yorkshire × Landrace] × Duroc) were used in the experiment. A sensor was installed at a height of 0.5 m in the center of the pig house. In experiment 1, temperature was changed every four days to 18℃ (T1), 22℃ (T2), 26℃ (T3), and then 30℃ (T4). In experiment 2, relative humidity was adjusted to 45% (low humidity [LH]), 60% (middle humidity [MH]), and then 75% (high humidity [HH]) for four days. In experiment 3, after cleaning the pig house just before experiment, only minimal ventilation was provided. VOCs and pig behaviors were observed for 7 days without cleaning the pig house. In experiment 4, three light bulbs of 40 W (470 lumens / 45 lx; low illuminance [LI]), 75 W (1,055 lumens / 103 lx; middle illuminance [MI]), and 100 W (1,521 lumens / 146 lx; high illuminance [HI]) were used for four days each. Pig behavior analysis was performed for following criteria : Feed intake, Standing, Lying, Sitting, Drink water, Rooting, Posture transition (lying-standing), Posture transition (standing-lying), Wallowing, and Biting. In experiment 1, feed intake time was lower (p < 0.05) for the T3 than other treatment groups. Standing time was highest (p < 0.05) for the T1 and lowest (p < 0.05) for the T3. Lying time was shorter (p < 0.05) in T1 and T2 compared to T3 and T4. Drinking frequency was higher (p < 0.05) for the T4 than other treatment groups. In experiment 2, the frequency of rooting and wallowing increased (p < 0.05) with increasing humidity. LH showed the lowest (p < 0.05) rooting frequency and HH showed the highest (p < 0.05) rooting frequency. In experiment 3, VOCs concentration did not (p > 0.05) change pig behavior. In experiment 4, lying time was the longest (p < 0.05) at LI and shortest (p < 0.05) at HI. Therefore, pig behavior is heavily influenced by the environment, especially temperature and humidity. However, correlation between pig behavior to VOCs and illuminance seems to be needed more research.
