Subacute Intravenous Toxicity Study of Recombinant Human Erythropoietin(rHu-EPO) in Rats (랫드에서 인체 재조합 적혈구 조혈인자, rHu-EPO의 아급성정맥독성시험)

  • This study was conducted to investigate the subacute intravenous toxicity of rHu-EPO in Sprague-Dawley rats. rHu-EPO was daily administered to male and female rats for 30 days with different dose levels(100, 500, 2,500 unit/ kg). After the administration period of 30 days, 5 males and 5females rats per each dose group were assigned for recovery period of additional 30 days. There were no clinical signs compared with control group but slight decrease in spontaneous motor activities and locomotions were observed at high dose groups of males and females. In males, feed consumption was reduced at 500 unit/kg and body weight gain was retarded. In the administration sites of tail, focal congestion was observed in a few animals treated with rHu-EPO. No deaths were occurred during the course of study. In hematological examination. a significant increase of hemoglobin and hematocrit was observed in the males and females rats receiving high dose of rHu-EPO. rHu-EPO administration for 30 days showed a marked decrease in glucose concentration. At the highest dose groups, there was a significant incerase in the weights of spleens in both sexes. but this was considered to be related to its pharmacological activity. These results indicate that rHu-EPO does not induce any significant toxic effect on Sprague-Dawley rats for 30 days.

Studies on the Changes in Serum Steroid Hormone Concentration and Serum Metabolite Contents During the Late Pregnancy in Korean Native Cow (한우에 있어서 임신말기 혈청중 Steroid Hormone 및 대사물질 수준의 변화에 관한 연구)

  • A series of experiment was conducted to determine the concentrations of progesterone and estradiol and metabolite contents in the serum of 26 Korean native cows raised at Alpine Experiment Station during the period of late pregnancy. Blood samples were collected by jugular puncture from individual cow at 5 day intervals from 30 days prepartum to 5 days prepartum and daily collected from 5 days prepartum to the day of parturition. Progesterone and estradiol concentrations in the serum were analyzed by Radioimmunoassay (R.I.A) method and serum metabolite contents were analyzed by autoanalzer MT II system. The following are summary of the results obtained: 1. Progresterone concentrations during the late preganacy were maintained at high level (5.12-11.70ng/$m\ell$) from 30 days prepartum to 8 days prepartum and fell rapidly from 5.12$\pm$1.07ng/$m\ell$ at 2 dyas prepartum to 1.48$\pm$0.32ng/$m\ell$ at 24 hrs prepartum. 2. Estradiol levles during the late pregancy increased gradually from 33.76$\pm$13.64pg/$m\ell$ at 30 days prepartum to 92.15$\pm$11.91pg/$m\ell$ at 11-15 days prepartum and increased thereafter sharply to a ranges of 161.76-238.4pg/$m\ell$ and were maintained at this increased levle until 24 hrs prepartum and decreased to 91.40pg/$m\ell$ at the parturition. 3. The correlation coefficients were found to be 0.2440 for cholesterol-progesterone relationship and -0.2552 for cholesterol-estradiol relationship, but there were statistically insignificant. 4. The changes in total protein contents during the late pregnancy were similar patterns to those of globulin and were maintained at high level only from 15 days to 5 days prepartum. 5. Glutamic oxaloacetate transaminase (GOT) levels were increased from 59.80$\pm$3.56u/$\ell$ at 90 days prepartum to 93.32$\pm$7.27u/$\ell$ at the day of parturition, but alkaline phosphatase levels were remained steady. 6. The levels of blood urea nitrogen, glucose and calcium remained almost constant during the late pregnancy. However, glucose concentration increased around the time of parturition.

Instant Tea Preparation from Cassia tora Seeds (결명자로부터 인스턴트차 제조)

  • In order to develop a type of instant tea cassia tora seed, changes of constiuent amounts before and after roasting $(3{\pm}0.5min\;at\;170{\pm}5^{\circ}C)$ effective extracting solvent and it's optimum concentration, dehydration method and sensory scores were investigated. Moisture, lipid, reducing sugar, emodin and rhein were decreased by roasting, whereas protein, and total sugar of cassia tora increased. Color intensity and extracting efficacy were also increased. Of three kinds of solvent (water, methanol and ethanol), ethanol was highest in redness and yellowness of exeracts, and filtrate yield. Effective extracting concentration of ethanol for cassia tora was 50%(V/V). Freeze dry product(FDP) and spary dry product(SDP) showed coffee and aloe-like smell, coffee like brown color, rapid soluble in cold and hot water, and fast caking in air during storage. Free sugar contents were high in order of raffinose>fructose >glucose>maltose>sucrose Metallic contents were high in order of sodium>calcium>potassium>magnesium>iron. In sensory score for color, taste and aroma, FDP and SDP were both above 8.0 point, however, of them FDP more or less higher than SDP In addion, score in developing value as an instant tea was above 8.0 point (valuable).

Optimal Culture Conditions for Production of Subtilisin-like Protease Inhibitor from Streptomyces thermocarboxydus C12 (Streptomyces thermocarboxydus C12에서 Subtilisin-like Protease Inhibitor 생산을 위한 최적배양조건)

  • The objective of this paper was to investigate optimal culture conditions for the production of an inhibitor against subtilisin-like protease from Streptomyces thermocarboxydus (S. thermocarboxydus) C12 isolated from sediments of Gwangyang coast. The optimal temperature and initial pH for the production of subtilisin-like protease inhibitor were $40^{\circ}C$ and pH 8.0, respectively. Inhibition activities were high for galactose, glucose and fructose. The best carbon source and its concentration were galactose and 1.6% (w/v), respectively. Inhibition activities were also high in medium containing polypeptone, proteose and peptone. Optimal nitrogen source and concentration were protease peptone and 0.5% (w/v), respectively. Optimal concentrations for inhibitor production were 1% (w/v) for NaCl and 1 mM LiCl for metal salts. The subtilisin-like protease inhibitor from S. thermocarboxydus C12 showed a maximum inhibitor activity after cultivation for 84 h under the optimized medium.

Development of a Beverage Using the Extracts from Bombyx mori L., Morus alba L., Dioscoreae rhizome and Inonotus obliquus (누에, 뽕나무, 산약 및 차가버섯 추출물을 이용한 음료 개발)

  • In this study, various antioxidant propertieswere evaluated by analyzing the proximate composition and free-sugar and DNJ contents of the Bombyx mori L., Mori fructus, Morus ramulus, Mori folium, Dioscoreae rhizome and Inonotus obliquus extracts, and the antidiabetic effect was evaluated through an in-vivo experiment. Product evaluation was conducted after preparing a beverage for the easy use of the mixed extract for biological activity, as a functional resource. The biochemical composition of the extracts was 0.31% crude protein, 0.114 g/100 mL free sugar and 161.02 mg/gdw DNJ, all of which showed excellent results in all the antioxidant ability and ${\alpha}$-glucosidase inhibition ability experiments. The beverage showed the following functionalities. The total-polyphenol content was 71.93%, but the electron-donating ability was highest in the 5% extract concentration. Moreover, when the TBARS values were experimented on, KO2 showed an especially high scavenging ability. During the five-week beverage supply after inducing diabetes with streptozotocin (STZ), the change in the blood sugar was measured, and the STZ-induced diabetes+oral-beverage group (C) showed a lower blood sugar level than the diabetes comparative group (B) in the second week. In the STZ-induced diabetes+free-diet/beverage group (D), the blood glucose level also slowly decreased in the second week. The lowest blood glucose level among the STZ-induced diabetes groups was shown in the fifth week.

Influence of flaxseed with rumen undegradable protein level on milk yield, milk fatty acids and blood metabolites in transition ewes

  • An experiment was conducted to determine the effects of two levels of rumen undegradable protein (RUP) without or with whole or extruded flaxseed on milk yield, milk component, milk fatty acids (FAs) profile and plasma metabolites in transition ewes. Three weeks before and after lambing, seventy-two Baluchi ewes were used in a completely randomized design with a 3 × 2 factorial arrangement of treatments. The treatments contained 1) no flaxseed + 20% RUP (no flaxseed, low RUP [NFLR]); 2) no flaxseed + 40% RUP (no flaxseed, high RUP [NFHR]); 3) 10% whole flaxseed + 20% RUP (whole flaxseed, low RUP [WFLR]); 4) 10% whole flaxseed + 40% RUP (whole flaxseed, high RUP [WFHR]); 5) 10% extruded flaxseed + 20% RUP (extruded flaxseed, low RUP [EFLR]), and 6) 10% extruded flaxseed + 40% RUP (extruded flaxseed, high RUP [EFHR]). Ewes fed 10% extruded flaxseed exhibited higher (p < 0.001) dry matter intake (DMI) and colostrum yield (p < 0.1) compared to other treatments. Two types of flaxseed and RUP levels had no significant effect on milk yield, but milk fat and protein contents decreased and increased in diets containing 40% RUP, respectively. Ewes fed extruded flaxseed produced milk with lower concentrations of saturated fatty acids (SFA) and higher α-linolenic and linoleic acids and also polyunsaturated fatty acids (PUFA) compared to other groups (p < 0.05). During post-lambing, the ewes fed diets containing flaxseed exhibited higher concentration of serum non-esterified FAs (NEFA) compared to diets without flaxseed (p < 0.01). The concentration of serum β-hydroxybutyric acid (BHBA) decreased in the diets containing flaxseed types at pre-lambing, but increased in diets containing extruded flaxseed at post-lambing (p < 0.01). The serum glucose concentration of ewes (pre and post-lambing) which consumed diets containing extruded flaxseed or 40% RUP increased, but blood urea concentration was elevated following supplementation of diet with whole flaxseed or 40% RUP (p < 0.001). In conclusion, utilization of 10% extruded flaxseed in the diets of transition ewes had positive effects on animal performance with favorable changes in milk FAs profile. However, there is no considerable advantage to supply more than 20% RUP level in the diet of transition dairy sheep.

Effect of Dietary Grape Pomace on Lipid Oxidation and Related Enzyme Activities in Rats Fed High Fat Diet (포도박이 고지방식이를 섭취한 흰쥐의 지질 산화와 항산화 효소 활성에 미치는 영향)

  • The present study was conducted to investigate the effect of dietary supplementation of grape pomace on lipid peroxidation and related enzyme activities of rats fed high fat diet. Male Sprague-Dawley rats weighing about 90 g were assigned to 4 experimental groups of 8 rats on the basis of their body weight. The high fat diet contained additional 15% lard to AIN 93-based diet. Rats were fed experimental diets containing 5% grape pomace for 4 weeks. Dietary supplementation of grape pomace reduced serum concentration of lipid peroxide in rats fed high fat diet. Hepatic concentration of lipid peroxide tended to be lower by feeding grape pomace. Hepatic total glutathione content and GSH/GSSG ratio were increased by grape pomace feeding in normal or high fat diet groups. Hepatic superoxide dismutase activity of grape pomace group with high fat diet was induced significantly compared with high fat diet group without grape pomace. Hepatic catalase activity of high fat fed rats was induced by feeding grape pomace. Grape pomace diet increased glutathione-S-transferase and glutathione peroxidase activities in rat liver fed high fat. Hepatic glucose-6-phosphatase activity was not affected by dietary supplementation of grape pomace in rats fed high fat. These results suggest that dietary supplementation of grape pomace may alleviate lipid peroxidation through antioxidant effect in rats fed high fat.

Nutrition Practice to Alleviate the Adverse Effects of Stress on Laying Performance, Metabolic Profile, and Egg Quality in Peak Producing Hens: I. The Humate Supplementation

  • This experiment was conducted to determine the effects of cage density (CD) and humate supplementation (HS) on laying performance, metabolic profile, and egg quality during the peak production period in hens. Lohman layers (n = 180, 46 wks of age) were blocked according to the location of cages and then allocated randomly to two levels of CD (4 or 6 hens per cage or 540 vs. 360 $cm^2$/hen) and three levels of HS (0, 0.15, and 0.30%). Egg production (EP) and feed consumption (FC) were measured daily; egg weight was measured bi-weekly; and BW was measured before and after the experiment. Blood and additional egg samples were obtained at the end of the experiment for determination of metabolic profile and egg quality. The data were analyzed using two-way ANOVA as repeated measures. Except for FC, CD did not affect laying performance parameters. Hens placed in high-density cages had lower FC than hens placed in normal-density cages. Increasing HS level linearly increased FC, EP, and feed conversion ratio (FCR). There was a CD by HS interaction effect on FC and EP. Hens placed in high-density cages had greater serum glucose, total protein, albumin, globulin, Ca, and P concentrations and tended to have greater serum corticosterone concentration than hens placed in normaldensity cages. Increasing HS level linearly increased serum glucose, total protein, albumin, globulin, creatine, and Ca concentrations and linearly decreased serum triglyceride and very low-density lipoprotein concentrations. There was a CD by HS interaction effect on serum glucose and albumin concentrations. There were no alterations in egg quality parameters in response to increasing CD. Albumen index and Haugh unit decreased linearly and other egg quality parameters did not change as HS level increased. In conclusion, increased caging density adversely affected metabolic profile, despite insignificantly deteriorating laying performance. Moreover, benefits from humate supplementation seem to be more noteworthy for hens housed in stressing conditions than for hens housed in standard conditions.

Zymological Properties of Foxtail Millet Wine-making by Isolated Strains from Nuruk (누룩에서 분리한 우수균주에 의한 좁쌀주의 양조특성)

  • In order to improve the quality of foxtail millet wine, zymological properties by isolated strains from Nuruk were investigated. Saccharomyces sp Y5-1 as brewing yeast, Aspergillus sp. M6-3, Aspergillus awamori 6970, and Aspengillus usamii mut. shirousamii 6959 (KCTC) as saccharifying molds were used, respectively. Acid content, soluble solids, color (b) and alcohol contents were increased during fermentation. Ethanol concentration of millet wine made with Nuruk by Aspergillus awamori 6970 and Aspergillus usamii mut. shirousamii 6959 were higher than the other, 10.6 and 10.1% respectively. Citric acid was only detected on $1{\sim}2$ day starting fermentation. Oxalic acid, lactic acid and acetic acid of millet wine were high in the wine made of Nuruk by Aspergillus usamii mot. shirousamii 6959, Aspergillus awamori 6970 and traditional Nuruk, respectively. During fermentation, glucose and xylose was higher than the others. Xylose was increased, but most of other sugar were decreased during fermentation. Acetone, ethyl acetate, methanol, ethanol, n-propanol, iso-buthanol and iso-amyl alcohol were detected In the wine made with Nuruk by Aspergillus awamori and Aspergillus usamii mut. shirousamii, iso-imyl alcohol and ethanol were high. On sensory evaluation, the wine made with Nuruk by Aspergillus usamii mut. shirousamii was the best on color and taste.

Investigation of the Cultural Characteristics of High Concentration Ehtanol Resistant Acetobacter sp. (고농도 에탄올내성 초산균의 개발 및 배양특성)

  • To increase the yield of acetic acid production, the author developed the bacterial strain which could brow well in high concentration of ehtanol from the seed culture using in conventional vinegar production factory. By attenuation of the isolated strain in the broth media containing 5-10% ethanol, we could get the strain which could grow in the broth medium containing 10% ethanol. This strain was identified and named as Acetobacter sp. FM-10, and it's cultural characteristics were also investigated. The medium containing 10% ethanol, 5% glucose and 1% yest extract was suitable for the acetic acid production with Acetobacter sp. FM-10. Optimum temperature and pH for the growth of Acetobacter sp. FM-10. were $30^{\circ}C$ and 5.0, respectively. The acidity of culture medium was reached to 9.0 % after 20 days static cultivation at $30^{\circ}C$.
