Effect of Tumeric (Curcuma longa) on Bile Acid and UDP-glucuronyl Transferase Activity in Rats Fed a High-fat and -cholesterol Diet (울금(Curcuma longa L.)이 고지방·고콜레스테롤 식이 흰쥐에서의 담즙산 및 UDP-glucuronyl transferase 활성에 미치는 영향)

  • The current study examined the effect of turmeric powder on bile acid and UDP-glucuronyl transferase activity in rats fed a high-fat and -cholesterol diet. Sprague-Dawley male rats weighing $120{\pm}10$ g were randomly assigned to a normal diet group (N group) and a high-fat and -cholesterol diet group (HF group), which was further divided into a high-fat and high-cholesterol with a 2.5% tumeric powder supplement group (TPA group) and 5% turmeric powder-supplemented group (TPB group). Body weight gain and food efficiency ratio were significantly increased in the N group as compared to the HF group, but they were significantly decreased in turmeric-supplemented groups as compared to the HF group. The total serum cholesterol and TG contents of the turmeric-supplemented groups were decreased as compared to those of the HF group. Especially, the TPB group was significantly decreased as compared to the HF group. The serum LDL-cholesterol and AI of the turmeric-supplemented groups were decreased as compared to the HF group. The hepatic triglyceride contents of all groups supplemented with the tumeric powder were significantly decreased as compared to the HF group. The hepatic UDP-glucuronyl transferase activity of the turmeric-supplemented groups was increased as compared to the HF group. In particular, the TPB group was significantly increased as compared to the HF group. The serum total bile acid contents of the turmeric-supplemented groups were increased as compared to the HF group. These results suggest that tumeric has powerful health benefits that are created via UDP-glucuronyl transferase activity, bile acid, and lipid metabolism.

Effect of Hog Millet Supplementation on Hepatic Steatosis and Insulin Resistance in Mice Fed a High-fat Diet (고지방식이로 유도한 지방간 마우스에서 기장 첨가식이가 지방간 및 인슐린 저항성에 미치는 영향)

  • The dietary intake of whole grains is known to reduce the incidence of chronic diseases such as obesity, diabetes, cardiovascular disease, and cancer. In our previous study, hog millet (HM, $Panicum$ $miliaceum$ L.) water extract showed the highest anti-lipogenic activity among nine cereal types in 3T3-L1 cells. In this study, the effect of hog millet water extract on hepatic steatosis and lipid metabolism in mice fed a high fat diet was investigated. Mice were fed a normal-fat diet (ND), high-fat diet (HFD) or HFD containing 1% or 2% (w/w) HM for 7 weeks. Body weight and food intake were monitored during the study period. Insulin resistance by homeostasis model assessment (HOMA-IR), fasting lipid profile, hepatic fatty acid metabolism-related gene expression determined, and intraperitoneal glucose tolerance test (IGTT) were performed at the study's end. The results indicated that 1% and 2% HM diets effectively decreased liver weights, blood TG and T-cholesterol levels (p<0.05), while the HDL-cholesterol level was increased (p<0.05) compared to HFD-induced steatotsis mice. Hepatic lipogenic-related gene ($PPAR{\alpha}$, L-FABP, and SCD1) expressions decreased, whereas lipolysis- related gene (CPT1) expression increased in animals fed the 2% PME diet (p<0.05). In addition, mice fed 1% or 2% HM diet had markedly decreased IGTT and HOMA-IR, compared to the those of the HFD-induced hepatic steatosis control group (p<0.05). These results indicated that HM inhibited hepatic lipid accumulation by regulating fatty acid metabolism, and suggested that HM is useful in the chemoprevention or treatment of high fat-induced hepatic steatosis and hepatic steatosis-related disorders including hyperlipidemia, glucose sensitivity, and insulin resistance.

Physiological Profile of Growing Rats: Effects of Cage Type and Cage Density

  • This experiment was conducted to examine the effects of cage type (CT) and cage density (CD) on physiological variables in growing rats. Sprague Dawley rats (n=108) weighing an average of 46 g were housed in metallic cage with woodchip bedding (MCWB), metallic cage with wire mesh (MCWM), and plastic shoebox with woodchip bedding (PCWB) separately by sex at normal ($160-cm^2/rat$, ND) and high ($80-cm^2/rat$, HD) CD from 3 to 10 wks of age. All cages were in dimension of $24{\times}40{\times}20$ cm ($W{\times}D{\times}H$). At the end of the experiment, blood samples were collected and 6 rats from each cage were sacrificed. No death was observed among rats at ND, whereas mortality rate at HD was 22.3% for males and 13.9% for females. Heart weight was affected by CT. Doubling CD caused 23, 11.8, 17.9, 8.6, 6.9, and 16.4% decreases in BW and weights of heart, liver, kidney, testis, and ovary, respectively. Except for adrenal gland, other organs for males were heavier than for females. Liver weight of males and females responded differently to CT and CD. Comparing with females, males had 7.3 and 5.2% heavier and 9.9% lighter liver weights in MCWB, MCWM, and PCWB, respectively. As CD doubled, liver weight for males and females decreased by 22.4 and 13.1%, respectively. Mean adrenal gland weight increased by 8.4% and decreased by 9.7% for males and females, respectively, with doubling CD. CT affected glucose, TG, Ca, and ALP levels. However, CD did not alter blood chemistry. Rats housed in metallic cages had greater neutrophil count and neutrophil:lymphocyte ratio than rats housed in plastic cages. Doubling CD caused a 24.2% increase in lymphocyte count. There were CT by CD, CT by sex, and CD by sex interaction effects on lymphocyte count. Doubling CD caused 0.1% decrease and 49.8 and 26.7% increases in lymphocyte count for rats housed in MCWB, MCWM, and PCWB, respectively. Comparing with females, lymphocyte count for males housed in MCWB, MCWM, and PCWB had 8.9 and 12.9% greater and 30.3% less lymphocyte counts, respectively. Lymphocyte count decreased by 4.12% for males, whereas it increased by 61.0% for females as CD doubled. Doubling CD resulted in 2.5 and 2.3% increases in erythrocyte count and hematocrit value. These data suggest that animals perform better in metallic cages than in plastic cages and that cage density had pronounceable effects on physiological parameters in a cage type and sex dependent-manner.

Study on the Eco-friend Frame Sheet with Improved Glasses Temple's Insertion-processibility by Blending Plasticizer of High Specific Heat (친환경 안경테 판재의 심입 가공성 향상을 위한 고비열 가소제 혼입에 관한 연구)

  • Purpose: To improve glasses temple's insert processibility of CA/PEG blend, triacetin with higher specific heat values in the processing temperature range is used as second plasticizer. Methods: The total amount of plasticizer is fixed at 30 wt% by CA. To determine optimal CA/PEG/triacetin blend for glasses frame, blends with different composition ratio were examined by various analysis: thermal properties, mechanical properties, glossiness. Results: Specific heat of the CA/PEG blend increased as the content of triacetin. In CA/PEG/triacetin blends, as triacetin concentration is increased, glass transition temperature is decreased and heat conservation rate of composites is increased. Furthermore, CA/PEG/triacetin blend exhibited higher mechanical properties and similar gloss characterization with CA/PEG blend. Conclusions: It is possible to improve the processibility inserting metal support to CA temple through varying the weight ratio of PEG/triacetin. The extruded sheets of CA/PEG/triacetin blend had better glossiness and mechanical properties than those of CA/PEG blend.

A STUDY ON THE PHYSICAL PROPERTIES AND VOLUMETRIC STABILITY OF SR-IVOCAP RESIN SYSTEM (SR-Ivocap resin system의 물리적 특성과 체적 안정성에 관한 연구)

  • This study helps to clarify conflicting reports by comparing the physical properties and accuracy of complete denture processed by the pack and press technique, continuous- pressure injection technique(SR-Ivocap system) and Mark press technique. The 6 different specimens have been evaluated using the SEM, Impact test, DSC (Differential Scanning Calorimetry) and DMTA (Dynamic Mechanical Thermal Analysis). Each sample was made of SR-Ivocap resin and QC-20 resin by different processing methods. The results were as follows ; 1. As the result of the observation on the fracture surface of resin by use of SEM, sample SR-Ivocap resin cured by continuous pressure injection method showed the most homogeneous structure. This is why molecules in SR-Ivocap resin have no orientation. 2. As the result of the Impact test in order to measure the deformity, fracture energy and impact resistance of resin, the samples with QC-20 acrylic resin and SR-Ivocap resin cured by continuous pressure injection method were exellent. 3. In consequence of measuring ${\alpha}$-glass transition temperature by use of DSC on the basis of temperature change, the glass transition temperatures of sample QC-20 resin cured by pack and press method and sample SR-Ivocap resin cured by continuous pressure injection method were very similar. Thus volumetric stability could not be evaluated only by glass transition temperature. 4. In comparing volumetric stability data by DMTA, the glass transition temperature(Tg) showed $137.88^{\circ}C$ at sample QC-20 resin cured by pack and press method and $139.78^{\circ}C$ at sample SR-Ivocap resin cured by continuous pressure injection method. Therefore sample SR-Ivocap resin cured by continuous pressure injection method seems to be superior to sample QC-20 resin cured by pack and press method in the dimensional stability at high temperature. 5. In comparing storage modulus data by DMTA, the storage modulus of sample SR-Ivocap resin cured by continuous pressure injection method was higher than that of sample QC-20 resin cured by pack and press method. So. sample SR-Ivocap resin cured by continuous pressure injection method seems to be superior to sample QC-20 resin cured by pack and press method in impact strength.

Effects of Dietary Thiazolidinedione Supplementation on Growth Performance, Intramuscular Fat and Related Genes mRNA Abundance in the Longissimus Dorsi Muscle of Finishing Pigs

  • The objective of this study was to investigate the effect of dietary supplementation with thiazolidinedione (TZD) on growth performance and meat quality of finishing pigs. In Experiment 1, 80 castrated finishing pigs (Large White${\times}$Landrace, BW = 54.34 kg) were randomly assigned to 2 treatments with 5 replicates of 8 pigs each. The experimental pigs in the 2 groups were respectively fed with a diet with or without a TZD supplementation (15 mg/kg). In Experiment 2, 80 castrated finishing pigs (Large White${\times}$Landrace, BW = 71.46 kg) were divided into 2 treatments as designed in Experiment 1, moreover, carcass evaluations were performed. The results from Experiment 1 showed that TZD supplementation could significantly decreased the average daily feed intake (ADFI) (p<0.05) during 0 to 28 d, without impairing the average daily gain (ADG) (p>0.05). In Experiment 2, the ADG was significantly increased by TZD supplementation during 14 to 28 d and 0 to 28 d (p<0.05) and the feed:gain ratio (F:G) was significantly decreased by TZD supplementation during 0 to 28 d (p<0.05). Compared with the control group, TZD group had significantly higher serum triglyceride (TG) concentration at 28h and serum high-density lipoprotein (HDL) levels at 14 d (p<0.05). Moreover, there was an apparent improvement in the marbling score (p<0.10) and intramuscular fat (IMF) content (p<0.10) of the longissimus dorsi muscle in pigs treated by TZD supplementation. Real-time RT-PCR analyses demonstrated that pigs of TZD group had higher mRNA abundance of $PPAR{\gamma}$ coactivator 1 (PGC-1) (p<0.05) and fatty acid-binding protein 3 (FABP3) (p<0.05) than pigs of control group. Taken together, these results suggested that dietary TZD supplementation could improve growth performance and increase the IMF content of finishing pigs through regulating the serum parameters and genes mRNA abundance involved in fat metabolism.

Influence of magnesium and calcium on the serum cholesterol level lowering (III) - Influence of Korea Ginseng - (마그네슘 및 칼슘이 혈청콜레스테롤 농도 저하에 미치는 영향 (III) - 인삼의 영향 -)

  • Influence of magnesium and calcium on the serum cholesterol concentration in rabbit blood serum was studied. The subjects were fed with the basal diet and magnesium, calcium and Korea Ginseng. The results obtained are summarized as follows: The subjects body weight fed a basal diet, magnesium, calcium and ginseng powder added did not increased, but the weight of liver was increased. Total cholesterol level of the subject fed ginseng powder 4mg. added diet group showed the lowest concentration. The subjects fed with a basal diet and magnesium/calcium(1:2) added diet showed the lower than anyother group. In case of free cholesterol and cholesterol ester, the subject fed with ginseng powder tap. magnesium/calcium(1:2) added diet showed the lowest concentration. The subjects of the lowest concentration of total cholesterol showed the lowest concentration of low density lipoprotein(LDL) and high density lipoprotein(HDL)/total cholesterol ratio was the higher level than any other group. In case of triglyceride and phospholipid, the subject fed with ginseng powder showed the triglyceride concentration gradually do creased, but phospholipid level was increased positively. In case of electrolytes, sodium and potassium of the subject fed with magnesium, calcium and ginseng powder 4mg added diet showed 120mEq/l 1 and 6.8mEq/l, respectively.

Study of Gene-gene Interaction within GNB3, ACE, ADRB3, ADRB2 among Korean Female Subject (한국인 비만 여성의 GNB3, ACE, ADRB3, ADRB2 유전자 다형성간의 상호관계에 관한 연구)

  • There have been several reports on the relationship between G protein β3 subunit gene (GNB3), angiotensin converting enzyme gene (ACE), β3-adrenergic receptor gene (ADRB3), and β2-adrenergic receptor gene (ADRB2) genotype and obesity or obesity related disease. The objective of this study was to examine the relationship between the combinations of these four genes' polymorphism and probability of obesity related disease in Korean female subjects. The experimental group was consisted of 85 obese Korean female subjects (body mass index, BMI≥27㎏/㎡). To determine the polymorphism, genomic DNA was isolated, and PCR was performed. Serological examinations (fasting plasma glucose, FPG; aspartate aminotranferase, AST; alanine aminotransferase, ALT; total cholesterol, TC; triglyceride, TG; high density lipoprotein-cholesterol, HDL; low density lipoprotein-choles terol, LDL) were carried by an autoanalyzer and serological methods. BMI, waist circumference (WC), hip circumference and waist hip ratio (WHR) were measured. Consequencely in the analysis with grouping of general genotyping and variant allele carrier/non-carrier, the result was not significantly different within all gene combinations and polymorphic pairings except higher waist circumference in Arg16Arg group of ADRB2 codon16 (P=0.024). And there was no significantly contrast result about age, height, weight, AST and ALT that are index feature of liver and gall bladder disease in polymorphic pairings of gene combinations. However, the statistical analysis of waist-hip ratio and waist circumference that could be recognized as the physical type of obesity showed T-Arg16 pairing carrier in GNB3-ADRB2 codon16 combination had increased WHR and WC significantly (P=0.046 and P=0.015 respectively). Futhermore, the levels of total cholesterol (TC) and low density lipoprotein choresteral (LDL) were significantly lower in C-I pairing of GNB3-ACE combination (P=0.032 and P=0.005). These results suggest that the T-Arg16 pairing carrier in GNB3-ADRB2 codon16 gene might have increased waist circumference and C-I pairing carrier in GNB3-ACE combination have lower possibility of contraction of cardiovascular disease related cholesterol and LDL despite of obese state.

Treatment of Ceroidosis for Cultured Flounder, Paralichthys olivaceus -Hematological characteristic of flounder with glutathione supplemented diets- (양식(養殖) 넙치의 Ceroid증(症) 치료(治療)에 대(對)하여 -Glutathione 첨가(添加)에 따른 혈액성상(血液性狀)-)

  • In order to investigate the effects of glutathione against ceroidosis on flounder, Paralichthys olivaceus, a serial experiment was carried out at indoor tanks in Cheju province, the southern island of R.O.Korea, from October 1 to November 4 of 1991. The choosed group of flounder for this experiment, ranged from 92 to 122g in body weight, already showed a distinct histological ceroidosis in liver, spleen, kidney about six months before this experiment. They were fed on formulated diets supplemented with glutathione of 0mg, 1mg, 2mg, 5mg, 10mg per kg of diet per day. Some flounders were fed on the decomposed diet as a control continuosuly. Two individuals fed on the experimental diets were cought at an interval 7 days for 35 days and analyzed the values of blood indexs. 1. While the values of RBC, Hb, Ht, MCHC, MCV of flounder fed on the decomposed diet and the glutathione non-supplemented diet were not changed compared with the beginning levels of experiment, those fed on the glutathione supplemented diets, showed markedly increased values from 7th day of experiment and from 21th day of experiment reached to the values of healthy flounder. 2. The variation of TP, Glu, Alp, BUN, TG, T-cho, GOT and GPT were not observed in the groups fed on the control diet and glutathione non-supplemented diet. In contrast, those values in the groups fed on the glutathione supplemented diets were markedly decreased from 7th day and become constant from 21th day. 3. The effect of glutathione supplementation with high concentration in diets showed much better improvements of symptoms concequently, compared with those of low concentration diets.

Research Related to Chronic Kidney Disease in BMI >25 Overweight Children (체질량 지수 25 이상의 과체중군에서 신장질환과 관련된 인자들에 대한 연구)

  • Purpose : Chronic kidney disease (CKD) and obesity are the worldwide public health problem. Obesity is an already well-established risk factor for CKD. The objective of this study is to evaluate the relationship between high BMI and increased risk for nephropathy by clinical data. Methods : Study group were 26 patients who had $BMI{\geq}25\;kg/m^2$ and control group were 49 patients with BMI<$25\;kg/m^2$. Both groups received renal biopsy in Kyung Hee Medical Center between 2003. Jan.-2007. Dec. BMI was calculated from measured weight and height when they were admitted to the hospital. We collected laboratory data such as CBC and blood chemistry. Results : Our hypothesis was that overweight and obesity are associated with incidence and progression of CKD. From kidney biopsy, we found IgAN 17, MesPGN 5, HSPN 2, Intestitial nephritis 1, IgMN 1 (total 26) in the study group whereas IgAN 22, MesPGN 17, HSPN 3, MGN 3, benign hematuria 2, MPGN 1, Intestitial nephritis 1, (total 49) were found in the control group. There was no significant difference between the two groups. Overweight patients demonstrated significantly higher platelet, TG, ALT, and uric acid level compared to control group. Conclusion : We identified a significant relationship between overweight and development of CKD. These results suggest that overweight children have an increased risk for CKD than those who are not obese. So, we should pay attention to children with overweight who have CKD and earlier weight management is crucial to prevent aggravation of CKD.
