Two Unrecorded Species, Globodera hypolysi and Sarisodera hydrophiza (Nematoda: Heteroderidae) from Korea (씨스트선충과 (Nematoda: Heteroderidae)의 한국 2 미기록종 Globodera hypolysi와 Sarisodera hydrophila의 기재)

  • Two unrecorded species of Heteroderinae, Clobodera hypolysi Ogawa, Ohshima and Ichinohe, 1983 and Sarisodera hydrophila Wouts and Sher, 1971 from Korea were firstly reported. The Korean specimens well correspond with the original description of Globodera hypolysi except stylet length of the juveniles, slightly shorter (21~24 vs. 24~25 $\mu$m) and shorter distance from the anus to the fenestra in cyst (22~50 vs. 25~80$\mu$m). The measurements of Korean specimen of Sarisodera hydrophila are slightly short compared with the type species; Female body length 367.2~597.6$\mu$m vs 359~688$\mu$m; Stylet length 37~44$\mu$m vs 38~46$\mu$m; Male body length 662~11987m vs 579~1405$\mu$m; stylet length 37~43.8$\mu$m vs 38~46$\mu$m. Juvnile body length 460~536$\mu$m vs 466~5587m. stylet length 32~387m vs 39~43$\mu$m.

  • During the survey on apple parasitic nematodes from Korea, Cryphodera coxi(Wouts, 1973) Luc, Taylor and Cadet, 1978 was discovered for the first time from Korea. The specimens preserved in the Nematology Laboratory, School of Bioresource Sciences, College of Natural Sciences, Andong National University.

  • Influence of susceptible and resistant soybean varieties to the development of the soybean cyst nematode(Heterodera glycines) was examined. It does not seem likly that there are any differences between varieties in attracting the Second Stage larvae of the nematode. In susceptible varieties, there were large populations and three generations during a ten·week examination period, while the resistant varieties maintained very low populations and had only two generations.

  • Meloidogyne marylandi Jepson & Golden, 1987 is described from Zoysia grass collected in Cheonan, Korea. The nematode did not produce galls, and females were generally surrounded by a massive egg sac. Perineal patterns of female were a high to rounded arch with shoulders, wavy striae usually forming a rough. Vulva and anus were sunken. Second-stage juvenile body length was 390.3 $\mu\textrm{m}$ (290-430), tail length 55.1 $\mu\textrm{m}$ (49-59) and lateral field with 4 incisures. Male stylet length was 18.1 $\mu\textrm{m}$ (15-24), spicules length 26.2 $\mu\textrm{m}$ (23-29) and lateral field with 4-5 incisures.

Identification of Heterodera glycines (Tylenchida; Heteroderidae) Using qPCR

  • The soybean cyst nematode, Heterodera glycines, is a major plant-parasitic nematode that has caused important economic losses to Korea's soybean production. Four species of cyst nematodes, H. schachtii, H. glycines, H. trifolii, and H. sojae, all belong to schachtii group are coexist in field soil in Korea. The rapid identification of the nematode is crucial for preventing crop damage and in decision making for controlling this nematode. This study aimed to develop a species-specific primer set for quantitative PCR (qPCR) assay of H. glycines. The specific primer set (HGF1 and HGR1) for H. glycines was designed based on the cytochrome c oxidase subunit I (COI) sequence of mitochondrial DNA. After optimization, it is possible to identify the H. glycines using a qPCR assay with DNA extracted from a single cyst and single second-stage juvenile (J2). The specificity was confirmed by the absence of SYBR fluorescent signals of three other Heterodera species. A serial dilution of DNA extracted from a single cyst was obtained for the sensitivity test. The result showed that the standard curve of the test had a highly significant linearity between DNA concentration and Ct value (R2 = 0.996, slope = -3.49) and that the detection limit concentration of DNA of the primer set was 10 pg of DNA per reaction. Our findings suggested that H. glycines could be distinguished from H. sojae and other Heterodera species when a qPCR assay is used with a specific primer set.
