Laboratory Investigation of Sexually Transmitted Infections in the Elderly Population of South Korea

  • This study analyzed the epidemiological trends of sexually transmitted infections (STIs) according to age and sex among individuals aged 50 years or older in South Korea from September 2018 to December 2020. We found that the positivity rate for Gardnerella vaginalis was the highest in the study group, followed by the positivity rate for Ureaplasma parvum (UP). Interestingly, the positivity rates for Mycoplasma hominis and UP were higher in female participants than in male participants. The positivity rate for Treponema pallidum was very low in the female participants. During the study period, the positivity rate for herpes simplex virus 2 increased in the female participants, while the positivity rate for Candida increased in the male participants. These results show that the STI positivity rate varies according to age and sex, and a difference was observed in the average age of positive participants according to the type of STIs. We found a clear pattern of infection in the elderly population and according to sex. Our findings are expected to be used as baseline data for future research, education, and prevention of STIs in the elderly population.

Evaluation of concurrent vaccinations with recombinant canarypox equine influenza virus and inactivated equine herpesvirus vaccines

  • Despite vaccination, equine influenza virus (EIV) and equine herpesvirus (EHV) infections still cause highly contagious respiratory diseases in horses. Recently, concurrent vaccination with EIV and EHV was suggested as a new approach; however, there have been no reports of concurrent vaccination with recombinant canarypox EIV and inactivated EHV vaccines. In this study, we aimed to compare the EIV-specific immune responses induced by concurrent administrations of a recombinant canarypox EIV vaccine and an inactivated bivalent EHV vaccine with those induced by a single recombinant canarypox EIV vaccine in experimental horse and mouse models. Serum and peripheral blood mononuclear cells (PBMCs) were collected from immunized animals after vaccination. EIV-specific serum antibody levels, serum hemagglutinin inhibition (HI) titers, and interferon-gamma (IFN-γ) levels were measured by enzyme-linked immunosorbent assay, HI assay, and quantitative polymerase chain reaction, respectively. Concurrent EIV and EHV vaccine administration significantly increased IFN-γ production, without compromising humoral responses. Our data demonstrate that concurrent vaccination with EIV and EHV vaccines can enhance EIV-specific cellular responses in horses.

  • Stevens-Johnson syndrome (SJS) produces a severe hypersensitivity reaction caused by Herpes simplex virus or mycoplasma infection, vaccination, systemic disease, or other agents. Several studies have investigated the genetic susceptibility involved in SJS. To provide further genetic insights into the pathogenesis of SJS, this study prioritized high-impact, SJS-associated pathogenic variants through integrating bioinformatic and population genetic data. First, we identified SJS-associated single nucleotide polymorphisms from the genome-wide association studies catalog, followed by genome annotation with HaploReg and variant validation with Ensembl. Subsequently, expression quantitative trait locus (eQTL) from GTEx identified human genetic variants with differential gene expression across human tissues. Our results indicate that two variants, namely rs2074494 and rs5010528, which are encoded by the HLA-C (human leukocyte antigen C) gene, were found to be differentially expressed in skin. The allele frequencies for rs2074494 and rs5010528 also appear to significantly differ across continents. We highlight the utility of these population-specific HLA-C genetic variants for genetic association studies, and aid in early prognosis and disease treatment of SJS.

The Effects of the Houttuynia Cordata Thunb Intake and Exercise Performance on Lipid Components and Abdominal Fat in Middle-Aged Women (어성초 복용과 운동 수행이 중년여성의 복부 지방 및 혈중지질 성분에 미치는 영향)

  • Houttuynia Cordata Thunb, well known as "E-Sung Cho" in Korea, has been used for the treatment of herpes simplex, chronic sinusitis and nasal polyps. Recently, some studies reported the antileukemic activity, anticancer activity, adjuvanticity, and antioxidant action of Houttuynia Cordata Thunb. In the present study, the anti-obesity activity of Houttuynia Cordata Thunb was investigated. Middle-aged women were examined over a period of 8 weeks (n=30). Participants were divided into four group: Houttuynia Cordata Thunb Exercise (HCE, n=7), Placebo Exercise (PE, n=8), Houttuynia Cordata Thunb Non Exercise (HCNE, n=8), and Placebo Non Exercise (PNE, n=7). Participants in Houttuynia Cordata Thunb groups were received 4.5 g Houttuynia Cordata Thunb and in placebo groups were received same dose of placebo thrice daily with meals. The result after 8 weeks demonstrated that HCE significant decreased visceral fat than PNE. Our present study provides the evidence that Houttuynia Cordata Thunb intake and exercise performance may reduce visceral fat and further studies with larger numbers of subjects are warranted to conclusively demonstrate effectiveness.

Unusual Acute Encephalitis Involving the Thalamus: Imaging Features

  • Objective: To describe the brain CT and MR imaging findings of unusual acute encephalitis involving the thalamus. Materials and Methods: We retrospectively reviewed the medical records and CT and/or MR imaging findings of six patients with acute encephalitis involving the thalamus. CT (n=6) and MR imaging (n=6) were performed during the acute and/or convalescent stage of the illness. Results: Brain CT showed brain swelling (n=2), low attenuation of both thalami (n=1) or normal findings (n=3). Initial MR imaging indicated that in all patients the thalamus was involved either bilaterally (n=5) or unilaterally (n=1). Lesions were also present in the midbrain (n=5), medial temporal lobe (n=4), pons (n=3), both hippocampi (n=3) the insular cortex (n=2), medulla (n=2), lateral temporal lobe cortex (n=1), both cingulate gyri (n=1), both basal ganglia (n=1), and the left hemispheric cortex (n=1). Conclusion: These CT or MR imaging findings of acute encephalitis of unknown etiology were similar to a combination of those of Japanese encephalitis and herpes simplex encephalitis. In order to document the specific causative agents which lead to the appearance of these imaging features, further investigation is required.

A Study on Antibacterial Effects of Ascorbic Acid against Mycobacterium tuberculosis (Ascorbic acid의 항결핵균 작용에 관한 연구)

  • Vitamin C is known as an integral element for the formation and maintenance of intercellular supporting structures. Ascorbic acid has been used for the treatment of certain poisonings and hypovitaminosis(scurvy) but also known as a powerful reducing agent, and can kill a variety of bacteria and detoxify bacterial exotoxins including various clostridial exotoxins in vitro. For viruses, vitamin C inactivates herpes virus, vaccinia virus and influenza virus and has been used for the prevention and treatment of the common cold. Thus ascorbic acid plays an important role in antimicrobial action. Scurvy also promotes the development of tubercles in experimentally infected guinea pig and the tuberculosis patients require more vitamin C than normal persons. However there is no reports that ascorbic acid could inhibit the growh of M. tuberculosis. In this paper, antibacterial effects of ascorbic acid against M. tuberculosis were studied. The results are as follows: 1. The single use of the ascorbic acid exhibited antibacterial effect in vitro against $5{\times}10^3/ml$ of M. tuberculosis $H_{37}$ Rv at the concentration of ascorbic acid 0.625mg/ml over 3 hours exposure and 0.05mg/ml over 9 hours exposure. 2. In vivo mice administered with ascorbic acid 50mg/day for 5, 10 and 15 days respectively were protected from M. tuberculosis $2LD_{50}$, $3LD_{50}$, $4LD_{50}$ and $5LD_{50}$ given intravenously.

Lack of Association between Herpes Simplex Virus Type 2 Infection and Cervical Cancer - Taq Man Realtime PCR Assay Findings

  • Background: About one third of the human population suffer cancer during their lifetime and more than 20% of total morbidity is related to neoplasia. Cervical cancer is generally the most common cancer in developing countries and the second most common in women globally. The role of human papilloma viruses viruses in its induction is clear. However, the involvement of hepres simplex virus type 2 (HSV-2) is controversial. Therefore a survey was conducted of the prevalence of HSV-2 in patients with cervical cancer and also healthy people with sensitive and quantitative Taq Man real-time PCR assay. Materials and methods: Seventy six formaldehyde fixed paraffin embedded tissue specimens from patients with histologically proven history of cervical cancer as well as 150 control blocks were sectioned for deparaffinization and DNA extraction. Results: There was no HSV-2 DNA in our patient specimens but four control samples were positive, all with a history of hysterectomy. Conclusion: Considering the absence of any positive viral HSV-2 DNA in our patients and also the presence of four positive specimens among our controls, we did not find any relationship between the presence of HSV-2 DNA and cervical cancer.

Partitioning and Inactivation of Viruses by Cold Ethanol Fractionation and Pasteurization during Manufacture of Albumin from Human Plasma

  • The purpose of the present study was to examine the efficacy and mechanism of the fraction IV cold ethanol fractionation and pasteurization ($60^{\circ}C$ heat treatment for 10h) steps, involved in the manufacture of albumin from human plasma, in the removal and/or inactivation of blood-born viruses. A variety of experimental model viruses for human pathogenic viruses, including the Bovine viral diarrhoea virus (BVDV), Bovine herpes virus (BHV), Murine encephalomyocarditis virus (EMCV), and Porcine parvovirus (PPV), were selected for this study. Samples from the relevant stages of the production process were spiked with the viruses, and the amount of virus in each fraction was then quantified using a 50% tissue culture infectious dose ($TCID_{50}$). The mechanism of reduction for the enveloped viruses (BHV and BVDV) during fraction IV fractionation was inactivation rather than partitioning, however, it was partitioning in the case of the non-enveloped viruses (EMCV and PPV). The log reduction factors achieved during fraction IV fractionation were ${\geq}6.9$ BHV, $\geq5.2$ for BBDV, 4.9 for EMC, and 4.0 for PPV. Pasteurization was found to be a robust and effective step in inactivating the enveloped viruses as well as EMCV. The log reduction factors achieved during pasteurization were $\geq7.0$ for BHV, $\geq6.1$ for BVDV, $\geq6.3$ for EMCV, and 1.7 for PPV. These results indicate that the production process for albumin has sufficient virus-reducing capacity to achieve a high margin for virus safety.

Inhibition of Herpesvirus-6B RNA Replication by Short Interference RNAs

  • RNA interference (RNAi) is a process of sequence-specific gene silencing, which is initiated by double-stranded RNA (dsRNA). RNAi may also serve as an antiviral system in vertebrates. This study describes the inhibition of herpesvirus-6B (HHV-6B) replication by short interference RNAs (siRNAs) that are targeted to the U38 sequence that encodes DNA polymerase. When virus-infected SupT1 cells were treated by siRNA, these cells blocked the cytopathic effect (CPE) and detected the HHV-6B antibody-negative in indirect immunofluorescence assays (IFA). Our result suggests that RNAi can efficiently block Herpesvirus-6B replication.
