일본 '고증파(考證派)' 의학에 관한 연구 (A Study on The 'Kao Zheng Pai'(考證派) of The Traditional Medicine of Japan)

  • 1.The 'Kao Zheng Pai'(考證派) comes from the 'Zhe Zhong Pai(折衷派)' and is a school that is influenced by the confucianism of the Qing dynasty. In Japan Inoue Kinga(井上金峨), Yoshida Koton(古田篁墩 $1745{\sim}1798$) became central members, and the rise of the methodology of historical research(考證學) influenced the members of the 'Zhe Zhong Pai', and the trend of historical research changed from confucianism to medicine, making a school of medicine based on the study of texts and proving that the classics were right. 2. Based on the function of 'Nei Qu Li'(內驅力) the 'Kao Zheng Pai', in the spirit of 'use confucianism as the base', researched letters, meanings and historical origins. Because they were influenced by the methodology of historical research(考證學) of the Qing era, they valued the evidential research of classic texts, and there was even one branch that did only historical research, the 'Rue Xue Kao Zheng Pai'(儒學考證派). Also, the 'Yi Xue Kao Zheng Pai'(醫學考證派) appeared by the influence of Yoshida Kouton and Kariya Ekisai(狩谷掖齋). 3. In the 'Kao Zheng Pai(考證派)'s theories and views the 'Yi Xue Kao Zheng Pai' did not look at medical scriptures like the "Huang Di Nei Jing"("黃帝內經") and did not do research on 'medical' related areas like acupuncture, the meridian and medicinal herbs. Since they were doctors that used medicine, they naturally were based on 'formulas'(方劑) and since their thoughts were based on the historical ideologies, they valued the "Shang Han Ja Bing Lun" which was revered as the 'ancestor of all formulas'(衆方之祖). 4. The lives of the important doctors of the 'Kao Zheng Pai' Meguro Dotaku(目黑道琢) Yamada Seichin(山田正珍), Yamada Kyoko(山田業廣), Mori Ritsi(森立之) Kitamura Naohara(喜多村直寬) are as follows. 1) Meguro Dotaku(目黑道琢 $1739{\sim}1798$) was born of lowly descent but, using his intelligence and knowledge, became a professor as a Shi Jing Yi(市井醫) and as a professor for 34 years at Ji Shou Guan(躋壽館) mastered the "Huang Di Nei Jing" after giving over 300 lectures. Since his pupil, Isawara Ken(伊澤蘭軒) taught the Lan Men Wu Zhe(蘭門五哲) and Shibue Chusai(澀江抽齋), Mori Ritsi(森立之), Okanishi Gentei(岡西玄亭), Kiyokawa Gendoh(淸川玄道) and Yamada Kyoko(山田業廣), Meguro Dotaku is considered the founder of the 'Yi Xue Kao Zheng Pai'. 2) The family of Yamada Seichin(山田正珍 $1749{\sim}1787$) had been medical officials in the Makufu(幕府) and the many books that his ancestors had left were the base of his art. Seichin learned from Shan Ben Bei Shan(山本北山), a 'Zhe Zhong Pai' scholar, and put his efforts into learning, teaching and researching the "Shang Han Lun"("傷寒論"). Living in a time between 'Gu Fang Pai'(古方派) member Nakanishi Goretada(中西惟忠) and 'Kao Zheng Pai' member Taki Motohiro(多紀元簡), he wrote 11 books, 2 of which express his thoughts and research clearly, the "Shang Han Lun Ji Cheng"("傷寒論集成") and "Shang Han Kao"("傷寒考"). His comparison of the 'six meridians'(3 yin, 3 yang) between the "Shang Han Lun" and the "Su Wen Re Lun"("素問 熱論") and his acknowledgement of the need and rationality of the concept of Yin-Yang and Deficient-Replete distinguishes him from the other 'Gu Fang Pai'. Also, his dissertation of the need for the concept doesn't use the theories of latter schools but uses the theory of the "Shang Han Lun" itself. He even researched the historical parts, such as terms like 'Shen Nong Chang Bai Cao'(神農嘗百草) and 'Cheng Qi Tang'(承氣湯). 3) The ancestor of Yamada Kyoko(山田業廣) was a court physician, and learned confucianism from Kao Zheng Pai's Ashikawa Genan(朝川善庵) and medicine from Isawa Ranken(伊澤蘭軒) and Taki Motokata(多紀元堅), and the secret to smallpox from Ikeda Keisui(池田京水). He later became a lecturer at the Edo Yi Xue Guan(醫學館) and was invited as the director to the Ji Zhong(濟衆) hospital. He also became the first owner of the Wen Zhi She(溫知社), whose main purpose was the revival of kampo, and launched the monthly magazine Wen Zi Yi Tan(溫知醫談). He also diagnosed and prescribed for the prince Ming Gong(明宮). His works include the "Jing Fang Bian"("經方辨"), "Shang Han Lun Si Ci"("傷寒論釋詞"), "Huang Zhao Zhu Jia Zhi Yan Ji Yao"("皇朝諸家治驗集要") and "Shang Han Ja Bing Lun Lei Juan"("傷寒雜病論類纂"). of these, the "Jing Fang Bian"("經方辨") states that the Shi Gao(石膏) used in the "Shang Han Lun" had three meanings-Fa Biao(發表), Qing Re(淸熱), Zi Yin(滋陰)-which were from 'symptoms', and first deducted the effects and then told of the reason. Another book, the "Jiu Zhe Tang Du Shu Ji"("九折堂讀書記") researched and translated the difficult parts of the "Shang Han Lun", "Jin Qui Yao Lue"("金匱要略"), "Qian Jin Fang"("千金方"), and "Wai Tai Mi Yao"("外臺秘要"). He usually analyzed the 'symptoms' of diseases but the composition, measurement, processing and application of medicine were all in the spectrum of 'analystic research' and 'researching analysis'. 4) The ancestors of Mori Ritsi(森立之 $1807{\sim}1885$) were warriors but he became a doctor by the will of his mother, and he learned from Shibue Chosai(澁江抽齋) and Isawaran Ken(伊澤蘭軒) and later became a pupil of Shou Gu Yi Zhai(狩谷掖齋), a historical research scholar. He then became a lecturer of medical herbs at the Yi Xue Guan, and later participated in the proofreading of "Yi Xin Fang"("醫心方") and with Chosai compiled the "Jing Ji Fang Gu Zhi"("經籍訪古志"). He visited the Chinese scholar Yang Shou Jing(楊守敬) in 1881 and exchanged books and ideas. Of his works, there are the collections(輯複本) of "Shen Nong Ben Cao Jing"("神農本草經") and "You Xiang Yi Hwa"("遊相醫話") and the records, notes, poems, and diaries such as "Zhi Yuan Man Lu"("枳園漫錄") and "Zhi Yuan Sui Bi"(枳園隨筆) that were not published. His thoughts were that in restoring the "Shen Nong Ben Cao Jing", "the herb to the doctor is like the "Shuo Wen Jie Zi"(說文解字) to the scholar", and he tried to restore the ancient herbal text using knowledge of medicine and investigation(考據), Also with Chosai he compiled the "Jing Ji Fang Gu Zhi"("經籍訪古志") using knowledge of ancient text. Ritzi left works on pure investigation, paid much attention to social problems, and through 12 years of poverty treated all people and animals in all branches of medicine, so he is called a 'half confucianist half doctor'(半儒半醫). 5) Kitamurana Ohira(喜多村直寬, $1804{\sim}1876$) learned scriptures and ancient texts from confucian scholar Asaka Gonsai(安積艮齋), and learned medicine from his father Huai Yaun(槐園), He became a teacher in the Yi Xue Guan in his middle ages, and to repay his country, he printed 266 volumes of "Yi Fang Lei Ju"("醫方類聚") and 1000 volumes of "Tai Ping Yu Lan"("太平禦覽") and devoted it to his country to be spread. His works are about 40 volumes including "Jin Qui Yao Lue Shu Yi"("金匱要略疏義") and "Lao Yi Zhi Yan"(老醫巵言) but most of them are researches on the "Shang Han Za Bing Lun". In his "Shang Han Lun Shu Yi"("傷寒論疏義") he shows the concept of the six meridians through the Yin-Yang, Superficial or internal, cold or hot, deficient or replete state of diseases, but did not match the names with the six meridians of the meridian theory, and this has something in common with the research based on the confucianism of Song(宋儒). In clinical treatment he was positive toward old and new methods and also the experience of civilians, but was negative toward western medicine. 6) The ancestor of the Taki family Tanbano Yasuyori(丹波康賴 $912{\sim}955$) became a Yi Bo Shi(醫博士) by his medical skills and compiled the "Yi Xin Fang"("醫心方"). His first son Tanbano Shigeaki(丹波重明) inherited the Shi Yao Yuan(施藥院) and the third son Tanbano Masatada(丹波雅忠) inherited the Dian You Tou(典藥頭). Masatada's descendents succeeded him for 25 generations until the family name was changed to Jin Bao(金保) and five generations later it was changed again to Duo Ji(多紀). The research scholar Taki Motohiro was in the third generation after the last name was changed to Taki, and his family kept an important part in the line of medical officers in Japan. Taki Motohiro(多紀元簡 $1755{\sim}1810$) was a teacher in the Yi Xue Guan where his father was residing, and became the physician for the general Jia Qi(家齊). He had a short temper and was not good at getting on in the world, and went against the will of the king and was banished from Ao Yi Shi(奧醫師). His most famous works, the "Shang Han Lun Ji Yi"("傷寒論輯義") and "Jin Qui Yao Lue Ji Yi"("金匱要略輯義") are the work of 20 years of collecting the theories of many schools and discussing, and is one of the most famous books on the "Shang Han Lun" in Japan. "Yi Sheng"("醫勝") is a collection of essays on research. Also there are the "Su Wen Shi"(素問識), "Ling Shu Shi"("靈樞識"), and the "Guan Ju Fang Yao Bu"("觀聚方要補"). Taki Motohiro(多紀元簡)'s position was succeeded by his third son Yuan Yin(元胤 $1789{\sim}1827$), and his works include works of research such as "Nan Jing Shu Jeng"(難經疏證), "Ti Ya"("體雅"), "Yao Ya"("藥雅"), "Ji Ya"(疾雅), "Ming Yi Gong An"(名醫公案), and "Yi Ji Kao"(醫籍考). The "Yi Ji Kao" is 80 volumes in length and lists about 3000 books on medicine in China before the Qing Dao Guang(道光), and under each title are the origin, number of volumes, state of existence, and, if possible, the preface, Ba Yu(跋語) and biography of the author. The younger sibling of Yuan Yin(元胤 $1789{\sim}1827$), Yuan Jian(元堅 $1795{\sim}1857$) expounded ancient writings at the Yi Xue Guan only after he reached middle age, was chosen for the Ao Yi Shi(奧醫師) and later became a Fa Yan(法眼), Fa Yin(法印) and Yu Chi(禦匙). He left about 15 texts, including "Su Wen Shao Shi"("素問紹識"), "Yi Xin Fang"("醫心方"), published in school, "Za Bing Guang Yao"("雜病廣要"), "Shang Han Guang Yao"("傷寒廣要"), and "Zhen Fu Yao Jue"("診腹要訣"). On the Taki family's founding and working of the Yi Xue Guan Yasuka Doumei(矢數道明) said they were "the people who took the initiative in Edo era kampo medicine" and evaluated their deeds in the fields of 'research of ancient text', the founding of Ji Shou Guan(躋壽館) and medical education', 'publication business', 'writing of medical text'. 5. The doctors of the 'Kao Zheng Pai' based their operations on the Edo Yi Xue Guan, and made groups with people with similar ideas to them, making a relationship 'net'. For example the three families of Duo Ji(多紀), Tang Chuan(湯川) and Xi Duo Cun(喜多村) married and adopted with and from each other and made prefaces and epitaphs for each other. Thus, the Taki family, the state science of the Makufu, the tendency of thinking, one's own interests and glory, one's own knowledge, the need of the society all played a role in the development of kampo medicine in the 18th and 19th century.

  • 본 연구에서 98종의 한약재로부터 인체에 무해한 물과 ethanol을 용매로 이용한 추출물에 대하여 xanthine oxidase(XOase)의 저해에 의한 통풍억제 효과를 측정한 결과 Chrysanthemum indicum L. (83.45%), Cuscuta chinensis (60.22%), Asiasarum sieboldi F. Maekawa (51.66%), Acorus gramineus (67.8%), Aconitum pseudo-laeve var. erectum (75.23%), Thuja orientalis (47.27%), Polygonum aviculare (53.98%), Carthami semen (63.99%), Syzygium aromaticum (40.22%) 등이 비교적 높은 XOase 저해율을 나타내었다. 감국의 경우 물과 ethanol 추출물 모두에서 XOase 저해활성이 가장 높게 측정되어 시료로 선발되었다. 감국의 유효성분을 확인한 결과 단순 고형분보다 고형분에 포함된 phenolic 성분이 XOase 저해 효과에 관여함이 입증되었다. 효율적인 추출을 위한 분쇄조건은 일반분쇄보다는 초미세 분쇄한 시료에서 추출 수율이 높아짐이 확인되었다. 추출된 감국 추출물의 XOase 저해 활성은 ethanol 추출물이 물 추출물에 비해 상대적으로 높은 저해 활성을 나타내었으며, phenolics의 첨가농도가 높아지면서 XOase에 대한 저해 활성은 농도 의존적으로 증가하는 것으로 확인되었다. 따라서 초미세 분쇄 기술은 감국으로부터 XOase 저해 물질의 추출 수율 증가를 목적으로 적용할 수 있는 기술로 개발이 가능할 것으로 판단되었다.

천마분말을 첨가한 식빵의 품질 특성 (Quality Characteristics of Bread Added with Gastrodia elata Blume Powder)

  • 혈압저하, 간질, 중풍, 두통에 효능이 있는 것으로 알려진 천마분말을 밀가루에 대하여 0%, (w/w) 0.5%, 1.0%, 1.5%, 2.0% 첨가하여 제조한 천마 빵의 제빵 적성을 조사하였다. Farinograph의 stability는 각각 57, 47, 38, 29, 20분으로 일정하게 감소하였다. 천마전분의 크기는 $36-60\;{\mu}m$ 정도였고 긴 타원형이었다. Amylograph 상의 최고점도는 대조구에 비해 천마분말 첨가 시 증가하는 것으로 나타났다. 식빵의 비용적은 $5.00{\sim}4.16$을 나타내었으며 0.5% 첨가시 대조구에 비해 비 용적이 10.2% 증가하였으나 2.0% 첨가시에는 오히려 16.8%의 감소율을 나타내었다. 천마 빵의 수분함량은 대조구에서는 43.57%였으나 천마분말 첨가에 따라 증가하는 경향이었다. 빵의 crust 및 crumb의 색도는 L, a, b 및 ${\Delta}E$값이 대조구와 유의적 차이를 나타내지 않았다. 천마 빵의 조직감에서도 대체로 대조구와 유의적 차이를 나타내지 않았으나 cohesiveness, chewiness, springness는 0.5% 첨가구에서 가장 높았으며 adhesiveness는 1.0%에서 가장 높게 나타났다. 내부특성과 외부특성 및 전반적인 기호도 향미 등에 대한 관능검사 결과 내부적 특성에서는 유의적 차이가 없었으며 전반적인 기호도, 향미, 겉색상, 쓴맛에서는 대조구가 가장 좋았고 부피, 껍질특성, 촉촉한 정도는 천마분말을 첨가가 좋은 것으로 나타났다. 대체로 대조구가 우수하나 천마분말 첨가 시 부드럽고 흰빵을 선호하는 사람들에게 좋은 기호도를 나타내고 0.5% 첨가구가 제빵 적성이 좋은 것으로 사료된다.처 특성간의 상관관계를 분석한 결과, 관능적으로 측정된 뭉치는 정도, 매끄러운 정도, 내부 촉촉함 특성은 기계적으로 측정된 부착성과는 양의 상관관계를 경도 및 껌성과는 음의 상관관계를 나타냈다. 그러나 경도 이외의 특성에서는 기계적 텍스처 특성과 관능적 텍스처 특성간에 상관관계가 매우 낮았다. 연구가 필요한 것으로 판단된다.키는 과정에서 산소 농도의 증가에 따라 formazan 생성량이 증가한 사실과 AsA 농도의 증가에 따라 formazan 생성량이 낮아지는 경향을 보인 사실, 그리고, AsA 농도에 따른 분해율을 조사한 결과, 본 실험에 사용된 $50\;{\mu}M$의 AsA 농도에서는 분해된 양이 거의 변화가 없는 것으로 보아 NBT가 아닌 AsA 자동산화반응 과정에서 생성된 $O_2\bar{{\bullet}}$에 의해 formazan이 생성된 것임이 확인되었다. 한편, Fe(III)이온 및 Cu(II)이온 존재 하에서 $0.01\;{\mu}M$이하의 낮은 농도에서는 $O_2\bar{{\bullet}}$ 생성이 중금속 이온 비존재 하에서의 경우보다 유의적으로 높은 것으로 밝혀졌지만, 그 이상의 농도에서는 낮은 수치를 나타내었다 이상의 결과로 보아 수용성계에서 뿐 만 아니라, 비수용성계에서도 중금속 이온 존재 하에서의 AsA 자동산화반응에서의 $O_2\bar{{\bullet}}$ 생성이 시사되었다.pm, $230^{\circ}C$에서 5.56 ppm으로 점진적으로 감소하였다. 이는 nonanoic acid, palmitic acid, stearic acid등이 고온가열에 의해 생성되었다가 이들이 열분해로 소실되었기 때문으로 생각된다.평균치는

인간 HepG2 Cell에서 항산화 효소의 mRNA 발현에 대한 잔대 에틸아세테이트 추출물 효과 (Effects of Adenophora triphylla Ethylacetate Extract on mRNA Levels of Antioxidant Enzymes in Human HepG2 Cells)

  • 잔대 뿌리는 우리나라에서 예로부터 민간약으로 이용되어 오고 있다. 본 연구에서는 인간 간세포인 HepG2에 잔대 뿌리의 에틸아세테이트 추출물을 처리했을 때 sodium nitroprusside(SNP)에 의해 유도된 세포 독성 및 항산화 유전자 발현에 미치는 영향력을 알아보았다. 먼저, 잔대 에틸아세테이트 추출물이 NO에 의해 유도된 세포 사멸을 저해할 수 있는지를 알아보기 위하여 HepG2 세포에 잔대 에틸아세테이트 추출물(각각 50과 100 $\mu$g/mL)을 24시간 먼저 처리한 후 세포내에서 NO을 생성시킬 수 있는 0.5 mM SNP를 처리하였다. NO에 의한 세포독성이 에틸아세테이트 추출물에 의해 저해되었다는 것을 mitochondrial dehydrogenase 활성을 알아보는 MTT assay를 실시하여 알아보았다. 더하여 우리는 잔대 에틸아세테이트 추출물이 세포내 항산화 방어 시스템인 Cu,Zn superoxide dismutase(SOD 1), Mn SOD(SOD 2), glutathione peroxidase(GPx), catalase와 glutathione metabolism과 관련되어져 있는 glutathione reductase(GR), $\gamma$-glutamyl-cystein synthetase(GCS), glutathione-S-transferase(GST), $\gamma$-glutamyltranspeptidase($\gamma$-GT), glucose-6-phosphate dehydrogenase(G6PD)의 mRNA 발현에 미치는 영향을 RT-PCR로 알아보았다. CAT, GCS 그리고 G6PD mRNA 수준이 잔대 에틸아세테이트 추출물 처리 후 증가하였으나, SOD 1, SOD 2, GPx, GST 그리고 $\gamma$-GT mRNA 수준은 변화지 않았다. 따라서 잔대 에틸아세테이트 추출물이 간접적 항산화 효과가 있고, 이들 효과는 아마 CAT, GCS, GR 그리고 G6PD 유전자 발현 증가에 의한 것이라고 추정되었다.

목향 헥산추출물이 대장암세포인 HT-29 세포의 증식에 미치는 영향 (Effect of the Hexane Extract of Saussurea lappa on the Growth of HT-29 Human Colon Cancer Cells)

  • 목향(Saussurea lappa)은 항암효과를 비롯하여 다양한 생리활성을 나타내는 것으로 알려지고 있으나 목향이 대장암에 미치는 영향에 대해 자세히 연구된 바가 없다. 본 연구에는 목향 헥산추출물이 인간의 대장에서 유래한 암세포인 HT-29 세포의 증식에 미치는 영향과 그 작용 기전에 대해 연구하였다. 헥산으로 목향을 추출하여 얻은 목향 헥산추출물을 HT-29 세포의 세포 배양액에 0, 1, 3, 5 ${\mu}g/mL$로 첨가하여 세포를 배양하였다. HT-29 세포의 증식은 목향 헥산추출물 처리 농도가 증가할수록 현저히 감소하였다. 인간의 대장에서 유래한 정상 상피세포인 FHC 세포의 증식은 목향 헥산추출물에 의해 변화하지 않았고, 인간의 피부에서 유래한 정상 섬유아세포인 CCD 1108Sk 세포 증식은 목향을 5${\mu}g/mL$ 농도로 72 시간 배양한 경우에만 감소하였다. 목향 헥산 추출물 처리에 의해 HT-29 세포의 세포주기 진행 중 sub G1기에 머무른 세포수가 증가하였고, 세포사멸 세포수가 현저히 증가하였다. 세포사멸의 주요한 조절인자인 Bcl-2 family 단백질 중 Bcl-2은 목향 헥산추출물에 의해 변화하지 않았으나 Bax는 유의적으로 증가하였다. Bcl-2 family 단백질과 더불어 세포사멸 조절에 중요한 역할을 하는 caspases의 활성형인 cleaved caspase-8, -9, -7, -3가 목향 헥산추출물에 의해 증가하였다. 또한 목향 헥산추출물 처리 농도가 증가할수록 cleaved PARP 단백질 수준도 현저히 증가하였다. 이 결과는 목향 헥산추출물이 세포사멸을 유도하여 대장암세포인 HT-29 세포의 증식을 억제함을 나타내며, 목향 헥산추출물에 의해 HT-29 세포의 세포사멸은 Bax 증가와caspases의 활성 증가에 의한 것임을 나타낸다. 본 연구는 목향을 독성과 부작용이 적은 암예방제나 항암제로 개발할 수 있는 가능성을 제시한다. 그러나 목향을 이용하여 제품 개발을 위해서는 유효 성분 동정 및 동물시험 등의 연구 수행이 필요할 것으로 사료된다.

홍삼추출액은 lipoteichoic acid로 자극된 소교세포에서 Akt 및 MAPK 의존적으로 heme oxygenase-1 발현을 유도함으로써 NO 생성을 억제함 (A Formulated Korean Red Ginseng Extract Inhibited Nitric Oxide Production through Akt- and Mitogen Activated Protein Kinase-dependent Heme Oxygenase-1 Upregulation in Lipoteichoic Acid-stimulated Microglial Cells)

  • 생삼을 쪄서 건조시킨 홍삼은 전통적으로 사용되고 있는 약재로서 면역, 내분비 및 중추신경계 작용을 증진시키며 염증을 억제하는 효과가 있는 것으로 알려져 있다. 본 연구에서는 그람 양성균의 세포벽 성분인 lipoteichoic acid (LTA)에 의한 염증반응에 홍삼추출액(RGE)이 항염증 효과를 가지는지 관찰하고 그 작용 기전을 연구하였다. BV-2 소교세포에서 RGE는 세포에 독성을 유도하지 않으면서 LTA로 인한 nitric oxide (NO)의 생성과 inducible nitric oxide synthase (iNOS) 발현을 억제하였으며, NF-kB p65의 핵으로의 이동과 IkB-a의 분해 또한 억제하였다. 한편, RGE는 농도의존적으로 heme oxygenase-1 (HO-1)의 발현을 유도하였으며, HO-1 siRNA를 처리했을 때는 RGE가 iNOS의 발현을 억제하지 못하였다. RGE는 HO-1의 발현에 관여하는 전사인자인 nuclear factor E2-related factor 2 (Nrf2)를 핵으로 이동을 촉진시켰다. 또한 RGE에 의한 HO-1의 발현은 phosphatidylinositol-3-kinase(PI-3K) 및 MAPK 억제제에 의해 감소되었으며, RGE가 Akt와 ERK, p38, JNK의 인산화를 유도하였다. 이상의 결과를 종합해보면, RGE는 PI-3K/Akt 및 ERK, p38, JNK 신호전달과정을 통해 HO-1의 발현을 유도함으로써 NO와 같은 염증매개물질의 생성을 억제한다는 것을 알 수 있다. 그러므로 홍삼추출액은 그람 양성균에 의한 신경염증과 염증관련 신경계 질환의 치료제로서 사용될 수 있을 것이라 사료된다.
