Hydrogen Peroxide에 의해 유도된 남성생식세포의 세포독성에 미치는 소양(銷陽)의 효과 (Protective effect of Cynomorii Herba extract on $H_2O_2$-induced cytotoxicity)

  • The purpose of this study is to examine the protective effect of Cynomorii herba on $H_2O_2$-induced cytotoxicity in male germ cells. The effects were studied by a modified MTT assay. Hydrogen peroxide significantly induced cytotoxicity in GC-1 spg cells, and co-treatment or pre-treatment of Cynomorii herba extract has reduced the cytotoxicity in a dose dependant manner. These results suggest that Cynomorii herba extract increases the survival rate of GC-1 spg cells through the antioxidant effect against $H_2O_2$-induced cytotoxicity.

대체의학으로서 백화사설초 급여가 돼지 생산성에 미치는 효과 (Effects of Oldenlandidae Herba as an Alternative Medicine on the Swine Productivity)

  • As control of swine consumptive disease is the key of success in pig farming business, the effect of Oldenlandidae Herba on prevention of swine diseases as an alternative medicine was investigated in-vivo. As it contains substances such as ${\beta}$-sitisterol, ursolic acid and 3-O-${\beta}$-D-giucoside, it is effective on suppression of colonic tumor in rats and also, is widely used as an preventative medicine of cancer and inflammation in Chinese medicine. In this experiment, sows and piglets on two different farms were treated with Oldenlandidae Herba, and they recoded weaning rate greater than 80%, which were higher than national average 70% (PSY 68%). Meanwhile, neither organ accumulation nor toxicity has been detected on the basis of hematology and serum chemistry. In conclusion, the extracts of Oldenlandidae Herba is expected to be an good alternative medicines for swine productivity.

  • Objectives The purpose of this study is to find out the effect of Taraxaci Herba Extract (THE), LPS, on pro-inflammatory mediatory. Methods After the treatment of Taraxaci Herba MeOH extract dissolved in EMEM for 1 hour prior to the addition of LPS ($1\;{\mu}g/ml$), cell viability was measured by MTT assay, Nitric Oxide production was monitored by measuring the nitrite content in culture medium. And levels of cytokine and PGE2 were analyzed by sandwich immunoassays. Results THE inhibited the production of nitrite and nitrate (0.03 and 0.1 mg/ml), TNF-$\alpha$, (0.03 and 0.1 mg/ml), IL-$1{\beta}$(0.03 and 0.1 mg/ml), IL-6 (0.01, 0.03 and 0.1 mg/ml) and PGE2(0.03 and 0.1 mg/ml) activated with LPS. In Raw 264.7 cells activated with lipopolysaccharide. Conclusions According to the results above, Taraxaci Herba can produce anti-inflammatory effect, which may play a role in adjunctive therapy in Gram-negative bacterial infections.

HPLC-UV에 의한 진득찰과 털진득찰의 Kirenol 정량분석 (Quantitative Analysis of Kirenol in Siegesbeckia glabrescens and S. pubescens by HPLC-UV)

  • Many diterpenoids from Siegesbeckia species (Compositae) and their anti-inflammatory actions have been examined. In this research, high-performance liquid chromatography-ultraviolet spectrophotometer (HPLC-UV) method was used to compare the quantitative level of kirenol (ent-pimarane-type diterpenoid) in the aerial parts of Korean S. glabrescens and S. pubescens and the Chinese Siegesbeckiae Herba. Fingerprints of the two HPLC chromatograms of Korean S. glabrescens and S. pubescens were similar, but considerably different from Chinese Siegesbeckiae Herba. The content of kirenol in S. pubescens ($16.51{\pm}0.10$ mg/ml dry weight as mean${\pm}$RSD) was higher than S. glabrescens ($13.48{\pm}0.12$ mg/g dry weight). These values were considerably higher than the Chinese Siegesbeckiae Herba ($1.55{\pm}0.74$ mg/g dry weight). Thin layer chromatography (TLC) analysis demonstrated the containing of kirenol in the three plant materials, but the presence of siegeskaurolic acid (entkaurane-type diterpenoid) only in the Chinese Siegesbeckiae Herba.

Peripubertal Administration of Icariin and Icaritin Advances Pubertal Development in Female Rats

  • Epimedii Herba is a traditional medicinal herb used in Korea and China and exerts estrogenic activity. In this study, we investigated the effect of peripubertal administration of Epimedii Herba on pubertal development in female rats using a modified protocol of the rodent 20-day pubertal female assay. Female Sprague-Dawley rats (21 days old after weaning, 10 rats per group) were divided into five groups: saline (Con), ethinyl estradiol (E2), Epimedii Herba ext (Ext), icariin (ICI), and icaritin (ICT), which were administered by oral gavage (E2 by subcutaneous injection) from postnatal day (PND) 21 through PND40. The time to vaginal opening (VO) was shorter for the Epimedii groups, particularly for the ICT group (p<0.05). Treatment with ICI and ICT significantly increased the duration of the estrus cycle (ICI, 2.78 days; ICT, 4.0 days; control, 1.78 days). Ovary weight was reduced by E2 treatment and increased by the Ext, ICI, and ICT treatments while the weight of the uterus and pituitary glands increased significantly only in the E2 and ICT groups. Although Epimedii Herba displayed relatively weak estrogenic activity, its repeated administration could affect pubertal development in female rats.

담죽엽의 충치균에 대한 항균활성 및 항염효과 (Antibacterial Activity against the Streptococcus mutans and Anti-inflammatory Effect of Lophatheri Herba)

  • Lophatheri Herba of Lophatherum gracile Bronghiart(Gramineae) has long been used for treatment of inflammation, fever and edema in Korea. In order to investigate antibacterial activity of the Lophatheri Herba against Streptococcus mutans ATCC27351, paper disc test and pH check were carried out with 80% ethanol extract of Lophatheri Herba(LEX). The LEX showed significant antibacterial activity. And at the dose of 50 mg/kg, LEX showed signifiant inhibition on the paw edema, vascular permeability and myeloperoxidase activity in rat's paw tissue. These results indicate that LEX has antibacterial activity against the Streptococcus mutans and anti-inflammatory effect.

  • The methanolic extract of Lysimachiae christinae Herba, which has been used for diuretic, calculi remover and jaundice remedy in oriental countries, was found to possess analgesic and antiinflammatory effect in Freund's complete adjuvant treated rat. From ethyl acetate fraction of the herb a phenyl propanoid isoferulic acid was isolated as an active principle.

  • In order to search for antioxidants from the plants, eighty-two kinds of herbal medicines were investigated. The MeOH extracts of Euryales Semen, Alpiniae Officinari Rhizoma. Drynariae Rhizoma, Sophorae Flos, Trachelospermi Caulis, Crassirhizomae Rhizoma, Euphorbiae lathyridis Semen, Lini Semen, Myristicae Semen, Epimedii Herba, Santali Lignum rubrum, Perillae Herba, Amomi Tsao-Ko Fructus and Garanii Herba showed potent antioxidative activities using the 1,1-diphenyl-2-picrylhydrazyl free radical generating system.   • Experimental studies were done to investigate analgesic and antiphlogistic actions of Violae Herba. Its water extract showed a significant dose dependent analgesic effect against writhing syndrome induced by acetic acid as well as in hot plate test in mice. The extract also showed a significant inhibitory effect against paw-edema induced by egg-white in rats.

  • The method extracts of Patriniae radix, Dianthii herba, Melandrii herba, Echinopii radix, Siegesbeckiae herba and Magnoliae cortex showed a significant elevation of serum transam inase activity accompanied by fatty degeneration and Kupffer accompanied by fatty degeneration and Kupffer cell activation in hepatic cells.
