Antioxidative and Hepatoprotective Effect of Compounds from the Flowers of Bidens bipinnata L. (도깨비바늘 꽃 성분의 항산화 및 간보호 효과)

  • Activity-guided fractionation of the EtOH extract of flowers of Bidens bipinnata L. have been furnished five flavonoids, sulfuretin(1), butein(2), 7,8,3',4'-tetrahydroxyflavanone(3), maritimetin(4) and okanin(5). All of the compounds showed significant activities on both linoleic acid peroxidation inhibition and DPPH radical scavenging effect. The evaluation for protective effect of isolated compounds against tacrine-induced cytotoxicity in human liver-derived Hep G2 cells was conducted. Compounds 1, 3, 4 and 5 showed significant protective effects with the $EC_{50}$ values of $36.1{\pm}0.9$, $23.3{\pm}0.7$, $41.0{\pm}1.0$ and $29.8{\pm}1.1{\mu}M$, respectively. Silybin, one of the well-known hepatoprotective agents, used as a positive control, and also showed protective effect with an $EC_{50}$ value of $84.3{\pm}0.7{\mu}M$.

Effect of Daewhang-whangryunhaedok-Tang on Carbon Tetrachloride-induced Hepatotoxicity and Acute Toxicity (대황황련해독탕의 사염화탄소 유발 간장해 보호효과 및 급성독성)

  • This study was performed to evaluate hepatoprotective effect of Daewhang-whangryunhaedok-Tang(DWT) on liver injured rats induced by $CCl_4$ and the acute oral toxicity of it in mice. The activities of serum transaminase(ALT/AST), alkaline phosphatase(ALP) and lactic dehydrogenase (LDH), the levels of serum total cholesterol(TC) and triglyceride(TG), change of liver enlargement, and inhibitory activities of lipid peroxidation, catalase and glutathione-S-transferase(GST) in liver microsome were determined in hepatotoxic rats induced by $CCl_4$ DWT DWT was significantly reduced the serum ALT, AST, ALP, LDH, TC and TG levels. And, the increase of lipid peroxidation, decrease of catalase and GST activities in the liver microsome of $CCl_4$-intoxicated rat were significantly improved by the treatment of DWT. Male and female mice were administered maximum dosages of 5,000 mg/kg b.w. of DWT. After single oral administration of DWT to mice, we observed them daily for 2 weeks. DWT did not induce any toxic signs in the mortalities, clinical signs, body weight changes, and gross necropsy findings of mice. Based on these results, it is concluded that DWT may have the hepatoprotective effect on $CCl_4$ induced hepatotoxicity in rats. Also, DWT may have no side effect and its $LD_{50}$ value may be over 5,000 mg/kg b.w. in mice.

Effect of Puerariae Radix Methanol Extract on Benzo(a)pyrenc -in - duced Hepatotoxicity in Rats (갈근 메탄올 엑기스가 흰쥐에 있어서 Benzo(a)pyrene에 의해 유도된 간장해에 미치 는 영향)

  • The present study was conducted to evaluate the hepatoprotective effect of puerariae Radix methanol extract on benzo(a) pyrene(B(a)P) - induced liver injuries in rats. In vitro experiment, primary cultured hepatocytes (5X105 cells/$m\ell$) were cultured for 20~24 hours after adding puerariae Radix mehtanol extract(32$\mu\textrm{g}$/$m\ell$) and B(a)P(50 uM). In vivo experiment, Puerariae Radix methanol extract(0.25 g/kg/day, per os) was administered for 7 days and B(a)P(0.1 mg/kg/day, intraperitoneally) was given after the last administration of extract. And then the hepatoprotective effect of Puerariae Radix methanol extract was investigated biochemically through in vitro and in vivo experiments. Namely, activities of enzymes (GOT, GPT and LDH) were measured and 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) assay were carried out in vitro cell culture study and GOT, GPT, LDH and ALP activities and HDL-cholesterol, total cholesterol and triglyceride contents were performed in vivo study. In vitro experiment, as a result of enzyme activity measurement(GOT, GPT and LDH) and MTT assay, GOT,GPT and LDH activities changed by B(a)P were recovered to normal levels and hepatocytes impaired by B(a)P were recovered to normal. In vivo experiment, Puerariae Radix methanol extract significantly decreased the enzyme activities(GOT, GPT, ALP and LDH in serum and GPT and ALP in tissue) and lipid contents in comparison to B(a)P-treated group.

Hepatoprotective effect of Ikwiseungyang-tang via Nrf2 activation (Nrf2 활성화를 통한 익위승양탕(益胃升陽湯)의 간세포 보호 효과)

  • Objectives : Oxidative stress is a important cause of liver disease, and regulation of oxidative stress is essential to maintain the normal metabolic function of the liver. Until a recent date, there has been no studies on the hepatoprotective effect of Ikwiseungyang-tang (IWSYT). Therefore, this study aims to demonstrate the hepatoprotective effect of IWSYT and its related molecular mechanisms on arachidonic acid (AA) + iron induced oxidative stress model in HepG2 cells. Methods : To determine the cytoprotective effect of IWSYT against AA + iron-induced oxidative stress, cell viability, apoptosis-related proteins, intracellular reactive oxygen species (ROS), GSH, and mitochondrial membrane potential (MMP) were measured. Nuclear factor erythroid 2-related factor 2 (Nrf2) activation was analyzed by immunoblot analysis. In addition, Nrf2 transcription activation through ARE binding was measured by reporter gene assays, and the expression of the Nrf2 target antioxidant genes were confirmed by immunoblot analysis. Results : IWSYT increased cell viability from cell death induced by AA + Iron, and inhibited apoptosis by regulating apoptosis-related proteins. Furthermore, IWSYT protected cells by inhibiting intracellular ROS production, GSH depletion, and MMP degradation. Nrf2 activation was increased by IWSYT, and Nrf2 target genes were activated by IWSYT too. Conclusions : These results suggest that IWSYT can protect hepatocytes from oxidative stress through Nrf2 activation and can be potentially applied in the prevention and treatment of liver damage.

  • Solanum lyratum (Solanaceae) has been used as a traditional analgesic, antipyretic and hepatoprotective agents in Korea. In this study, we investigated the hepatoprotective effect of ethylacetate extract of Solanum lyratum (SL) on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of SL (150, 300 mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase levels.   • The protective effect of Bifidobacterium breve K-110, K-111 and B. infantis K-525 on carbon tetrachloride induced hepatotoxicity in rats was investigated. Among them, B. infantis K-525 had the most potent hepatoprotective activity. It reduced serum aspartate aminotransferase and alanine aminotransferase levels to 51% and 80% of the $CCl_{4}-treated$ groups, respectively. In rat liver homogenate intoxificated with $CCl_{4}$, B. infantis K-525 inhibited in vitro as well as in vivo lipid peroxidation more than the other Bifidobacteria.

Effect of Ganoderma lucidum Mycelial Fractions on the Functional Recovery of Primary Cultured Hepatocytes. (영지 균사체 분획이 일차배양 간세포 기능회복에 미치는 영향)

  • The cultured mycelia of Ganoderma lucidum were extracted and the extract was separated into six fractions by organic solvent fractionation. The antihepatotoxic activity of all the fractions was evaluated by measuring activities of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT). Among the fractions tested, the high-polarity fractions such as aqueous and n-butanol fractions significantly reduced activities of GPT and GOT in CCl4- and galactosamine-intoxicated rat primary hepatocytes. When intracellular synthetic activities were measured by pulsing the rate primary cultured hepatocytes with [3H]-uridine and [3H]-leucine, activities of DNA, RNA and protein. When direct toxicities of the fractions were measured against human hepatoma(SK-Hep-1), the non-polarity fractions such as n-butanol and ethyl acetate fractions showed potent direct cytotoxicities even at the concentration of 1 $\mu\textrm{g}$/ml. These data showed that Ganoderma lucidum has hepatoprotective and hepatotoxic recovery principles in its mycelia.

  • The effect of oral treatment with Ficus racemosa leaf extract (400 mg/kg for seven days) were studied on hepatic damage induced by paracetamol (750 mg/kg, i.p.) in rats. Biochemical parameter like SGOT, SGPT, serum bilirubin and alkaline phosphatase were estimated to assess liver function. These biochemical observations were supplemented by histopathological examination of liver sections. The activity of extract was also comparable to Neutrosec a known hepatoprotective formulation.

Protective effect of Jageum-Jung on chlorpyrifos-induced acute toxicity in ICR mice

  • Chlorpyrifos (CPF) is one of the most heavily used organophosphate pesticides and is useful as an insecticide drug. However, CPF also causes toxic effects in nontarget organisms, including humans and animals. Jageum-Jung (JGJ) is a traditional oriental medicine, composed of five specific herbs with antioxidant and hepatoprotective properties, used for detoxification. In the present study, highly concentrated CPF was orally administrated to male Institute of Cancer Research mice to produce acute toxicity, and the protective effects of JGJ administration were investigated through statistical analysis of changes in body and organ weights and serum biochemical parameters. JGJ caused body and organ weights to recover and reduced the levels of serum biochemical parameters indicative of liver damage, such as glutamic oxalate transaminase, glutamic pyruvate transaminase, alkaline phosphatase, lactic dehydrogenase, urea, glucose, total cholesterol, and triglyceride, that had been increased by CPF treatment. Our results demonstrated that JGJ ameliorates the effects of acute chlorpyrifos-induced toxicity. Therefore, JGJ has the potential to be used as a traditional medicine to alleviate insecticide toxicity.

  • Zingiber officinale is a spice which is having a strong historical medicinal background and used by different systems of medicine for various ailments. It is also used to maintain the characteristic sharpness and tangy essence of food and beverages and used in carbonated drinks, as a conserve in sugar syrup. Ginger considered as an incredible drug for inflammation, gastrointestinal problems, pain and sexual problems since centuries. There are many studies carried out in recent years, which proves that the active constituents of ginger shows a variety of pharmacological effects like antiemetic, hepatoprotective, anti hyperlipidemic and antibacterial. The most momentous among all the nutraceutical aspects of ginger are its positive control on gastrointestinal tract including digestive stimulant action, anticancer effect and anti-inflammatory effect.
