Objectives : In Traditional Korean medicine, Daucus carota L. has been used for treating dyspepsia, diarrhea, dysentery and cough. Recent pharmacognosic evidence showed D. carota has anti-oxidant, anti-cancer, anti-fungal, and hypotensive effects. Present study investigated hepato-protective effect of D. carota ethanol extract (DCE) against oxidative stress in HepG2 cells. Methods : After HepG2 cells were pretreated with different concentrations of DCE, the cells were exposed to tert-butyl hydroperoxide (tBHP) for inducing oxidative stress. Cell viability, hydrogen peroxide production, glutathione concentration, and mitochondrial membrane potentials were measured to explore hepato-protective effect of DCE. Phosphorylation of AMP-activated protein kinase (AMPK) and effect of compound C on cell viability were determined to investigate the role of AMPK on DCE-mediated cytoprotection. Results : DCE significantly decreased the tBHP-mediated cytotoxicity in a concentration dependent manner and reduced the changes on apoptosis-related proteins by tBHP in HepG2 cells. In addition, DCE significantly prevented hydrogen peroxide production, glutathione depletion, and mitochondrial membrane impairment induced by tBHP. Treatment with DCE increased phosphorylation of AMPK, and the DCE-mediated cytoprotection was abolished by pretreatment with compound C. Conclusions : These results demonstrate that DCE can protect hepatocytes from oxidative stress through activation of AMPK.

The hepato-protective effects of the extract from Protaetia brevitarsis against hepatotoxicity by carbon tetrachloride ($CCI_4$) were studied in rats. The rats were orally treated with $CCI_4$ (50% in corn oil) at initial dose of $1\;m{\ell}/kg$ followed by $0.5m{\ell}/kg$ four times during 2-week period. The extract of P. brevitarsis (50, 100 or 200 mg/kg) or its vehicle was administered day after day from 1 week before $CCI_4$ Injection during five weeks. $CCI_4$ induced hepato-celluar degeneration and necrosis induced to increase in serum aspartate amintransferase (AST) and alanine aminotransferase (ALT) levels. In biochemical analyses, thiobarbituric acid-reactive substances (TBARS) and antioxidant enzymes such as superoxide dismutase (SOD) and catalase in hepatic tissues were remarkably increased by $CCI_4$ treatment. Not only increases in serum AST and ALT, but also induction of lipid peroxidation and antioxidant enzymes in hepatic tissues caused by $CCI_4$ were significantly attenuated by the P. brevitarsis extract in a dose-dependent manner. Such hepato-protective effects of P. brevitarsis extract were confirmed by histopathological examinations, wherein only mild hepatocytic vacuolations were observed in the liver of rats treated with a high dose (100 mg/kg) of P. brevitarsis extract in comparison with severe hepatocytic degenerations administered with $CCI_4$ alone. From these results, it is suggested that the extract of Protaetia brevitarsis could be a promising candidate for the protection of liver injury, based on the preventive effects against morphological cellular injuries, lipid peroxidation and serum biochemical parameters.
In order to evaluate the protective effects of galangin on $CCl_4$-induced hepatotoxicity, GOT, GPT and malondialdehyde(MDA) values were measured in ICR mice. Galangin,a flavonoid compound, was administered orally for six days and immediately $CCl_4$ was injected intraperitoneally after the last dose of galangin. Mice were sacrificed at 24h after the administration of $CCl_4$. In the multiple pre-treatments for 6 consecutive days, galangin showed more potent protective effects than silymarin as reference active compound in serum GOT, GPT and MDA values in the liver at all doses tested. Antioxidative activity was determined by measuring the amounts of MDA formed from ethyl linoleate by $H_2O_2$ in vitro. Galangin showed higher inhibition than silymarin. These results demonstrate a possible hepato-protective role of galangin against $CCl_4$-induced hepatotoxicity in vivo and $H_2O_2$-induced lipid peroxidation in vitro. Therefore, galangin may be capable of protecting hepatotoxicity.
The protective effect of Carthamus tinctorius L. Semen on the carbon tetrachloride induced liver damaged rats were studied. First, methanol extract was prepared and the extract was fractionated with hexane, $CHCl_3$, BuOH and $H_2O$ respectively. Animals were divided into 6 groups and each group was treated with each fraction respectively. To investigate the hepato-protective effect of Carthamus tinctorius L. Semen AST, ALT, albumin, TP, cholesterol, TG, creatinine and total bilirubin values were measured in each treated group and compared with those of control group. GST activity was increased in BuOH group compared with the control group. In malondialdehyde levels, all fractions was decreased compared with the control group. In histopathologic examination, hexane and $H_2O$ fractions of Carthamus tinctorius L. Semen observed mild degree of ballooning degeneration. The results show the protective effect of Hexane,$CHCl_3$, BuOH and $H_2O$ fractions on hepatotoxicity of $CHl_4$ by decreasing ALT, AST, bilirubin, cholesterol, TG and BUN. It seems that the decrease of MDA are related to the recovery effect. The protective effects of Carthamus tinctorius L. Semen fractions in hepatotoxic pathogenesis by $CHl_4$ was suggested in blood chemistry analysis and histopathologic examination.
In this study, the ameliorating effect of soybean embryos on the impact of alcohol consumption was investigated on rat hepatocytes and in reducing total serum cholesterol levels and total serum lipid levels. Liver histology and two clinically important enzyme markers, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), of rats administered with both alcohol and soybean embryo were compared with a control group. The treatment regimen of soybean embryo significantly reduced the serum ALT and AST levels of the subjects, demonstrating the hepato-protective effects of soybean embryo. Electron microscopy indicated that the administration of soybean embryo preserved the important hepatocyte structures and prevented the presence of lipid droplets and secondary lysosomes. Furthermore, total cholesterol and total lipid levels were significantly reduced. These results indicate that treatment with soybean embryo can positively mediate the effects of alcohol on hepatocytes and general liver functions.

Effects of the extract of oriental herbal composition, Jang Baek Union (JBU), on hangover and liver function were studied in rats. To investigate anti-hangover effect, alcohol concentration and acetaldehyde concentration of blood were measured after oral ethanol treatment. Blood alcohol concentration was significantly reduced by pre-treatment of JBU extract. The effect of JBU extract on the blood acetaldehyde concentration was more significant than commercial product used as a positive control. To investigate hepato-protective effect, serum GOT and GPT levels and histological changes of liver tissue were analyzed in $CCl_4$-treated rats. JBU extract adminstration significantly inhibited the increase of serum GPT and GOT levels induced by $CCl_4$-treatment. Moreover, histological injuries of liver tissue such as fatty changes and sinusoidal leukocytosis were inhibited by JBU extract.
The hepatoprotective activity of six extracts (BE, EE, HH, PS-1, PS-2, KP) from Artimisia iwayomogi was investigated against experimentally produced hepatic damages. Silymarin, DDB and UDCA were used as reference compounds. Treatment with PS-1 extract reduced hepatic demages induced by $CCl_4$, acetaminophen and ANIT but it did not alter ethionine-induced hepatotoxicity In addition, PS-1 extract showed a protective effect against chronic $CCl_4$-induced hepatotoxicity as well as liver regeneration. PS-2 and KP extracts exhibited significant antihepatotoxic effects on D-galactosamine-induced hepatitis. Treatment with EE extract inhibited ethionine-induced fatty liver. These data indicate that the PS-1 extract is the roost hepato-protective constituent and rationalize the traditional use of this plant in hepatobiliary disorders.
The Hovenia extracts showed a significant hepato-protective activity against alcohol and CC4-induced hepatotoxicity. Injection of CC4 and alcohol induced liver injury and increased serum GOT (glutamic oxaloacetic transaminase) and GPT (glutamic pyuvic transaminase) levels in injured rats. The ADH (alcohol dehydrogenase)-like and ALDH (aldehyde dehydrogenase)-like activities in the Hovenia extracts were studied. Their activities in the extracts of Hovenia wood were higher than the extracts of other parts of Hovenia. And then Hovenia wood extracts have high effects on the alcohol and acetealdehyde degradation. The Hovenia extracts were decreased concentration of fusel oil. The ADH-like and ALDH-like activities in the Hovenia wood extract were 1.14 $unit/{\mu}l$, 0.33$unit/{\mu}l$ respectively.

Objectives : Jasminum officinale L. var. grandiflorum is used as a traditional or folk remedy in China to treat arthritis, hepatitis, duodenitis, conjunctivitis, gastritis, and diarrhea. In this study, we aimed to study the hepatocyte protective activity and molecular mechanism of Jasminum officinale L. var. grandiflorum aqueous extract (JGW) using HepG2 hepatocyte cell lines. Methods : HepG2 cells were pretreated with diverse concentrations of JGW, and then the cells were exposed to tert-butyl hydroperoxide (tBHP) for inducing oxidative stress. Hydrogen peroxide (H2O2) production, glutathione (GSH) concentration, mitochondrial membrane potential (MMP) and cell viability were measured to investigate hepato-protective effects of JGW. Phosphorylation of AMP-activated protein kinases (AMPK), acetyl coenzyme A carboxylase (ACC) and effects of compound C on cell viability were examined to observe the role of AMPK on JGW-mediated cytoprotection. Results : Pretreatment with JGW (10-300 ㎍/mL) significantly suppressed cytotoxicity induced by tBHP in a concentration dependent manner and reduced the expression of cleaved PARP and cleaved caspase-3 proteins related to apoptosis in HepG2 cells. In addition, pretreatment with JGW significantly prevented the increase in H2O2 production, GSH depletion, and lower MMP induced by tBHP. Treatment with JGW (30 minutes of incubation and concentrations of 100 and 300 ㎍/mL) increased the phosphorylation of AMPK and ACC and treatment with compound C, a chemical inhibitor of AMPK, inhibited the cytoprotective effect of JGW. Conclusions : Our results demonstrated that JGW may protect hepatocytes from oxidative stress via activation of AMPK.
This study was conducted to improve the useful components and biological activity of Lentinula edodes fermented by lactic acid bacteria (LAB). Three LAB strains (Lactobacillus brevis KCCM 11904, L. plantarum KCCM 354469, and L. fermentum KCCM 12116) were inoculated and used for L. edodes hot water extract (10%, 20%, 30%) fermentation. LAB fermentation of L. edodes hot water extracts decreased pH and thus were more acidic than non-fermented L. edodes hot water extract. β-glucan and ergothioneine contents were increased by L. edodes in a concentration-dependent manner. The ergothioneine and β-glucan contents were highest in fermented with 30% L. edodes hot water extract fermented by L. plantarum and L. brevis (40.48 mg/100 g and 13.94%, respectively). The hepato-protective effect of fermented L. edodes hot water extracts by the three LAB were tested using Sprague-Dawley rat primary hepatocytes. In primary hepatocytes obtained following liver injury induced by acetaminophen, fermented L. edodes hot water extracts by the three LAB showed protective effects, as evident by reduction of the aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase liver markers. The collective results indicate that the fermented L. edodes hot water extracts obtained using LAB are potentially valuable in preventing or treating liver disease.
