The hepato-protective effects of the extract from Protaetia brevitarsis against hepatotoxicity by carbon tetrachloride ($CCI_4$) were studied in rats. The rats were orally treated with $CCI_4$ (50% in corn oil) at initial dose of $1\;m{\ell}/kg$ followed by $0.5m{\ell}/kg$ four times during 2-week period. The extract of P. brevitarsis (50, 100 or 200 mg/kg) or its vehicle was administered day after day from 1 week before $CCI_4$ Injection during five weeks. $CCI_4$ induced hepato-celluar degeneration and necrosis induced to increase in serum aspartate amintransferase (AST) and alanine aminotransferase (ALT) levels. In biochemical analyses, thiobarbituric acid-reactive substances (TBARS) and antioxidant enzymes such as superoxide dismutase (SOD) and catalase in hepatic tissues were remarkably increased by $CCI_4$ treatment. Not only increases in serum AST and ALT, but also induction of lipid peroxidation and antioxidant enzymes in hepatic tissues caused by $CCI_4$ were significantly attenuated by the P. brevitarsis extract in a dose-dependent manner. Such hepato-protective effects of P. brevitarsis extract were confirmed by histopathological examinations, wherein only mild hepatocytic vacuolations were observed in the liver of rats treated with a high dose (100 mg/kg) of P. brevitarsis extract in comparison with severe hepatocytic degenerations administered with $CCI_4$ alone. From these results, it is suggested that the extract of Protaetia brevitarsis could be a promising candidate for the protection of liver injury, based on the preventive effects against morphological cellular injuries, lipid peroxidation and serum biochemical parameters.
Matricaria Chammomillal L., Foemiculum Vulgare mill, and Plantago Psylium L. have been screened for their hepato protective activities against liver damge induced by $CCl_4$ intoxication in mice. Hydroalcoholic extractions (2:8) of herbal drugs were concentrated in vacuo and concentrated crude extracts of Matrica Chammomilla L. and Foeniculum Vulgare mill were orally administered at doses of 100 mg/kg, 200 mg/kg, 400 mg/kg, and 800 mg/kg. Plantago Psyllium was given at doses of 50 mg/kg, 100 mg/kg, 200 mg/kg, and 400 mg/kg. Liver protective activities of these herbs were determined after administration of $CCl_4$ Liver size, serum enzyme activities, sleeping time, and histopatology of the liver were examined one hour after administration of $CCl_4$. ALT and AST activities, liver weight and sleeping time decreased in groups that received 400 mg/kg of Matricaria Chammomilla L. or Foeniculum Vulgare. Histological investigation showed significant increase in hepatic cell regeneration and reduction in liver injury. The group that received 100 mg/kg Plantago Psylium showed liver protection but protection was not significant in other doses.
The aim and objective of this study are to carry out the liver protective activities against the $CCl_4$ intoxication in mice with some Iranian medicinal plants traditionally used for liver injuries. The methanol extracts of Cichorium intybus, Lactuca scarJoia, Eucalyptus camadulensis were evaluated. With various doses of these plants, liver protective activities were performed after $CCl_4$ administration to mice. The serum aminotransferases activites, liver sizes, and histopatological examinations of liver were studied. At a dose of 50 mg/ kg, all three plants were able to protect liver damages induced by $CCl_4$.
Objectives : This study was to examine the efficacy of GCT on the hepatoprotective effect in the liver function and immune octivity. Methods : The experiment to investigate the hepatoprotective effect of GCT on the liver damage was conducted with D-galactosamine. The experiments to verify the effects of GCT on the immune activity were conducted by carbon clearance assay, plaque-forming cell SRBC assay of IgM, lymphoproliferation assay of T and B cells, and adherence and phagocytosis of mocrophages. Results: In the damage of liver induced by D-galactosamine, GCT carried hepatoprotective effect on AST. In carbon clearance assay GCT showed significant effect on phagocytosis of Kuffer cells. In the plaque-forming cell assay, GCT improved the formation of IgM. In the lymphoproliferation assay, GCT activated the formation of T and B lymphocytes. In macrophages, GCT activated adherence and phagocytosis. Conclusion : Though further study is needed, our findings suggest that GCT could be recommended as hepatoprotector and immune modulator for liver disease.
In this study, the ameliorating effect of soybean embryos on the impact of alcohol consumption was investigated on rat hepatocytes and in reducing total serum cholesterol levels and total serum lipid levels. Liver histology and two clinically important enzyme markers, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), of rats administered with both alcohol and soybean embryo were compared with a control group. The treatment regimen of soybean embryo significantly reduced the serum ALT and AST levels of the subjects, demonstrating the hepato-protective effects of soybean embryo. Electron microscopy indicated that the administration of soybean embryo preserved the important hepatocyte structures and prevented the presence of lipid droplets and secondary lysosomes. Furthermore, total cholesterol and total lipid levels were significantly reduced. These results indicate that treatment with soybean embryo can positively mediate the effects of alcohol on hepatocytes and general liver functions.

This study sought to investigate the ameliorating effects of a Korean medicinal herb (KMH) complex on the impacts of alcohol consumption in rat hepatocytes and in reducing the total cholesterol levels and the total lipid levels in the serum. We compared the body weight gain and ratio of the liver, the kidney to body weight, and also the serum biochemistry of the rats administered with both the alcohol and the KMH complex to the control rats treated with alcohol alone. The clinically important enzyme markers (Aspartate Aminotransferase, AST, and Alanine Aminotransferase, ALT) of rats, administered with both the alcohol and the KMH complex treatments, were compared with those in the control group. The treatment regimen (KMH complex) significantly reduced the serum AST and ALT levels, indicating the hepato-protective effects of the KMH complex. Furthermore, total cholesterol and total lipid levels were significantly reduced. These results indicate that the KMH complex may positively mediate the effects of alcohol on hepatocytes and the general liver functions.
Oxidative stress is a common mechanism contributing to initiation and progression of hepatic damage in a variety of liver disorders. Hence there is a great demand for the development of agents with potent antioxidant effect. The aim of the present investigation is to evaluate the efficacy of Moringa oleifera as a hepatoprotective and an antioxidant against 7, 12-dimethylbenz[a]anthracene induced hepatocellular damage. Single oral administration of DMBA (15 mg/kg) to mice resulted in significantly (p<0.001) depleted levels of xenobiotic enzymes like, cytochrome P450 and b5. DMBA induced oxidative stress was confirmed by decreased levels of reduced glutathione (GSH) and glutathione-S-transferase (GST) in the liver tissue. The status of hepatic aspartate transaminase (AST), alanine transaminase (ALT), and alkaline phosphatase (ALP) which is indicative of hepatocellular damage were also found to be decreased in DMBA administered mice. Pretreatment with the Moringa oleifera (200 and 400 mg/kg) orally for 14 days significantly reversed the DMBA induced alterations in the liver tissue and offered almost complete protection. The results from the present study indicate that Moringa oleifera exhibits good hepatoprotective and antioxidant potential against DMBA induced hepatocellular damage in mice that might be due to decreased free radical generation.
Objectives : Positive effects of Ginseng has great research attentions such as anticancer, anti-diabetic, antiaging, liver, immune function, CNS, etc. In this study, we investigated Hydroponic-cultured Ginseng Folium fermented byBacillus subtilisto establish fermentation conditions for enhancing functionality.Methods : Ginseng Folium were cultivated hydroponic-cultured and were extracted with methanol. We inoculateBacillus subtilisfor fermentation by adding to 0%, 3% and 5% sugar respectively and checked antioxidant activities, total phenolic content and total saponin content in 2 days intervals during 11 days. The antioxidant activities were studied by the 1,1-diphenyl-2-picryl hydrazyl(DPPH) radical, 2, 2'-Azino-bis(3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt(ABTS) radical scavenging assay and Reducing power assay. We analyzed the Total phenol content, crude saponin content and ginsenoside content. Moreever, Hepatoprotective effects by Glutamic oxaloacetic transaminase(GOT) and Glutamic pyruvic transaminase(GPT) in Sprague-Dawley rat.Results : The results of DPPH and ABTS were 66.89% and 96.72%, respectively. The reducing power was resulted in optical density of 0.7312 with 3% sugar after 9 days of fermentation. and the concentration at 200 ㎍/㎖. Total phenol content was 36.92㎎/g with 3% sugar after 9 days of fermentation, in which crude saponin content wasn't changed, and ginsenoside content such as Rg3, Re and Rb was increased. Activities of GOT and GPT concentration were decreased in rat.Conclusions : This study suggests that hydroponic-cultured Ginseng Folium fermented byBacillus subtilisin 9 days showed significant efficacy of hepato-protection as well as antioxidant compared to the others. In addition, it shows not only improved value but also utilized hydroponic-cultured Ginseng Folium by fermentation.
본 연구에서는 멀꿀 열매 추출물의 세포수준에서 항산화 활성과 아세트아미노펜(APAP)으로 유도된 간 독성 동물모델에서의 간 기능 보호효과를 연구하였다. 멀꿀 열매 추출물의 총 폴리페놀 함량과 플라보노이드 함량은 각각 $16.13{\pm}0.27mg$ gallic acid equivalent/g 및 $4.7{\pm}0.80mg$ catechin equivalent/g이었다. 또한 DPPH 라디컬 소거능과 ORAC도 각각 $63.62{\pm}4.10{\mu}g/ml$ 및 $90.63{\pm}5.29{\mu}M$ trolox equivalent/g이었다. 과산화수소로 유도된 산화적 손상에 대한 멀꿀 열매 추출물의 간세포 보호효과를 실험한 결과, $200{\mu}g/ml$에서 세포생존율이 증가하고 증가된 LDH 활성이 감소함을 확인하였다. 간세포에서 과산화수소로 산화적 스트레스를 유도하여 감소된 항산화 효소(SOD, CAT, GR, GPx)의 활성은 멀꿀 열매 추출물 처리로 활성이 증가하여 간세포를 보호하였다. 아테트아미노펜(APAP) 유도 간 손상 생쥐 모델에서 간 보호활성을 평가하였다. 간 손상 혈청 표지지표인 ALT 및 AST수준이 APAP 단독 처리군에 비해 멀꿀 열매 추출물 200 mg/kg군에서 유의적으로 감소되었으며, 간의 과산화지질함량도 감소한 것으로 나타났다. 간 조직의 병리학 검사에서도 간조직이 정상 회복되는 형태를 보였다. 이상의 결과를 토대로 멀꿀 열매 추출물이 항산화 기전을 통해 간기능 보호 효과를 나타냄을 확인할 수 있었다.
한인진에 대한 항산화 효과와 항균 효과를 알아보기 위하여 추출용매별 고형분 함량을 측정하여 본 결과 물 추출물에서 18. 1%로 가장 많은 고형분을 얻을 수 있었으며 메탄올, 에탄을 추출물에서는 12. 8%, 12. 6%로 별 차이가 없었다. 항산화 효과를 알아보는 방법 중 DPPH 소거효과를 이용하여 측정하여 메탄올 추출물이 $36.2{\mu}g/ml$로 가장 효과 좋았지만, 대두유에 대한 한한인진 추출물의 항산화 효과를 $60^{\circ}C$로 20일간 저장하면서 과산화물가(POV)를 측정하여 대두유를 기질로 한 저장시험 에서 1,000ppm 농도의 물추출물과 메탄올 추출물이 저장 기간이 20일 되었을 때 46.8(meq/kg, 50.8(meq/kg)의 과산화물가를 나타내 대조구의 과산화물가 79.1 (meq/kg)에 비해 낮았다. 식품의 부패균이나 식중독의 원인균으로 알려진 세균 4종을 대상으로한 항균실험에서는 추출 용매에 따른 항균력은 에탄올 추출물이 가장 좋았으며 세균에 대한 항균력으로는 Vibrio parahaemolyticus에 대해 가장 좋았다. 한인진의 간보호 효과 시험에서는 한인진 추출물을 경구투여한 V, IV군의 간무게, 체중에 대한 간무게의 비율이 생리 식염수만을 투여한 III군보다는 낮았으나 시료 채취전후로 한인진 추출물을 경구투여한 V군과 IV군사이에는 별다른 차이점이 없었다. 예방효과를 알아보기 위해 10일간 실험을 실시한 결과 한인진 물추출물만을 단독 투여한 VI군의 체중은 계속 증가하였으나 한인진 물추출물을 계속 투여한 다음 시료 채취전에 사염화탄소를 투여한 VIII군의 간무게, 체중에 대한 간비율이 대조군 VII보다 다소 감소하였다. 병리학적인 소견으로 한인진추출물을 투여 한 군에서는 사염화탄소만 투여한 군보다 간세포의 괴사와 지방적의 축적이 감소하여 사염화탄소에 의한 간독성에 효과가 있는 것으로 사료된다.
