Aloe-Emodin-Mediated Photodynamic Therapy Attenuates Sepsis-Associated Toxins in Selected Gram-Positive Bacteria In Vitro

  • Sepsis is an acute inflammatory response that leads to life-threatening complications if not quickly and adequately treated. Cytolysin, hemolysin, and pneumolysin are toxins produced by gram-positive bacteria and are responsible for resistance to antimicrobial drugs, cause virulence and lead to sepsis. This work assessed the effects of aloe-emodin (AE) and photodynamic therapy (PDT) on sepsis-associated gram-positive bacterial toxins. Standard and antibiotic-resistant Enterococcus faecalis, Staphylococcus aureus, and Streptococcus pneumonia bacterial strains were cultured in the dark with varying AE concentrations and later irradiated with 72 J/cm-2 light. Colony and biofilm formation was determined. CCK-8, Griess reagent reaction, and ELISA assays were done on bacteria-infected RAW264.7 cells to determine the cell viability, NO, and IL-1β and IL-6 pro-inflammatory cytokines responses, respectively. Hemolysis and western blot assays were done to determine the effect of treatment on hemolysis activity and sepsis-associated toxins expressions. AE-mediated PDT reduced bacterial survival in a dose-dependent manner with 32 ㎍/ml of AE almost eliminating their survival. Cell proliferation, NO, IL-1β, and IL-6 cytokines production were also significantly downregulated. Further, the hemolytic activities and expressions of cytolysin, hemolysin, and pneumolysin were significantly reduced following AE-mediated PDT. In conclusion, combined use of AE and light (435 ± 10 nm) inactivates MRSA, S. aureus (ATCC 29213), S. pneumoniae (ATCC 49619), MDR-S. pneumoniae, E. faecalis (ATCC 29212), and VRE (ATCC 51299) in an AE-dose dependent manner. AE and light are also effective in reducing biofilm formations, suppressing pro-inflammatory cytokines, hemolytic activities, and inhibiting the expressions of toxins that cause sepsis.

만룡(蔓龍)의 생약학적(生藥學的) 연구(硏究) (Pharmacognostical Studies on ‘Manryong’)

  • The pharmacognostical and pharmacological studies of 'Manryong' were carried out, which has been widely used in Chun-Ra Province, Korea and the following results were obtained. 1) The original plant of the bulb MANRYONG is Erythronium japonicum DECNE. (Liliaceae). 2) The internal structure consists of mostly parenchyma containing numerous starch grains, vascular bundles and substituted fibers. 3) The external surface occurs as a yellowish white and slightly bented ovoidal shape. 4) 'Manryong; contains 11 kinds of free amino acids, such as asparagin, tryptophan, cystine, glutamic acid, threonine, glycine, leucine, proline, histidine, methionine, and alanine. 5) Hemolytic action of the crude saponin isolated from 'Manryong' is weaker than that of digitonin. 6) Effects of 'Manryong' extracts on ceruloplasmin were studied and an antidotal activity of the extracts was found in liver intoxicated with carbon tetrachloride.

모기유층에 대한 살충성 Bacillus thuringiensis H9B 균주의 특성 (Characterization of Mosquitocidal Bacillus thuringiensis Strain H9B)

  • One strain of mosquitocidal Bacillus thuringiensis, H9B, was isolated from soil. The biochemical characteristics and flagella antigenicity of the strain H9B is similar to that of B. thuringiensis subsp. darmstadiensis. The delta-endotoxin of the strain H9B coincided with that of B. thuringiensis subsp. darmstadiensis strain 73E10-2 on agarose double immunodiffusion test. The delta-endotoxin of B. thuringiensis subsp. israelensis contains hemolysin fragment (28 kb) on SDS-PAGE when the delta-endotoxin was solubilized in alkali, while that of the strain H9B does not contain 28 kb protein.

  • Cloning and expression of two different crystal protein genes from transformed Bacillus thuringiensis were investigated. B. thuringiensis NT0423 is toxic to both Lepidopterous and Dipte-rous larvae. The pCG5 vector carrying crystal protein genes (mosquitocidal and hemolytic activity) of B. thurigiensis subsp. morrisoni PG-14 was transformed into B. thurigiensis NT0423. Transformant has expressed two type crystals of bipyramid from NT0423 and ovoid from pCG5 in one cell.

  • A protein-polysaccharide fraction was prepared from the carpophores of Laetiporus sulphureus. This fraction suppressed growth of sarcoma 180 in A-strain mice when administered i. p. To investigate the mechanism of antitumor action of this fraction, plaque assay was conducted by administrating i. p. to the mise at a dose level of 50mg/kg for five days. Ten days later, the mice were immunized with 1 * 10$^{7}$ sheep red blooc cells. The number of hemolytic plaque forming cells was significantly greater than that of the control mice. Three monosaccharides and fifteen amino acids were identified in the protein-polysaccharide fraction.

  • Bioassay-directed chromatographic fractionation of a methylene chloride extract of Ailanthus altisshima indicated the presence of 20(S). 24(R), epoxy-25-hydroxydammarane-3-one(compound 1. ocotillone). which was isolated from this plant for the first time.   • 형방지황탕은 한의학에서 중요한 약처방으로 인간의 기력 회복에 약효가 탁월한 것으로 알려져 있다. 본 연구에서는 형방지황탕으로부터 항생 물질(HTI)을 분리하여 항진균활성 및 그 작용기작을 검토하였다. 그 결과 각종 병원성 진균에 대해 항진균 활성을 나타내었으며, 인간 적혈구에 대해서는 세포독성이 나타나지 않은 것으로 확인되었다. 또한 캔디다 알비칸스를 대상으로 스트레스 방어기작과 세포 이형성 유도에 HTI이 미치는 영향을 검토한 결과, 트리 할로즈의 증가와 이형성 유도에 영향을 미치는 것으로 나타나 이러한 현상이 항진균활성에 상관관계가 있음을 확인하였으며, 또한 진균의 세포주기에 미치는 영향도 항진균 활성의 요인이 됨을 밝혔다.

  • 본 연구는 probiotics mixture(Lactobacillus acidophilus PBS066(DSM 24936), Lactiplantibacillus plantarum PBS067(DSM 24937), Limosilactobacillus reuteri PBS072(DSM 25175))의 안전성 및 탄수화물 소화 효소 저해 효과를 in vitro로 평가하는 것을 목적으로 한다. 모든 균주는 유럽식품안전처(EFSA) 지침에서 권장하는 항생제 내성 프로필을 충족시켰으며, 모든 균주는 용혈 활성 및 세포 독성을 나타내지 않았다. Probiotics mixture, L. plantarum PBS067 그리고 L. reuteri PBS072는 α-amylase 활성을 억제하는 것으로 나타났으며, probiotics mixture와 이의 균주 3종 모두 α-glucosidase 활성을 억제하는 것으로 나타났다. 이로써 본 연구에 사용한 probiotics mixture 및 이의 단일 균주 3종의 안전성과 탄수화물 소화 억제 효과를 확인하였으며, 따라서 probiotics mixture 이의 단일 균주 3종은 섭취하여도 안전하며 섭취 시 체내 탄수화물 대사를 잠재적으로 조절하는 것을 확인하였다.

Analogs of Periplanetasin-4 Exhibit Deteriorated Membrane-Targeted Action

  • Periplanetasin-4 is an antimicrobial peptide with 13 amino acids identified in cockroaches. It has been reported to induce fungal cell death by apoptosis and membrane-targeted action. Analogs were designed by substituting arginine residues to modify the electrostatic and hydrophobic interactions accordingly and explore the effect of periplanetasin-4 through the increase of net charge and the decrease of hydrophobicity. The analogs showed lower activity than periplanetasin-4 against gram-positive and gram-negative bacteria. Similar to periplanetasin-4, the analogs exhibited slight hemolytic activity against human erythrocytes. Membrane studies, including determination of changes in membrane potential and permeability, and fluidity assays, revealed that the analogs disrupt less membrane integrity compared to periplanetasin-4. Likewise, when the analogs were treated to the artificial membrane model, the passage of molecules bigger than FD4 was difficult. In conclusion, arginine substitution could not maintain the membrane disruption ability of periplanetasin-4. The results indicated that the attenuation of hydrophobic interactions with the plasma membrane caused a reduction in the accumulation of the analogs on the membrane before the formation of electrostatic interactions. Our findings will assist in the further development of antimicrobial peptides for clinical use.

Vibrio fluvialis 유래의 hemolysin 정제와 생화학적 특성 (Purification and characterization of biochemical properties of hemolysin from Vibrio fluvialis)

  • 병원성 미생물인 Vibrio fluvialis로부터 hemolysin을 분리정제 하였다. 즉, hemolysin의 정제는 균 배양액을 황산암모늄, ion exchange chromatography를 행하였으며 SDS-PAGE를 통해 그 단백질의 크기가 약 79kDa임을 알게되었다. 정제된 hemolysin (VFH)는 35$^{\circ}C$에서 최적활성을 나타내었고, 4$0^{\circ}C$에서는 그 활성이 감소되었다. RTG-2 어류유래 세포주에 hemolysin (VFH)의 처리로 세포독성을 측정한 결과 50$\mu\textrm{g}$의 VFH가 약 80%의 cell line을 사멸시켰다. 또한 현미경을 통한 관찰에서도 세포의 형태변화를 관찰할 수 있었다.
