Molecular Characterization of Survival and Toxigenesis of Vibrio vulnificus

  • Understanding the molecular pathogenesis of the multifaceted host-pathogen interaction is critical in the development of improved treatment and prevention, as well as elucidating how certain bacteria can circumvent host defenses, multiply in the host, and cause such extensive damage. Disease caused by infection with V. vulnificus is remarkable for the invasive nature of the infection, ensuing severe tissue damage, and rapidly fulminating course. The characterization of somatic as well as secreted products of V. vulnificus has yielded a large list of putative virulence attributes, whose known functions are easily imagined to explain the pathology of disease. These putative virulence factors include a carbohydrate capsule, lipopolysaccharide, a cytolysin/hemolysin, elastolytic metalloprotease, iron sequestering systems, lipase, and pili. However, only few among the putative virulence factors has been confirmed to be essential for virulence by the use of molecular Koch's postulates. This presentation describes molecular biological characterization of the virulence factors contributing to survival as well as to toxigenesis of V. vulnificus.

Characterization of Mosquitocidal Bacillus thuringiensis Strain H9B (모기유층에 대한 살충성 Bacillus thuringiensis H9B 균주의 특성)

  • One strain of mosquitocidal Bacillus thuringiensis, H9B, was isolated from soil. The biochemical characteristics and flagella antigenicity of the strain H9B is similar to that of B. thuringiensis subsp. darmstadiensis. The delta-endotoxin of the strain H9B coincided with that of B. thuringiensis subsp. darmstadiensis strain 73E10-2 on agarose double immunodiffusion test. The delta-endotoxin of B. thuringiensis subsp. israelensis contains hemolysin fragment (28 kb) on SDS-PAGE when the delta-endotoxin was solubilized in alkali, while that of the strain H9B does not contain 28 kb protein.

  • The object of thes research is to lucidate the clinical effects of Gami-Cheongshimyeonjayeum on the anti-allergic and gimmune response to rats and mice. The obtaind results of Gami-Cheongshimyeonjayeum administration are as follows : 1. The cytotoxic index increased slightly at over concentration $1{\times}10^{-2}/ml$, but it is thought that the cytotoxic effect show no significant value under the concentration $1{\times}10^{-3}/ml$. 2. Gami-Cheongshimyeonjayeum water extract seems to suppress the leakage volume of Evans blue to decrease the vascular permeability. 3. Gami-Cheongshimyeonjayeum show the inhibitory effect of footpad edema. 4. Protein leakage is inhibited by the administration of Gami-Cheongshimyeonjayeum. 5. The delayed type hypersensitivity to SRBC is suppressed at the 24hours after the administration of Gami-Cheongshimyeonjayeum on the experimental group 1, 2. 6. Hemolysin and hemagglutinin titer, and RFC increase to bulid up the immune function.

  • In order to investigate the effects of api-toxin therapy on depression of immune response, mice were exposed to cold stress $(-20^{\circ}C)$) in 30 minutes twice a day and api-toxin(Bee Venom from Korea and U.S.A) therapy was treated to the Kihae $(CV_6)$ locus of human body every two days with 4 sessions of treatment for 8 days. The mice were sensitized i.v. with $5{\times}10^8$ sheep red blood cells (SRBC) on the 4th day and challenged i.d. with $2{\times}10^9$ SRBC 4 days later. Immune Response for delayed type hypersensitivity (DTH), lympocyte transformation(LTT), hemolysin titer(HL), plaque forming cells (PFC) were measured.

  • The effects of Asiasarum Root on the cell mediated and humoral immune responses were investigated in ICR mice. ICR mice received intraperitoneal injection of methanol extracts (ether fraction, butanol fraction, water fraction) for 5 days before sensitization. The change of body weight (%), the organ weight ratio (%), the numbers of RBC and WBC, the ear swelling by dinitrofluorobenzene, the titers of hemagglutination (HA) and hemolysin (HY) to SRBC were determined. The results were as follows; 1. The change of body weight (%) showed a tendency of increasing. 2. The weight of spleen (%) decreased. 3. Ear swelling was maximum at 48 hours after challenge and was significantly decreased in the groups. 4. The numbers of RBC and WBC were decreased. 5. HA and HY titers were decreased.

  • In order to investigate the effect of Mori Folium on immunologic control function, the various immune responses were studied. The results were obtained as follows : 1. The transformation of lymphocyte was increased significantly in all concentration of Mori Folium treated group as compared with negative and positive control. 2. In FACS analysis, the population of helper T cell and T cell were increased significantly in Mori Folium treated group as compared with control. 3. IL-12(p35) and INF-${\gamma}$ were upregulated in Mori Folium treated group 4. NO and carbon clearance were significantly increased in Mori Folium treated group as compared with control. 5. The hemagglutinin titer and hemolysin titer were increased but insignificantly in Mori Folium treated group as compared with control.

  • In order to investigate the effects of shipjuntaepotang (少陰人 十全大補湯) on immune response, the author performed this experimental study. Delayed type hypersensitivity (DTH) and rosette forming cells (RFC) for cell-mediated immune response, hemagglutinin (HA) titers, hemolysin (HL) titers, and carbon clearance for phagocytic function of MPS (mononuclear phagocyte system) were measured in ICR mice. The results were summarized as follows. 1. DTH in the experimental group was increased, as compared with the control group, with statistical significance. 2. RFC in the experimental group was increased, as compared with the control group, with statistical significance. 3. HA-titers were increased in the experimental group, as compared with the control group, with statistical significance. 4. HL-titers were increased in the experimental group, as compared with the control group, with statistical significance. 5. Carbon clearance was increased in the experimental group, as compared with the control group, with statistical significance. Through invivo experimental study in ICR mice, these findings suggest that shipjuntaepo-tang enhance both cellmediated and humoral immune responce.

Charaterization of an Escherichia coli O157:H7 Strain Producing Verotoxin 2Isolated from a Patient in Korea

  • Nine hundred patients diagnosed with diarrhea or hemorrhagic uremic syndrome in the Kyungpook Province, Korea, were examined from November 1998 to February 2000. One patient in Kumi appeared to possess the Escherichia coli O157:H7 strain, which is very important in clinical decision making and public health action. The isolated strain, an E. coli O157:H7 KM, contained a 60 MDa plasmid and typical virulence genes including the verotoxin 2 gene, ehxA gene (encoding enterohemorrhagic hemolysin), and eae (encoding attaching and effacing protein-intimin) gene. This strain produced only verotoxin 2. Pulsed field gel electrophoretic analysis showed that the genomic organization of the E. coli O157:H7 KM strain may differ greatly from those of representative strains previously reported in the United States and Japan.

Identification of intestinal microflora in rainbow trout

  • Although trout farming is well established in Korea, very little information is available on the composition of intestinal microflora in rainbow trout (Salmo gairdnerii). In 1994, from October through November, we investigated the composition and succession of intestinal bacteria. As fish grew, total viable counts increased dramatically until 45 days after fertilization when anaerobes started to appear on the media. After this time, they increased steadily. Ten aerobic generic were identified and Gram negative bacteria constituted 85% of total isolates. Among these, Pseudomonas, Eikenella, and Alcaligenes were the three major genera. Six anaerobic genera were isolated and identified. The ratio of anaerobes to aerobes was about 1 : 1 in adult trout and the composition of genera was changed under different conditions.

Rapid Identification of Vibrio vulnificus in Seawater by Real-Time Quantitative TaqMan PCR

  • In order to identify Vibrio vulnificus in the Yellow Sea near Gunsan, Korea during the early and late summers, the efficiency of the real-time quantitative TaqMan PCR was compared to the efficiency of the conventional PCR and Biolog identification system^TM. Primers and a probe were designed from the hemolysin/cytolysin gene sequence of V. vulnificus strains. The number of positive detections by real-time quantitative TaqMan PCR, conventional PCR, and the Biolog identification system from seawater were 53 (36.8%), 36 (25%), and 10 strains (6.9%), respectively, among 144 samples collected from Yellow Sea near Gunsan, Korea. Thus, the detection method of the real-time quantitative TaqMan PCR assay was more effective in terms of accuracy than that of the conventional PCR and Biolog system. Therefore, our results showed that the real-time TaqMan probe and the primer set developed in this study can be applied successfully as a rapid screening tool for the detection of V. vulnificus.
