실험계획 법에 의한 혈중 글루코즈 측정 시 타 성분의 영향 분석 (Influence of Other Blood Components in Predicting Glucose Concentration using Design of Experiment)

  • 혈액 속의 글루코즈 농도를 측정 시 타 성분이 미치는 영향을 분석하였다. 이를 위하여 헤모글로빈을 포함한 총 5 가지의 주요 성분을 이용하여 혈액 팬텀을 만들었으며 글루코즈의 배음대역을 포함하는 1500 ~ 1850nm의 흡수스펙트럼을 측정하여 농도 추정 모델을 세웠다. 농도 예측은 다변량통계분석 모델인 부분최소자승회귀분석을 이용하였다 다른 성분들이 글루코즈 측정에 있어 미치는 영향은 실험계획법을 적용한 인자 분석에 의하여 조사하였다. 트리 글라이세라이드는 영향을 미치지 않는 것으로 나타났으며. 알부민과 글로불린의 경우 적은 범위의 영향을 주는 것으로 나타났다 하지만 헤모글로빈의 영향은 큰 것으로 나타나 글루코즈 측정에 있어 헤모글로빈의 농도 보상이 필요한 것으로 판명되었다.

Enhanced Production of ${\varepsilon}$-Caprolactone by Coexpression of Bacterial Hemoglobin Gene in Recombinant Escherichia coli Expressing Cyclohexanone Monooxygenase Gene

  • Baeyer-Villiger (BV) oxidation of cyclohexanone to ${\varepsilon}$-caprolactone in a microbial system expressing cyclohexanone monooxygenase (CHMO) can be influenced by not only the efficient regeneration of NADPH but also a sufficient supply of oxygen. In this study, the bacterial hemoglobin gene from Vitreoscilla stercoraria (vhb) was introduced into the recombinant Escherichia coli expressing CHMO to investigate the effects of an oxygen-carrying protein on microbial BV oxidation of cyclohexanone. Coexpression of Vhb allowed the recombinant E. coli strain to produce a maximum ${\varepsilon}$-caprolactone concentration of 15.7 g/l in a fed-batch BV oxidation of cyclohexanone, which corresponded to a 43% improvement compared with the control strain expressing CHMO only under the same conditions.

Partial Least Squares-discriminant Analysis for the Prediction of Hemodynamic Changes Using Near Infrared Spectroscopy

  • Using continuous wave near-infrared spectroscopy, we measured time-resolved concentration changes of oxy-hemoglobin and deoxy-hemoglobin from the primary motor cortex following finger tapping tasks. These data were processed using partial least squares-discriminant analysis (PLS-DA) to develop a prediction model for a brain-computer interface. The tasks were composed of a series of finger tapping for 15 sec and relaxation for 45 sec. The location of the motor cortex was confirmed by the anti-phasic behavior of the oxy- and deoxy-hemoglobin changes. The results were compared with those obtained using the hidden Markov model (HMM) which has been known to produce the best prediction model. Our data imply that PLS-DA makes better judgments in determining the onset of the events than HMM.

Inhibitory Effects of Total Extract and Flavonols from Hardy Rubber Tree (Eucommia ulmoides Oliver) Leaves on the Glycation of Hemoglobin

  • Our previous study demonstrated that aqueous ethanolic extract of hardy rubber tree (HRT; Eucommia ulmoides Oliver) leaves potently inhibited glycation, and that two known flavonols and one new flavonol were the active compounds in protein glycation in vitro using the model system of bovine serum albumin and fructose. The aim of this study was to examine the effectiveness of HRT extract and its flavonols against the glycation of hemoglobin (Hb) in primary cultured rat erythrocytes. The extract at $40-400\;{\mu}g/mL$ and the flavonols at $0.2-20\;{\mu}g/mL$ significantly inhibited Hb glycation in a concentration-dependent manner. They were more effective than aminoguanidine, a well-known inhibitor of glycation. Three flavonols seemed to be responsible for the inhibitory activity of the extract, furthermore, the extract of HRT leaves showed advantages over single isolated flavonols in the inhibition of Hb glycation.

한국인(韓國人) 빈혈빈도(貧血頻度)에 관(關)한 연구(硏究) (A Study on the Anemia Prevalence in Korea)

  • The purpose of the present study was to evaluate the nutritional status of the Korean population and its relationship with the age, sex, economic status and geographic characteristics. Taking in consideration the geographic location and socioeconomic states, ten regions out of the twenty-four regions where the Ministry of Health and Social Affairs had executed a programme (National Nutrition Survey) on evaluation of the community nutritional status were selected. Hemoglobin concentration hematocrit, and mean corpuscular hemoglobin concentration were tested from a total of 906 subjects in a month of November, 1979. The results of this study suggest that the anemia prevalences of young children in Seoul and rural areas were remarkedly decreased during last ten years, but the prevalence of low income group in provincial cities are still high and particularly in 7-l4yr. and female adult group.

유방염에 이환된 젖소에 있어서 혈액 동태학에 관한 연구 (Study on the hemodynamics in cows with naturally mastitis)

  • Hemodynamic values were assessed in cows with naturally mastatis. hemodynamic tests included WBC, RBC, PCV, Hemoglobin, Monocyte, Eosinophil, Neutrophil, Lymphocyte, and prothrombin time, activated partial thromboplastin time, thrombin time, fibrinogen, platelet, antithrombin-III, and plasminogen activities. Significant changes were observed in the mean values of most analytses : WBC, monocytes, eosinophil, neutrophil were increased and Iymphocyte were decreased. prothrombin time was increased: activated partial thromboplastin time, thrombin time. increased : activated partial thromboplastin time, thrombin time, fibrinogen concentration, plasminogen activity and platelet concentration were decreased : and RBC, PCV, hemoglobin and antithrombin-III activity were unchanged, compared with normal mean values. Thesse data indicated activation of hemodynamic mechanisms, initiated either directly by bacteria produced endotoxin of secondaly inflammatory mediators produced in response to caused bacteria and naturally acquired mastitis was very similar to the experimental endotoxin-induced mastitis.

  • We measure the hemodynamic responses during the 4 kinds of tasks on the motor cortex in the right and left human brain by using near-infrared spectroscopy. The experimental results show that the change of concentration of oxy-hemoglobin is larger than that of deoxy-hemoglobin and the change of concentration of chromophores induced by finger and arm related task show more activations than that of leg.

Optimization of Microwave-Assisted Method for Accelerated Glycated Hemoglobin Quantification from Amino Acids to Proteins

  • Glycated hemoglobin ($HbA_{1c}$) has been commonly used to screen and diagnose for patients with diabetes mellitus. Here the accelerated procedure of microwave-assisted sample treatment from acid hydrolysis to enzyme digestion followed by isotope dilution liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) was optimized and applied to measure $HbA_{1c}$ in an effort to speed up analysis time. First, two signature peptides of $HbA_{1c}$ and hemoglobin $A_0$ were certified with amino acid analysis by setting optimized acid hydrolysis conditions to $150^{\circ}C$, 1.5 h and $10{\mu}M$ sample concentration in 8 M hydrochloric acid. Consequently, the accurate certified peptides above were used as calibration standards to implement the proteolytic procedure with endoproteinase Glu-C at $37^{\circ}C$, 700 W for 6 h. Compared to the traditional method, the microwave heating not only shortened dramatically sample preparation time, but also afforded comparable recovery yields. The optimized protocol and analytical conditions in this study are suitable for a primary reference method of $HbA_{1c}$ quantification with full SI-traceability and other similar proteins in complex biological samples.

운동선수들의 혈액분석을 통한 Etrythropoietin 간접도핑검사 (Blood Analysis for Indirect Doping Control of Erythropoietin in Sports)

  • The use of recombinant human erythropoietin (rhEPO), a stimulator of erythropoiesis, banned in sports because of the medical risk associated with thrombosis. Due to analytical difficulties to differentiate between natural human EPO (hEPO) and rhEPO, blood parameters of erythropoiesis such as contents of hemoglobin (cut-off value <17.5 g/d l for man, and < 16.0 g/dl for women), hematocrit and reticulocytes (cut-off value <2.0%) were measured to focus the misuse of rhEPO. We conducted anti-doping test for 122 blood samples of the World Cup athletes. The mean values of key parameters are as follows; 14.5$\pm$1.0 g/dl for hemoglobin, 41.7$\pm$2.8% for hematocrit, and 1.3$\pm$0.4% for reticulocyte. Blood sample was found to be stable up to 8 hours for the reticulocyte measurement. In addition, the soluble transferrin receptor and ferritin levels were measured by immunoassay methods using plasma samples (n=28) in which the mean value was 0.8$\pm$0.5 $\mu\textrm{g}$/$m\ell$ and 54.6$\pm$33.7 ng/$m\ell$, respectively. The results indicate that all samples tested were negative for the blood parameters of indirect anti-doping test for hEPO misuse. The statistical evaluation suggest that several other parameters such as red blood cell, mean corpuscular hemoglobin concentration, mean corpuscular volume, mean corpuscular hemoglobin and white blood cell could be considered as factors influencing hEPO function in addition to five parameters mentioned.

흰쥐에 있어서 PEG-헤모글로빈 SBI의 단회정맥투여 약물동태 (Pharmacokinetics of a New PEG-hemoglobin SBI for Single Intravenous Administration to Rats)

  • An intravenous pharmacokinetics for a new red cell substitute, PEG-hemoglobin SB1, was studied in SD rats. Total-hemoglobin and its metabolite methemoglobin in the plasma were determined using a spectrophotometer. The limit of quantitation was 0.01g/dL and the C.V for interday assay reproducibility was less than 6%. Upon intravenous administration of anticipated clinical dose, 10 ml(0.7 gHb)/kg, plasma concentration curve of total-hemoglobin was well described by one-compartment model. The $t_{\frac{1}{2}},{\;}CL_{t}$, Vd and $AUC^{0-48hr}$ were $8.23{\pm}0.96{\;}hr,{\;}0.06{\;}{\pm}{\;}0.01 {\;}dL/hr/kg,{\;}0.66{\pm}0.05{\;}dL/lg{\;}and{\;}13.6{\;}{\pm}{\;}1.01g{\cdot}hr/dL$, respectively, in male rats(n=5, $mean{\;}{\pm}{\;}SD$). Those parameters in female rats were $9.21{\;}\pm{\;}2.31{\;}hr,{\;}0.06{\;}{\pm}{\;}0.01{\;}dL/hr/kg,{\;}0.79{\pm}{\;}0.08{\;}dL/kg{\;}and{\;}13.0{\;}{\pm}{\;}2.36{\;}g{\cdot}hr/dL$, respectively. Similar kinetic profiles between males and females were also obtained from other parameters. Small amount of methemoglobin, an oxidative metabolite of SB1, was detected in the plasma of both sexes, where the $AUC^{0-48{\;}hr,m}$ and $t_{{\frac{1}{2}},m}$ were approximately $1.5{\;}g{\cdot}hr/dL$ and 20 hr, respectively. The present work provides a critical kinetic data for the effective clinical applications of PEG-hemoglobin SB1.
